더덕으로부터 면역세포 활성 성분의 분리 (Isolation of Active Components on Immunocytes from Codonopsis Lanceolatae)

  • The purpose of this research was to isolate an active component on immunoytes from 70% MeOH extract of Codonopsis lanceolatae Radix(CLE). CLE was fractionated successively with hexane, methylene chloride, n-butyl alcohol and water, and then the water fraction was separated with molecularporous membrane tubing(m.w. 3,500). Each fraction(50mg/kg) was administered p.o. once a day for 7 days in BALB/c mice respectively. None of these fractions affected the apoptosis and mitochondrial transmembrane potential in thymocyte. Hexane and methylene chloride fractions decreased CD4$^{[-10]}$ CD8$^{+}$ single-positive cells, and the water fraction enhanced CD4$^{+}$ CD8$^{[-10]}$ single-positive cells in thymocyte. The proliferation of thymocytes was decreased by the fraction hexane, but was enhanced by the water fraction. Hexane, methylene chloride and butyl alcohol fractions suppressed the production of nitric oxide, which was not affected by the water fraction. Hexane and butyl alcohol fraction suppressed the phagocytic activity, but water fraction enhanced it. The components(m.w. 3,500 above) separated from the water fraction enhanced the proliferation of thymocyte, the population of CD4$^{+}$ CDB$^{[-10]}$ single-positive cells, and phagocytic activity in macrophage. These results suggest that the stimulative components of proliferation, TH population and phagorytc activity is in the water fraction, and the molecular weight is 3,500 above.   • We have identified a T cell-activating material in the culture supernatant of Streptomyces species. The factor in microbial culture supernatant (MCS) induced thymocyte proliferation in a does dependent fashion and it could be detected by immunoblot analysis using anti-interleukin-1(IL-1) antibody. The factor in MCS was slightly larger(about 21 kd) in its molecular weight than IL-1 on SDS-PAGE. When 125I-MCS was covalently coupled with homo-bifunctional cross-linking agent, disuccinimidyl-propionate to IL-1 receptor(IL-1R) on mouse thymoma cell(EL-4) and immunoprecipitated with anti-IL-1R antibody the molecular weight of this complex of 110 kd was observed.

당귀용회환의 세포독성(細胞毒性)에 관(關)한 연구(硏究) (A Study on Cytotoxicity of Dangkwi-Yonghoe-Hwan)

  • The purpose of this research was to investigate cytotoxicity of DangkwiYonghoe-Hwan(DYH) and the constitutive crude drugs on several cancer cell-lines, thymocytes, splenocytes and 3T3 cells. The DYH consists of Coptidis Rhizoma, Scutellariae Radix, Phellodendri Cortex, Gardeniae Fructus, Gentianae scabrae Radix, Indigo pulverata Levis, Aloe, Rhei Rhizoma, Moschus, Saussureae Radix and Angelicae Gigantis Radix. The cytotoxicity was determined by MTT method. The DYH inhibited the proliferation of MOLT-4, K562, HL-60, Jurkat, L1210, P815, S180 and Yac-1, thymocyte, splenocyte and 3T3 cells. The cytotoxicity of Coptidis Rhizoma on the cancer cell-lines was the most potent in the constitutive crude drugs. The proliferation of cancer cell-lines was partly inhibition and partly increase by the treatment of Scutellariae Radix, Gardeniae Fructus, Gentianae scabrae Radix, Indigo pulverata Levis, Aloe, Rhei Rhizoma, Moschus and Angelicae Gigantis Radix. Phellodendri Cortex and Saussureae Radix had a poor cytotoxicity on cancer cell-lines. Coptidis Rhizoma and Phellodendri Cortex inhibited the proliferation of thymocyte, splenocyte and 3T3 cells.

