Protective Effects of Pyrus pyrifolia NAKAI Leaf Extracts on UVB-induced Toxicity in Human Dermal Fibroblasts (자외선B 노출로 인해 손상된 피부세포에 대한 돌배나무잎 추출물의 보호효과)

  • Skin damage is mainly caused by environmental factors such as ultraviolet light, heat, and smoking. It is known that reactive oxygen species production is commonly involved in the pathogenesis of skin damage induced by these factors, causing skin aging. Pyrus pyrifolia Nakai continues to be a popular and highly consumed fruit in many countries with known beneficial effects including antitumor, antioxidative, and anti-inflammatory effects. However, there is no evidence of a therapeutic effect of Pyrus pyrifolia extract (PPE) against skin aging via inhibition of mitochondria-mediated apoptosis. In this study, we investigated PPE protective effect against photoaging induced by UVB ($50mJ/cm^2$) in HS68 human dermal fibroblasts. Lactate dehydrogenase assay showed that PPE significantly protected HS68 cells against UVB-induced damage in a dose-dependent manner. Other assays using DCF-DA demonstrated that PPE protected HS68 cells by regulating reactive oxygen species production. PPE also regulated mitochondrial dysfunction and mitochondrial membrane potential induced by UVB, and inhibited UVB-induced caspase-3 activity. These results indicate that PPE protects human dermal fibroblasts from UVB-induced damage by regulating the oxidative defense system.

AtERF11 is a positive regulator for disease resistance against a bacterial pathogen, Pseudomonas syringae, in Arabidopsis thaliana (애기장대 AtERF11 유전자에 의한 Pseudomonas syringae에 대한 병 저항성 유도)

  • AvrRpt2 protein triggers hypersensitive response (HR) and strong disease resistance when it is translocated from a bacterial pathogen Pseudomonas sp. to host plant cells containing a cognate RPS2 resistance protein through Type III Secretion System (TTSS). However, AvrRpt2 protein can function as the effector that suppresses a basal defense and enhances the disease symptom when functional RPS2 resistance protein is absent in the infected plant cells. Using Affymetrix Arabidopsis DNA chip, we found that many genes were specifically regulated by AvrRpt2 protein in the rps2 Arabidopsis mutant. Here, we showed that expression of AtERF11 that is known as a member of B1a subcluster of AP2/ERF transcription factor family was down regulated specifically by AvrRpt2. To determine its function in plant resistance, we also generated the Arabidopsis thaliana transgenic plants constitutively overexpressing AtERF11 under CaMV 355 promoter, which conferred an enhanced resistance against a bacterial pathogen, Pseudomonas syringae pv. tomato DC3000. Thus, these results collectively suggest that AtERF11 plays a role as a positive regulator for disease resistance against biotrophic bacterial pathogen in plant.

Alleviation of Low and High Temperature Injury in Tomato Plants by Uniconazole (Uniconazole처리가 토마토의 저온 및 고온 피해 경감에 미치는 효과)

  • This study was carried out to determine the effectiveness of uniconazole in ameliorating low and high temperature injury in tomato plants(Lycopersicon esculentum Mill. cvs. Fireball and Patio). Plants were given a soil drench of 0, 0.001, 0.01 or $0.1mg{\cdot}pot^{-1}$ uniconazole, and after 14 days, were treated with 12-h day/12-h night cycles at $25/25^{\circ}C$, $2.5/25^{\circ}C$, $25/2.5^{\circ}C$ or $40/40^{\circ}C$ for 4 days in controlled-environment chamber. Number of damaged leaves per plant, reduction of stem elongation, and overall injury were high at $2.5/25^{\circ}C$, but more reduction of leaf elongation, delay of flowering, and abortion of floral bud were observed in plants at $40/40^{\circ}C$. There was difference in degree of injury between cultivars, thus, 'Fireball' was much affected by unfavorable temperature regimes. All concentrations of uniconazole reduced leaf and stem elongation, increased total chlorophyll concentration, delayed flowering, and significantly provided protection against low and high temperature injury in two cultivars. In general, the application of uniconazole did not inhibit flowering delay and floral bud abortion induced by high and low temperature exposure. Our results support the hypothesis that the role of uniconazole is related to defense system against oxidative stress induced by low temperature stress. Further research is required to clarifu the phytoprotective mechanism of this compound agaist high temperature stress.

