Micro-CT Analysis of Cranial Bone and Tooth Density in Mice Deficient for GDF11 or Myostatin

  • Purpose: Growth differentiation factor 11 (GDF11) and myostatin (MSTN) are closely-related transforming growth factor β family members reported to play crucial roles in bone formation. We previously reported that, in contrast to MSTN, GDF11 promotes osteogenesis of vertebrae and limbs. GDF11 has been also reported as an important regulator in tooth development by inducing differentiation of pulp stem cells into odontoblasts for reparative dentin formation. The goal of this study was to investigate the differential roles of GDF11 and MSTN in dental and cranial bone formation. Methods: Micro-computed tomography analysis was performed on cranial bones, including frontal, parietal, and interparietal bones, and lower incisors of wild-type, Gdf11 knockout (Gdf11-/-), and Mstn knockout (Mstn-/-) mice. Tissue volume, thickness, and mineral density were evaluated for both cranial bone and lower incisors. Lower incisor lengths were also measured. Because Gdf11-/- mice die shortly after birth, analysis was performed on newborn (P0) mice. Results: Compared to those of Mstn-/- mice, cranial bone volume, thickness, and mineral density levels were all significantly diminished in Gdf11-/- mice. Tissue mineral density of Gdf11-/- mice were also significantly decreased compared to wild-type mice. Likewise, lower incisor length, tissue volume, thickness, and mineral density levels were all significantly reduced in Gdf11-/- mice compared to Mstn-/- mice. Incisor length was also significantly decreased in Gdf11-/- mice compared to wild-type mice. Mstn-/- mice exhibited mildly increased levels of tissue volume, thickness, and density in cranial bone and lower incisor compared to wild-type mice although statistically not significant. Conclusions: Our findings suggest that GDF11, unlike MSTN, endogenously promotes cranial bone and tooth development.

  • Amoving aeration-membrane (MAM) bioreactor was employed for the production of 2$\mu$g/mL of tissue type Plasminogen Activator (tPA)in serum free medium from normal human fibroblast cells. This system could maintain high cell density for long periods of steady state conditions in perfusion cultivation. Under normal operating condition, shear stress was as low as 0.65 dynes/$\textrm{cm}^2$ at the agitation speed of 80 rpm. Even though cell density gradually decreased with increasing agitation speed, tPA production increased linearly with increasing shear stress within a moderate range. This culture system allowed production of 2$\mu$g tPA/mL while maintaining a high cell denisty of 1.0$\times$107 viable cell/mL.

  • This study was carried out to investigate ontogeny of stomata and aerenchyma tissue in Trapa natans L. var. bispinosa Makino, an aquatic plant. Ontogeny of stomata in this plant was an aperigenous type surrounding with 5 to 8 epidermal cells without subsidiary cells. Stomata were distributed abundantly on the upper surface of leaf, however, no stoma was found on the lower surface of leaf, and on the epidermis of reproductive organ, petiole and stem. Ontogency of aerenchyma tissue was progressed with five steps; 1) formation of angular cells by division of cortex cells, 2) development of small and large globular cells in accompany with schizogenous intercellular space, 3) enlargement of globular cells and more expansion of intercellular space, 4) cell induction of long elliptic and triarmed shape, 5) completion of the largest intercellular space from endodermis toepidermis. During the growth period two types of leaf were appeared at each node of stems; one type was a submerged and early-fallen leaf, the other was a floating leaf on water surface.

Preparation of PHBV/Collagen Nanofibrous Mats and their Tissue Compatibility Compatibilscaffolds for tissue engineering

  • The nanofibrous scaffolds were obtained by co-electrospinning PHBV and collagen Type I in HIFP. The resulting fiber diameters were in the range between 300 and 600 nm. The nanofiber surfaces were characterized by ATR-FTIR, ESCA and AFM. The PHBV and collagen components of the PHBV-Col nanofibrous scaffold were biodegraded by PHB depolymerase and a collagenase Type I aqueous solution, respectively. It was found, from the cell-culture experiment, that the PHBV-Col nanofibrous scaffold accelerated the adhesion of the NIH 3T3 cell compared to the PHBV nanofibrous scaffold, thus showing a good tissue engineering scaffold.

  • Vertebrates regenerate tissues in three ways: proliferation of cells that maintain some or all of their differentiated structure and function, redifferentiation of mature cells followed by proliferation and redifferentiation into the same cell type or transdetermination to another cell type, and activation of restricted lineage stem cells, which have the ability to transdetermine to different lineages under the appropriate conditions. The behavior of the cells during regeneration is regulated by growth factors and extracellular matrix molecules. Some non-regenerating tissues are now known to harbor stem cells which, though they form scar tissue in vivo, are capable of producing new tissue-specific cells in vitro, suggesting that the injury environment inhibits latent regenerative capacity. Regenerative medicine seeks to restore tissues via transplantation of stem cell derivatives, implantation of bioartificial tissues, or stimulation of regeneration in vivo. These approaches have been partly successful, but several research issues must be addressed before regenerative medicine becomes a clinical reality.

