The aim of this study was to screen the polymorphisms and distribution of each genotype of insertion/ deletion (indel) markers which were found in a preliminary comparative study of bovine genomic sequence databases. Comparative bioinformatic analyses were first performed between the nucleotide sequences of Bovine Genome Project and those of expressed sequence tag (EST) database, and a total of fifty-one species of indel markers were screened. Of these, forty-two indel markers were evaluated, and nine informative indel markers were ultimately selected for population analysis. Nucleotide sequences of each marker were re-sequenced and their polymorphic patterns were typed in six cattle breeds: Holstein, Angus, Charolais, Hereford, and two Korean native cattle breeds (Hanwoo and Jeju Black cattle). Cattle breeds tested in this study showed polymorphic patterns in eight indel markers but not in the Indel-15 marker in Charolais and Holstein. The results of analysis for Jeju Black cattle (JBC) population indicated an observed heterozygosity (Ho) that was highest in HW_G1 (0.600) and the lowest in Indel_29 (0.274). The PIC value was the highest in HW_G4 (0.373) and lowest in Indel_6 (0.305). These polymorphic indel markers will be useful in supplying genetic information for parentage tests and traceability and to develop a molecular breeding system for improvement of animal production in cattle breeds as well as in the JBC population.
Liriope platyphylla has traditionally been used in Korea and China as a therapeutic drug for the treatment of coughing, sputum, neurodegenerative disorders, obesity, and diabetes. In an effort to assess the functions of a novel extract from Liriope platyphylla in diabetes therapy, the insulin secretion abilities of 10 extracts were screened via measurements of insulin concentration in the culture supernatant using an Insulin ELISA kit. The results of this assay showed the highest levels of insulin in the LP9M80-H treated group, followed by the LP-H, LP-M, LP-E and LP9M80-C treated groups, whereas other extracts did not induce insulin secretion in the HIT-T15 cells. However, the extracts capable of stimulating insulin secretion simultaneously evidenced high apoptotic activity as compared with other extracts. Therefore, one of these extracts, LP9M80-H, was initially selected as the optimal candidate for a therapeutic drug and its optimal concentration was determined. The results of the ELISA and MTT assay demonstrated that a concentration of approximately 100-125 ug/ml of LP9M80-H was optimal with regards to cell viability and insulin secretion in the HIT-T15 cells. These results suggest that LP9M80-H could be considered as an excellent candidate for a diabetes-therapeutic drug that could induce insulin secretion in pancreatic $\beta$-cells.
Treatment of hepatitis B virus (HBV) with lamivudine is effective in suppressing virus replication and results in reduced inflammatory activity. However the most troublesome problem of lamivudine treatment is the emergence of lamivudine-resistant strains with amino acid substitution in the YMDD motif of DNA polymerase gene during the treatment. The aim of this study was to determine the mutation of YMDD motif (codon 552) and codon 528 in chronic HBV patients with lamivudine therapy using PCR-direct sequencing and to investigate the relationship between lamivudine mediated HBV mutation and HBeAg. HBV DNA was extracted from serum samples of HBV patients and amplified by nested PCR with two sets of primer pairs selected in HBV DNA polymerase gene. Amplified PCR product was analyzed by 2% agarose gel electrophoresis and direct sequencing. HBV mutation was detected in 124 out of 207 samples (60%). Single mutation was 50.8% for M552I, 43.5% for M552V, 5.7% for M552I/V and the L528M mutation was 67.0%. Double mutation was 43.6% for M552V/L528M, 33.1% for M552I/L528(wild type), 17.7% for M552I/L528M and 5.6% for M552I/V/L528M. Serine mutation at YMDD motif (M552S) was not found and the L528M mutation frequently accompanied M552V type. In this study, the typical difference of frequencies for HBV mutation depending on HBeAg was not found. Moreover, the PCR-direct sequencing method used in this study might be a powerful tool for the mutation study in clinical reference laboratories with high volume.

Purpose: The aim of this study was to evaluate the correlation between inter-condylar width and inter-maxillary first molar width to present the criteria for prosthetic reconstruction of dental arch width in maxillary and mandibular fully edentulous patients. Materials and Methods: 120 Koreans (60 males and 60 females) who underwent the cone beam computerized tomography (Cone-beam CT) were selected. The Cone-beam CT images were analysed using Invivo 5.1. After reorientation of axis, inter-maxillary first molar width was measured by clicking both mesio-buccal cusp tip of maxillary first molar. And inter-condylar width was measured by clicking both middle points of condyles. The collected data were analysed with SPSS Version 20.0 and statistical significance of the correlation between inter-condylar width and inter-maxillary first molar width was verified by Pearson's correlation analysis. Results: The mean inter-condylar width of Korean was 105.9 mm, and that of male (108.3 mm) was statistically significantly wider than the female (103.4 mm). The inter-maxillary first molar width of Korean was 57.1 mm, and that of male (57.9 mm) was statistically significantly wider than the female (56.2 mm). Pearson's correlation analysis between inter-condylar width and inter-maxillary first molar width showed a Pearson correlation coefficient of 0.614 and statistically significantly positive correlation. Conclusion: Intercondylar width and inter-maxillary first molar width showed positive correlation and the average ratio of inter-condylar with and inter-maxillary first molar width was 1:0.54. Based on the results of this limited study, inter-condylar width can be used as a guide for setting up dental arch width in fully edentulous patient.

