The effect of heat stress on frame switch splicing of X-box binding protein 1 gene in horse

  • Objective: Among stress responses, the unfolded protein response (UPR) is a well-known mechanism related to endoplasmic reticulum (ER) stress. ER stress is induced by a variety of external and environmental factors such as starvation, ischemia, hypoxia, oxidative stress, and heat stress. Inositol requiring enzyme $1{\alpha}$ ($IRE1{\alpha}$)-X-box protein 1 (XBP1) is the most conserved pathway involved in the UPR and is the main component that mediates $IRE1{\alpha}$ signalling to downstream ER-associated degradation (ERAD)- or UPR-related genes. XBP1 is a transcription factor synthesised via a novel mechanism called 'frame switch splicing', and this process has not yet been studied in the horse XBP1 gene. Therefore, the aim of this study was to confirm the frame switch splicing of horse XBP1 and characterise its dynamics using Thoroughbred muscle cells exposed to heat stress. Methods: Primary horse muscle cells were used to investigate heat stress-induced frame switch splicing of horse XBP1. Frame switch splicing was confirmed by sequencing analysis. XBP1 amino acid sequences and promoter sequences of various species were aligned to confirm the sequence homology and to find conserved cis-acting elements, respectively. The expression of the potential XBP1 downstream genes were analysed by quantitative real-time polymerase chain reaction. Results: We confirmed that splicing of horse XBP1 mRNA was affected by the duration of thermal stress. Twenty-six nucleotides in the mRNA of XBP1 were deleted after heat stress. The protein sequence and the cis-regulatory elements on the promoter of horse XBP1 are highly conserved among the mammals. Induction of putative downstream genes of horse XBP1 was dependent on the duration of heat stress. We confirmed that both the mechanisms of XBP1 frame switch splicing and various binding elements found in downstream gene promoters are highly evolutionarily conserved. Conclusion: The frame switch splicing of horse XBP1 and its dynamics were highly conserved among species. These results facilitate studies of ER-stress in horse.

Proper Monitoring Methods for Safety Management of Tailings Dam (광물찌꺼기적치장의 안전관리를 위한 적정 모니터링 방안 연구)

  • This study has focused on analysis factors affecting safety monitoring system at tailings sites, and the evaluation equipment to monitor the factors. Twenty sites at eighteen mines with unsafe conditions were selected to examine the equipment. There were three main factors influenced safety in the sites including surface erosion, piping, and slope instability. In detail, the surface erosion was divided into three sub-factors (planting, soil-topping layer, and tailings), piping into three sub-factors (liner, rain protection facility and leachate), and slop instability was also divided into three sub-factors (slop, concrete wall, and reinforcing wall). As results of in-field measurement, a CCTV was the most effective facility, and electrical resistivity survey, acoustic sensing, thermal liner sensor, structure inclinometer, rainfall meter, and flowmeter were also highly effective. According to applications of the facilities in the unstable tailings, structural defects were mainly found in the piping, which was the most important monitoring factor for safety management of tailings sites.

  • In this study, in order to secure the high reliability of the memristor, we adopted a patterned lithium filament seed layer as the main agent for resistive switching (RS) characteristic on the 30 nm thick ZrO2 thin film at the device manufacturing stage. Lithium filament seed layer with a thickness of 5 nm and an area of 5 ㎛ × 5 ㎛ were formed on the ZrO2 thin film, and various electrode areas were applied to investigate the effect of capacitance on filament type memristive behavior in the parallel memristive circuit of memristor and capacitor. The RS characteristics were measured in the samples before and after 250℃ post-annealing for lithium metal diffusion. In the case of conductive filaments formed by thermal diffusion (post-annealed sample), it was not available to control the filament by applying voltage, and the other hand, the as-deposited sample showed the reversible RS characteristics by the formation and rupture of filaments. Finally, via the comparison of the RS characteristics according to the electrode area, it was confirmed that capacitance is an important factor for the formation and rupture of filaments.

