托裡消毒散이 抗腫瘍 및 免疫作用에 미치는 效果 (The effect of TakliSodoksan extract on anti-tumor action and immune-function)

  • The purpose of this Study was to investigate effect of TakliSodokSan(TSS) on the anti-tumor, immunocytes and nitric oxide(NO) production from mice peritoneal macrophages. This Study estimated the proliferation of L1210 cell lines, A431 cell lines, Hep-G2 cell lines, K562 cell lines, 3T3 cell lines, mouse thymocytes and mouse splenocytes and NO production from pcritoneal macrophages in vitro, and estimated the proliferation of L1210 cells, thymocytes and splenocytcs, NO production from peritoneal macrophages and body weight in L1210 cells-transplanted mice in vivo. The results were obtained as follows; 1. TSS inhibited significantly the proliferation of L1210, A431, Hep-G2, K562 cell lines in vitro. 2. TSS accelerated the proliferation of mice thymocytes and splenocytes in vitro. 3. TSS was not increased the nitric oxide production from mice peritoneal macrophages in vitro. 4. TSS inhibited significantly the proliferation of L1210 cells in Ll210 cells∼transplanted mice. 5. TSS accelerated the proliferation of mice thymocytes and splenocytes In L1210 cells-transplanted mice. 6. TSS was increased significantly the nitric oxide production from mice peritoneal macrophages in L1210 cells-transplanted mice. 7. TSS was increased the body weight as comparing with control group in Ll210 cells-transplanted mice.

보폐원탕(補肺元湯)이 면역반응에 미치는 영향 (Effect of Bopaewon-tang on Immune Reaction)

  • The purpose of this research was to investigate the effects of Bopaewon-tang (BT) on immune reaction in mice. BT (500 mg/kg, p.o.) increased the viability of thymocytes, but decreased the viability of splenocytes in vivo. BT enhanced the population of $CD4^+$cells in thymocytes and enhanced the population of $Thy1^+$ cells in splenocytes and splenic $CD8^+$ cells. Also, BT decreased the production of IL-4 from splenocytes. Furthermore, BT enhanced the production of nitric oxide and the phagocytic activity of peritoneal macrophages. These results suggest that BT enhances the immune response via activation of thymocytes and macrophages.

Ethanol Eluted Extract of Rhus verniciflua Stokes Showed both Antioxidant and Cytotoxic Effects on Mouse Thymocytes Depending on the Dose and Time of the Treatment

  • For a long time Rhus verniciflua Stokes (RVS) has traditionally been used as a herbal plant. It is known to contain various biological activities. Previously, a crude ethanol extract from RVS was reported to have antioxidant effects, and antiproliferative activities, on human cancer cell lines. In this report, we prepared a highly purified ethanol extract from RVS, which did not contain the urushiol derivatives, named REEE-1 ($\underline{R}$hus $\underline{e}$thanol $\underline{e}$luted $\underline{e}$xtract-1), to investigate the mechanisms of the scavenging activity of hydroxyl radicals using mouse thymocytes. The results from the deoxyribose, DNA nicking, and glucose/glucose oxidase enzyme assays showed that REEE-1 contained a strong scavenging activity of oxygen free radicals, especially of hydroxyl radicals. However, interestingly, REEE-1 also showed cytotoxicity against the thymocytes, although the effect was variable, depending on the concentrations and times of treatment. The REEE-1-mediated cytotoxicity against thymocytes, which has been used as one of the well-characterized models for apoptosis studies, was verified to be apoptotic. This was proven by the following: the appearance of DNA laddering, increases in DNA fragmentation, low fluorescence intensity in the nuclei after propidium iodide staining, and positive Annexin V staining of the cells. These results suggested that REEE-1 had both antioxidative activity and cytotoxicity against the thymocytes, although the effect of the cytotoxicity was variable, depending on the dose and time of the treatment.