증손오적환(增損五積丸) 비적방(脾積方)이 생쥐의 복강암에 미치는 영향 (Effects of Jeungson-Ojeok-Hwan-Bijukbang on Solid Tumor in Mice)

  • Objective : Jeungson-Ojeok-Hwan Bijukbang(JOH) has been used to treat patients with tumor etc. In the theory of Korean medicine, JOH can treat Juk-Chui. JOH is known to have treat Juk-Chui. Juk-Chui is analogous to tumor. Methods : For these reasons, the present study was designed to investigate the effects of JOH on solid tumor in mice in terms of immune-potentiating and direct cytotoxic action of JOH in vitro and vivo study. The present author investigated thymocyte and splenocyte proliferation to confirm immune-potentiating activity of JOH and also investigated tumor/body weight ratio and survival rates in tumor bearing mice. Results : In this study, administration of JOH decreased tumor/body weight ratio significantly, and prolonged survival duration compared to non-treated control. In addition, treatment with JOH suppressed proliferation rate of Sarcoma 180 (S-180) cells significantly, and elevated proliferation rates of thymocytes isolated from normal mice. These results were co-related with in vivo study. Conclusion : these results suggest that JOH is useful to treat patient with solid tumor, because JOH has direct toxic action for tumor cell and immune-potentiating action for T cells. Further study will need to elucidate exact mechanisms related in anti-cancer action of JOH.

  • To effectively purify of IL-1 inhibitor from human febrile urine, we have established monoclonal antibody that reacts with human recombinant interleukin l$\beta$(IL-1$\beta$). The antibody, designated ON-1, was highly specific to IL-1$\beta$ and no cross-reaction with other cytokines(IL-l$\alpha$ and IL-4) was observed. As the results of ELISA inhibition assay and Western blotting method, it was further identified that ON-1 had high binding specificity with IL-1$\beta$. IL-1 receptor binding material from febrile patient urine was effectively purified with affinity column chromatography which conjugated with ON-1. This urinary material inhibited the thymocyte proliferation in a dosedependent manner. IL-l$\beta$ induced thymocyte proliferation activity was inhibited to 67.3% at 6 $\mu\textrm{g}$ of the purified urinary material. The result may suggest that this urinary material the purified urinary material. The result may suggest that this urinary material will have antagonic effect on IL-1 action mechanism and act IL-l$\beta$ inhibitor.

Effects of Acanthopanax sessiliflorus on Immune Cells such as Thymocytes, Splenocytes and Macrophages in Mice

  • Objective : Immune potentiation including activation of T cells, B cells, macrophages, and dendritic cells is known to play a key role in prevention and treatment of patients with cancer. In this study, we investigated the effects of Acanthopanax sessiliflorus (AR) on the immune system, especially on thymocytes, splenocytes, and macrophages. Methods : We investigated the effects of AR on proliferation of splenocytes in normal mice, and the effects on proliferation of splenocytes and thymocytes in tumor-bearing mice. In addition, the effect of AR on NO production using macrophages was investigated. Results : Treatment with AR accelerated proliferation of splenocytes in vitro. AR also accelerated thymocyte proliferation, but did not affect splenocytes proliferation in normal mice. In contrast, AR accelerated proliferation of splenocytes and thymocytes significantly in tumor bearing mice. In addition, NO production level from macrophages was elevated by treatment with AR. Conclusion : These results demonstrate that AR has anti-cancer activities and related mechanisms are involved in immune potentiation such as acceleration of immune cell proliferation and elevation of NO production level in macrophages. In addition, we also demonstrate the possibilities of AR as complementary and alternative medicine to standard anti-cancer drugs.

울금이 MDA-MB-231 세포 및 DMBA로 유발된 흰쥐의 유방암에 미치는 영향 (Effects of Curcuma longa L. on MDA-MB-231 Human Breast Cancer Cells and DMBA-induced Breast Cancer in Rats)