  • Recently, as the fishery resources are depleted, expectations for productivity improvement by 'rearing fishery' in land farms are greatly rising. In the case of land farms, unlike ocean environments, it is easy to control and manage environmental and breeding factors, and has the advantage of being able to adjust production according to the production plan. On the other hand, unlike in the natural environment, there is a disadvantage in that operation costs may significantly increase due to the artificial management for fish growth. Therefore, profit maximization can be pursued by efficiently operating the farm in accordance with the planned target shipment. In order to operate such an efficient farm and nurture fish, an accurate growth prediction model according to the target fish species is absolutely required. Most of the growth prediction models are mainly numerical results based on statistical analysis using farm data. In this paper, we present a growth prediction model from a stochastic point of view to overcome the difficulties in securing data and the difficulty in providing quantitative expected values for inaccuracies that existing growth prediction models from a statistical point of view may have. For a stochastic approach, modeling is performed by introducing a Gaussian process regression method based on water temperature, which is the most important factor in positive growth. From the corresponding results, it is expected that it will be able to provide reference values for more efficient farm operation by simultaneously providing the average value of the predicted growth value at a specific point in time and the confidence interval for that value.

  • The aim of this study was to evaluate and establish of emission test method for liquid phase building materials such as paint, adhesive, sealant by emission cell. A small-scale emission chamber and emission cell were used to evaluate emission of TVOC from paint, adhesive, sealant. The quantity of TVOC emission were measured by a gas chromatography/mass spectrometry (GC/MS). Background concentration of TVOC was below $10{\mu}g/m^3$ in the emission chamber and cell. Air tightness and recovery in chamber and cell showed good results. The recovery of thermal desorber for toluene and n-dodecane were about 120%. The repeatability of response factor and retention time in GC/MS below 30%. The method detection limit of VOCs ranged 0.04~8.82 ng. The concentration of TVOC emission using emission cell was 1.35~1.41 times higher than emission chamber. The correlation of TVOC emission using chamber and cell method was significantly high (r=0.91~0.97).

Twelve-Month Volume Reduction Ratio Predicts Regrowth and Time to Regrowth in Thyroid Nodules Submitted to Laser Ablation: A 5-Year Follow-Up Retrospective Study

  • Objective: Laser ablation is a therapeutic modality used to reduce the volume of large benign thyroid nodules. Unsatisfactory reduction and regrowth are observed in some treated nodules. The aim of the study was to evaluate the long-term outcomes of laser treatment for solid nodules during a 5-year follow-up period, the regrowth rate, and the predictive risk factors of nodule regrowth. Materials and Methods: We retrospectively evaluated patients with benign, solid, cold thyroid nodules who underwent laser ablation and were followed-up for 5 years. According to the selection criteria, 104 patients were included (median baseline nodule volume, 12.5 mL [25.0-75.0%, 8-18 mL]; median energy delivered, 481.5 J/mL [25.0-75.0%, 370-620 J/mL]). Nodule volume, thyroid function test results, and ultrasound were evaluated at baseline and then annually after the procedure. Results: Of 104 patients, 31 patients (29.8%) had a 12-month volume reduction ratio (VRR) < 50.0% and 39 (37.5%) experienced nodule regrowth. Of these 39 patients, 17 (43.6%) underwent surgery and 14 (35.9%) underwent a second laser treatment. The rate of nodule regrowth was inversely related to the 12-month VRR, i.e., the lower the 12-month VRR, the higher the risk of regrowth (p < 0.001). The mean time for nodule regrowth was 33.5 ± 16.6 months. The 12-month VRR was directly related to time to regrowth, i.e., the lower the 12-month VRR, the shorter the time to regrowth (p < 0.001; R2 = 0.3516). Non-spongiform composition increased the risk of regrowth with an odds ratio of 4.3 (95% confidence interval [CI] 1.8-10.2; p < 0.001); 12-month VRR < 50.0% increased the risk of regrowth with an odds ratio of 11.7 (95% CI 4.2-32.2; p < 0.001). Conclusion: The VRR of thyroid nodules subjected to similar amounts of laser energy varies widely and depends on the nodule composition; non-spongiform nodules are reduced to a lesser extent and regrow more frequently than spongiform nodules. A 12-month VRR < 50.0% is a predictive risk factor for regrowth and correlates with the time to regrowth.