Genomic DNAs(gDNAs) were isolated from the venus clam(Gomphina aequilatera) from Samcheok(venus clam from Samcheok; VCS) and Wonsan(venus clam from Wonsan; VCW) located in the East Sea of the Korean Peninsula. The amplified products were generated by agarose gel electrophoresis(AGE) with oligonucleotides primer, detected by staining with ethidium bromide and viewed by ultraviolet ray. The seven arbitrarily selected primers BION-21, BION-23, BION-25, BION-27, BION-29, BION-31 and BION-33 generated the shared loci, polymorphic, and specific loci, with the molecular sizes ranging from 150 bp to 2,400 bp. In this study, 147 polymorphic loci(147/954 loci, 15.41%) in VCS population and 274(274/996 loci, 27.51%) in VCW population were generated with seven primers. These results suggest the genetic variation in VCW population is higher than in VCS population. Especially, the 700 bp bands generated by the primer BION-21 were identified commonly in two Gomphina populations, which identified populations and/or species. This specific primer was found to be useful in the identification of individuals and/or population, resulting from the different DNA polymorphism among individuals/species/population. Two Gomphina populations between the individual SAMCHEOK no. 03 and WONSAN no. 22 showed the longest genetic distance(0.696) in comparison with other individuals used. The complete linkage cluster analysis indicating three genetic groupings and dendrogram revealed close relationships among individual identities within two geographical populations of venus clam(G. aequilatera) from the Samcheok and Wonsan. The intra-species classification and clustering analyses inferred from molecular markers supported the traditional taxonomy of the species based on morphological characters such as shell size, shape and color. Accordingly, as mentioned above, RAPD analysis showed that VCS population was more or less separated from VCW population.
The eating quality of rice is one of the main concerns of rice breeding programs in many countries, especially in japonica rice cultivation areas. To select new resources with high eating quality from Korean native japonica rice, we evaluated a total of 76 varieties, including 47 native rice resources (26 landraces + 21 weedy rice) of Korea. In this study, all eating quality traits varied widely among the native resources, and some of the native resources revealed a high evaluation score in the palatability, expected eating quality, and physicochemical traits among the tested whole-plant materials. From the results, we selected two landraces (Sangdo and Waebyeo) and three weedy rice varieties (Hoengseongaengmi3, Namjejuaengmi6, and Wandoaengmi6) as promising resources for improvement of rice eating quality. Specifically, Wandoaengmi6 presented potential as a key breeding material for improving the eating quality of Korean rice cultivars, having the best evaluation results in palatability score (PS 0.83) from the sensory test and glossiness value (GV 81.8) from the Toyo taste meter of cooked rice. Given the urgent need to overcome the constraint of the narrow genetic background of Korean japonica rice, the results could be a practical solution for exploring new opportunities for improving rice eating quality through the expansion of genetic resources.
To select the effective fungicides for the control of leaf spot disease of jujube tree (Zizyphus jujuba) caused by Phoma sp., inhibitory effects of 26 fungicides for mycelial growth were investigated at $250{\mu}g\;a.i./m{\ell}$. In the test, eight fungicides were selected and minimum inhibitory concentration (MIC) for mycelial growth and an inhibitory effect for spore germination were investigated. Among the fungicides, myclobutanil, hexaconazole, and triflumizole were excluded in control effect tests because of their relatively high MICs. MICs were ranged $10-50{\mu}g\;a.i./m{\ell}$ for benomyl, carbendazim + kasugamycin (CK), and thiophanate-methyl. triflumizole (TT), and $50-250{\mu}g\;a.i./m{\ell}$ for iprodione + propineb (IT) and iminoctadine-triacelate (IT). However, benomyl and IP showed very low inhibitory effect on conidial germination. When the fungicides were sprayed on the seedlings before the leaves were inoculated with conidial suspension of Phoma sp., the protective values of CK and TT were around 70% at 1,000 ppm and around 90% at 2,000 ppm. The protective values were around 70% at 2,000 ppm (benomyl), 4,000 ppm (IP), and 8,000 ppm (IT). When the fungicides were sprayed after inoculation, benomyl showed the highest curative values of over 90% at 1,000 ppm and the values of CK and TT ranged $70{\sim}80%$ at 1,000 ppm. However, IP and IT had little or no effect on therapy of the disease. IT caused necrotic phytotoxicity on the leaves of jujube seedlings. As results, the best fungicides for the protection of jujube trees from leaf spot disease were CK (2,000 ppm) and TT (2,000 ppm) and for the remedy of the tree, benomyl (1,000 ppm) was the best. Therefore, alternate application of benomyl and CK or TT will be effective in the disease control.
