Agriculture Pollution and its Countermeasures with Special Consideration of Pesticides (작물생산과 농업공해 및 그 대책)

  • Because of the continuing rapid increase in pesticide usage in Taiwan, much attention has been focus on pesticide contamination of food and effect of pesticides on human and environmental health. The Plant Protection Center (PPC) conducts safety evaluation of pesticides usee! in Taiwan. The pesticides are classified into different groups based on their acute toxicities. Pesticides which are classified into extremely toxic group are not allow to used on short term crops or the continuously harvest crops. The acute toxicity of pesticides to the beneficial insects are also studied, special attention has been paid to the two predators of rice brown planthopper. 60% of cultivated land in Taiwan are paddy field; therefore, acute fish toxicity was taken into consideration when a pesticide was applied for registration to be used in the paddy. Fish toxicities were evaluated by the dangerous rating value which is the amount of pesticide residue in the field water over the TLM value. Mutagenicity of pesticides was continuously evaluated by using Arne's microbial testing method. Island wide survey of residual levels of pesticides of known pollutants such as chlorinated hydrocarbon . insecticides, mercurial compounds in soil, water and biological samples were carried out constantly. The potential of a new1y Imported esticides to pollute the environment were studied by using model ecosystem. Ecological magnification (EM) of a chemical was calculated from model ecosystem. A chemical was considered as a pollutant when its EM value over 5000. In order to ensure the levels of pesticides residue of the crop within the safety limit. The 'tolerance' of pesticides on different crop groupings were established base on 1) acceptable daily intake value of individual pesticides, 2) average daily consumption of each crop groupings by Chinese person, 3) Actual residues of pesticides. on different crops obtained from supervised trials. Total about 79 pesticides for which the tolerances have been established on different crop groupings. Because the intensive agricultural system was adopted in Taiwan. The phytotoxicity of pesticides to the non-target crops was therefore become one of the important factor in the safety evaluation of pesticide usage. These will include 1) direct injury, 2) injury caused by pesticide polluted irrigation water, 3) injury caused by the pesticide polluted soil, 4) reduction of growth caused by the effect of pesticide on the soil microorganisms. This paper will reviewed all the aspects mentioned in the previous .paragraphs. Most the works have done in Taiwan by the PPC.y the PPC.

  • To investigate the possibility of the BPH control by systematic application of insecticides, biological effects of buprofezin and isoprothiolane upon the BPH population was evaluated in the laboratory and pot. Both chemicals did not affect the developmental period of the nymphal stage with the applied dosages. When the buprofezin treated instar nymph was developed to the adult, adult's longevity was fairly reduced and such an effect of buprofezin was greater upon the younger instar nymphs. Residual effect of buprofezin was about 30 days in the pot. And it could suppress the BPH population when the younger instar nymphs were dominant age group in the population at the time of treatment. Isoprothiolane was also more effective for the suppression of the BPH population when the younger instar nymphs were dominant at the time of treatment. And it was expected that by buprofezin treatment in July the control effect of isoprothiolane against the BPH population could be maximized when it was treated to control the rice neck blast in late July or early August, because buprofezin could induce the biased population age distribution in which younger instar nymphs were main group at the time of isoprothiolane treatment.

Screening method of varietal resistance to planthoppers labeled with radioisotope $^{32}P$ ( I ) (방사성동위원소 $^{32}P$를 이용한 멸구 류에 대한 품종저항성 검정방법에 관한 연구(I))

  • The screening method of varietal resistance on the plant hoppers has generally been evaluated as a reaction of plant after infesting insects. However, feeding amount of insects to the varieties was investigated in this experiment. The new method using isotope $^{32}P$ for rice varietal resistance to plant hoppers was carried out through the following method. Insects tested were caged for a few hours on the plants which had absorbed $^{32}P$ solution in small vials for $24\~48\;hours$. After feeding, insects were killed in the refrigerator with formalin solution, and then were measured by the feeding amount as a count per minute (CPM) with the G.M. Counter. The results obtained were summarized as follows; 1. The apparatus of Type D(Fig. 2) was most effective and the safest among four others. 2. The optimum amount of $H_3PO_4$ solution was found to be $2\~3m1$. 3. Radioactivity of $0.7\mu\;Ci.\;^{32}P$ was sufficient to check varietal difference of feeding amount by the brown planthopper. 4. Radioisotope was found from the body of insects but not in the cuticular layer nymphs cast off.

  • 'Mogwoo', a new high yield and whole crop silage rice (Oryza sativa L.) cultivar, was developed by the rice breeding team of the National Institute of Crop Science, RDA, Suwon, Korea, from 1999 to 2009, and was released in 2010. It was derived in 1999 from a cross between Dasanbyeo, having a high yield, and Suweon431/IR71190-45-2-1. A promising line, SR25848-C99-1-2-1, selected by the pedigree breeding method, was designated the name of 'Suweon 519' in 2007. This cultivar has about 155 days of growth period from seeding to heading, and is tolerance to lodging, with erect pubescent leaves as well as a long and thick culm. This cultivar has the same number of tillers per hill and higher spikelet numbers per panicle compared to Nokyang. 'Mogwoo' has longer leaves compared with other Tongil-type varieties. This new variety is resistant to grain shattering, leaf blast, bacterial leaf blight, and small brown planthopper. The biomass yield of 'Mogwoo' was 1,956 kg/10a in a regional test over three years. The result shows that 'Mogwoo' is adaptable to central and south-east plain areas of Korea.

