Effect of Bleaching and Permanent Wave Manipulation on the Amino Acid Composition of Hair (모발 탈색 및 퍼머넌트 웨이브가 모발의 아미노산 조성에 미치는 영향)

  • Variations of the seventeen amino acids(aspartic acid, threonine, serine, glutamic acid, proline, glycine, alanine, cystein, valine, methionine, isoleucine, leucine, tyrosine, phenylalanine, histidine, lysine, arginine) were analyzed in human hair sample by amino acid auto analyzer(AAA). The effect of bleaching and permanent wave manipulation on the amino acid composition of hair were investigated. Hair samples were collected from 10 males in their thirties. Hair samples were treated with 10 mL of 6 N hydrochloric acid at $110^{\circ}C$ for 24 h and analysed by AAA. The results showed that the amino acid content of normal hair(73.9%) decreased to 71.5% and 69.3% after bleaching and permanent wave treatment, respectively. Furthermore, mean contents of lysine and tyrosine in amino acids showed obviously decreased about 25% by permanent wave and bleaching treatment. On the other hand, serine, cystein, leucine and histidine were not changed.

Characteristics of Hericium erinaceus and its Extracts (노루궁뎅이 버섯 및 추출물의 특성)

  • The functional properties of Hericium erinaceus were analyzed. The crude protein content was 8.01%. free sugars were mainly composed of glucose(47.09 mg%) and fructose(34.65 mg%), but sucrose and maltose were not detected. The free amino acids were mainly glutamic acid(1,468.12 mg%), alanine(716.07 mg%) and threonine(643.95 mg%) in Hericium erinaceus. It doesn't difference between water and ethanol extract on soluble solid content. Comparing minerals of extracts from Hericium erinaceus, water extract showed higher contents than ethanol extract except for I This tendency is similar to superoxide scavening activity and electron donating activity. But phenolic compounds, ethanol extract was higher than water extract. In comparison of water and ethanol extract of Hericium erinaceus, as a whole water extract was excellent.

Downregulation of Cyclin D1 by Sophorae Flos through Proteasomal Degradation in Human Colorectal Cancer Cells

  • Although Sophorae Flos (SF) has been reported to exert an anti-cancer activity, molecular targets and mechanisms associated with anti-cancer activity of SF have been unclear. Because cyclin D1 has been regarded as an important regulator in the cell proliferation, we focused cyclin D1 and investigated the effect of SF on the cyclin D1 regulation in light of elucidating the molecular mechanism for SF’s anti-cancer activity. The treatment of SF decreased cellular accumulation of cyclin D1 protein. However, SF did not change the level of cyclin D1 mRNA. Inhibition of proteasomal degradation by MG132 attenuated SF-mediated cyclin D1 downregulation and the half-life of cyclin D1 was decreased in the cells treated with SF. In addition, a point mutation of threonine-286 to alanine attenuated SF-mediated cyclin D1 downregulation. Inhibition of ERK1/2 by a selective inhibitor, PD98059 suppressed cyclin D1 downregulation by SF. From these results, we suggest that SF-mediated cyclin D1 downregulation may result from proteasomal degradation through its threonine-286 phosphorylation via ERK1/2. SF-induced proteasomal degradation of cyclin D1 might inhibit proliferation in human colorectal cancer cells.   • Raf protein kinases and protein kinase C belong to serine/threonine-specific proteins in the cytoplasin, and are similar to each other in functional structure and the aspect of the distribution of celI. The distribution of raf protein kinase in the cerebrum of Geoclemys reevesfi as studied by using the antibodies against a-raf and c-raf protein kinase which induce the expression of raf fainily oncogenes. In general, raf protein kinases were distributed in such restricted regions as the general pallium, hippocampal formation, pdmordiuin hippocampi,nucleus of lateral olfactory tract, basal amygdaloid nucleus, and bed of stria terminalis. Immunological labeling of c-raf protein kinase was more widespread than that of a-raf. However, the intensity of the labeling of c-raf was lower than that of a-raf. The spherical cells of basal amygdaloid nucleus is a ring-like form, because only the cytoplasm was imunolabeled. Especially, c-raf protein kinase occurred in the cells which contained protein kinase C abundandy such as pyramidal cells and Purkinje cells. This suggests that a- and e-raf protein kinases may synegistically induce carclnoma with myc gene which is activated by protein kinase C.

Analysis of Amino Acids, Vitamins and Minerals of Fruiting Body of Fomitopsis pinicola (소나무 잔나비버섯의 아미노산, 비타민 및 무기물 분석)

  • The nutritional composition of Fomitopsis pinicola (F. pinicola) fruiting body has been analyzed for medicinal and edible uses. The contents of crude fibers, carbohydrates, crude protein, moisture, crude fats and ashes were 43.3%, 26.3%, 12.8%, 12.6%, 3.3% and 1.7%, respectively. Eighteen amino acids were found in F. pinicola. Among total amino acids, glutamate content was the highest (457 mg/100 g dry mushroom) and arginine, glycine, valine, aspartate and isoleucine were followed. Concerning free amino acids, glutamine, arginine, trytophan, and glutamate were dominant. The vitamin E content was the highest (276 mg/100 g dry mushroom), then vitamin H and vitamin B, were followed. The mineral contents were as follows: K 165.06 mg, P77.57 mg, Mg 46.11 mg, Fe 21.56 mg, and Ca 16.90 mg based on 100 g dry mushroom.

