  • A bacterial strain J-59 was isolated from a humus soil, which produced simultaneously a thermostable glucose isomerase as well as xylanase. The morphological, cultural and physiological characteristics of the isoisomerase strain J-59 were detemined by the use of the media and methods described in International Streptomyces Project. The chemotaxonomic characteristics of the isolated strain J-59 were determined by the analysis of G+C molar % of DNA, diaminipimelic acid, composition of fatty acid and menaquinone. As the results of various examinations, the strain J-59 was identified to be Streptomyces chibaensis. This strain produced glucose isomerase intracellularly and xylanase extracellularly when grown in a medium containing xylan, but it was not able to utilize the xylose or xylan as a carbon source. The glucose isomerase of S. chibaensis J59 was highly thermostable, which retained more than 75% activity in the presence of Co$^{2+}$ at 80$\circ $C for 72 h.

  • A thermostrable ${\beta}-galactosidase$ (${\beta}-galactoside$ galactohydorlase, EC 3.2.1.23) was inducible in Bacillus coagulans by lactose and D-glactose. The enzyme was purified 87 fold, and the optimum temeprature and pH for actiivity were determined to be $60^{\circ}C$ and pH 7.5, respectively. Kinetic determinations at $55^{\circ}C$ established a Km of 3.3mM for the chromogenic substrate onitorphenyl ${\beta}-D-galactopyranoside$ (ONPG). Galactose and lactose were competitive inhibitors with Ki of 6.1mM and 4.9mM, respectively. The enzyme ws relatively thermostable. The crude enzyme was inactivated about 20% after 20 min of exposure at $60^{\circ}C$ and the purified was about 50%. Maximal enzyme activity required $Mn^{++}$, and for the thermal stabilization $Fe^{++}\;and\;Ca^{++}$ were necessary.

  • A gene (GenBank AF096282) coding for a $\alpha$-glucosidase (TcaAG, EC 3.2.1.20) from Thermus caldophilus GK24 was expressed in Saccharomyces cerevisiae, a generally recognized as safe (GRAS) host. The thermostable $\alpha$-glucosidase was produced inside of the GRAS host at 0.04 unit/mg-dry cell by the constitutively expressing ADH1 promoter and at 1.2 unit/mg-dry cell by the inductively expressing GALl0 promoter, respectively. No $\alpha$-glucosidase activities were found in the medium when the MF-alpha signal sequence from S. cerevisiae or $\alpha$-amylase signal sequence from Aspergillus oryzae were fused before the $\alpha$-glucosidase gene for the secretion.

Characterization of the Intact Form of Thermotoga maritima Pectinase TmPecN Expressed in Escherichia coli

  • The thermostable pectinase gene TmPec isolated from Thermotoga maritima was introduced into the NdeI site of pRSET-B vector and expressed in its intact form in Escherichia coli BL21. The overexpressed intact form of pectinase (TmPecN protein) was partially purified by heat-denaturation procedure. TmPecN showed the highest activity between 85 and $95^{\circ}C$, and at approximately pH 6.5. Enzyme activity was stably maintained at temperatures below $85^{\circ}C$. In the presence of $Ca^{2+}$, pectinase activity of TmPecN increased to 128.4% of normal level. In contrast, $Ba^{2+}$, $Zn^{2+}$, and $Mn^{2+}$ strongly inhibited TmPecN activity. We conclude that the biochemical properties of the intact form of TmPecN are comparable to those of the recombinant protein TmPec reported previously.

Engineering Recombinant Streptomyces coelicolor Malate Synthase with Improved Thermal Properties by Directed Mutagenesis

  • Streptomyces thermovulgans malate synthase (stMS) is known to be more thermostable and thermoactive than S. coelicolor malate synthase (scMS). Therefore, based on the amino acid sequence of stMS, 3 scMS mutants, namely P186R, T8PL9P, and T8PL9PP186R, were created by site-directed mutagenesis in an attempt to engineer a more thermoactive and thermostable enzyme. An enzymatic analysis of the wild-type and mutant MS revealed that P186R and T8PL9PP186R were more thermoactive than the wild-type scMS and T8PL9P. Furthermore, all 3 mutants exhibited a greater thermo stability than scMS, thereby suggesting that both R186 and P8P9 can cause increased thermo stability in scMS.

Cellulase-Free Thermostable Alkaline Xylanase from Thermophilic and Alkalophilic Bacillus sp. JB-99

  • The characterization of a partially purified, cellulase-free, thermostable alkaline xylanase from thermoalkalophilic Bacillus sp. JB-99 was investigated. The xylanase production was the highest when birchwood xylan was added to a medium containing finely powdered rice bran, showing 4,826 IU$ml^-1$ of activity for 15 h of incubation. The partially purified xylanase exhibited an optimum temperature and pH at $70^C{\circ}$ and 10, respectively. The enzyme was stable at pH 5-11 at $50^C{\circ}$. The xylanase activity was strongly inhibited by $Hg^2+$, while dithiothreitol, cysteine, and ${\beta}$-mercaptoethanol enhanced the activity.

