Growth of Seeded Escherichia coli in Rewetted Cattle Waste Compost of Different Stages

  • Compost is used mainly as an organic fertilizer, but it is also used as bedding material for cattle. Dairy cattle have been identified as a main reservoir of pathogenic Escherichia coli O157:H7. Further, E. coli is regarded as an environmental pathogen that causes bovine clinical mastitis. Hence, its growth in compost spread or compost bedding should be avoided. Physical and chemical conditions, available nutrients and microflora in compost change greatly during the composting process. Since pathogen growth in compost seems to be related to these changes, we assessed the possibility of E. coli growth in compost samples collected at 0, 7, 13, 22, 41, 190 and 360 d. Cattle waste composts with and without added tofu residue were collected from static piles and immediately air-dried. Compost samples were inoculated with a pure culture of E. coli, the moisture content was adjusted to 50%, and the samples were incubated for 5 d at $30^{\circ}C$. The numbers of E. coli in compost before and after incubation were determined by direct plating on Chromocult coliform agar. Almost all compost samples supported E. coli growth. Samples collected during or immediately after the thermophilic phase (day 7) showed the highest growth. Growth in samples more than 13 d old were not significantly different from those of aged compost samples. The addition of tofu residue gave a higher growth than its absence in younger samples collected prior to 13 d. To minimize the risk of environmental mastitis, the use of compost in the initial stage of the process is better avoided.

Application of TAO System and RDF for Treatment of Cattle Manure (우분뇨의 고형연료화와 고온호기산화 공정 적용 가능성에 관한 연구)

  • We studied the possibility to produce solid fuel using cattle manure and to apply TAO (Thermophilic Aerobic Oxidation) process of solid-liquid separation fraction. The physiochemical compositions of cattle manure solid fuel chip were analyzed as water 0.12%, low calorific value 3,510 kcal/kg, ashes 11.9%, chlorine 0.82%, sulfur dust 0.5%, mercury non-detection, cadmium 1.0 mg/kg, lead 2 mg/kg, arsenic non-detection. In treating cattle manure with TAO reactor the internal temperature of the reactor was increasing higher and $50^{\circ}C$ and over was maintained after 20 hours on. The physiochemical compositions of liquids increased from pH 7.3 to pH 9.18 and EC decreased from 4.6 to 3.48 mS/cm in treating process of cattle manure with TAO reactor. COD and SCOD decreased from 16,800 to 10,400 mg/L, from 4,600 to 2,040 mg/L respectively, which showed about 38% and 56% of remove efficiency respectively.

  • The composting process is a suitable to dispose the livestock manure in terms of resources recovery. However the performence of composting process is greatly affected by the environmental conditions such as characteristics of manure, type of the bulking agent, initial moisture contents, temperature, recycle and so on. The purpose of this study is to evaluate the optimum environmental conditions of composting process for livestock manures. The analytical results indicated that no bulking agent was necessary for the cow manure because of the proper C/N ratio. However the pig manure required a bulking agent since the pig manure had not only low in C/N ration but poor ventilation characteristics. In addtion, the initial miosture content for optimum composting appeared to be about 60%. The temperature control was also an essential factor to enhance the activity of thermophilic microorganisms in the laboratory composting unit. It was further found that the recycle of composts may contributed the completion of composting precess as well as C/N ratio reduction and moisture control.

  • Most of small- scale livestock facility have difficulties to treat organic wastewater by activated sludge process as distinguished feces and urine. The organic wastewater discharged have characteristics of slurry, high concentration of BOD, 55 etc. This study was performed to estimate suitable condition for the application of packing materials, air flow, pollutant load and so on as important parameter to treat organic wastewater by thermopile oxlc process. As a result obtained at this experiments the most suitable condition for BOD load, air flow indicate 3.0kg · m$^{-3}$ day$^{-1}$,50 ∼ 100L· min$^{-1}$ m$^{-3}$, respectively, and we knew that it was necessary to add waste contained high calories to degrade about 80% of pollutant among waste- water. It showed that plastic material can be used as packing media because it can be provided as inhabitation for microorganism owing to intensity of material and characteristic of keeping moisture.

  • Ground bed cultivation method for shiitake mushroom (Phogo; Lentinula edodes) has been advanced to use the oak-sawdust composted by microorganisms. This method has some advantages in saving the labors and improving productivity. The aim of this study is to analyze microbial community in oak sawdust and to provide the information for efficient cultivation of shiitake mushroom. In process of composting, the proportions of thermophiles increased from 10% to 80%. This results show the microbial community succession from mesophilic bacteria to thermophilic bacteria. T-RFLP analysis and nucleotide sequencing of 16S rRNA gene demonstrated that the change of dominant bacteria coupled with shifts in each stages. The family of Enterobacteriaceae as a mesophilic bacteria were dominant (100%) in oak sawdust sample, but Amycolatopsis (49.0%) and Saccharopolyspora (26.5%) as a thermophilic Actinomycetes were dominant in the last day of composting. In hyphal growth stage, Leuconostoc (psychrophilic bacteria) was dominant (75.0%). The composting process of sawdust is very important for growth of mushroom mycelium. Therefore, there is need for the further study for the Amycolatopsis as thermophilic Actinomycetes.

