Effects of exploration and molecular mechanism of CsV on eNOS and vascular endothelial functions

  • This study aimed to investigate the effects and potential mechanisms of Chikusetsusaponin V (CsV) on endothelial nitric oxide synthase (eNOS) and vascular endothelial cell functions. Different concentrations of CsV were added to animal models, bovine aorta endothelial cells (BAECs) and human umbilical vein endothelial cells (HUVECs) cultured in vitro. qPCR, Western blotting (WB), and B ultrasound were performed to explore the effects of CsV on mouse endothelial cell functions, vascular stiffness and cellular eNOS mRNA, protein expression and NO release. Bioinformatics analysis, network pharmacology, molecular docking and protein mass spectrometry analysis were conducted to jointly predict the upstream transcription factors of eNOS. Furthermore, pulldown and ChIP and dual luciferase assays were employed for subsequent verification. At the presence or absence of CsV stimulation, either overexpression or knockdown of purine rich element binding protein A (PURA) was conducted, and PCR assay was employed to detect PURA and eNOS mRNA expressions, Western blot was used to detect PURA and eNOS protein expressions, cell NO release and serum NO levels. Tube formation experiment was conducted to detect the tube forming capability of HUVECs cells. The animal vasodilation function test detected the vasodilation functions. Ultrasonic detection was performed to determine the mouse aortic arch pulse wave velocity to identify aortic stiffness. CsV stimulus on bovine aortic cells revealed that CsV could upregulate eNOS protein levels in vascular endothelial cells in a concentration and time dependent manner. The expression levels of eNOS mRNA and phosphorylation sites Ser1177, Ser633 and Thr495 increased significantly after CsV stimulation. Meanwhile, CsV could also enhance the tube forming capability of HUVECs cells. Following the mice were gavaged using CsV, the eNOS protein level of mouse aortic endothelial cells was upregulated in a concentration- and time-dependent manner, and serum NO release and vasodilation ability were simultaneously elevated whereas arterial stiffness was alleviated. The pulldown, ChIP and dual luciferase assays demonstrated that PURA could bind to the eNOS promoter and facilitate the transcription of eNOS. Under the conditions of presence or absence of CsV stimulation, overexpression or knockdown of PURA indicated that the effect of CsV on vascular endothelial function and eNOS was weakened following PURA gene silence, whereas overexpression of PURA gene could enhance the effect of CsV upregulating eNOS expression. CsV could promote NO release from endothelial cells by upregulating the expression of PURA/eNOS pathway, improve endothelial cell functions, enhance vasodilation capability, and alleviate vessel stiffness. The present study plays a role in offering a theoretical basis for the development and application of CsV in vascular function improvement, and it also provides a more comprehensive understanding of the pharmacodynamics of CsV.

ENACT Project: Promoting Pre-Service Science Teachers' Views on the Social Responsibility of Scientists and Engineers (ENACT 프로젝트에 참여한 예비 과학교사들의 과학기술자의 사회적 책임에 대한 인식 변화 탐색)

  • This study aims to investigate the effects of the ENACT project on promoting pre-service science teachers (PSTs)' views on the social responsibility of scientists and engineers. The ENACT project was designed to cultivate the social responsibility by integrating the theoretical framework of socioscientific issues (SSIs) education with problem-based inquiry approaches for the resolution of the issues. Thirty-two PSTs voluntarily participated in the project and completed the five stages over three months. Data was collected through a questionnaire to measure PSTs' view of the social responsibility of scientists and engineers (VSRoSE) and focus-group interviews. Results indicate that the PSTs presented statistically significant changes in their views of the social responsibility after the ENACT project. The mean scores of the five sub-dimensions of VSRoSE significantly increased. The interviews also supported that the PSTs had opportunities to seriously consider the social responsibility of scientists and engineers through epistemological exploration of science and technology (Cycle I), and problem-solving and action-taking (Cycle II). In particular, they agreed more on such responsibilities as consideration of societal needs and demands, pursuit of the common good, civic engagement and services using their expertise, communications with the public regarding potential risks, and participation in policy decision-making related to science and technology advances. Educational implications for SSI education and teacher education were suggested.