야관문이 생쥐의 면역세포 활성에 미치는 영향 (Effect of Lespedeza Cuneata G. Don on the Activity of Murine Immune Cells)

  • The purpose of this research was to investigate the effects of 50% ethylalcohol extracts of Lespedeza cuneata G. Don (LE) on the activity of murine immune cells. LE was administered orally once a day for 7 days at the dose of 250 mg/kg. LE increased the cell viability of thymocytes, splenocytes and peritoneal macrophages in vitro and in vivo system, but decreased the cell viability of thymocytes and splenocytes in the presence of concanavalin A in vivo system. Also, the administration of LE was increased the production of ${\gamma}$-interferone, but did not affect the production of interleukin-2 and interleukin-4. Furthermore, LE decreased the phagocytic activity of peritoneal macrophages in vitro and in vivo system, but enhanced the production of nitric oxide. These results suggest that LE has a immuno-regulative action via stimulation of immune cells proliferation.

황기와 행인(杏仁)이 면역세포(免疫細胞)의 Apoptosis 및 Nitric Oxide에 미치는 효과(效果) (The effect of Astragali Radix and Armeniacae Semen on the apoptosis of immunocytes and nitric oxide production from peritoneal macrophages)

  • The purpose of this research was to investigate effects of Astragali Radix(AR) and Armeniacae Semen(AS) on T-lymphocytes and peritoneal macrophages in mice. The proliferation of thymocytes and splenocytes were teated using macroplate-reader. The apoptosis and sub-population of T-lymphocytes were tested using a flow cytometer. Nitric oxide production was tested using a Griess reagents. The result were obtained as follow; 1. AR increased the proliferation of thymocytes and splenocytes. 2. AS decreased the proliferation of thymocytes and splenocytes. 3. AR and AS decreased No production fron peritoneal macrophages 4. AR and AS were accelerate T-lymphocytes apoptosis. 5. AR and AS increased $T_C$ cells population, but decreased $T_H$ cells population of T-lymphocyte.

Hydrocortisone 이 흉선세포(胸線細胞)에 미치는 영향(影響) (Effect of Hydrocortisone on Thymus Cells)

  • On day 2 after treatment of Lewis rat with 25mg of hydrocortisone, the cell number in the thymus was reduced to less than 10% of the matched control. By day 12 after hydrocortisone treatment the thymic cell population was recovered almost its original value and on day 24 after treatment the number of thymocytes was equivalent to that of normal thymocytes. The density distribution profile of hydrocortisone treated rats as compared with normal rats showed a marked decrease in the denser fraction D while the lighter fractions. (A plus B, and C) showed a considerable proportional increase. The proportion of dead cells in thymus suspension from hydrocortisone treated rats was higher than that from their normal counterparts. On separation, the dead cells accumulated selectively in the pellet and A fraction. The response of the thymocytes from hydrocortisone treated rats to PHA was increased compared to that from normal rats. Among the subpopulations, D fraction, which was relatively unresponsive in normal rats, showed a marked increase in PHA response and C fraction showed some increase in the response.

A small carbohydrate fraction from Artemisia Folium suppresses death of the mouse thymocytes in vitro by down-regulating the Fas death receptor gene

  • Artemisia Folium is a preparation of dried leaves from Artemisia species and has been used traditionally to prevent or treat various kinds of woman's diseases. A similar preparation called Chinese Moxa has been used to treat rheumatism by moxibustion in Chinese medicine. A small carbohydrate fraction of approximately 1,000 dlaton from the water-soluble extract of the Artemisia Folium promoted survival of the mouse thymocytes in culture. A mouse gene array study suggested that the fraction might modulate Fas/FasL dependent apoptotic cell death and thus had influence on the survival of the thymocytes in culture. (omitted)

소음인(少陰人) 승양익기탕(升陽益氣湯)의 면역조절작용(免疫調節作用) (Immunoregulatory Action of Soeumin Seungyangikkitang)

