단세포군항체(monoclonal antibody)를 이용한 항암제치료법이 다각적인 방면으로 실험되고 있으나, 환자에 주입된 단세포군항체중에서 실제의 암전달량은 언제나 기대치에 미치지 못하고 있다. 그러므로, 효과적인 항암제치료와 조기암진단을 위하여 암내부에서의 물질전달에 미치는 인자들의 포괄적인 측정이 필요하였다. 본 실험에서는 면역결핍누드쥐에 이식된 neurobastoma의 물질전달환경을 이해하기 위하여 성장특성과 포도당대사속도(GMR)를 측정하였으며, 세포간체액압력(IFP), 국부혈액속도(BPR)와 pH를 높이 2cm의 암에서 반경방향으로 측정하였다. 성장특성으로써 체적배가시간을, 세포활동성으로써 GMR를 측정하였더니, 각각 8.1일 (SD 0.44일), 23.53 mg/min/100g(SD 3.54 mg/min/100 g)이었다. 암중심 IFP는 암부피가 커짐에 따라 증가하여 높이 3cm의 암에서는 12.3mmHg(SD 2.6mmHg)이 되었다. 반경방향에 따른 IFP, BPR, pH를 특정한 결과, 암중심에 접근할수록 IFP는 증가되었고, 반면에 BPR과 pH는 감소되었다. 이로써 암내부에서의 증가된 IFP, 감소된 BPR과 pH가 단세포군항체와 그 공합체의 내부전달을 저해하는 원인으로 작용할 수 있다는 사실이 제시되었으며, 이들 인자들을 적당한 기술로 조절하여 더 많은 단세포군항체를 암내부로 전달시킬 수 있을 것으로 판단된다.

BACKGROUD/OBJECTIVES: Neuroinflammation plays critical role in neurodegenerative disorders, such as Alzheimer's disease (AD). We investigated the effect of three licorice varieties, Glycyrhiza uralensis, G. glabra, and Shinwongam (SW) on a mouse model of inflammation-induced memory and cognitive deficit. MATERIALS/METHODS: C57BL/6 mice were injected with lipopolysaccharide (LPS; 2.5 mg/kg, intraperitoneally) and orally administrated G. uralensis, G. glabra, and SW extract (150 mg/kg/day). SW, a new species of licorice in Korea, was combined with G. uralensis and G. glabra. Behavioral tests, including the T-maze, novel object recognition and Morris water maze, were carried out to assess learning and memory. In addition, the expressions of inflammation-related proteins in brain tissue were measured by western blotting. RESULTS: There was a significant decrease in spatial and objective recognition memory in LPS-induced cognitive impairment group, as measured by the T-maze and novel object recognition test; however, the administration of licorice ameliorated these deficits. In addition, licorice-treated groups exhibited improved learning and memory ability in the Morris water maze. Furthermore, LPS-injected mice had up-regulated pro-inflammatory proteins, such as inducible nitric oxide synthase (iNOS), cyclooxygenase-2, interleukin-6, via activation of toll like receptor 4 (TLR4) and nuclear factor-kappa B ($NF{\kappa}B$) pathways in the brain. However, these were attenuated by following administration of the three licorice varieties. Interestingly, the SW-administered group showed greater inhibition of iNOS and TLR4 when compared with the other licorice varieties. Furthermore, there was a significant increase in the expression of brain-derived neurotrophic factor (BDNF) in the brain of LPS-induced cognitively impaired mice that were administered licorice, with the greatest effect following SW treatment. CONCLUSIONS: The three licorice varieties ameliorated the inflammation-induced cognitive dysfunction by down-regulating inflammatory proteins and up-regulating BDNF. These results suggest that licorice, in particular SW, could be potential therapeutic agents against cognitive impairment.