In this paper it is outlined the methodology of estimating the parameters of water balance analysis method for calculating recharge, using ground water level rises in monitoring well when values of specific yield of aquifer are not available. This methodology is applied for two monitoring wells of the case study area in northern area of the Jeiu Island. A water balance of soil layer of plant rooting zone is computed on a daily basis in the following manner. Diect runoff is estimated by using SCS method. Potential evapotranspiration calculated with Penman-Monteith equation is multiplied by crop coefficients($K_c$) and water stress coefficient to compute actual evapotranspiration(AET). Daily runoff and AET is subtracted from the rainfall plus the soil water storage of the previous day. Soil water remaining above soil water retention capacity(SWRC) is assumed to be recharge. Parameters such as the SCS curve number, SWRC and Kc are estimated from a linear relationship between water level rise and recharge for rainfall events. The upper threshold value of specific yield($n_m$) at the monitoring well location is derived from the relationship between rainfall and the resulting water level rise. The specific yield($n_c$) and the coefficient of determination ($R^2$) are calculated from a linear relationship between observed water level rise and calculated recharge for the different simulations. A set of parameter values with maximum value of $R^2$ is selected among parameter values with calculated specific yield($n_c$) less than the upper threshold value of specific yield($n_m$). Results applied for two monitoring wells show that the 81% of variance of the observed water level rises are explained by calculated recharge with the estimated parameters. It is shown that the data of groundwater level is useful in estimating the parameter of water balance analysis method for calculating recharge.
Purpose : Epidemic keratoconjunctivitis (EKC) caused by adenovirus is a highly contagious disease, which has been reported as outbreaks involving adults in the community. However, there has been no report on EKC outbreak by adenovirus in a neonatal intensive care unit (NICU) in Korea. Aims of this study were to investigate the EKC outbreak by adenovirus type 8 in NICU and to confirm an effectiveness of polymerase chain reaction (PCR) for diagnosis. Methods : Conjunctival swab or nasopharyngeal aspirate specimens were taken from all patients and tested by viral culture and PCR. Adenovirus serotype was determined by sequencing of PCR product of selected region of hexon gene using the virus isolates or specimens. Results : An outbreak of EKC occurred which was involving 12 preterm infants in the NICU of the Seoul National University Children's Hospital between July 12th and August 1st, 2005. Three hospital staffs and one family member of the neonate were also affected. Adenovirus was detected in 12/12 (100%), 6/11 (54.5%) by PCR and virus culture, respectively. Eleven PCR-positive neonates were identified as serotype 8 by sequencing. The first affected 4 babies have had routine ROP (retinopathy of prematurity) examinations one week ago. While previous outbreaks were sustained for a few months, the event in our unit was controlled without complications in 3 weeks. Conclusion : We analyzed the EKC outbreak by adenovirus type 8 in NICU. Adenovirus serotype was identified by PCR and sequencing with high sensitivity for the first time in Korea, so we suggest this method can be very useful for rapid diagnosis and infection control.

This study was carried out to compare the agronomic characteristics, forage yield, and feed value of nine sorghum ${\times}$ sudangrass hybrids at paddy field cultivation. The nine recommended sorghum ${\times}$ sudangrass hybrids used in this study were P877F, Cow pow, Turbo gold, Maxi graze, Jumbo, G7, Sweet home, Honey chew BMR and Green star. Plant height, leaf number, stem diameter, and stem hardness were higher in Jumbo than other varieties (P<0.01). Sugar content (brix) was higher in order of P877F > Sweet home > maxi graze > Green star > Cow pow > G7 (P<0.01). Fresh (86,199kg/ha) and dry matter yield (16,206 kg/ha) of Jumbo were higher than other varieties (P<0.01). Crude protein was the highest in Maxi graze (6.5%), but crude fat was the highest in Honey chew BMR as 2.1% (P<0.01). NDF and ADF of Jumbo and G7 were higher than other varieties (P<0.01). TDN was higher in order of Maxi graze > Sweet home > P877F > Honey chew BMR > Green star, but no significant differences were found among the varieties. Minerals were the highest in Cow pow (15,020.5 mg/kg), and Sweet home (6,222.6 mg/kg) was the lowest as compared to other varieties (P<0.01). Total amino acids were higher in order of Maxi graze > Sweet home > Turbo gold > Honey chew BMR > Jumbo (P<0.01). Crude protein yield and crude fat yield were the highest in Turbo gold (814.6 kg/ha) and Honey chew BMR (309.8kg/ha), respectively (P<0.01). Mineral yield was the highest in Cow pow as 207.6 kg/ha (P<0.01). Amino acid yield (592.2 kg/ha) and TDN yield (10,194 kg/ha) were the highest in Sweet home. Based on the above results, sorghum ${\times}$ sudangrass hybrid varieties should be selected on the basis of the purpose of use. Because, each varieties showed various growth characteristics and nutrition yield (protein, fat, mineral, amino acid, TDN).
