Washed mycelia of Aspergillus nidulans FGSC159 were incubated in CMC minimal liquid medium and the culture filtrate which contained induced extracellular cellulase was fractionated by a three-step procedure including chromatography on Bio-Gel P-150, chromatography on DEAE-Sephadex A-50 and chromatography on Sephadex G-100. Three CMCase components ; F-I-Ia, F-I-Ib and F-II-Ia were prepared. No enzyme activity toward avicel could be detected in these components. Similarly, there was no ${\beta}-glucosidase$ activity. pH-optima of the three components were all 5.0 in acetate buffer. Temperature-optima for the activities of F-I-Ia, F-Ib and F-II-Ia were $45^{\circ}C,\;40^{\circ}C\;and\;50^{\circ}C$, respectively. F-II-Ia was shown to be more thermostable than the other two components. F-II-Ia was proved to have quite a different substrate specificity and action property and action property from those of F-I-Ia and F-I-Ib by product analysis on liquid chromatography.
Effect of Fe and Mn contents on the tensile properties has been studied in Al-9wt%Si-0.3wt%Mg alloy. As Fe content of Al-9wt%Si-0.3wt%Mg-0.5wt%Mn alloy increased from 0.15wt% to 0.45wt%, tensile strength of as-cast alloy decreased from 192 MPa to 174 MPa, and elongation of the alloy also decreased from 4.8% to 4.2%. Decrease of these properties can be explained as the formation of plate shape, ${\beta}-Al_5FeSi$ phase with high Fe/Mn ratio of the alloy. However when Mn content of Al-9wt%Si-0.3wt%Mg-0.45wt%Fe alloy increased from 0.3wt% to 0.5wt%, tensile strength of T6 aged alloy increased from 265 MPa to 275 MPa, and elongation of the alloy increased from 2.3% to 3.6%. These improvements attribute to chinese script, ${\alpha}-Al_{15}(Mn,Fe)_3Si_2$ phase shape-modified from ${\beta}-Al_5FeSi$ phase with low Fe/Mn ratio of the alloy.

Higenamine, a benzyltetrahydroisopuinoline analog isolated from aconite tuber, has potent isotropic action. Recent studies suggest it may have beta receptor agonistic property in that its inotropic action is blocked by propranolol in isolated rabbit heart. However, no study has been carried out on other organs than heart. Higenamine is expected to have pharmacological actions on smooth muscle on the ground that it has catecholamine moiety and tetrahydrosioquinoline nucleus in its chemical structure, both of which are well known to have smooth muscle relaxation effects. Therefore present study was aimed at determining whether higenamine has bronchodilating effect in isolated guinea pig trachea smooth muscle rich in adrenergic beta receptor and if any, it has agonistic effect on beta receptor. The results were summarized as follows : 1. Higenamine had remarkable bronchodilating effect in guinea pig tracheal smooth muscle in a dose-dependent manner. 2. Bronchodilator effect of higenamine in isolated guinea pig tracheal smooth muscle was blocked competitively by propranolol. The $pD_2$ value of higenamine in isolated guinea pig tracheal smooth muscle was 5.65 and the $pA_2$ value of propranolol against higenamine in the same preparation was 7.97.
Poly(trimethylene terephthalate-co-trimethylene 2,6-naphthalate)s (P(TT-co-TN)s) with various copolymer composition were synthesized, and their chain structure, thermal property and crystalline structure were investigated by using $^1$H-NMR spectroscopy, differential scanning calorimetry (DSC) and wide-angle X-ray diffraction (WAXD), respectively. It was found from sequence analysis that all the P(TT-co-TN) copolymers synthesized have a statistical random distribution of TT and TN units. It was also observed from DSC thermograms that the glass transition temperature increases linearly with increasing the TN comonomer content, whereas the melting temperature of copolymer decreases with increasing the corresponding comonomer content in respective PTT- and PTN-based copolymer, showing pseudo-eutectic melting behavior. All the samples melt-crystallized isothermally except for P(TT-co-66 mol % TN) exhibit multiple melting endotherms and clear X-ray diffraction patterns. The multiple melting behavior originates from the dual lamellar population and/or the melting-recrystallization-remelting. The X-ray diffraction patterns are largely divided into two classes depending on the copolymer composition, i.e., PTT and PTN $\beta$-form diffraction patterns, without exhibiting cocrystallization.
This study was conducted to investigate the effect of dietary supplementation of vitamin A on the membrane property and ultrastructure in ethanol-administered rat livers. Male Sprague-Dawley rats weighing of 130 ~150g were fed with experimental diets for 7 weeks. The diets contained different types of vitamin A which were $\beta$-carotene, retinyl acetate and retinoic acid. After feeding theexperimental diets for 7 weeks, a dose of 3.0g ethanol (30%, W/V)/kg B.W was injected to rats intraperitoneally. Control rats received 0.9% saline containing isocaloric sucrose instead of ethanol. Plasma membrane fluidity of liver decreased in rats fed with vitamin a -Deficient diet with ethanol as compared to that of control rats. Fluidity change of liver plasma membrane that ethanol had induced was influenced by dietary supplementation of vitamin A, but not influenced by the type of supplemented vitamin. A . The ultrastructural changed of hepatic mitrochondria were observed in some rats such as vitamin A-deficient rats with ethanol. Inadequate consumptionof vitamin A contributed to ultrastructural changes such as swelled mitochondria occurred by ethanol-induced hepatotoxicity. Although accurate mechanism involved in the plasma membrane-stabilizing effect of vitamin A is still unclear, dietary supplementation of vitamin A such as retinyl acetate is neede to modulate this change. The direct involvement of membrane property on the cell damage caused by ethanol treatment remains to be established.
