This experiment was conducted to investigate varietal responses to cool temperature (18℃) and short photoperiod (8 hours a day) in a controlled room of phytotron. Five burley tobacco varieties (Nicotiana tabacum L.), Awelius (very early flowering type), Burley 21 and KB 103 (medium), Banket Al and MB 1 (slightly late) and TC 681 (late) were compared in number of leaves per plant and day to flower in different treatment periods (10, 15, 20, 25 and 30 days). The days to flower decreased significantly than the control (in the phytoron with 14hr photoperiods and a minimum temperature 20'c) when the treatment periods were over 15 days for the varieties Aurelius, Burley 21 and KB 103, and 20 days for TC 681, and 30 days for Banket Al, respectively. And also the number of leaves decreased significantly than that of the control when the periods of treatment were over 10 days for Burley 21 and Aurelius, 15 days form 103, and 20 days for TC 681 and Banket Al, and 30 days for MB 1, respectively. The decreasing ratio in the number of leaves per plant under cool temperature and short photoperiod treatments was the lowest in Banket Al and MB 1. A CAB cDNA clone(pKGCAB) encoding the light harvesting chlorophyll a/b binding protein of the semi-shade plant, Korean ginseng(Panax ginseng C. A. Meyer) was isolated by the one-way path random sequencing of ginseng cDNA library clones and transgenic tobacco plants(Nicotiana tabacum NC82) were produced by the transformation of this ginseng CAB gene in use of Agrobacterium tumefaciens LBA4404. The CAB gene showed type 1 structure of LHCP-II, 84% similarity in nucleotide sequence and 92% in amino acid sequence to that of Nicotiana tabacum CAB40, respectively. Seed germination and initial growth of the transgenic tobacco plants transformed with the cDNA fragment were accelerated under low light intensity compared with those of normal tobacco plant, that may result from the higher light sensitivity of the transgenic plants than that of the normal.
Field experiment was conducted to find out the chemical characteristics of 12 Korean local varities, Local varities were compared with oriental, flue-cured and burley tobacco. Among tested varities, Hyangcho and Matskawa were desirable in respect of flavor, leaf color and texture. Hyangcho and Matskawa were higher in the content of nicotine and petroleum ether extract. Gajacho, Byeolcho and Kukjuo were higher in the content of reducing sugar, but lower nicotine, total nitrogen and petroleum ether extract. The nicotine and total nitrogen content of Kwangcho, Daruma and Catterton were lower than that of Br. 21. In the content of volatile organic acid which were major characteristic component of oriental tobacco, the Korean local varities were lower in the content of 3-methyl butanoic and 3-methyl pentanoic acid, but hirities, Hyangcho, Matskawa and Daruma were higher in the content of volatile organic acid. Korean local varities, especially small leaf group, were higher in the content of benzyl alcohol and furfural than any other type tobacco.
Burley tobacco (Nicotiana tabacum L.) KB 108 was developed from a single cross between KB 104 and TC 591 which was developed from a cross between Burley 49 and Tobacco Introduction 1406. It was tested for its resistance to black shank, potato virus Y(PVY), TMV and agronomic characteristics under field conditions. KB 108 has resistance to tobacco mosaic virus(TMV) and necrotic strain of potato virus Y(PVY-VN) with secreting glandular trichomes. It has also moderate resistance to black shank caused by phytophthora parasitica val. Nicotianae. KB 108 has an up-right plant growth habit similar to Burley 21. It flowers about 1-2 days later than Burley 21. The leaf width and length of KB 108 are approximately 3 cm wider and longer than those of Burley 21. The yield of KB 108 was higher 4%, nearly equal in value per kg compared to Burley 21.
Recently the method of basal compound fertilizer (N : P : K = 18.2 : 9.8 : 35.0) Placement has been changed from banding to broadcasting in tobacco cultivation. The effects of the compound fertilizer placement, level of fertilizer and additional urea application on the agronomic characteristics, two chemical compositions and physical Properties of burley tobacco(Nicotiana tabacum L. cv. KB 108) were investigated at Chonju Experiment Station, Korea Ginseng and Tobacco Research Institute in 1995-1996. Six treatments consisted of (1) band (method of fertilizer Placement) + In kg/10a(compound fertilizer) + 0(additional urea applied), (2) broadcast+140+0, (3) broadcast+140+25, (4) band+180+0, (5) broadcast+180+0, and (6) broadcast +180+25. The additional urea was applied at hilling. No significant differences were detected between banding and broadcasting method of compound fertilizer Placement in field 9rowth, wield, organoleptic qualify(price Per kilogram), chemical composition and Physical properties of cured leaf, The increased compound fertilizer by 30%(40k/10a) or the additional urea application by 25kg per 10a produced slightly higher yield than the recommended amount of basal compound fertilizer without additional urea application did. It also increased the total alkaloid content of cured leaf. It is recommended that no more basal fertilizer above the recommended amount and no additional urea application are needed in burley tobacco fertilization, even though the method of basal fertilizer placement being chanced from banding to broadcasting. To classify the inheritance of resistance to potato virus Y, crosses between susceptible flue-cured tobacco variety NC 95 and resistant variety McNair 30 were conducted. The parents, $F_1$ plants, $F_2$ populations, and haploid plants derived from anthers of $F_1$ plants were screened for a resistance of two potato virus Y strains (PVY-VB and PVY-VN) isolated in Korea. The Chi-square values for the $F_3$ populations and haploids of $F_1$ fitted 1 :3 and 1 :1 ratios of resistant to susceptible for two strains, respectively. Therefore, it was found that the resistance of McNair 30 for the potato virus Y was controlled by a single recessive gene. Moreover the resistance to two strains screened was inherited dependently.