Ukrainian Students' Analysis of Abuse Treatment by Parents: Retrospective and Perspective in Virtual and Real Environments

  • Given the prevalence of violence in Ukrainian families, measures to prevent parental abusive treatment against children are urgent. It is important to study today's youth awareness about violence within families in order to enhance a culture of engagement with spouses and children in future. The aim of the study is to examine students' reflective experiences and their attitudes towards forms, frequency, causes and effects of parental abusive treatment. During the research the following methods were used step-by-step: theoretical analyses of scientific resources, anonymous questionnaire, quantitative and correlation analyses. According to result of survey 98 students who were interviewed, none of them fell victim of sexual abuse in their families. However, more than a half (51%) of the students surveyed experienced some forms of parental physical punishment. All the interviewed students encountered psychological cases of parental violence. The mostprevalent forms of parental abuse among the interviewees are criticism, negative comparison, emotional detachment, arrogance, intimidation, blackmail and humiliation. The most prevalent consequences of parental abuse among students are anxiety, low self-esteem, insecurity, impatience, suspiciousness, constraint in communication. Students agree that budget limitations, forced labor, criticism, spanking, emotional detachment, ignoring type of communication, reproach, blackmail are acceptable methods of punishment to use when raising their own children. These results clearly demonstrate the problem of the impact of parental abuse on children and its consequences in the future. A virtual dimension of the actualized problem is considered, namely: - virtualization of aggression and cruelty in the postmodern world. - the level of determining the factor of cruelty from the space of virtual culture. - the mirror image of everyday cruelty in the virtual environment; - the phenomenon of video games as a source and context of representation of the factor of cruelty in behavioral realities; - cybercrime as a virtualized result of cruelty in family and everyday realities. - futurological perspectives of virtualization of cruelty in communicative culture in general and in family relations in particular. The postmodern world is fundamentally different from the traditions and culture of the past, primarily due to the development of computer technologies and the virtualization of life in general. So, for example, virtual communities have become, in a certain way, another world, a second reality of life in general. And certain behavioral factors, in particular cruelty in the private environment, became a projection of such a phenomenon as cybercrime. Video games are a unique modern phenomenon, which multipolarly absorb all facets of human potential, communicative tendencies, behavioral and characterological factors, from the warmth of interpersonal relationships to the extreme degree of cruelty.

  • The main purpose of the work is to analyze modern innovative educational practices in the field of culture and art and their effectiveness in the context of the spread of digitalization trends. The study used general scientific theoretical methods of analysis, synthesis, analogy, comparative, induction, deduction, reductionism, and a number of others, allowing you to fully understand the pattern of modern modernization processes in a long historical development and demonstrate how the rejection of the negativity of progress allows talented artists to realize their own potential. The study established the advantages and disadvantages of involving innovative technologies in the educational process on the example of European experience and outlined possible ways of implementing digitalization processes in Ukrainian institutions of higher education, formulated the main difficulties encountered by teachers and students in the use of technological innovation in the pandemic. The rapid development of digital technologies has had a great impact on the sphere of culture and art, both visual, scenic, and musical in all processes: creation, reproduction, perception, learning, etc. In the field of art education, there is a synthesis of creative practices with digital technologies. In terms of music education, these processes at the present stage are provided with digital tools of specially developed software (music programs for composition and typing of musical text, recording, and correction of sound, for quality listening to the whole work or its fragments) for training programs used in institutional education and non-institutional learning as a means of independent mastering of the theory and practice of music-making, as well as other programs and technical tools without which contemporary art cannot be imagined. In modern stage education, the involvement of video technologies, means of remote communication, allowing realtime adjustment of the educational process, is actualized. In the sphere of fine arts, there is a transformation of communicative forms of interaction between the teacher and students, which in the conditions of the pandemic are of two-way communication with the help of information and communication technologies. At this stage, there is an intensification of transformation processes in the educational industry in the areas of culture and art.

