목 적: 본 연구는 대한방사선종양학회지 게재 논문의 통계 오류 현황을 파악하고 이에 근거한 문제 제기를 통해 학회지의 학술적 발전에 기여하고자 하였다. 대상 및 방법: $2006{\sim}2007$년 사이에 대한방사선종양학회지에 게재된 총 77편의 논문을 연구 대상으로 하였다. 각각의 논문에 적용된 통계 방법론의 적정성 평가는 통계점검표를 활용하였다. 통계점검표에는 연구의 종류, 통계 기법의 사용 범위, 각 논문에 적용된 통계 기법의 종류, 통계적 기법 적용의 타당성 항목이 포함되었다. 통계 오류는 '생략의 잘못'과 '시행의 잘못' 항목으로 나누었다. 한 논문에서 서로 다른 항목이 여러 가지 관찰된 경우 각각 횟수로 측정하였다. 같은 항목이 2회 이상 측정된 경우 1회로 횟수를 측정하였다. 통계 전문가가 개별 논문을 대상으로 통계점검표를 작성하였다. 일차 평가자가 방사선종양학 전문가가 아닌 것에서 올 수 있는 평가 오류가 있을 수 있으므로 개별 통계점검표는 간행위원회에서 한 차례 더 세부 점검을 하였다 작성된 통계점검표의 통계 분석은 SAS (version 9.0, SAS Institute, NC, USA) 소프트웨어를 이용하였고 빈도분석을 시행하여 각 항목의 빈도와 백분율을 산출하였다. 결 과: 총 77편 중 원문을 다운로드 할 수 없었던 4편을 제외한 73편의 게재 논문을 대상으로 평가하였다. 증례보고는 5편, 원저 논문은 58편이었다. 대상 논문 중 46편의 논문에서 통계적 추론을 사용하였고, 16편의 논문에서 단순한 기술통계를 사용했으며, 11편의 논문에서는 통계적 기법을 사용하지 않았다. 추론통계를 사용한 46편의 논문에서 사용된 추론통계의 횟수는 각각 분할표분석 17회(37.0%), 비교통계분석 23회(50.0%), 회귀분석 7회(15.2%), 상잔분석 5회(10.9%), 생존분석이 27회(58.7%)였다. 통계 기법을 활용하여 연구 결과를 분석할 때 통계적용의 오류가 없는 논문은 19%였다. '생략의 잘못'은 34편(50.0%)의 논문에서 총 50회 관찰되었다. '시행의 잘못'은 35편(51.5%)의 논문에서 총 47회 관찰되었다. '생략의 잘못'과 '시행의 잘못'이 모두 발견된 논문은 21편(30.9%)이었다. 결 론: 대한방사선종양학회지에 게재된 논문에서 통계 분석 과정의 다양한 영역에 걸쳐 크고 작은 통계적 오류가 있음을 확인하였다. 향후 대한방사선종양학회지 투고 논문의 심사과정에서 통계 오류에 관한 적절한 심사를 추가하는 것이 필요하다고 판단된다.
활성산소종으로 유도되는 연골 세포의 apoptosis는 퇴행성 관절염의 발병 기전에 중요한 역할을 한다. Schisandrin 속의 과일에서 발견되는 생체 활성 화합물인 schisandrin A는 여러 가지 약리학적 작용을 하는 것으로 보고되고 있다. 현재까지 schisandrin A의 유도체들의 항산화 효과에 대해서는 여러 연구가 보고되었지만, schisandrin A의 항산화 효능의 분자 기전은 아직 미해결 상태로 남아 있다. 본 연구는 SW1353 인간 연골세포에서 산화적 스트레스($H_2O_2$)에 대한 schisandrin A의 세포 보호 여부를 조사하였다. 본 연구의 결과에 의하면 schisandrin A는 PARP 단백질의 분해와 caspase-3의 활성 차단을 통해 $H_2O_2$에 의해 유도된 성장 억제와 apoptosis를 유의적으로 억제하였다. 이러한 schisandrin A의 anti-apoptotic 효과는 미토콘드리아 기능 손상의 억제와 pro-apoptotic Bax의 발현 증가 및 anti-apoptotic Bcl-2의 발현 감소의 차단과도 관련이 있었다. 또한, schisandrin A는 ROS의 생성과 DNA 손상 마커인 H2AX의 인산화도 효과적으로 저해하였다. 따라서 SW1353 연골세포에서 schisandrin A는 산화적 스트레스에 의한 ROS 생성의 억제를 통하여 apoptosis와 DNA 손상을 보호하였음을 알 수 있었다. 결론적으로 본 연구의 결과는 schisandrin A가 ROS의 과잉 생산으로 인한 산화적 장애에 치료적 잠재력이 있음을 보여준다.