Bacterial Community Dynamics during Composting of Food Wastes (음식물 쓰레기 퇴비화 과정에 따른 세균군집 구조의 변화)

  • Composting is a biological process converting solid organic waste into valuable materials such as fertilizer. The change of bacterial populations in a composting reactor of food waste was investigated for 2 months. Based on shifts in temperature profile, the composting process could be divided into the first phase ($2^{\circ}C\sim55^{\circ}C$), the second phase ($55^{\circ}C\sim97^{\circ}C$), and the third phase ($50^{\circ}C\sim89^{\circ}C$). The number of total bacteria was $1.66\times10^{11}$ cell/g, $0.29\times10^{11}$ cell/g, and $0.28\times10^{11}$ cell/g in the first, second, and third stages, respectively. The proportions of thermophiles increased from 33% to 89% in the second stage. T-RFLP analysis and nucleotide sequencing of 16S rRNA gene demonstrated that the change of bacterial community structure was coupled with shifts in composting stages. The structure of bacterial community in the ultra-thermophilic second stage reflected that of seeding starter. The major decomposers driving the ultra-thermophilic composting were identified as phylotypes related to Bacillus and Pseudomonas.

Effects of diverse Pre-treatment methods on the sludge digestion and methane production in combined mesophilic anaerobic and thermophilic aerobic sludge digestion process (다양한 전처리에 따른 중온혐기-고온호기 복합 슬러지 처리공정의 슬러지 처리효율 및 메탄 생성량 변화)

  • In this study, various influent sludge pre-treatment methods were adopted to investigate their effects on the sludge digestion and methane production in combined mesophilic anaerobic and thermophilic aerobic sludge digestion process. A lab-scale sewage sludge digestion process was operated during 4 phases using different feed sludge pre-treatment strategies. In phase 1, feed sludge was supplied without any pre-treatment. In contrast, in phases 2, 3 and 4, thermal, thermal-alkaline and long time alkaline treatment (7 days) were applied to influent sludge, respectively. With sludge pre-treatment, TCOD removal was drastically increased from 44% to 76% from phases 1 to 4, respectively. Also, pre-treatment of feed sludge significantly improved the methane production rate of MAD, showing an increment from 101 to 165-256mL/L/day. Meanwhile, TCOD removal and methane production at phase 4 were not increased, compared to those at phase 3. Based on the experimental results, it was concluded that pre-treatment of feed sludge significantly increases the efficiency of sludge digestion and thermal-alkaline method was the most effective method among the pre-treatment methods examined.

Bacterial Communities Developing during Composting Processes in Animal Manure Treatment Facilities

  • We analyzed succession of the bacterial communities during composting of animal manure in three individual facilities. Polymerase chain reaction (PCR) and denaturing gradient gel electrophoresis (DGGE) targeting for the bacterial 16S rRNA gene were used to clarify the changes of bacterial community throughout each composting process. Our study revealed that the bacterial community structures differed during the composting process. The bacterial community in composting of facility A showed little change throughout the process. In the compost sample from facility B, its community had a small shift as the temperature increased. In compost from facility C, the temperature dynamically changed; it was shown that various bacterial communities appeared and disappeared as follows: in the initial phase, the members of phylum Bacteroidetes dominated; in the thermophilic phase, some bacteria belonging to phylum Firmicutes increased; towards the end, the community structure consisted of three phyla, Bacteroidetes, Firmicutes, and Proteobacteria. This study provides some information about the bacterial community actually present in field-scale composting with animal manure.

Comparative Effects of Υ-irradiation and Ethylene Oxide Fumigation on the Quality of Carrot Powder (Υ-선 및 훈증제 처리가 당근분말의 품질에 미치는 영향)

  • For the purpose of improving microbiolgical quality of carrot powder, which is being used as minor ingredients of convenience foods, comparative influence of Υ-irradiation and ethylene oxide(E.O) fumigation was investigated with emphasis on sterilizating effect and physicochemical propertied Carrot powder sample was contaminated with thermophiles 4.0$\times$103/g, acid tolerant bacteria 3.7$\times$103/g and coliforms 1.6$\times$102/g, respectively. And thus sanitation process was required. E.O fumigation in commercial practice was not enough to destroy thermophilic bacteria, while Υ-irradiation at below 5 kGy could reduce all microorganisms up to undectected levels. Radiosensitivity(D10) of thermophiles was shown to be 2.25 kGy. The applicable dose of irradiation to the improvement of microbiological quality showed insignificant influences on the physicochemical quality of the sample.

Comparative Studies on Static Windrow and Aerated Static Pile Composting of the Mixtures of Cattle Manure and Rice Hulls -I. Variation of Physico-chemical Parameters (우분뇨와 왕겨 혼합물의 퇴비화에서 정치식과 통기퇴적식의 비교연구 -I. 퇴비재료의 이화학적 환경변화)

  • Variations of temperature and physicochemical environment during composting of a mixtures(2:1, v/v) of cattle manure and rice hulls(CMR) in two different composting methods, static windrow(SW) and aerated static pile system(ASPS), were monitored for evaluating the efficient composting system in greenhouse. The pH of composting materials increased to around 8.9 initially, then decreased and stabilized slowly to the neutral value. Composting materials in ASPS showed a rapid stabilization in pH value from the 4th week comparing to the speed in SW. Thermophilic stage for ASPS Lasted at 3 week whereas 6 weeks for WS. Required time to get thermophilic zone in compost was shorter in ASPS than in WS. Reduction rate in total carbon(T-C) was higher in ASPS than in WS. Organic matter was reduced more rapidly in ASPS than in SW showing 9 percent difference after the 6th week. Total nitrogen(T-N) increased while composting process, showing 9 percent after 6th week in WS and 1.8 percent after 7th week in ASPS. C/N ratio was stabilized after 6th week showing 17 and 21 level in WS and ASPS each. Quantity of ash and mineral content increased during composting in both system, showing higher content in ASPS. Composting process by intermittent, aerated static pile system in greenhouse had a significant effect on the reduction of required period for composting.