  • Actinomycetes CS0707 has been isolated in soil sample from location in the Jeju province. Korea, and produces thermostable extracellular proteases. Actinomycetes CS0703 showed the highest protease activity at late exponential phase when grown in OSYM medium (oatmeal 2.0%, soybean meal 1 %, dried yeast 1 %, mannitol 1 %) at $48^{\circ}C$. Three forms of protease(Ta-1, TA-2 and Ta-3) were fractionated by Ultrogel AcA 54 column chromatography, and further purified through ammonium sulfate fractionation, ultramembrane filtration, and DEAE-sepharose CL-6B column chromatography. Production of L-DOPA by Thermostable Tyrosine Phenol-lyase of a Thermophilic Symbiobacterium Species Overexpressed in Recombinant Escherichia coli

  • A thermostable tyrosine phenol-lyase gene of a thermophilic Symbiobacterium species was cloned and overexpressed in Escherichia coli in order to produce the biocatalyst for the synthesis of 3, 4-dihy-droxyphenyl-L-alanine (L-DOPA). The substrates used for the synthetic reaction were pyrocatechol, so-dium pyruvate, and ammonium chloride. The enzyme was stable up to $60^{\circ}C$, and the optimal temperature for the synthesis of L-DOPA was $37^{\circ}C$ . The optimal pH of the reaction was about 8.3. Enzyme activity was highly dependent on the amount of ammonium chloride and the optimal concentration was estimated to be 0.6 M. In the case of pyrocatechol, an inactivation of enzyme activity was observed at con-centrations higher than 0.1 M. Enzyme activity was increased by the presence of ethanol. Under op-timized conditions, L-DOPA production was carried out adding pyrocatechol and sodium pyruvate to the reaction solution intermittently to avoid substrate depletion during the reaction. The concentration of L-DOPA reached 29.8 g/l after 6 h, but the concentration didn t increase further because of the formation of byproducts by a non-enzymatic reaction between L-DOPA and pyruvate.

Purification and Characterization of a Thermostable Xylose (Glucose) Isomerase from Streptomyces chibaensis J-59

  • Xylose (glucose) isomerase was purified to homogeneity from cell-extracts of Streptomyces chibaensis J-59 via ammonium sulfate precipitation followed by chromatography on DEAE-cellulose, and gel filtration on Sephacryl S-300. The purified enzyme is a homotetramer with a native molecular mass of 180 kDa and a subunit molecular mass of 44 kDa. The amino acid N-terminal sequence of glucose isomerase from S. chibaensis J-59 was determined to be Ser-Tyr-Gln-Pro-Thr-Pro-Glu-Asp-Arg-Phe-Thr-Phe-Gly-Leu. The first 14 amino acids of the N-terminal sequence of the enzyme showed strong analogies with N-terminal sequences of glucose isomerase produced by other Streptomyces spp. The optimum pH and temperature for activity were 7.5 and 85, respectively. The purified enzyme required $Mg^{2+}$, $Co^{2+}$, and $Mn^{2+}$ for the activity, $Mg^{2+}$ being the most effective. The enzyme was not inhibited by $Ca^{2+}$, but was inhibited by $Hg^{2+}$, $Ag^+$, and $Cu^{2+}$. The $K_m$, $V_{max}$, and $k_{cat}$ values of S. chibaensis J-59 isomerase for glucose were 83 mM, 40.9 U/mg, and $1,843min^{-1}$, respectively. In the presence of $Co^{2+}$, cell-free enzymes retained 100% without loss of activities by the heat-treatment at $70^{\circ}C$ for 7 days. The enzyme retained 50% residual activity after heating at $85^{\circ}C$ for 13.5 h, at $90^{\circ}C$ for 126 min. The enzyme is more thermostable than any other glucose isomerases of Streptomyces spp.

Purification and Characterization of a Thermostable Protease from Pseudomonas aeruginosa NS-83

  • A bacterial strain NS-83 isolated from soil was able to produce an extracellular thermostable protease. The strain was identified as Pseudomonas aeruginosa based on its morphological and physiological characteristics. A thermostable protease from this strain has been purified to homogeneity as judged by SDS-PAGE and isoelectric focusing. The purification procedures included hydrophobic interaction, ion exchange, and gel filtration chromatography. The $M_r$ and the pl of the enzyme were 32,000 and 5.9, respectively. The optimal pH at 55$^{\circ}C$ and the optimal temperature at pH 7.0 were 8.0 and 60$^{\circ}C$, respectively. The D-values of the enzyme at 60, 65, and 70$^{\circ}C$ were 22, 2.1, and 0.75 hrs, respectively. The enzyme activity was significantly inhibited in the presence of 1 mM o-phenanthroline or EDTA, suggesting that the enzyme is metalloprotease. The $K_m$, and $V_{max}$ for Hammarsten casein were found to be 3.2 mg/ml and 0.918 unit/ml, respectively. These enzymatic properties were similar to those of elastase produced from P. aeruginosa IFO 3455, but the enzyme was clearly different from the reported elastase, in respect to $Ca^{++}$ effects on enzyme-thermostability. This property, together with amino acid composition analysis, confirmed that the enzyme differs from the known P. aeruginosa elastase.
