The Performance of Anion Exchange Expanded Bed Adsorption Chromatography on the Recovery of G6PDH from Unclarified Feedstock with High Biomass Concentration

  • The bed stability of Streamline DEAE (p = 1.2 g/mL) in a 20mm (i.d.) glass expanded bed contactor, and its performance on the recovery of glucose 6-phosphate dehydrogenase (G6PDH) from unclarified yeast homogenate were investigated. A residence time distribution study showed that a stable expanded bed was achieved. The theoretical plate and Bodenstein numbers determined were 25 and 53, respectively. A recovery yield of 87% and purification factor of 4.1 were achieved in the operation using 5% (w/v) biomass concentration feedstock. The performance of the anion exchange EBAC was still considerable good at a biomass concentration as high as 15% (w/v).

  • This research is, first of all, a theoretical study concerning 'conflict.' Only then, we could obtain ways in which we manage and resolve various problems arising from conflicts in business between Korean and Chinese companies. In doing this, we also tried to grasp cultural characteristics, or factors, in Chinese ways of carrying out negotiations that lead to conflicts with us. On the basis of these preliminary considerations, we developed practical techniques of conflict management and types of negotiation strategy. We thereby could suggest broader strategic implications for better performance in international business. Concretely, this research investigates techniques of conflict management and types of negotiation strategy. For such techniques and types, we suggest, (1) Sharing technique or reconciliatory compromising negotiation and its compromise strategy, (2) collaborative technique or cooperative negotiation and its win-win strategy, (3) competitive technique or competitive negotiation and its profit-seeking attack strategy, (4) accommodative technique or receptive negotiation and its relation-maintaining yield strategy, (5) avoidant technique or evasive negotiation and its indifference-showing avoidance strategy. This research contributes to promote understanding on negotiation culture of chinese corporate. and we provide the guideline of the conflict management and the insight for the efficiency strategy of chinese business negotiation. But, empirical data and statistical examinations should be added to our present research for the future prospective ones.

  • Polymethylphenylsilane(PMPS) was synthesized with methylphenyldichlorosilane using metal sodium. Various sample coated on quartz plate were exposed and the yield of residual film was calculated. To obtain the fine image forming after developing and drying, optical transmittance characterization have to be considered. Exposure is described by three optical parameters X, Y, and Z. There parameters are normally determined from optical transmittance measurements of exposed PMPS films. Z parameter was determined from the initial slope of a transmittance according to exposure time. This set of function parameters provided a complete description of photoresist exposure and development and became the basis for the theoretical process models.

Experimental and analytical study in determining the seismic performance of the ELBRF-E and ELBRF-B braced frames

  • In this article the seismic demand and performance of two recent braced steel frames named steel moment frames with the elliptic bracing (ELBRFs) are assessed through a laboratory program and numerical analyses of FEM. Here, one of the specimens is without connecting bracket from the corner of the frame to the elliptic brace (ELBRF-E), while the other is with the connecting brackets (ELBRF-B). In both the elliptic braced moment resisting frames (ELBRFs), in addition to not having any opening space problem in the bracing systems when installed in the surrounding frames, they improve structure's behavior. The experimental test is run on ½ scale single-story single-bay ELBRF specimens under cyclic quasi-static loading and compared with X-bracing and SMRF systems in one story base model. This system is of appropriate stiffness and a high ductility, with an increased response modification factor. Moreover, its energy dissipation is high. In the ELBRF bracing systems, there exists a great interval between relative deformation at the yield point and maximum relative deformation after entering the plastic region. In other words, the distance from the first plastic hinge to the collapse of the structure is fairly large. The experimental outcomes here, are in good agreement with the theoretical predictions.

  • The purpose of this study is to examine the effects which applies teaching program that search solution to prevent global warming in middle school gifted class. This program of total 10 class was theoretical and experimental class and the students presented after collecting the synthetic data about climate-change and roleplayed what they debated to firm their country's opinion about Kyoto Protocol. Finally, students as policymaker made mock Kyoto Protocol by discussion. Impression papers and images about climate-changes was made by students in last class. In discussion of making up mock Kyoto Protocol, developed and developing nations had an argument about carbon dioxide emissions because of their economic benefits. In the course of allocating mutual $CO_2$ reduction, they needed yield and negotiation among nations. Although they mainly took passive position about $CO_2$ reduction, atmosphere that all the country make greater effort for the future global environment was formed. Like this, students had more concepts over climate change by making mock Kyoto Protocol. And they could equipped global scientific literacy and responsibility by participating in social decision making process of solving global environmental issues.