Acquiring a selective growth advantage by breaking the proliferation barrier established by gatekeeper genes is a centrally important event in tumor formation. Removal of the mammalian Hippo kinase Mst1 and Mst2 in hepatocytes leads to rapid hepatocellular carcinoma (HCC) formation, indicating that the Hippo signaling pathway is a critical gatekeeper that restrains abnormal growth in hepatocytes. By rigorous genetic approaches, we identified an interacting network of the Hippo, Wnt/${\beta}$-catenin and Notch signaling pathways that control organ size and HCC development. We found that in hepatocytes, the loss of Mst1/2 leads to the activation of Notch signaling, which forms a positive feedback loop with Yap/Taz (transcription factors controlled by Mst1/2). This positive feedback loop results in severe liver enlargement and rapid HCC formation. Blocking the Yap/Taz-Notch positive feedback loop by Notch inhibition in vivo significantly reduced the Yap/Taz activities, hepatocyte proliferation and tumor formation. Furthermore, we uncovered a surprising inhibitory role of Wnt/${\beta}$-catenin signaling to Yap/Taz activities, which are important in tumor initiation. Genetic removal of ${\beta}$-catenin in the liver of the Mst1/2 mutants significantly accelerates tumoriogenesis. Therefore, Wnt/${\beta}$-catenin signaling, known for its oncogenic property, exerts an unexpected function in restricting Yap/Taz and Notch activities in HCC initiation. The molecular interplay between the three signaling pathways identified in our study provides new insights in developing novel therapeutic strategies to treat liver tumors.
Polyvinylidene fluoride (PVDF) as a representative polymer with the piezoelectric property has been studied since the 1960s. Crystalline structure of poly(vinylidene fluoride) polymer is composed of five different crystal structure of the polymer as a semi-crystalline. Among the various crystal structures, ${\beta}-type$ crystal exhibits a piezoelectricity because the permanent dipoles are aligned in one direction. Generally ${\beta}-form$ crystal structure can be obtained through the transformation of the ${\alpha}-form$ crystal structure by the stretching and it can increase the amount through the after treatment as poling process after stretching. ${\beta}-form$ crystal structure the PVDF fibers produced by wet spinning is formed through a diffusion mechanism of a polar solvent in the coagulation bath. However, it has a disadvantage that the diffusion path of the solvent remains as pores in the fiber because the fiber solidification occurs simultaneously with the diffusion of the polar solvent. These pores play a role in reducing effect of poling process owing to effect of disturbances acting on the polarization by the electric field. In this work, the drying method using the microwave was introduced to remove more effectively the residual solvent and the pore within PVDF fibers produced through wet-spinning process and piezoelectric PVDF fibers was produced by transformation of the remaining ${\alpha}$ form crystal structure into ${\beta}-crystal$ structure through the stretching process.
PMAP-23 is a 23-residue antimicrobial peptide derived from porcine myloid cells. In order to investigate the effects of two Pro residues at positions 12 and 15 of PMAP-23 on antibiotic activity, two analogues in which Ala was substituted for Pro residue at position 12 or 15 were synthesized. $Pro^{12}{\rightarrow}Ala$ (PMAPl) or $Pro^{15}{\rightarrow}Ala$(PMAP2) substitution in PMAP-23 caused a significant reduction on antitumor and phospholipid vesicle-disrupting activities, but did not cause a significant effect on antibacterial activity. PMAP-23 displayed the type I ${\beta}-turn$ structure with a negative ellipticity at near 205 om in SDS micelle, whereas PMAP1 and PMAP2 had a somewhat ${\alpha}-helical$ propensity in TFE solution, as compared to PMAP-23. These results suggest that two Pro residues of positions 12 and 15 in PMAP-23 play important roles in the formation of ${\beta}-turn$ structure on lipid membrane and its ${\beta}-turn$ structure may be essential for antibiotic activity including phospholipid vesicle-disrupting property.
Chitosan is a dietary fiber because of a linear polysaccharide composed of $\beta-(1{\rightarrow}4)$-linked 2-amino-2-deoxy-D-glucopyranose. In this study, control release system of vitamin C has been estimated in chitosan molecular network as a vitamin C carrier of controlled release. The amount of released vitamin C were decreased in higher amount of chitosan concentration. Especially, vitamin C were slowly released from chitosan solution in dialysis membrane when compared with vitamin C solution alone in dialysis membrane. These result assumed that chitosan driving force is dependent on chitosan molecular weight and cationic property of amino group with anionic property of vitamin C.