이 연구는 Fluorouracil(5-FU)가 병원에서 암치료제로 쓰이는 세포독성약제의 한 종류이기 때문에 병원내부에 5-FU의 폭로 정도와 그 관리를 위한 가장 효율적인 세척약제를 조사하기 위한 것이다. 이러한 세포독성 약제실이나 종약학 병동에서 근무하는 실무자들은 세포독성약제로 오염된 표면에 빈번하게 접촉하게 되면 세포유전성이나 DNA손상에 대한 위험이 높게 된다. 따라서 이러한 약제의 세척제실험을 위하여 4가지 약제로서 시행한분해시험에서 0.5%(w/v) NaClO 용액만이 5-FU를 즉시 분해시켰으므로 이 용액은 오염표면의 잔유물을 분해시키는데 사용될 수 있다. 세포독성 약제실의 오염표면에서 확인된 농도범위는 2.0에서 $13.8{\mu}g/m^2$까지이고, 종약학 병동의 오염표면에서 측정한 농도범위는 5.39에서 $11.53{\mu}g/m^2$이었다. 5-FU와 같은 세포독성약제로부터 피부오염을 피하기 위하여 작업장의 노출허용기준과 같은 법적인 조치, 완벽한 표면세척제 및 보호구사용과 같은 엄격한 관리기준이 마련되어야 할 것이다.
HeLa 세포의 spheroid를 배 양시켜 cis-platinum과 carboplatin으로 처리한 후 그 반응을 세포의 생존분획으로 분석하였다. 체외실험 model인 spheroid를 사용하여 platinum유사체의 약효와 방사선 감수성을 평가하고 약제에 대한 단층 세포와 spheroid의 감수성 차이를 세포-생존곡선에서 규명하기 위하여 본 실험을 시행하였다. Cis-platinum 농도-곡선에서 spheroid의 $Cq=3.4{\mu}M$이고 $Co=1.2{\mu}M$이었다. 이 것은 단층세포에 비하여 Co는 큰 변화가 없으나 Cq가 증가되어 cis-platinum이 저산소층 세포보다 활동적으로 분화하는 표면세포에 주로 작용하였으며, 반대로 carboplatin의 효과는 spheroid에 대한 $(Co=15.0{\mu}M)$ 감수성이 단층세포$(Co=32.5{\mu}M)$에 비하여 크게 증가되어, spheroid의 심층 세포에 주로 작용하였다. 방사선과 carboplation의 병용효과를 세포 생존분획이 0.01 수준에서 isobologram으로 분석한 결과 상호작용으로 supra-additive 효과를 나타내었다. 따라서, carboplatin은 cis-platinum에 비하여 신장과 위장에 대한 독성작용이 적고, 방사선과 병용함으로써 향후 더욱 효과적 인 종양 치료에 중요한 역할을 할 것으로 기대한다.
Background: In this study, we evaluated the anti-cancer activity and potential molecular mechanism of 70% ethanol extracts of the root of Aralia cordata var. continentalis (Kitagawa) Y. C. Chu (RAc-E70) against human colorectal cancer cells. Methods and Results: RAc-E70 suppressed the proliferation of the human colorectal cancer cell lines, HCT116 and SW480. Although RAc-E70 reduction cyclin D1 expression at the protein and mRNA levels, RAc-E70-induced reduction in cyclin D1 protein level occurred more dramatically than that of cyclin D1 mRNA. The RAc-E70-induced downregulation of cyclin D1 expression was attenuated in the presence of MG132. Additionally, RAc-E70 reduced HA-cyclin D1 levels in HCT116 cells transfected with HA-tagged wild type-cyclin D1 expression vector. RAc-E70-mediated cyclin D1 degradation was blocked in the presence of LiCl, a $GSK3{\beta}$ inhibitorbut, but not PD98059, an ERK1/2 inhibitor and SB203580, a p38 inhibitor. Furthermore, RAc-E70 phosphorylated cyclin D1 at threonine-286 (T286), and LiCl-induced $GSK3{\beta}$ inhibition reduced the RAc-E70-mediated phosphorylation of cyclin D1 at T286. Conclusions: Our results suggested that RAc-E70 may downregulate cyclin D1 expression as a potential anti-cancer target through $GSK3{\beta}$-dependent cyclin D1 degradation. Based on these findings, RAc-E70 maybe a potential candidate for the development of chemopreventive or therapeutic agents for human colorectal cancer.
