  • This study shows that lignocellulosic biomass saccharification work has been carried out with rice-straw by the extracellular enzyme from KMU001, and the enzymes produced in 5%(w/v) wood biomass were characterized by protein and various enzyme activity measurements. Several cellulases such as Endoglucanase(EG), $\beta$-D-1,4-Glucosidase(BGL), Cellobiohydrolase(CBH), and $\beta$-D-1,4-Xylanase (BXL) were detected. Saccharification of rice-straw by the enzyme yielded about 233mg/g of glucose after 48hrs.

Screening for α-amylase Inhibitory Activities of Woody Plants

  • Inhibitors of α-amylase are important for the treatment of diabetes and obesity. Using enzyme inhibitor's activity, ethanolic extracts of 87 species in 12 families were screened and compared their inhibitory effect on α-amylase, As a results, we can find that extracts of Distylium racemosum, Acer tegmentosum, Corylapsis veitchiana, Cornus walteri and Corylapsis spicata showed higher α-amylase inhibitory activities than the others and have potential possibility of using control agents for carbohydrate-dependent disease.

유아참여 워크숍을 통한 숲놀이 활동 및 공간 요소의 도출에 관한 연구 (A Study on the Deduction of the Forest Play Activity and Space through Preschooler Participatory Workshop)

  • 최근 사용자의 필요와 요구가 반영된 조경공간계획을 위한 방법으로 사용자 참여 워크숍이 적용되고 있다. 또한, 빠른 속도로 증가하고 있는 유아숲체험원의 조성에 대한 질적 향상이 요구되고 있다. 따라서 본 연구의 목적은 유아숲체험원을 조성함에 있어 사용자의 기본적인 필요와 요구를 담은 계획요소(숲놀이 활동과 공간)를 유아참여 워크숍을 통해 도출하는 것에 있다. 이를 위해 유아발달상의 요구가 충족된 유아 참여디자인 주요 도구를 선정하고, 단계별 워크숍을 수행하였다. 첫 번째, 사전 워크숍단계에서 유아참여(K유치원 6, 7세 통합반 41명) 결과를 통해 계획요소의 표준유형을 도출하였다. 두 번째, 본 워크숍단계에서 대상지(송산물빛유아숲체험원)에 도입할 계획요소를 참여유아(사전워크숍 참여유아)들의 선택을 통해 도출하고, 그 과정과 결과를 분석하였다. 유아 참여과정에서 사용된 도구는 사전워크숍에서 '그림그리기', 본 워크숍에서 계획요소 표준유형 차트(숲놀이활동 픽토그램 차트, 숲놀이공간 실사이미지 차트)이다. 그 결과, 첫 번째 도출된 숲놀이 활동의 표준유형은 38개이다. 인지놀이 27개(기능 16, 구성 4, 상징 4, 규칙 있는 게임 3), 기타 놀이 9개(감각(탐색) 5, 기타 4), 사회놀이 2개(혼자, 집단) 유형으로 구성된다. 숲놀이 공간의 표준유형은 21개이다. 놀이시설 공간 8개(시설 5개, 자연 3개), 수공간 2개 외 5 공간 11개로 구성된다. 두 번째, 대상지에 적용한 결과, 대상지 내 도입할 숲놀이 활동은 전체적으로는 기능놀이가, 단위활동으로는 오르기와 물놀이가 가장 많이 선택되었다. 또한 유아발달놀이 측면에서는 기능, 구성, 상징, 규칙 있는 게임, 기타 놀이가 고르게 선택되었다. 대상지 내 도입할 숲놀이 공간의 경우, 유아의 성별 나이별 선택결과는 유아의 전체 선택 결과와 비슷한 경향을 보이나, 남아는 여아보다 신체, 모험 등의 기능놀이공간을, 여아는 남아보다 쉼터공간을 더 선호하는 것으로 분석되었다. 이 결과는 유아의 숲놀이 행태에 대하여 직접 관찰한 선행연구의 결과와 유사하였으며, 유아들이 대상지를 인지하고, 대상지 내에 도입할 계획요소를 선택한 것으로 분석된다. 따라서 본 연구에서의 참여디자인 워크숍 과정과 과정별 도구들은 연구의 목적에 맞게 구축 적용된 것으로 파악되며, 향후 유아숲체험원 조성계획 시 적용할 수 있는 사례로서 가치가 있다.