A Review on the Deposition/Dissolution of Lithium Metal Anodes through Analyzing Overpotential Behaviors (과전압 거동 분석을 통한 리튬 금속 음극의 전착/탈리 현상 이해)

  • Lithium metal is the most promising anode for next-generation lithium-ion batteries due to its lowest reduction potential (-3.04 V vs. SHE) and high specific capacity (3860 mAh/g). However, the dendritic formation under high charging current density remains one of main technical barriers to be used for commercial rechargeable batteries. To address these issues, tremendous research to suppress lithium dendrite formation have been conducted through new electrolyte formulation, robust protection layer, shape-controlled lithium metal, separator modification, etc. However, Li/Li symmetric cell test is always a starting or essential step to demonstrate better lithium dendrite formation behavior with lower overpotential and longer cycle life without careful analysis. Thus, this review summarizes overpotential behaviors of Li/Li symmetric cells along with theoretical explanations like initial peaking or later arcing. Also, we categorize various overpotential data depending on research approaches and discuss them based on peaking and arcing behaviors. Thus, this review will be very helpful for researchers in lithium metal to analyze their overpotential behaviors.

Differentiating Contents for Rural Art Spaces through Place Assets (장소자산을 활용한 농촌 문화예술공간의 콘텐츠 차별화 방안)

  • This paper discusses a way to differentiate contents and enhance the competitiveness of art spaces in rural regions through place assets, noting the growing number of rural art spaces and their great potential as cultural spaces in recent years. First, we surveyed existing research and resources to understand the current status and roles of rural art spaces in the context of theoretical discussions on place assets. Based on this, we classified place assets into physical, human, and symbolic elements. After that, we conducted two case studies that apply this classification. For the case studies on the Ami Art Museum and the Potato Flower Studio, we investigated the sites and conducted in-depth interviews with the directors following a qualitative research method. Our findings through the case studies show that Ami Art Museum and Potato Flower Studio remodeled closed school buildings, which are known as physical place assets in rural areas. However, there was a difference in utilizing other place asset elements, and this resulted in different types of differentiated contents. We found three types of rural art spaces from two case studies: inhabitant-friendly, artist-friendly, and tourist-friendly. Finally, in our research, we presented a method of utilizing place assets that differentiate rural art places along with a method for developing differentiated contents. To conclude, the significance of our study is that it attempted to find out a way to make rural art spaces sustainable, and sought cultural regeneration in rural areas through revitalization of rural art spaces.

A Study on the Establishment of Design and Construction Process Standardization through Building BIM Application Case (건축물 BIM 적용사례를 통한 설계 및 시공프로세스 표준화 수립에 대한 연구)

  • In order to satisfy the extraction and use of information such as estimates and processes required in the design and construction stages of BIM, which is an expectation of overall construction operation for the design and construction stage of domestic buildings, it is insufficient to supply and apply mobile technologies or terminals. In this paper, standardization of BIM-based processes from the design stage to the construction stage is proposed as an efficient construction system method through mobile-based simulation and test-bed case analysis review. The current status and potential of BIM application were identified through theoretical review of BIM and case studies at home and abroad. In addition, the overall flow of the project and the direction of effective process construction were investigated through each process by 3D, 4D, and 5D execution stage and the role of each collaborator. 4D building process BIM simulation system using mobile was implemented by applying a visualization engine that simulates process information, object information connection module, and related object information. Therefore, it was possible to minimize the possibility of re-construction of the BIM design and construction process model through the visualization of 2D drawings based on the 3D model of the building and the review of errors and interferences in the drawings. In addition, in the implementation of simulation for each process of the construction process through mobile devices, it was possible to support construction progress and process management according to the optimal option selected by the user.