Objective : The etiology of chronic prostatitis is likely multifactorial, resulting from either a cascade of events after an initiating factor or from a variety of etiologic mechanisms. There is substantiating evidence to support the role of the inflammatory responses in its pathogenesis, and the clinical value in the evaluation of therapeutic efficacy. Forsythiae Frucus has been traditionally used in treatment of inflammatory diseases, including of prostatitis and urinary tract inflammation. In this study, we investigated the effects of Forsythiae Frucus on inflammatory cytokines and cyto-pathological alternation in the rat model of chronic non-bacterial prostatitis induced by castration and $17{\beta}$-estradiol treatment. Methods : Two-month-old rats were treated with $17{\beta}$-estradiol after castration for induction of experimental non-bacterial prostatitis. which is similar to human chronic prostatitis in histopathological profiles. Forsythiae Frucus as an experimental specimen, and testosterone as a positive control, were administered orally. The prostates were evaluated by histopathologlcal parameters including the epithelial score and epithelio-stromal ratio for glandular damage. and the expression of inflammatory cytokine genes including interleukin (IL)-$1{\beta}$, IL-5, IL-12, tumor necrosis factor (TNF)-$\alpha$. eotaxin, inducible nitric oxide synthase (iNOS) and cyclooxygenase-2(cox-2). Results : While prostates of control rats revealed severe acinar gland atrophy and stromal proliferation. the rats treated with Forsythiae Frucus showed a diminished range of tissue damage. Epithelial score was improved in the Forsythiae Frucus group over that of the control (P<0.05). The epithelia-stromal ratio was lower in the Forsythiae Frucus group when compared to that of the control (P<0.05). In the reverse transcription-polymerase chain reaction (RT-PCR) of inflammatory cytosine genes. Forsythiae Frucus inhibited the expression of IL-$1{\beta}$, TNF-$\alpha$, iNOS, cox-2 genes, while it modulated the expression of IL-5, which is an anti-inflammatory cytokine. Conclusions : These findings suggest that Forsythiae Frucus may protect the glandular epithelial cells and also inhibit stromal proliferation in association with the immune modulation including the suppression of inflammatory cytokines and increase of anti-inflammatory cytokines. From theses results. we suggest that Forsythiae Frucus could be a useful remedy agents for treating chronic non-bacterial prostatitis.

Lygodium japonicum has been traditionally used in treatment of urinary tract inflammation and voiding disturbance. In this study, we investigated the therapeutic effects and action mechanism of Lygodium japonicum in the rat model of non-bacterial prostatitis induced by castration and testosterone treatment. Methods : Five-month-old rats were treated with 17$\beta$-estradiol after castration for induction of experimental non-bacterial prostatitis, which is similar to human chronic prostatitis in histopathological profiles. Lygodium japonicum and testosterone were administered as an experimental specimen and a positive control. respectively. The prostates were evaluated by histopathological parameters including the epithelial score and epithelio-stromal ratio for glandular damage. PCNA labeling index for cyto-proliferation and a TUNEL(deoxyuridine triphosphate biotin nick end-labeling) assay for cell apoptosis. Results : While prostates of control rats revealed severe acinar gland atrophy and stromal proliferation, the rats treated with Lygodium japonicum showed a diminished range of tissue damage. Epithelial score was improved in the Lygodium japonicum group over that of the control (P<0.05). The epithelio-stromal ratio was lower in the Lygodiutn japonicum group when compared to that of the control (P<0.05). Although there was no difference in PCNA and TUNEL positive cells of the glandular epithelia. we found an decreased number of PCNA positive cell and concurrent increase of TUNEL positive cells in the stroma of Lygodium japonicum treated rats (P<0.01). Conclusions : These findings suggest that Lygodium japonicum may protect the glandular epithelial cells and also inhibit stromal proliferation in association with suppression of cyto-proliferation and stimulation of apoptosis. We concluded that Lygodium japonicum could be a useful remedy agents for treating chronic non-bacterial prostatitis.