  • Fire caused by heater has various causes as many as the types of heater. also, lots of damage of human life and property loss are caused, since annually continuous fire accident by heater in traditional market is frequently occurring. There are not many cases of fire due to heater in most of residential facilities such as general house, apartments, etc., because they are supplied with heating boiler, however the restaurant, store and office of the market, sports center, factory, workplace, etc. still use heater, e.g. oilstove, electric heater, etc., so that they are exposed to fire hazard. Also, when investigating the number of fire due to heater, it was analyzed to occur in order of home boiler, charcoal stove, oilstove, gas heater/stove, electric stove/heater, the number of fire per human life damage was analyzed in order of gas heater/stove, oil heater/stove, electric heater/stove, briquette/coal heater. Also, gas and oil related heater were analyzed to have low frequency, however, with high fire intensity. Therefore, this research aimed at considering more scientific fire inspection and identification approach by reenacting and reviewing fire outbreak possibility caused by combustibles' contact and conductivity under the normal condition and abnormal condition in respect of ignition hazard, i.e. minimum ignition temperature, carbonization degree and heat flux along with it, due to oilstove and electric stove, which are still frequently used in public use facility, traditional market, and, of which actual fire occurrence is the most frequent. As the result of reenact test, ignition hazard appeared very small, as long as enough heat storage condition is not made in both test objects(oilstove/electric stove), however carbonization condition was analyzed to be proceeded per each part respectively. Eventually, transition to fire is the ignition due to heat storage, so that it was analyzed to ignite when minimum heat storage temperature condition of fire place is over $500^{\circ}C$. Particularly, in case of quartz pipe, the heating element of electric stove, it is rapidly heated over the temperature of $600^{\circ}C$ within the shortest time(10sec), so that the heat flux of this appears 6.26kW/m2, which was analyzed to result in damage of thermal PVC cable and second-degree burn in human body. Also, the researcher recognized that the temperature change along with Geometric View Factor and Fire Load, which display decrease of heat, are also important variables to be considered, along with distance change besides temperature condition. Therefore, the researcher considers that a manual of careful fire inspection and identification on this is necessary, also, expects that scientific and rational efforts of this research can contribute to establish manual composition and theoretical basis on henceforth fire inspection and identification.

A Quantification Method for the Cold Pool Effect on Nocturnal Temperature in a Closed Catchment (폐쇄집수역의 냉기호 모의를 통한 일 최저기온 분포 추정)

  • Cold air on sloping surfaces flows down to the valley bottom in mountainous terrain at calm and clear nights. Based on the assumption that the cold air flow may be the same as the water flow, current models estimate temperature drop by regarding the cold air accumulation at a given location as the water-like free drainage. At a closed catchment whose outlet is blocked by man-made obstacles such as banks and roads, however, the water-like free drainage assumption is no longer valid because the cold air accumulates from the bottom first. We developed an empirical model to estimate quantitatively the effect of cold pool on nocturnal temperature in a closed catchment. In our model, a closed catchment is treated like a "vessel", and a digital elevation model (DEM) was used to calculate the maximum capacity of the cold pool formed in a closed catchment. We introduce a topographical variable named "shape factor", which is the ratio of the cold air accumulation potential across the whole catchment area to the maximum capacity of the cold pool to describe the relative size of temperature drop at a wider range of catchment shapes. The shape factor is then used to simulate the density profile of cold pool formed in a given catchment based on a hypsometric equation. The cold lake module was incorporated with the existing model (i.e., Chung et al., 2006), generating a new model and predicting distribution of minimum temperature over closed catchments. We applied this model to Akyang valley (i.e., a typical closed catchment of 53 $km^2$ area) in the southern skirt of Mt. Jiri National Park where 12 automated weather stations (AWS) are operational. The performance of the model was evaluated based on the feasibility of delineating the temperature pattern accurately at cold pool forming at night. Overall, the model's ability of simulating the spatial pattern of lower temperature were improved especially at the valley bottom, showing a similar pattern of the estimated temperature with that of thermal images obtained across the valley at dawn (0520 to 0600 local standard time) of 17 May 2011. Error in temperature estimation, calculated with the root mean square error using the 10 low-lying AWSs, was substantially decreased from $1.30^{\circ}C$ with the existing model to $0.71^{\circ}C$ with the new model. These results suggest the feasibility of the new method in predicting the site-specific freeze and frost warning at a closed catchment.