A fibrinolytic protease (PoFE) was purified from the cultured mycelia of the edible oyster mushroom Pleurotus ostreatus, using a combination of various chromatographies. The purification protocol resulted in an 876-fold purification of the enzyme, with a final yield of 6.5%. The apparent molecular mass of the purified enzyme was estimated to be 32 kDa by SDS-PAGE, fibrin-zymography, and size exclusion using FPLC. The optimal reaction pH value and temperature were pH 6.5 and $35^{\circ}C$, respectively. PoFE effectively hydrolyzed fibrinogen, preferentially digesting the $A{\alpha}$-chain and the $B{\beta}$-chain over the ${\gamma}$-chain. Enzyme activity was enhanced by the addition of $Ca^{2+},\;Zn^{2+},\;and\;Mg^{2+}$ ions. Furthermore, PoFE activity was potently inhibited by EDTA, and it was found to exhibit a higher specificity for the chromogenic substrate S-2586 for chymotrypsin, indicating that the enzyme is a chymotrypsin-like metalloprotease. The first 19 amino acid residues of the N-terminal sequence were ALRKGGAAALNIYSVGFTS, which is extremely similar to the metalloprotease purified from the fruiting body of P. ostreatus. In addition, we cloned the PoFE protein, encoding gene, and its nucleotide sequence was determined. The cDNA of cloned PoFE is 867 nucleotides long and consists of an open reading frame encoding 288 amino acid residues. Its cDNA showed a high degree of homology with PoMEP from P. ostreatus fruiting body. The mycelia of P. ostreatus may thus represent a potential source of new therapeutic agents to treat thrombosis.
Low-density lipoprotein (LDL) induces cell proliferation in human aortic smooth muscle cells (hAoSMCs), which may be involved in atherogenesis and intimal hyperplasia. Recent studies have demonstrated that $Cl^-$ channels are related to vessel cell proliferation induced by a variety of stimuli. In this study, we investigated a potential role of $Cl^-$ channels in the signaling pathway of LDL effects on hAoSMC proliferation with a focus on the activation of Erk1/2-PI3K/Akt and the subsequent upregulation of Egr-1. $Cl^-$ channel blockers, DIDS, but neither NPPB nor Furosemide, completely abolished the LDL-induced DNA synthesis and cell proliferation. Moreover, DIDS, but not NPPB, significantly decreased LDL-stimulated $Cl^-$ concentration, as judged by flow cytometry analysis using MQAE as a $Cl^-$-detection dye. DIDS pretreatment completely abolished the activation of Erk1/2 and PI3K/Akt in a dose-dependent manner that is the hallmark of LDL activation, as judged by Western blot and proliferation assays. Moreover, pretreatment with DIDS ($Cl^-$ channel blockers) but not LY294002 (PI3K inhibitors) completely abolished the LDL-induced upregulation of Egr-1 to the same extent as PD98059 (MEK inhibitors to inhibit Erk), as judged by Western blot and luciferase reporter assays. This is the first report, to our knowledge, that DIDS-sensitive $Cl^-$-channels play a key role in the LDL-induced cell proliferation of hAoSMCs via the activation of Erk1/2 and PI3K/Akt and the upregulation of Egr-1.
Uterine leiomyoma is the most common tumor in the female genital tract. Although the tumor is benign, it is a matter of paramount importance since it often causes profuse menstrual bleeding, pressure symptoms and infertility. Nevertheless, the etiology and pathophysiology of this abnormality remain poorly understood. The traditional definitive treatment for uterine leiomyomas is hysterectomy and, even today, symptomatic leiomyomas are the leading cause of hysterectomy in Korea. Clearly, the development of a safe, effective, and nonsurgical method of treatment for leiomyoma would be of great benefit to many women. This study demonstrated growth inhibition of uterine leiomyoma cells using Haeohyultang (HT). When human leiomyoma cells were treated with Haeohyultang, cells showed dose-dependent growth inhibitory effect. Cell growth was inhibited by over 40% as determined by both cell counts and MTS assay. Reduction of cellular viability as a consequence of exposure to Haeohyultang resulted from induction of apoptosis, as assessed by DNA fragmentation, PARP cleavage, caspase 9 and caspase 3 assay. Flow cytometry analysis with uterine leiomyoma cells demonstrated sub G1 cell cycle arrest after treatment with drug Haeohyultang. But, the expression levels of p27 and p21 were not changed in Haeohyultang treated cells compared with control. However, the expression levels of clAP1 were reduced by Haeohyultang compared with control. This reduction of clAP1 data means activation of the caspase family, and then induction of PARP cleavage and apoptosis. These results suggest that Haeohyultang may be potential therapeutic approach in the clinical management of uterine leiomyoma.