Transgenic Tomato Plants That Overexpress Superoxide Dismutase in Fruits (토마토 과실에서 Superoxide Dismutase를 고발현하는 형질전환 식물체)

  • Superoxide dismutase (SOD) plays an important role in cellular defense against oxidative stress in plants. We have developed transgenic tomato plants overexpressing a cassava SOD in fruits. Three transgenic tomato plants (one from cv. Pink forcer and two from cv. Koko) using a new vector system, ASOp :: . mSOD1/pBI101, harboring ascorbate oxidase promoter (ASOp) expressing dominantly in cucumber fruits, CuZnSOD cDNA (mSOD1) isolated from cultured cells of cassava, and nptll gene as a selectable marker were successfully developed. SOD specific activity (units/mg protein) in transgenic fruits of both cultivars was increased with maturation of the fruits. SOD specific activity of well-mature fruits in transgenic Pink forcer and Koko showed approximately 1.6 and 2.2 times higher than control fruits, respectively. The strength of SOD isoenzyme bands well reflected the SOD activity during the fruit maturation. These results suggested that SOD gene was properly introduced into tomato fruits in a fruit-dominant expression manner by ASO promoter.

  • Cyberattack technology is evolving to an unpredictable degree, and it is a situation that can happen 'at any time' rather than 'someday'. Infrastructure that is becoming hyper-connected and global due to cloud computing and the Internet of Things is an environment where cyberattacks can be more damaging than ever, and cyberattacks are still ongoing. Even if damage occurs due to external influences such as cyberattacks or natural disasters, intelligent self-recovery must evolve from a cyber resilience perspective to minimize downtime of cyber assets (OS, WEB, WAS, DB). In this paper, we propose an intelligent self-recovery technology to ensure sustainable cyber resilience when cyber assets fail to function properly due to a cyberattack. The original and updated history of cyber assets is managed in real-time using timeslot design and snapshot backup technology. It is necessary to secure technology that can automatically detect damage situations in conjunction with a commercialized file integrity monitoring program and minimize downtime of cyber assets by analyzing the correlation of backup data to damaged files on an intelligent basis to self-recover to an optimal state. In the future, we plan to research a pilot system that applies the unique functions of self-recovery technology and an operating model that can learn and analyze self-recovery strategies appropriate for cyber assets in damaged states.

Luteolin-loaded Phytosomes Sensitize Human Breast Carcinoma MDA-MB 231 Cells to Doxorubicin by Suppressing Nrf2 Mediated Signalling

  • Nuclear factor erythroid 2-related factor 2 (Nrf2) has been recognized as a transcription factor that controls mechanisms of cellular defense response by regulation of three classes of genes, including endogenous antioxidants, phase II detoxifying enzymes and transporters. Previous studies have revealed roles of Nrf2 in resistance to chemotherapeutic agents and high level expression of Nrf2 has been found in many types of cancer. At physiological concentrations, luteolin as a flavonoid compound can inhibit Nrf2 and sensitize cancer cells to chemotherapeutic agents. We reported luteolin loaded in phytosomes as an advanced nanoparticle carrier sensitized MDA-MB 231 cells to doxorubicin. In this study, we prepared nano phytosomes of luteolin to enhance the bioavailability of luteolin and improve passive targeting in breast cancer cells. Our results showed that cotreatment of cells with nano particles containing luteolin and doxorubicin resulted in the highest percentage cell death in MDA-MB 231cells (p<0.05). Furthermore, luteolin-loaded nanoparticles reduced Nrf2 gene expression at the mRNA level in cells to a greater extent than luteolin alone (p<0.05). Similarly, expression of downstream genes for Nrf2 including Ho1 and MDR1 were reduced significantly (p<0.05). Inhibition of Nrf-2 expression caused a marked increase in cancer cell death (p<0.05). Taken together, these results suggest that phytosome technology can improve the efficacy of chemotherapy by overcoming resistance and enhancing permeability of cancer cells to chemical agents and may thus be considered as a potential delivery system to improve therapeutic protocols for cancer patients.

Antioxidant Activity in Water and Methanol Extracts from Korean Edible Wild Plants (국내산 산채류의 물 및 메탄올 추출물에 대한 항산화 활성)

  • The present study was performed to investigate antioxidant activities of Korean edible wild extracts. In water and methanol extracts of edible wild plants, total polyphenol content of water and methanol extracts ranged 4.6~183.8 and 8.2~270.1 mg/g, respectively. Radical scavenging activity against DPPH and ABTS radicals was observed to be high in Lysimachia barystachys, Aceriphyllum rossii, Securinega suffruticosa, Sedum kamtschaticum, and Pedicularis resupinata. Pedicularis resupinata, Securinega suffruticosa, Patrinia villosa, and Solidago virga-aurea var. asiatica showed effective SOD-like activity. There was significant correlation between polyphenol content and DPPH and ABTS radical scavenging activities, and SOD-like activity. Plant extracts such as Pedicularis resupinata, Securinega suffruticosa, Rhapontica uniflora, Lysimachia barystachys, and Aceriphyllum rossii had higher polyphenol content, radical scavenging, and SOD-like activity. These results indicate that Korean edible wild plants may be useful as potential antioxidant sources for improving human antioxidant defense system.