숲유치원에서의 유아의 경험 세계 (Childrens' Experience in the Forest Kindergarten)

  • 본 연구는 숲유치원에서의 유아의 경험 세계를 살펴보고 그 안에 내재된 유아교육적 함의를 도출하고자 하였다. 더불어 이를 통해 앞으로 숲유치원이 나아가야 할 긍정적 방향성을 모색할 수 있는 기초자료로 삼고자 하였다. 자료 수집은 2012년 3월 넷째 주부터 2012년 11월 셋째 주까지 참여관찰, 면담, 문서 수집, 사진과 비디오 촬영을 통해 이루어졌다. 연구참여자는 부산시 H유치원 만5세반 24명의 유아와 유아들의 어머니 7인, 원장과 담당교사 2인, 숲해설가 2인이다. 연구자는 본 연구의 목적을 실현하기 위해 현상학적 질적 연구 방법을 통해 본 연구의 현장인 숲유치원의 상황적인 맥락을 고려하며 유아들을 관찰하였다. 연구결과 숲유치원에서의 유아들의 경험 세계를 본성의 장, 심미의 장, 참 놀이의 장으로 나타낼 수 있었다. 이를 통해 숲유치원이 유아들에게 타고난 본성을 드러내어 아름다움을 느끼고 참 놀이에 깊이 몰입할 수 있는 공간으로서 의미를 지니고 있음을 확인할 수 있었다.

Modification of ginsenoside saponin composition via the CRISPR/Cas9-mediated knockout of protopanaxadiol 6-hydroxylase gene in Panax ginseng

  • Background: The roots of Panax ginseng contain two types of tetracyclic triterpenoid saponins, namely, protopanaxadiol (PPD)-type saponins and protopanaxatiol (PPT)-type saponins. In P. ginseng, the protopanaxadiol 6-hydroxylase (PPT synthase) enzyme catalyses protopanaxatriol (PPT) production from protopanaxadiol (PPD). In this study, we constructed homozygous mutant lines of ginseng by CRISPR/Cas9-mediated mutagenesis of the PPT synthase gene and obtained the mutant ginseng root lines having complete depletion of the PPT-type ginsenosides. Methods: Two sgRNAs (single guide RNAs) were designed for target mutations in the exon sequences of the two PPT synthase genes (both PPTa and PPTg sequences) with the CRISPR/Cas9 system. Transgenic ginseng roots were generated through Agrobacterium-mediated transformation. The mutant lines were screened by ginsenoside analysis and DNA sequencing. Result: Ginsenoside analysis revealed the complete depletion of PPT-type ginsenosides in three putative mutant lines (Cr4, Cr7, and Cr14). The reduction of PPT-type ginsenosides in mutant lines led to increased accumulation of PPD-type ginsenosides. The gene editing in the selected mutant lines was confirmed by targeted deep sequencing. Conclusion: We have established the genome editing protocol by CRISPR/Cas9 system in P. ginseng and demonstrated the mutated roots producing only PPD-type ginsenosides by depleting PPT-type ginsenosides. Because the pharmacological activity of PPD-group ginsenosides is significantly different from that of PPT-group ginsenosides, the new type of ginseng mutant producing only PPD-group ginsenosides may have new pharmacological characteristics compared to wild-type ginseng. This is the first report to generate target-induced mutations for the modification of saponin biosynthesis in Panax species using CRISPR-Cas9 system.