  • Objectives: Breast cancer is the most common cancer among women and has rapidly increasing rate annually. At present, western cancer therapies by surgery, radiation, and anticancer drug have not been fully effective. So many interests are given to herbal medicine on cancer treatment recently. This study was designed to investigate the effects of Curcuma longa L. (CL) on MDA-MB-231 human breast cancer cells and DMBA-induced breast cancer in rats. Methods: In this experiment, MDA-MB-231 cells were cultured in cell culture plates. 0.0625, 0.125, 0.25, 0.5, 1.0 mg/ml of CL extract were tested for their anti-proliferative effects on MDA-MB-231 cells by MMT assay. And we induced breast cancer in rats. The changes in tumor's weight, and the effects on proliferations of splenocyte and thymocyte were investigated. Results: CL showed anti-proliferative effects on MDA-MB-231 cells in proportion to concentration of the CL. DMBA-induced breast cancer in rats, tumor's weight of the rat was not statistically significant, but showed a tendency to be reduced in the groups treated with CL. Proliferation rate of the rat's splenocyte and thymocyte increased in proportion to CL. In breast cancer tissue, expression of ER-${\alpha}$ was weakened proportionately to the concentration of the CL. Conclusions: These data suggest that CL can prevent the proliferation of breast cancer, then CL is useful to treat patient with breast cancer.

내소옥설탕(內消沃雪湯)이 항암 및 면역에 미치는 영향 (Effects of NaisoOkseol-Tang (NOT) on solid tumor and immunity)

These results were co-related with in vivo study. In addition, NO production from abdominal macrophages isolated from normal mice was not affected by treatment with NOT. Conclusion : NOT is useful to treat patient with solid tumor, because NOT has direct toxic action for tumor cell and immune-potentiating action for T cells.

아토피피부염이 유발된 마우스에 대한 3종 가감방풍통성산(활석 제외)의 효과 비교 (Effects of Three Types of GagamBangpungtongseong-san(Except Talcum) on the Atopic Dermatitis in Mice)

  • Objectives : This study was to examine the effects of 3 types of BTS which were excluded talcum only or replaced talcum to Lonicera japonicae Flos or Kochiae Fructus on the DNCB-induced atopic dermatitis in mice. Methods : In this study, Balb/c mice were divided into five groups: normal, control, GBT(BTS except talcum), GBTG(GBT added Lonicera japonicae Flos), and GBTJ(GBT added Kochiae Fructus). And the effects on atopic dermatitis were evaluated by weight change, ear's thickness and weight, thickness of dorsal skin, severity scale of dorsal skin, histopathologic findings of dorsal skin by H&E and toluidine blue stain, proliferation of splenocyte and thymocyte in vitro, proliferation of splenocyte in vivo, IL-4, TNF-α, IgE in serum. Results : There were no significantly changes in body weight and effect of ear's weight in GBT, GBTG, and GBTJ group. The thickness of ear of GBT and GBTJ group showed significant decrease. And the thickness of dorsal skin of GBTJ group significantly decreased compared to the control, GBT, and GBTG group. All the treated groups significantly decreased in severity scale, histopathologically reduced epidermal thickness, and mast cell infiltration. In vitro, all the treated groups increased in the proliferation rates of splenocyte. However, in vivo study, it showed a falling tendency and GBT group significantly decreased compared to control, GBTG, and GBTJ group. In vitro study, GBTG group significantly decreased in the proliferation rates of thymocyte. There was no IgE contents chnage in GBT, GBTG, and GBTJ groups but IL-4 and TNF-α contents were significantly decreased. Conclusions : GBT, GBTG, and GBTJ are expected to improve symptoms of atopic dermatitis and further studies are needed for development of BTS's transformation.

  • Objective : Phyllostachyos Folium (PF) has been used to treat patients with febrile disease consuming the body fluids marked by fever with restlessness, thirst etc. In the theory of herbology, PF can clear away heat and promote the production of the body fluids, relieve restlessness. Recently PF is known to have anti-bacterial, anti-oxidantic effects. Methods : The present study was carried out to investigate the effects of PF on solid tumor in mice in terms of immune-potentiating and direct cytotoxic action of PF in vitro and vivo study. The present author investigated thymocyte and splenocyte proliferation to confirm immune-potentiating activity of PF and also investigated tumor/body weight ratio and survival rates in tumor bearing mice. Result : In this study, administration of PF decreased tumor/body weight ratio significantly, and prolonged survival duration compared to non-treated control. In addition, treatment with PF suppressed proliferation rate of Sarcoma 180 (S-180) cells significantly, and elevated proliferation rates of thymocytes isolated from normal mice. These results were co-related with in vivo study.