본 연구에서는 참도박 에탄올 추출물(GEHEE)의 항염증 효과를 알아보기 위해 lipopolysaccharide에 의해 활성화된 대식세포로부터 분비되는 염증매개인자들의 발현량과 마우스 모델을 이용한 귀 부종 및 조직학적 관찰 실험을 진행하였다. 그 결과 염증을 유발하는 대표적 물질인 NO 및 사이토카인[interleukin(IL)-6, $IL-1{\beta}$ 및 tumor necrosis factor $receptor-{\alpha}$]의 분비량이 GEHEE 농도 의존적으로 유의적 감소를 보였다. 또한 전사인자인 nuclear $factor-{\kappa}B$의 활성과 인산화효소인 mitogen-activated protein kinases(p38, JNK 및 ERK)의 발현량이 억제됨을 확인함에 따라 염증작용 기전에서 이들의 활성 억제가 NO 및 사이토카인 분비량 조절에 영향을 미침을 확인하였다. 동물모델을 이용한 실험에서는 croton oil로 부종을 유발한 마우스 귀 조직에 GEHEE를 처리하였을 때 항염증제인 prednisolone 처리구와 유사한 수준으로 경피 및 진피의 두께가 감소한 것을 확인하였으며, 조직 내 침윤되는 mast cell의 수도 현저히 억제됨을 확인하였다. 따라서 본 연구 결과는 참도박 에탄올 추출물의 항염증 효능을 가진 기능성 소재로의 이용 가능성을 제시한다.
${\beta}$-lapachone은 남미에 자생하는 lapacho 나무(Tabeuia avellanedae)의 수액에 함유된 quinone계열의 일종으로 많은 인체암세포에서 apoptosis를 유발하는 것으로 알려져 있다. 본 연구는 A549 인체폐암세포를 대상으로 ${\beta}$-lapachone에 의한 apoptosis 유발 과정에서 나타나는 또 다른 현상들을 조사하기 위하여 실시되었다. ${\beta}$-lapachone이 처리된 A549 세포는 처리 농도의 증가에 따라 생존율이 감소되었으며, 이는 apoptosis 유발과 연관이 있음을 MTT assay와 flow cytometry 분석을 통하여 확인하였다. ${\beta}$-lapachone에 의한 A549 세포의 증식억제는 종양억제유전자 p53과 cyclin-dependent kinase 저해제인 p21의 발현을 전사 및 번역 수준에서 증가시켰으며, p53 단백질의 인산화 증가와 연관성이 있었다. 또한 ${\beta}$-lapachone은 caspase-3과 -9를 활성화시켰으며, 활성화된 caspase-3의 기질 단백질들[poly(ADP-ribose) polymerase, ${\beta}$-catenin 및 phospholipase C-$\gamma$1]의 단편화를 유도하였다. 아울러 ${\beta}$-lapachone은 cyclooxygenase (COX)-2의 mRNA 및 단백질의 발현을 억제하였으나 COX-1의 발현에는 영향을 미치지 않았으며, ${\beta}$-lapachone에 의한 COX-2의 발현억제는 prostaglandin E2의 생성 저하에 관련이 있었다. 본 연구의 결과는 ${\beta}$-lapachone의 항암활성 기전의 이해와 더불어 ${\beta}$-lapachone이 폐암세포에서 강력한 항암활성이 있음을 보여 주는 것이다.