Evaluating Pre-silicon Treatment to Alleviate Drought Stress and Increases Antioxidative Activity in Zoysia japonica

  • This study was performed to determine the effects of silicon on zoysiagrass after the application of drought stress. The daily amount of water or scilicon solution was 150 ml per a pot. For 14 days, plants were treated with 0.1 and 1.0 mM silicon (Si) and with distilled water for control and the drought only-treatment. Afterward, the plants in Si and drought treatment were exposed to a 21-day under drought stress condition but the plants in control received water. The results indicated that the growth and the moisture and chlorophyll contents decreased in the drought only-treatment and 0.1 mM Si compared to the control. However, 1.0 mM Si showed an increase in the growth with a significant increase of water and chlorophyll contents. The MDA and $H_2O_2$ concentrations and electrolyte leakage decreased, while the radical scavenging capacity increased in 1.0 mM Si. 1.0 mM Si showed little to no differences in the growth and no differences in water and chlorophyll contents, electrolyte leakage, MDA and $H_2O_2$ concentrations and antioxidant capacity compared to the control. These results suggested that application of silicon is useful for drought tolerance improvement of zoysiagrass under drought that is occurring in turf fields.

Demethoxylation of Milled Wood Lignin and Lignin Related Compounds by Laccase from White-rot Fungus, Cerrena unicolor

  • Highly purified Cerrena unicolor laccase (benzenediol:oxygen oxidoreductase, EC 1.10.3.2) caused the demethoxylation of milled wood lignin and several lignin related substances. The constitutive form of the enzyme produced extracellularly by C. unicolor fermenter culture was isolated and purified by ion-exchange chromatography on the DEAE-Toyopearl column and by affinity chromatography on a ConA-Sepharose and Syringyl-AH-Sepharose 4B columns. The enzyme was further immobilized on functionalized porous glass (CPG) and keratin coated CPG. The demethylating activity was monitored both by estimation of released methanol and by detection of the level of methoxyl groups (also in some water miscible solvents) after incubation of lignin materials with laccase preparations (free and immobilized). The effects of the incubation time and temperature on the demethoxylating activity of immobilized laccase preparations were also studied.

  • Bioprospecting has been widely used to assess the economic potential of different plant species and their value-addition. Prospecting for biological material like plants with medicinal or other economically valuable properties like fibre or oil is becoming a dynamic activity. Our folklore with embedded cultural heritage has tremendous possibilities and potential for bioprospecting. This forest region of Western Rajasthan is enriched with diverse vegetational wealth, if subjected to bioprospecting may prove to be a boon for the society.

독활 에탄올 추출물의 대장암 세포에서 Cyclin D1 단백질 분해 유도를 통한 세포 생육 억제활성 (Anti-proliferative Activity of Ethanol Extracts of Root of Aralia cordata var. continentalis through Proteasomal Degradation of Cyclin D1 in Human Colorectal Cancer Cells)

  • Background: In this study, we evaluated the anti-cancer activity and potential molecular mechanism of 70% ethanol extracts of the root of Aralia cordata var. continentalis (Kitagawa) Y. C. Chu (RAc-E70) against human colorectal cancer cells. Methods and Results: RAc-E70 suppressed the proliferation of the human colorectal cancer cell lines, HCT116 and SW480. Although RAc-E70 reduction cyclin D1 expression at the protein and mRNA levels, RAc-E70-induced reduction in cyclin D1 protein level occurred more dramatically than that of cyclin D1 mRNA. The RAc-E70-induced downregulation of cyclin D1 expression was attenuated in the presence of MG132. Additionally, RAc-E70 reduced HA-cyclin D1 levels in HCT116 cells transfected with HA-tagged wild type-cyclin D1 expression vector. RAc-E70-mediated cyclin D1 degradation was blocked in the presence of LiCl, a $GSK3{\beta}$ inhibitorbut, but not PD98059, an ERK1/2 inhibitor and SB203580, a p38 inhibitor. Furthermore, RAc-E70 phosphorylated cyclin D1 at threonine-286 (T286), and LiCl-induced $GSK3{\beta}$ inhibition reduced the RAc-E70-mediated phosphorylation of cyclin D1 at T286. Conclusions: Our results suggested that RAc-E70 may downregulate cyclin D1 expression as a potential anti-cancer target through $GSK3{\beta}$-dependent cyclin D1 degradation. Based on these findings, RAc-E70 maybe a potential candidate for the development of chemopreventive or therapeutic agents for human colorectal cancer.