Dosimetry of the Low Fluence Fast Neutron Beams for Boron Neutron Capture Therapy (붕소-중성자 포획치료를 위한 미세 속중성자 선량 특성 연구)

  • Purpose : For the research of Boron Neutron Capture Therapy (BNCT), fast neutrons generated from the MC-50 cyclotron with maximum energy of 34.4 MeV in Korea Cancer Center Hospital were moderated by 70 cm paraffin and then the dose characteristics were investigated. Using these results, we hope to establish the protocol about dose measurement of epi-thermal neutron, to make a basis of dose characteristic of epi-thermal neutron emitted from nuclear reactor, and to find feasibility about accelerator-based BNCT. Method and Materials : For measuring the absorbed dose and dose distribution of fast neutron beams, we used Unidos 10005 (PTW, Germany) electrometer and IC-17 (Far West, USA), IC-18, ElC-1 ion chambers manufactured by A-150 plastic and used IC-l7M ion chamber manufactured by magnesium for gamma dose. There chambers were flushed with tissue equivalent gas and argon gas and then the flow rate was S co per minute. Using Monte Carlo N-Particle (MCNP) code, transport program in mixed field with neutron, photon, electron, two dimensional dose and energy fluence distribution was calculated and there results were compared with measured results. Results : The absorbed dose of fast neutron beams was $6.47\times10^{-3}$ cGy per 1 MU at the 4 cm depth of the water phantom, which is assumed to be effective depth for BNCT. The magnitude of gamma contamination intermingled with fast neutron beams was $65.2{\pm}0.9\%$ at the same depth. In the dose distribution according to the depth of water, the neutron dose decreased linearly and the gamma dose decreased exponentially as the depth was deepened. The factor expressed energy level, $D_{20}/D_{10}$, of the total dose was 0.718. Conclusion : Through the direct measurement using the two ion chambers, which is made different wall materials, and computer calculation of isodose distribution using MCNP simulation method, we have found the dose characteristics of low fluence fast neutron beams. If the power supply and the target material, which generate high voltage and current, will be developed and gamma contamination was reduced by lead or bismuth, we think, it may be possible to accelerator-based BNCT.

Evaluation of Air Ion According to the Distance in Large Flat Park - Focused on Olympic Park in Songpa-gu, Seoul - (대규모 평지형 공원 내 거리별 공기이온 평가 - 서울시 송파구 올림픽공원을 사례로 -)

  • Increasing impervious surfaces can cause various physical and mental illnesses as well as further problems like deterioration of the thermal environment and disconnection of the water circulation system in the downtown area. This study is intended to suggest baseline data for the future creation or remodelation of large-scaled urban parks in consideration of human health on the aspect of climate by analyzing air ions condition and correlation analysis between air ions and meteorological elements depending on distances from the center in the urban park. The study site was divided into four types by distances from the center of Olympic Park in Songpa-gu, Seoul, then classified as A and B. After selecting 80 points to record, land coverage and vegetation status were investigated, and then meteorological elements and air ions were measured. First, the green space rate by section is A section Center (90%)> Inner (70%)> Outer-inner (10%) = Outer (10%), B section Center (100%)> Inner (60%)> Outer- In order of inner (30%)> Outer (0%). Second, for weather factors, the temperature was analyzed by Outer (24.18℃) > Outer-inner (22.90℃) > Inner (22.43℃) > Center (21.719℃). For relative humidity, center (46.49%) > Inner (43.36%) > Outer-inner (42.41%) > Outer (38.53%) was analyzed. Third, In the case of cations in the air ions, Outer(435.71ea/㎤) > Outer-inner(415.16ea/㎤) > Inner (389.46ea/㎤) > Center (352.64ea/㎤). In the case of negative ions, Center (569.72ea/㎤) > Inner (499.83ea/㎤) > Outer-inner(410.58ea/㎤) > Outer (292.02ea/㎤). It was analyzed to increase toward Center, and analyzed to decrease with increasing distance from road. For the Air ion index, it was analyzed as Center (1.67) > Inner (1.31) > Outer-inner (1.08) > Outer (0.71). Negative irons was analyzed to increase when its measuring points close to the center and decrease with shortening distance to the road. Fourth, as a result of correlation analysis, positive(+) correlation is negative ions and ion index. These were measured to increase as the recording points closing to the center of the park from the road. The negative(-) correlation is temperature, solar radiation, and positive ions. These increased as getting closer to the road.