  • Warts, or verrucae, are benign epithelial proliferations of the skin and mucosa caused by infection with human papillomaviruses (HPV). It is now recognized that there are many different HPV types. Especially type3 is most frequently observed in flat wart. Other types, such as type2, 10, 14, 27, 28, 29, 38, and 41 are rarely encounted in flat wart. We describe here a simple and economic method for detection and identification of epidermodysplasia verruciformis-associated HPV. The method is based on polymerase chain reaction (PCR) amplification and restriction analysis. The method has been developed with cloned HPV DNA and DNA from clinical samples. Clinical samples are from either frozen tissue or paraffin-embedded tissue. Genomic fragments were obtained from two different HPV types (3 and 10). The amplification fragments were identified by a form of miniature fingerprinting, with a set of restriction enzymes that gave a unique digestion pattern for each HPV type. We have tested 74 clinical samples. Only type3 among these clinical samples is detected, and one sample is involved in neither type3 nor type10.

  • Obesity is a worldwide problem that is associated with metabolic disorders. Obesity is caused by the accumulation of an abnormal amount of body fat in adipose tissue. Adipose tissue is a major metabolic organ, and it has been classified as either white adipose tissue (WAT) or brown adipose tissue (BAT). WAT and BAT are characterized by different anatomical locations, morphological structures, functions, and gene expression patterns. WAT is mainly involved in the storage and mobilization of energy in the form of triglycerides. On the other hand, BAT specializes in dissipating energy as heat through uncoupling protein-1 (UCP-1)-mediated non-shivering thermogenesis. Novel type of brown-like adipocyte within WAT called beige/brite cells was recently discovered, and this transdifferentiation process is referred to as the "browning" or "britening" of WAT. Recently, Brown fat and/or browning of WAT have been highlights as a new therapeutic target for treatment of obesity and its related metabolic disorders. Here, we describe recent advances in the study of BAT and browning of WAT, focusing on proteomic approaches.

  • Background : Constitution is related to allergic patients. Objective : The aim of this study was to analyzed allergy Material and Method : We investigated 50 patients suffering from allergy and analyzed the relationship iris constitution and allergy. Conclusion : We analyzed allergy patients, among of them, Abdominal connective tissue weakness constitution is 33(66%), neurogenic type and hydrogenic constitution is 10(20%), bronchopulmonary connective tissue weakness constitution is 5(10%). We know that Allergic patients are abdominal connective tissue weakness constitution, neurogenic type, hydrogenic constitution and bronchopulmonary connective tissue weakness constitution.

양식 넙치(Paralichthys olivaceus)에서 발생한 랩도바이러스성 질병의 병리조직학적 특징 (Histopathology of a rhabdoviral disease of farmed flounder Paralichthys olivaceus)

  • On April of 2000 and February of 2001, a disease characterized by darkening, ascites and enlargement of the spleen occurred in flounder, Paralichthys olivaceus, farmed near northern and eastern coasts of Korea. Various degree of necrosis including liquefactive-type was observed mainly in hemopoietic tissue of spleen and kindney. Certain tissue such as branchial soft tissue including epithelium showed a similar necrotic findings. But some tissues such as intentine had several necrotic clusters of wandering cells, being possibly leukocytes. Necrotic evidences of blood cells were evident in various tissues including hemopoietic tissues. Necrobiotic bodies were frequently encountered in blood smear preparations. Bullet type-virious were detected in the spleenic cells. Each virion was about 500 nm in diameter and 170nm in length. From these results, this disease was thought to be a rhabdoviral disease.

A Retrospective Analysis of Ruptured Breast Implants

  • Background Rupture is an important complication of breast implants. Before cohesive gel silicone implants, rupture rates of both saline and silicone breast implants were over 10%. Through an analysis of ruptured implants, we can determine the various factors related to ruptured implants. Methods We performed a retrospective review of 72 implants that were removed for implant rupture between 2005 and 2014 at a single institution. The following data were collected: type of implants (saline or silicone), duration of implantation, type of implant shell, degree of capsular contracture, associated symptoms, cause of rupture, diagnostic tools, and management. Results Forty-five Saline implants and 27 silicone implants were used. Rupture was diagnosed at a mean of 5.6 and 12 years after insertion of saline and silicone implants, respectively. There was no association between shell type and risk of rupture. Spontaneous was the most common reason for the rupture. Rupture management was implant change (39 case), microfat graft (2 case), removal only (14 case), and follow-up loss (17 case). Conclusions Saline implants have a shorter average duration of rupture, but diagnosis is easier and safer, leading to fewer complications. Previous-generation silicone implants required frequent follow-up observation, and it is recommended that they be changed to a cohesive gel implant before hidden rupture occurs.