A novel modeling of settlement of foundations in permafrost regions

  • Settlement of foundations in permafrost regions primarily results from three physical and mechanical processes such as thaw consolidation of permafrost layer, creep of warm frozen soils and the additional deformation of seasonal active layer induced by freeze-thaw cycling. This paper firstly establishes theoretical models for the three sources of settlement including a statistical damage model for soils which experience cyclic freeze-thaw, a large strain thaw consolidation theory incorporating a modified Richards' equation and a Drucker-Prager yield criterion, as well as a simple rheological element based creep model for frozen soils. A novel numerical method was proposed for live computation of thaw consolidation, creep and freeze-thaw cycling in corresponding domains which vary with heat budget in frozen ground. It was then numerically implemented in the FISH language on the FLAC platform and verified by freeze-thaw tests on sandy clay. Results indicate that the calculated results agree well with the measured data. Finally a model test carried out on a half embankment in laboratory was modeled.

Structural Effects on the Tensile and Morphological Properties of Zeolite-filled Polypropylene Derivative Composites

  • We have studied the effects that inorganic zeolite powder have on structurally different copolymer [poly(propylene-co-ethylene)] and terpolymer [poly(propylene-co-ethylene-co-l-butene)] systems and the possibility of preparing suitable porous composite films. The impact strength and yield stress of the composites did not improve upon any further loading of zeolite, but the modulus increased gradually with respect to the filler loading. The experimental modulus of each of the two systems was compared with theoretical models. We performed a morphological study of the filler mixing efficiency and image analysis. The number-, weight-, and z-average air hole diameters were compared with respect to the draw ratio as well as the zeolite loading. The experimental results suggest that these two matrices can provide a new choice for preparing future multiphase polymeric porous films by stretching them unidirectionally. In particular, we suggest that a 40 wt% zeolite loading at a draw ratio of 4 is useful for porous film applications.

A Study on the Reaction-Stoichiometry of Autotrophic Denitrification based on Growth Characteristic of Microorganism (미생물 성장 특성에 기초한 독립영양탈질의 화학양론식 연구)

  • It is necessary to supply external carbon source for enhancement of biological nitrogen removal from domestic wastewater with low influent C/N ratio. Sulfide was chosen as a cost effective electron donor and reaction stoichiometry for autotrophic denitrification was investigated by conducting bench-scale experiments in this study. Higher sulfur to nitrogen (S/N) ratio than the calculated value from theoretical reaction stoichiometry was required when the anoxic reactor was operated at open condition because dissolved oxygen introduced by surface aeration reacted with sulfide with ease. In addition, higher sulfate production and lower yield of microorganism could be observed under the same condition. It was possible to obtain reliable reaction stoichiometry for autotrophic denitrification by establishing pure anoxic condition. Linear relationship between bacterial growth and consumption of nitrate, sulfide, alkalinity, and sulfate production enabled to derive a relatively correct reaction stoichiometry for autotrophic denitrification when sulfide was used as an electron donor.

  • This rape deals with the experimental and theoretical investigations on the aerodynamic performance of the Wells turbine. The two-dimensional cascade theory is used to estimate the thrust and torque of turbine, and finally to yield an efficiency of turbine. The turbine is assumed to rotate with a constant rotational speed in a sinusoidally varying unsteady flow field. Experimental approach is made in a wave simulator, producing a sinusoidally reciprocating air flow corresponding to the wave motion in an Oscillating Water Column(OWC) chamber. Performance data of turbine measured at various operating conditions are analyzed and compared to numerical results in order to understand the overall features of a Wells turbine.

Development of Thermostable Fusant, CHY1612 for Lignocellulosic Simultaneous Saccharification and Fermentation (섬유질계 동시당화발효를 위한 내열성 융합 효모, Kluyveromyces marxianus CHY1612의 개발)

  • To develop thermostable ethanol fermentative yeast strain for lignocellulosic simultaneous saccharification and fermentation, high ethanol producing yeast, Saccharomyces cerevisiae CHY1012 and thermostable yeast, Kluyveromyces marxianus CHY1703 were fused by protoplast fusion. The thermostable fusant, CHY1612 was identified as a Kluyveromyces marxianus by phenotypic and physiological characteristics, as well as molecular analysis based on the D1/D2 domains of the large subunit (26S) rDNA gene and the internally transcribed spacer (ITS) 1 + 2 regions. For lignocellulosic ethanol production, AFEX pretreated barley straw at $150^{\circ}C$ for 90 min was used in a simultaneous saccharification and fermentation (SSF) process using thermotolerant CHY1612. The SSF from 16% pretreated barley straw at $43^{\circ}C$ gave a saccharification ratio of 90.5%, a final ethanol concentration of 38.5 g/L, and a theoretical yield of 91.2%. These results show that K. marxianus CHY1612 has potential for lignocellulosic ethanol production through simultaneous saccharification and fermentation with further development of process.
