Decolorization of Poly R-478 Dye by Coriolus versicolor IFO 30388 (구름버섯(Coriolus versicolor IFO 30388)에 의한 Poly R-478 염료의 탈색)

  • Effects of nitrogen and carbon sources on the decolorization rate of poly R-478 dye by a white rot basidiomycete Coriorus versicolor IFO 30388 were examined. The fungus exhibited 87.2% of decolorization rate when it was cultured in the state of stationary in a nitrogen-limited medium (pH 4.5) which contained 2.0% glucose, 0.04% ammonium tartrate, 0.02% poly R-478 dye, 2% $KH_2PO_4$, 0.5% $MgSO_4{\cdot}7H_2O$, 0.1% $CaCl_2{\cdot}2H_2O$, 0.002% thiamine-HCl and 10 mM 2,2 dimethylsuccinate (sodium) at $28^{\circ}C$ for 10 days. Decolorization of the dye occurred in the presence of nitrogen source in the medium and decolorization rate increased rapidly after depletion of $NH_4^+$ from the medium.

Isolation, Identification and Culture Condition of Microorganism Producing Glutathione (Glutathione 생산균주의 분리 동정 및 생산조건)

  • The strain which produced glutathione was isolated from soil samples. The isolated strain was identified as the genus Candida through its morphological. cultural and physiological characteristics. The highest production of glutathione by Candida sp. was obtained after cultivation with shaking for 36 hours in the culture medium containing fructose 1.0%, yeast extract 4.0%. NaCI 0.04%, thiamine-HCI $5{\mu}g$/ml, and L-cysteine 0.04%. The optimal pH and temperature for the glutathione production were pH 6.0 and $25^{\circ}C$, respectively. The glutathione production by Candida sp. under the optimal culture condition was $92{\mu}g/ml$.

  • Nutritional requirements for the growth of Flammulina velutipes were studied. Mannitol, glutamic acid and ammonium nitrate were chosen for the maximum mycelial growth when various carbon and nitrogen sources tested. Optimum C : N ratio for the mycelial growth was 20 : 1. Potassium dihydrogen phosphate was selected among the phosphate sources. Magnesium sulphate and thiamine HCl stimulate mycelial growth. Final compositions of optimized chemically defined medium were 1.5% mannitol, 0.082% $NH_4NO_3$, 0.312% glutamic acid, 0.25% $KH_2PO_4$, 0.06% $MgSO_4{\cdot}7H_2O\;and\;0.3\;{\mu}g/l$ thiamin HCl. This medium not only support mycelial growth but also induce fruit body formation.

  • Coriolus versicolor, is one of the most popular medicinal mushrooms due its various biologically active components. This study was conducted to obtain basic information regarding the mycelial culture conditions of C. versicolor. Based on the culture, and MCM media were suitable for the mycelial growth of the mushroom. The optimum carbon and nitrogen sources were dextrin and yeast extract, respectively, and the optimum C/N ratio was 10 to 2 when 2% glucose was used. Other minor components required for optimal growth included thiamine-HCl and biotin as vitamins, succinic acid, lactic acid and citric acid as organic acids, as well as $MgSO_4{\cdot}7H_2O$ as mineral salts.

  • This study was carried out to determine the basic mycelial culture conditions for Poria cocos growth. According to colony diameter and mycelial density, suitable media for mycelial growth were Malt yeast extract, Potato dextrose agar, Yeast extract agar, and Yeast malt agar. The optimum temperature for mycelial growth was between 25 and $35^{\circ}C$, and the optimum pH value was between 4 and 7. Carbon and nitrogen sources were fructose and yeast extract. The optimum C/N ratio was about 10 to 1 with 2% glucose. Other minor components for optimal growth were thiamine-HCl and nicotinamide as vitamins, acetic and lactic acid as organic acids, and $MgSO_4{\cdot}7H_2O$ and $FeSO_4{\cdot}7H_2O$ as mineral salts.