Effects of Coenzyme Q10 on the Antioxidant System in Blood and Liver of Laying Hens (산란계에서 Coenzyme Q10 급여가 혈액 및 간 조직의 항산화 작용에 미치는 영향)

  • The objective of this study was to investigate the effects of dietary coenzyme Q10 (CoQ10) sources on the antioxidant defense system in the blood and liver of laying hens. Thirty-six 40-wk old Lohmann Brown hens were randomly assigned to three groups based on body weight, with four cages with three layers each. Laying hens were divided into one of the following groups: control (CON), powdered CoQ10 (PCoQ, 100 mg/kg diet), and emulsified CoQ10 (ECoQ, 100 mg/kg diet). All hens were fed a control diet or a control diet supplemented with powdered or emulsified CoQ10 ad libitum for five weeks. There were no differences in body weight, weight gain, and organ weights among the treatment groups, including the liver and spleen. The blood total antioxidant power (TAP) in the ECoQ group increased (P<0.05) by approximately 2-fold compared to that in the CON group. However, there was no significant difference in blood TAP levels between the PCoQ and ECoQ groups, although a decreasing trend (P<0.13) was observed for levels of TAP in the ECoQ group. The mRNA expression and specific activities of superoxide dismutase, glutathione peroxidase, and catalase in the liver were not affected by dietary CoQ10 or type of CoQ10. However, hepatic lipid peroxidation in the ECoQ group was lower (P<0.05) than in the CON group. In conclusion, emulsified CoQ10 increased blood TAP and decreased hepatic lipid peroxidation without affecting antioxidant enzymes, suggesting that emulsified CoQ10 might be more applicable as an active antioxidant supplement than powdered type in laying hens.

Analysis of the Eyeglasses Supply System for Ametropes in ROK Military (한국군 비정시자용 안경의 보급체계 분석)

  • Purpose : To analyze the eyeglasses supply system for ametropic soldiers in ROK military. Methods : We investigated and analyzed the supply system of eyeglasses for the ametropic soldiers provided by the Korean military. The refractive powers and corrected visual acuity were measured for 37 ametropic soldiers who wear insert glasses for ballistic protective and gas-masks supplied by the military based on their habitual prescriptions. Full correction of refractive error was prescribed for subjects having less than 1.0 of distance visual acuity, and comparison was held for inspecting the changes in corrected visual acuity. Suggestions were provided for solving the issues regarding current supplying system, and this study investigated the applicabilities for utilizing professional optometric manpower. Results : The new glasses supplied by army for ametropic soldiers were duplicated from the glasses they worn when entering the army. The spherical equivalent refractive powers of the conventional, ballistic protective and gas-mask insert glasses supplied for 37 ametropic soldiers were $-3.47{\pm}1.69D$, $-3.52{\pm}1.66D$ and $-3.55{\pm}1.63D$, respectively, and the spherical equivalent refractive power of full corrected glasses was $-3.79{\pm}1.66D$, which showed a significant difference(p<0.05). The distant corrected visual acuity measured at high and low contrast(logMAR) of conventional, ballistic protective and gas-mask insert glasses were $0.06{\pm}0.80$, $0.21{\pm}0.82$, $0.15{\pm}0.74$, $0.34{\pm}0.89$, $0.10{\pm}0.70$ and $0.22{\pm}0.27$, respectively, while the corrected visual acuity by full corrected glasses were increased to $0.02{\pm}1.05$, $0.10{\pm}0.07$, $0.09{\pm}0.92$, $0.26{\pm}0.10$, $0.04{\pm}1.00$ and $0.19{\pm}1.00$, respectively. There was a significant difference(p<0.05) except for the case of the low contrast corrected visual acuity of the conventional and gas-mask insert glasses. The procedure for ordering, dispensing, and supplying military glasses consists of 5 steps, and it was found that approximately two weeks or more are required to supply from the initial examination. Conclusion : The procedure of supplying the military glasses showed three issues: 1) a lack of refraction for prescription system, 2) relatively long length of time required for supplying the glasses, 3) an inaccurate power of supplied glasses. In order to solve those issues, in the short term, education is necessarily required for soldiers on the measurement of the refractive powers, and in the near future, further standard procedures for prescription of glasses as well as the securement of optometric manpower are expected.