  • The purpose of this research was to investigate the effects of Seungyangikkitang (SIT) on the immune cells in BALB/c mice. SIT (500mg/kg) was administerd p.o. once a day for 7 days. SIT enhanced the proliferation of thymocytes, but decreased the proliferation of splenocytes. SIT enhanced the subpopulation of cytotoxic T cells in thymocytes and helper T cells in splenocytes, but did not affect the subpopulation of B220/Thy1 cells. SIT enhanced the production of γ-interferon and interleukin-2 in thymocytes, splenocytes and serum, but did not affect the production of interleukin-4. SIT suppressed the production of nitric oxide, but enhanced the lucigenin chemiluminescence and the engulfment of FITC-conjugated E. coli particles in peritoneal macrophages. These results suggest that SIT has a potent activity on the specific immunity via the cytokine secretion of Th1 cells and the non-specific immunity via the phagocytic activity of macrophages in vivo.

천문동(天門冬)이 항암(抗癌) 및 면역세포(免疫細胞)에 미치는 영향(影響) (The effect of Asparagi Tuber on Anti-cancer and Immunocytes)

  • 동의학(東醫學)에서는 질병의 발생에 대하여 정기(正氣)의 강약(强弱)에 그 관건(關鍵)이 있다하였다. 그리하여 모든 질병의 치료에 있어서도 정기(正氣)의 회복에 중점을 두는 부정법(扶正法)이나 거사법(祛邪法)을 사용하면서도 정기(正氣)에 손상(損傷)을 입히지 않도록 많은 관심을 기우리고 있다. 암(癌)의 치료 또한 정기(正氣)가 얼마만큼 보충되는가에 따라 항병능력(抗病能力)이 항진(亢進)된다할 수 있을 것이다. 현재 국내외적(國內外的)으로 악성종양(惡性腫瘍)을 치료하기 위하여 다방면으로 연구를 하고 있으며, 또한 실제로 사용하고 있는 치료방법들로는 수술요법(手術療法) 방사선요법(放射線療法) 면역료법(免疫療法) 화학요법(化學療法)등이 있지만 아직까지는 미흡한 상태라 할 수 있을 것이다. 현재 일반적으로 항암제(抗癌劑)를 이용한 화학요법(化學療法) 등이 사용되고 있고, 면역료법(免疫療法)을 위한 많은 방법들이 연구되고 있어 본 저자(著者)들은 천문동(天門冬)이 면역세포(免疫細胞)의 증식(增殖)을 촉진시켜주면서 항암작용(抗癌作用)이 있을 것으로 사료(思料)되는 바 피부암세포(皮膚癌細胞)인 A431 cell line과 골수암세포(骨髓癌細胞)인 KHOS-NP cell line에 천문동(天門冬)을 투여(投與)하여 암세포(癌細胞)의 증식(增殖)을 살펴보고, 면역세포(免疫細胞)인 T cell과 B cell의 증식(增殖)을 살펴보았다. 또한 복강내(腹腔內) macrophage에서 분비되는 NO의 양(量)을 In vitro와 In vivo실험(實驗)을 통하여 살펴보았고, T cell의 apoptosis 및 subpopulation의 양(量)을 관찰하였다. 이 결과(結果) 천문동(天門冬)은 A431 cell 및 KHOS-NP cell에 항암작용(抗癌作用)을 보였고, T-cell의 증식(增殖)을 촉진시켰으며, 마우스 복강내(腹腔內) macrophage에서 분비(分泌)되는 NO의 양(量)을 감소시켜고, T cell의 apoptosis에 있어서는 대조군(對照群)에 비하여 유의성(有意性)있게 증가(增加)시켰으며, subpopulation에서는 $T_H$ cell을 증가(增加)시켰다. 그리하여 천문동(天門冬)은 항암작용(抗癌作用) 뿐만아니라 면역세포(免疫細胞)의 증식(增殖)에도 관여하는 약물(藥物)로 인정(認定)할 수 있을 것으로 사료(思料)된다.