  • Auricularia auricula-judae is an edible mushroom, which is known as wood ear, free ear, black ear mushroom, and free jelly fish. This study was carried out to obtain the basic information for mycelial culture conditions of Auricularia auriculajudae. According to colony diameter and mycelial density, the media for suitable mycelial growth were PDA and MCM. The optimum temperature for mycelial growth was $25{\sim}30^{\circ}C$.   • The optimal cultural conditions for production of the bluish purple prgment by the cultivation of Streptomyces californicus KS-89 were determined with various substrates. The carbon and nitrogen sources on the production of pigment indicated that soluble strarch and glycerol as carbon sources and sodium glutamate sodium nitrate as nitrogen sources given a mzimum yield of the pigment at 3$0^{\circ}C$ for 7 days. The addition of ferrous sulfate was essential. The highest production of pigment was observed with cultivation in a medium containing 2.0% soluble starch 1% glycerol 0.5% sodium glutamate 0.05% sodim nitrate 0.001% L-proline 0.025% K2HPO4 0.005% MgSO4 .7H2O, 0.04% FeSO4.7H2O, 0.001% thiamine.HCl and pH7.0.

  • The production of cellulolytic enzymes by Pleurotus sajor-caju JAFM 1017 was stimulated by folic acid and thiamine-HCl. Among the inorganic salts, optimum concentrations of $KH_2PO_4$ and $MgSO_4{\cdot}7H_2O$ were 0.2% (w/v) and 0.04% (w/v), respectively, but other inorganic salts were not effective for the production of the enzymes. The optimum culture temperature and pH for the production were $25^{\circ}C$ and 5.5 for avicelase, and $30^{\circ}C$ and 5.0 for CMCase, and $30^{\circ}C$ and 6.5 for ${\beta}-glucosidase$, respectively.

  • This study was carried out to obtain the basic data for artificial culture of Grifola umbellata. The optimal condition for the mycelial growth was obtained at $20^{\circ}C$ and pH 4, respectively. G. umbellata showed the most favorable growth on the Hoppkins media. Carbon sources such as glucose, fluctose and manitol were favorable for stimulating a mycelial growth of G. umbellata. Valine, one of nitrogen sources also appeared to be favorable to a mycelial growth. The optimum C/N ratio was about 30:1 in case that 1% glucose as carbon source was mixed with the basal media. Lactic acid as organic acid was most favorable to the mycelial growth. Also, thiamine-Hcl as vitamin source was favorable. The mineral nutrient of $FeSO_4$ or $MgSO_4$ was most favorable to G. umbellata, and its optimal concentration was about 0.01% in $FeSO_4$ and 0.1 % in $MgSO_4$ respectively. Among 4 different cereal extract media, polished rice extract medium which was mixed with silkworm pupae was most suitable for a favorable growth of G. umbellata.

  • Over 90 of lysine producing auxotrophs were obtained from Corynebacterium sp. S-27-12, Brevibacterium flavum ATCC 15168 and Micrococcus glutamicus ATCC 13032 by UV light, $Co^{60}$ irradiation and N-methyl-N'-nitro-N-nitrosoguanidine treatment. One of the mutant, Brev. flavum U46-N59, was identified as a leucine auxotroph and accumulated lysine during flask (500 ml) cultivation (180 strokes/min.) up to 21.6 mg per ml of broth at pH 7.5 and $28^{\circ}C$ after 4 days. The medium consisted of glucose, 100; urea, 10; corn steep liquor, 40; $KH_2PO_4,\;2;\;K_2HPO_4,\;0.5;\; MgSO_4.\;7H_2O,\;0.4;\;antifoam\;S-57,\;1g;\;Fe_2(SO_4)_3.XH-2O,\;10;\; MnCl_2,\;4H_2O,\;10mg;\;biotin,\;30;\;thiamine-HCl,\;100{\mu}g$in 1l of distilled water, and 40 U/ml of penicillin was added after 36 hrs fermentation.