Objectives : Citrus is the fruit that is readily available around us. Therefore, we investigated the anti-inflammatory effects of fraction isolated from the Citrus hassaku pericarp in RAW264.7 macrophage cells. Methods : The effects of fraction from Citrus hassaku pericarp on cell viability on RAW264.7 cells were measured by the MTT assay. The mRNA levels of iNOS and COX-2, its protein level by fraction of Citrus hassaku pericarp treatment in RAW264.7 macrophage cells were investigated by RT-PCR and immunoblots. Nitrite accumulation in the culture was measured colorimetrically by the Griess reaction using a Griess reagent. The amount of IL-6 and TNF-${\alpha}$ production was determined using an enzyme-linked immunosorbent assay (ELISA) kit. Results : The results indicated that the fraction of Citrus hassaku pericarp concentration highly suppressed lipopolysaccharide (LPS)-induced nitric oxide (NO) and IL-6 productions without a cytotoxic effect on RAW264.7 cells. fraction of Citrus hassaku pericarp inhibited the expressions of LPS-induced iNOS and COX-2 protein and their mRNA in a dose-dependent manner. Particularly, fraction of Citrus hassaku pericarp suppressed the level of nuclear factor-${\kappa}B$ (NF-${\kappa}B$) activity, which was linked with the suppression of LPS-induced phosphorylation of p65 at serine 276 and p65 translocation into nuclei, but not MAPK signaling. In addition, treatment with fraction of Citrus hassaku pericarp inhibited the production of IL-6 and TNF-${\alpha}$ in LPS-stimulated RAW264.7 cells. Conclusion : Our results indicate that fraction of Citrus hassaku pericarp potentially inhibits the biomarkers related to inflammation through the blocking of NF-${\kappa}B$ p65 activation, and it may be a potential therapeutic candidate for the treatment of inflammatory diseases.
BACKGROUND/OBJECTIVES: The present study aimed to further investigate the potential health beneficial effects of long-term seaweed supplementation on lipid metabolism and hepatic functions in DIO mice. MATERIALS/METHODS: Four brown seaweeds (Undaria pinnatifida [UP], Laminaria japonica [LJ], Sargassum fulvellum [SF], or Hizikia fusiforme [HF]) were added to a high fat diet (HFD) at a 5% ratio and supplemented to C57BL/6N mice for 16 weeks. Triglycerides (TGs) and total cholesterol (TC) in the liver, feces, and plasma were measured. Fecal bile acid (BA) levels in feces were monitored. Hepatic insulin signaling- and lipogenesis-related proteins were evaluated by Western blot analysis. RESULTS: Fasting blood glucose levels were significantly reduced in the LJ, SF, and HF groups compared to the HFD group by the end of 16-week feeding period. Plasma TG levels and hepatic lipid accumulation were significantly reduced in all 4 seaweed supplemented groups, whereas plasma TC levels were only suppressed in the UP and HF groups compared to the HFD group. Fecal BA levels were significantly elevated by UP, LJ, and SF supplementation compared to HFD feeding only. Lastly, regarding hepatic insulin signaling-related proteins, phosphorylation of 5'-AMP-activated protein kinase was significantly up-regulated by all 4 types of seaweed, whereas phosphorylation of protein kinase B was up-regulated only in the SF and HF groups. Lipogenesis-related proteins in the liver were effectively down-regulated by HF supplementation in DIO mice. CONCLUSIONS: Brown seaweed consumption showed hypotriglyceridemic effects in the prolonged DIO mouse model. Specifically, combinatory regulation of BA excretion and lipogenesis-related proteins in the liver by seaweed supplementation contributed to the reduction of plasma and hepatic TG levels, which inhibited hyperglycemia in DIO mice. Thus, the discrepant and species-specific functions of brown seaweeds provide novel insights for the selection of future targets for therapeutic agents.
Introduction: The esophagus squamous cell carcinoma (ESCC) is one of the most deadly malignances, and a current challenge is the development of effective therapeutic agents. Our present work addressed the effect of HIF-$1{\alpha}$ siRNA alone or in combination with cisplatin on the growth of ESCC in nude mice. Materials and Methods: Xenografts were established by inoculating ESCC TE-1 cells in nude mice, and transplanted tumors were treated with HIF-$1{\alpha}$ siRNA, cisplatin alone or together. Growth was assessed by measuring tumor volume. HIF-$1{\alpha}$ mRNA and protein expression were detected using RT-PCR and immunohistochemistry, respectively. Apoptosis of ESCC TE-1 cells was analyzed by flow cytometry. Results: In our nude mice model, HIF-$1{\alpha}$ siRNA effectively inhibited the growth of transplanted ESCC, downregulating HIF-$1{\alpha}$ mRNA and protein expression, and inducing ESCC TE-1 cell apoptosis. Notably when combinated with cisplatin, HIF-$1{\alpha}$ siRNA showed synergistic interaction in suppressing tumor growth. Furthermore, the proportion of apoptotic cells in HIF-$1{\alpha}$ siRNA plus cisplatin group was significantly higher than that in cisplatin or HIF-$1{\alpha}$ siRNA-treated groups (P<0.05). Conclusions: Down-regulated HIF-$1{\alpha}$ expression induced by siRNA could effectively suppress the growth of transplanted ESCC $in$$vivo$. HIF-$1{\alpha}$ siRNA could enhance the cytotoxicity of cisplatin, which suggests that a combination of these two agents may have potential for therapy of advanced ESCC.

제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