Effect of Viscosity and Clogging on Grout Penetration Characteristics (점도 변화와 폐색 현상을 고려한 그라우트재의 침투 특성)

  • Many construction projects adopt grouting technology to prevent the leakage of groundwater or to improve the shear strength of the ground. Recognition as a feasible field procedure dates back to 1925. Since then, developments and field use have increased rapidly. According to improvement of grout materials, theoretical study on grout penetration characteristics is demanded. Fluid of grout always tends to flow from higher hydraulic potential to lower one and the motion of grout is also a function of formation permeability. Viscosity of pout is changed by chemical action while grout moves through pores. Due to the increment of viscosity, permeability is decreased. Permeability is also reduced by grout particle deposits to the soil aggregates. In this paper, characteristics of new cement grout material that has been developed recently are studied: injectable volume of new grout material is tested in two different grain sizes of sands; and the method to calculate injectable volume of grout Is suggested with consideration of change in viscosity and clogging phenomena. The calculated values are compared with injection test results. Viscosity of new grout material is found to increase as an exponential function of time. And lumped parameter $\delta$ of new grout material to be used for assessing deposition characteristics is estimated by comparing deposit theory with injection test results considering different soil types and different injection pressures. Injection test results show that grout penetration rate is decreased by the increase of grout viscosity and clogging phenomena.

A Qualitative Study for Building up the Concept of Culture Welfare (문화복지 개념 정립을 위한 질적 연구 - 휴먼서비스 실천가들의 인식을 중심으로 -)

  • This study aimed to build up the concept of culture welfare. For this purpose, we analyzed the practitioners' understanding about culture welfare in culture and social welfare by Modified Grounded Theory Approach. The results showed that the practitioners did not understand the concept of culture welfare clearly, and revealed being in confusion. Their understandings about culture welfare were classified into five types, 1) a type denying a terminology, culture welfare itself, 2) a type without awareness about culture welfare, 3) a type regarding culture welfare as sub realm in the special domain of culture and social welfare, 4) a type undividing and integrating culture and social welfare, 5) a type recognizing an independent domain of culture welfare. But despite of large deviation like this, the practitioners were common to give emphasis on giving opportunities to everyone in the enjoyment of culture, for heightening the quality of life. Summing up, culture welfare is defined as 'all direct and indirect effort in culture and arts, in a narrow sense to economical vulnerable class, and in a broad sense to everyone, to raise cultural susceptibility, creative thinking, and potential competence for realization a life worthy of man. The significance of this study is in establishment the definite and practical concept of culture welfare through a deep analysis the practitioners' understanding. This will give the theoretical basis to the study and the development of various programs in culture welfare.

An Environmental Design of the Catholic Retreat Center Adopting the Theoretical Aesthetics - In the Case of the Suncheon Jesuits Spiritual Center - (신학적 미학을 적용한 가톨릭 피정시설 환경설계 - 순천 예수회 영성센터를 대상으로 -)

  • Nowadays, the increasing number of religious travelers seeking inner healing has led to the popularity of Buddhist 'temple stays', Protestant and Catholic 'retreats' as alternative vacations. However, unlike Buddhist temples, which are inculturated, Christian retreat centers follow the formula of general recreational facilities and lack an overall framework due to relying on sponsorship. This study attempts to design appropriate outdoor environments for a retreat by reflecting 'theological aesthetics' based on the process of 'Spiritual Exercises', the main method of retreat. First, as the Spiritual Exercises are often described as an 'inner pilgrimage', the images and theological aesthetics experienced in prayer themes were examined. Next, the characteristics of the spaces required for the facility were analyzed, and the composition of religious spaces was proposed. Also the landscape resources of the site were investigated, and the retreatants' behavior to derive spatial preferences was analyzed. Overall, outdoor spaces were planned to induce a shift in consciousness and broaden the spectrum of the retreatants' experience through a physical environment consistent with the mechanism of prayer. This study aimed to implement theological aesthetics by selecting a retreat center with high potential for landscape resources. As a result, it was possible to reveal the 'poetic landscape' to effectively evoke images during 'contemplative prayer', and to support religious experience. The significance of this study lied in deriving various design vocabularies for outdoor spaces of religious facilities.