본 연구진은 신규 미생물인 Nocardia sp. CS682 균주에서 항균물질인 nargenicin을 확보하고, 그 유도체 5종을 확보하여 그람양성, 음성 및 다약제 내성균에 대한 항균 효능 및 LPS로 자극된 대식세포에서의 nitric oxide 생성 억제능을 확인하였다. Nargenicin 유도체들은 nargenicin 및 vancomycin에 비교하여 우수한 항균 활성을 보여주었으며, compound 4 및 5는 광범위한 항균 효능 외에 nitric oxide 생성 억제능을 보여 항균-항염효과를 가지는 dual effector로써 감염, 면역 질환에 응용 가능성을 시사하였다. Nargenicin 유도체들은 향후 염증반응에서의 면역 조절 기작에 대한 추가 연구가 필요할 것으로 사료된다.
이상의 연구 결과로 미루어 볼 때, 도깨비부채 잎(RPL)은 ${\beta}-catenin$의 분해 유도를 통해 대장암, 유방암, 폐암, 전립선암 및 췌장암 세포의 생육을 억제하는 것으로 나타났다. 본 결과는 도깨비부채 잎의 항암을 위한 대체보완소재 및 천연 항암제 개발을 위한 소재로 활용이 가능할 것으로 판단된다. 그러나 추가적 연구를 통해 도깨비 부채 잎의 항암 활성물질의 분석연구가 필요할 것으로 사료된다.
천연 benzophenanthridine alkaloid의 일종인 sanguinarine에 의한 인간 암세포에서의 세포사멸 유도는 암 치료를 위한 잠재적 치료 가능성으로 여겨져 왔으나 기본적인 항암 기전은 여전히 불분명하다. 종양 억제제 p53의 결실 또는 돌연변이는 결장암세포의 항암제 내성에 대한 주요 원인으로 작용하다. 따라서, 본 연구에서는 정상 p53을 가진 HCT116 (p53+/+) 및 p53이 결여된 HCT116 (p53-/-) 결장암세포를 대상으로 sanguinarine에 의해 유도되는 세포사멸에서 p53의 역할을 조사하였다. 본 연구의 결과에 의하면, sanguinarine은 HCT116 (p53-/-) 세포에 비하여 HCT116 (p53+/+) 세포의 생존력을 현저히 감소시켰다. 아울러 sanguinarine은 HCT116 (p53-/-) 세포보다 HCT116 (p53+/+) 세포에서 p53 및 cyclin-dependent kinase 억제제 p21WAF1/CIP1의 발현을 증가시키면서 DNA 손상 및 세포사멸의 유도를 증가시켰다. Sanguinarine은 HCT116 (p53+/+) 세포에서 외인성 및 내인성 세포사멸의 개시에 관여하는 caspase-8 및 caspase-9의 활성을 증가시켰으며, 전형적인 효과기 caspase인 caspase-3을 활성화시켰다. 또한, sanguinarine은 HCT116 (p53+/+) 세포에서 Bax/Bcl-2의 발현 비율을 증가시키고 미토콘드리아 손상을 유발하였지만, HCT116 (p53-/-) 세포에서는 이러한 현상이 관찰되지 않았다. 결론적으로 본 연구의 결과는 sanguinarine은 HCT116 결장암세포에서 p53 의존적으로 외인성 및 내인성 세포사멸의 경로 활성을 통하여 세포사멸을 유도하였음을 의미한다.