  • This study was carried out in order to investigate the browning reaction of fish oil-amino acid model system at different temperatures and watger activities. The 23 amino acids, induced during dehydration in the presence of oil and avicel(5 to $45^{\circ}C$), Aw 0.33 to 0.95, were resulted in three types of browning patterns : Type I showed high browning rates at Aw 0.33, 0.95 than at Aw 0.52, 0.75(phenylalanine, trans-4-hydroxy-L-proline, methionine, valine). Type II showed high browning rates decreased with increasing water activity(poline, leucine, isoleucine, arginine). Type III showed high browning rates at Aw 0.52, 0.75 than at Aw 0.33, 0.95(tryptophan, cystein, threonine, lysine). The temperature effect on the browning development of the four most active amino acids : phenylalanine, valine, trnas-4-hydroxy-proline and methionine are shown to represent the 23 amino acids. Above $25^{\circ}C$ the browning rate began to increase. Activation energy of the amino acids-fish oil was 8 to 40kcal/mole, and $Q_{10}$ were 2 to 10.

Production of theobromine from caffeine by Pseudomonas sp. (Pseudomonas sp.에 의한 Caffeine으로부터 Theobromine의 생산)

  • For the production of theobromine from caffeine, 5 strains of bacteria capable of producing theobromine were isolated from soil. Among them, the strain CT-017 showed the best ability of producing theobromine, and was partially identified as a Pseudomonas sp. For the production of theobromine, fructose was the most effective carbon source at an optimum concentration of 5 g/l. The most effective nitrogen source was 5 g/l of beef extracts. And 0.02 g/l of $Fe^{2+}$, 1.0 g/l of threonine were effective for the production of theobromine. The optimum temperature and initial pH were $28^{\circ}C$ and 6.5, respectively. After 23 hr cultivation, 7.98 g/l of theobromine was produced from 15 g/l of caffeine which corresponds to a conversion yield of 53.2%.

  • 1) The Moisture and glycogen contents of salted-oyster (Zin Suk Hwa Jut) were much less than those of raw oyster, while the free sugar content was dominent. 2) The amino acids were detected and identified as aspartic acid, glutamic acid, cystine, serine, glycine, lysine, threonine, histidine, alanine, arginine, valine, methionine, tyrosine, phenylalanine, leucine, iso-leucine and tryptophan in the juice of salted-oyster by using the method of paper chromatography, and the all essential amino acids from nutritional view point was admitted in this juice. 3) The juice contained 0.264mg/ml of tyrosine, 0.304mg/ml of arginine, 0.046mg/ml of tryptophan, 0.460mg/ml of histidine, 0.350mg/ml of methionine, 0.440mg/ml of cystine and 0.115mg/ml of phenylalanine.

  • To investigate changes of components in salt-fermented sardine, Sardinops melanostictus sauce during 18 months fermentation, various chemical properties were examined at 2$\~$3 months intervals. The degree of hydrolysis increased sharply until 5 months of fermentation and showed the gentle increasement after that. On the other hand, the contents of total and amino nitrogens, total ATP related compounds increased gradually during 18 months of fermentation. The hypoxanthine and uric acid were abundant in ATP related compounds, ranging from $75\%$ to $87\%$. The contents of inosine+hypoxanthine and uric acid were crossed at 13.9 months of fermentation. After 18 months of fermentation, sauce was rich in free amino acids, glutamic acid, aspartic acid, lysine, alanine, threonine in that order.

Anti-Proliferative Effect of Naringenin through p38-Dependent Downregulation of Cyclin D1 in Human Colorectal Cancer Cells

  • Naringenin (NAR) as one of the flavonoids observed in grapefruit has been reported to exhibit an anti-cancer activity. However, more detailed mechanism by which NAR exerts anti-cancer properties still remains unanswered. Thus, in this study, we have shown that NAR down-regulates the level of cyclin D1 in human colorectal cancer cell lines, HCT116 and SW480. NAR inhibited the cell proliferation in HCT116 and SW480 cells and decreased the level of cyclin D1 protein. Inhibition of proteasomal degradation by MG132 blocked NAR-mediated cyclin D1 downregulation and the half-life of cyclin D1 was decreased in the cells treated with NAR. In addition, NAR increased the phosphorylation of cyclin D1 at threonine-286 and a point mutation of threonine-286 to alanine blocked cyclin D1 downregulation by NAR. p38 inactivation attenuated cyclin D1 downregulation by NAR. From these results, we suggest that NAR-mediated cyclin D1 downregulation may result from proteasomal degradation through p38 activation. The current study provides new mechanistic link between NAR, cyclin D1 downregulation and cell growth in human colorectal cancer cells.