ITS2 DNA Sequence Analysis for Eight Species of Delphacid Planthoppers and a Loop-mediated Isothermal Amplification Method for the Brown Planthopper-specific Detection (멸구과 8종의 ITS2 DNA 염기서열 비교 분석과 고리매개등온증폭법(LAMP)을 이용한 벼멸구 특이 진단법)

  • Estimates of evolutionary sequence divergence and inference of a phylogenetic tree for eight delphacid planthopper species were based on the full-length nucleotide sequence of the internal transcribed spacer 2 (ITS2) region. Size of the ITS2 DNA sequence varied from 550 bp in Sogatella furcifera to 699 bp in Nilaparvata muiri. Nucleotide sequence distance ($d{\pm}S.E.$) was lowest between N. muiri and N. bakeri ($0.001{\pm}0.001$), and highest between Ecdelphax cervina and Stenocranus matsumurai ($0.579{\pm}0.021$). Sequence distance between N. lugens and other planthoppers ranged from $0.056{\pm}0.008$ (N. muiri) to $0.548{\pm}0.021$ (S. matsumurai). In the neighbor-joining phylogenetic tree, all planthoppers were clustered separately into a species group, except N. muiri and N. bakeri. The ITS2 nucleotide sequence of N. lugens was used to design four loop-mediated isothermal amplification (LAMP) primer sets (BPH-38, BPH-38-1, BPH-207, and BPH-92) for N. lugens species-specific detection. After the LAMP reaction of three rice planthoppers, N. lugens, S. furcifera, and Laodelphax striatellus, with the four LAMP primer sets for 60 min at $65^{\circ}C$, LAMP products were observed in the genomic DNA of N. lugens only. In the BPH-92 LAMP primer set, the fluorescence relative to that of the negative control differed according to the amount of DNA (0.1 ng, 10 ng, and 100 ng) and incubation duration (20 min, 30 min, 40 min, and 60 min). At $65^{\circ}C$ incubation, the difference was clearly observed after 40 min with 10 ng and100 ng, but with a 60-min incubation period, the minimum DNA needed was 0.1 ng. However, there was little difference in fluorescence among all DNA amounts tested with 20 or 30 min incubations.

Biological activity of shizukanols isolated from Chloranthus japonicus roots (홀아비꽃대(Chloranthus japonicus) 뿌리로부터 분리한 shizukanol들의 생물활성)

  • The methanol extract of Chloranthus japonicus roots effectively controlled the development of rice blast(Magnaporthe grisea), rice sheath blight(Corticium sasaki), tomato pay mold(Btrytis cinerea), tomato late blight(Phytophthora infestans), and wheat leaf rust(Puccinia recondita). From the methanol extract of C. japonicus roots, three antifungal substances were isolated. Their chemical structures were determined to be shizukanols B, C, and D mainly by mass and NMR spectral data. Among the three substances, shizukanol C showed the strongest inhibitory activity against mycelial growth of the plant pathogenic fungi tested; it completely inhibited mycelial growth of M. grisea. Colletotrichum gloeosporioides, and C. acutatum at concentrations of more than $12.5{\mu}g$/ and P. infestans at concentrations of more than $3.13{\mu}g/m\ell$. They also controlled effectively the development of rice blast and wheat leaf rust. On the other hand, they caused phytotoxic symptoms on barley leaves and inhibited the growth of duckweed (Lemna paucicostata) with $EC_{50}$ values of $30.0{\mu}g/m\ell$ for shizukanol B, $49.9{\mu}g/m\ell$ for shizukanol C, and $154{\mu}g/m\ell$ for shizukanol D. In addition, shizukanol C showed an insecticidal activity against brown planthopper (Nilaparavata lugens), peen peach aphid (Myzus persicae), diamond-back moth (Plutella xylostella), and tobacco cutworm (Spodoptera litura) of the 5 arthropod pests tested with mortality values of more than 60% at a concentration of $1,000{\mu}g/m\ell$.

Temperature-dependent Development Model and Forecasting of Adult Emergence of Overwintered Small Brown Planthopper, Laodelphax striatellus Fallen, Population (애멸구 온도 발육 모델과 월동 개체군의 성충 발생 예측)

  • The developmental period of Laodelphax striatellus Fallen, a vector of rice stripe virus (RSV), was investigated at ten constant temperatures from 12.5 to $35{\pm}1^{\circ}C$ at 30 to 40% RH, and a photoperiod of 14:10 (L:D) h. Eggs developed successfully at each temperature tested and their developmental time decreased as temperature increased. Egg development was fasted at $35^{\circ}C$(5.8 days), and slowest at $12.5^{\circ}C$ (44.5 days). Nymphs could not develop to the adult stage at 32.5 or $35^{\circ}C$. The mean total developmental time of nymphal stages at 12.5, 15, 17.5, 20, 22.5, 25, 27.5 and $30^{\circ}C$ were 132.7, 55.9, 37.7, 26.9, 20.2, 15.8, 14.9 and 17.4 days, respectively. One linear model and four nonlinear models (Briere 1, Lactin 2, Logan 6 and Poikilotherm rate) were used to determine the response of developmental rate to temperature. The lower threshold temperatures of egg and total nymphal stage of L. striatellus were $10.2^{\circ}C$ and $10.7^{\circ}C$, respectively. The thermal constants (degree-days) for eggs and nymphs were 122.0 and 238.1DD, respectively. Among the four nonlinear models, the Poikilotherm rate model had the best fit for all developmental stages ($r^2$=0.98~0.99). The distribution of completion of each development stage was well described by the two-parameter Weibull function ($r^2$=0.84~0.94). The emergence rate of L. striatellus adults using DYMEX$^{(R)}$ was predicted under the assumption that the physiological age of over-wintered nymphs was 0.2 and that the Poikilotherm rate model was applied to describe temperature-dependent development. The result presented higher predictability than other conditions.