The experiment was conducted to obtain basic information on breeding of flue-cured tobacco varieties. Nine cultivars and partial diallel set of 36 F1 hybrides were grown at Taegu Experiment Station, Korea Ginseng & Tobacco Research Institute in 1983. Estimated heritability in the narrow-sense ranged from 66.21% to 94.12% for yield, leaves per plant, days to flower, leaf weight, leaf width, leaf shape, nicotine content and reducing sugar content, while that for stalk height, leaf length, midrib weight and midrib width ranged from 28.12% to 56.25%. The genotypic correlations were positive among yield, leaf weight, leaf length, leaf width, midrib weight and midrib width in the middle leaves, days to flower, nicotine content and reducing correlated with leaf length than leaf width. At the path coefficient analysis of the effect of leaf and midrib characters upon yield according to stalk position, leaf weight, leaf width and midrib width in the middle leaves, and leaf weight, and leaf width in the top leaves showed high direct effects.
Six flue- cured cultivars of Nicotiana tabacum L., 15 possible $F_1$ hybrids and $F_2$ Populations among them, and 15 haploid Populations from Fl hybrids and haploids from Parents, were evaluated. Comparisons of the $F_1$, hybrids and $F_2$ Populations with the Parents indicated that heterosis values were small but significant for yield, plant height, days to flower, leaf length and width and total alkaloids from-6.0% to 5.4% in $F_1$ hybrids, and from -3.4% to 3.6% for Plant height days to flower leaves per plant in $F_2$ populations, respectively. There were positive correlations for yield, plant height, days to flower, leaves per plant and total alkaloids between diploid and haploid populations. Increase or decrease ratio of haploids to diploids of total alkaloids was appeared to 30.3%. Those for yield, leaf length and width, value arid reducing sugar were ranged from -4.1% to -27.6%.
Phenylalanine ammonia-lyase (PAL), a key enzyme of the phenylpropanoid biosynthesis pathway, is activated by a number of developmental and environmental cues. The coding region of the NtPAL4 gene was 2,154 bp in length, and its deduced protein was composed of 717 amino acids. Sequence analysis of NtPAL4 cDNA from tobacco (Nicotiana tabacum L.) revealed high structural similarity to PAL genes of other plant species. The NtPAL4 gene exists as a single copy in the tobacco plant, and its transcripts were strongly expressed in flowers and leaves. NtPAL4 expression was significantly induced in response to NaCl, mannitol, and cold treatments, but it was not induced by abscisic acid (ABA). NtPAL4 expression decreased gradually after treatment with ABA and $H_2O_2$; however, NtPAL4 transcripts accumulated after treatment with methyl viologen (MV). Our results suggest that the NtPAL4 gene may function in response to abiotic stresses.
연초(N. tabacum L.)의 세포질 웅성불임 F$_1$ 식물체와 N. africana의 종간교배에 의한 웅성불임 반수체식물의 육성 및 특성을 조사한 결과는 다음과 같다. 1. (CMS NC82$\times$burley 21) F$_1$과 N. africana의 종간교배에 의한 세포질 웅성불임 반수체 식물의 출현빈도는 삭당 0.5주로 나타났다. 2. 종간교배으로 얻은 웅성불임 반수체식물 중 잎과 줄기의 색이 green(황색종)과 yellow(버어리종)의 분리비는 3 : 1로 나타났다. 3. 반수체 식물 중 잎과 줄기의 색이 yellow(버어리종 type)인 개체에 TMV저항성을 검정한 결과 저항성과 이병성의 비는 1 : 1로 나타났다. 4. (CMS MC82 Burley 21)F$_1$과 N. africana의 종간교배으로 세포질 웅성불임 isogenic Burley 21 반수체 식물의 육성이 가능하였고 이 반수체 식물의 염색체를 배가함으로써 여교배육종법에 비하여 조기에 세포질 웅성불임 Burley 21을 육성할 수 있을 것으로 생각된다.
