Vertical profiles of dissolved Cu and Ni at the upper 400 m water layer have been determined at two stations in the central East Sea in October 1995. This is the first report on the vertical distribution of trace metals in Korea. Copper concentrations are in the range of 2.1~5.8 nmol/kg and 1.6~2.4 nmol/kg for stations S and N, respectively. The vertical profile of Cu at S is found a scavenging type (i.e., drastic decrease with increasing depth). Concentrations of Ni range from 4.3 to 7.1 nmol/kg and from 3.4 to 5.4 nmol/kg for stations Sand N, respectively. At station S, Ni is best correlated with phosphate, but not at stations N. Such difference between two stations are probably due to their different vertical distribution of water masses. Station S has a strongly stratified water column with 6 distinct water masses, but station N with a well-mixed subsurface water layer extending from 50 to 300 m depth. Extremely low salinity (31.87~31.96 psu) found at the surface water of station S was interpreted as a result of the Yangtze River effluents which were probably fed into the East Sea through the Korea Strait during the late summer. Such seasonal appearance of low salinity in southern part of the East Sea was reported previously. The concentrations of Cu and Ni at two sites are comparable to those reported in the North Pacific. It was found that Ni mostly exist as dissolved phase.

Ventilation shafts are pathways in mines and tunnels for the removal of dust or smoke during underground space construction and operation. In mines, blasting with long blast holes is preferred for the excavation of a ventilation shaft in the 10~20m long crown pillar section. In this case, the bottom part of the blast hole is completely drilled in order to determine the drilling error, and this causes a problem of lowering the explosive charge and blasting efficiency. It is possible to solve the problem of explosive loading and to increase the blast efficiency by covering the curb of the blasthole by using stemming material. In this study, simulations for the blasting of a ventilation shaft were performed with various stemming lengths and the blasthole diameters(45, 76mm) using AUTODYN 2D SPH(Smooth particle hydrodynamics) analysis technique. Also the optimal bottom stemming column was derived by checking the size of the boulder and burden line according to blasting. Analysis result, blasting efficiency is lessened in case of stemming length less than 30cm and the optimal length of the stemming material should be 30cm or higher to achieve high efficiency of blasting.
Accumulation, elimination and subcellular distribution of heavy metals in Littorina brevicula exposed to cadmium and zinc separately and concurrently were investigated. When the winkles had been exposed to 400 ㎍/L CdCl₂ and 3000 ㎍/l ZnSO₄ separately for 90 days, each of the metal body burden in the whole sofl parts increased in proportion to time of exposure until 70 days. But it didn't increase after 70 days. But when the winkles had been exposed to cadmium and zinc simultaneously, cadmium body burden decreased but zinc body burden increased as compared to the winkles exposed to each of the metal. We also found that cadmium accumulated in the winkles was not depurated for 42 days, but zinc accumulated in them was depurated. Especially, zinc was depurated faster when they had been exposed to mixture of cadmium and zinc. After the winkles had been exposed to cadmium and zinc separately for 70 days, about 60% cadmium of the total body burden was associated with the soluble fraction, while about 75% zinc of the total body burden was associated with insoluble fraction. And these trends of metal partitioning did not alter when the winkles had been exposed to metal mixture. After the soluble fraction applied to gel-filtration chromatography column, the distribution patterns of cadmium and zinc associated with proteins or ligands were different each other. Most of cadmium (>90%) in the soluble fraction was bound to MBP-1 (Metal-binding protein-1), about 6.5 kDa), while zinc was distributed evenly to HMW (High molecular weight fraction, >60 kDa), MBP-1, MBP-2 (about 5 kDa), LMW (Low molecular weight fraction, <1 kDa).

The down-core distribution of chlorophyll a, organic carbon contents and ${\delta}^{13}C$ in the bottom sediments were measured to understand the evolution of eutrophication in Masan Bay. Bottom sediment were collected in January 1994. The chlorophyll a and organic carbon contents in the sediment core decreased with increasing sediment depth, respectively. Bottom sediments (0~20 cm) in Masan Bay was rich in chlorophyll a (avg. 9.6 ${\mu}g\;g^{-1}$ dryweight) and organic C (avg. 2.5%). The down-core distribution of chlorophyll a suggests that the inner part of Masan Bay has experienced the acceleration of chlorophyll a supply since 1960s. Flux of organic carbon to the sea floor is in the range of 10 $gCm^{-2}\;yr^{-1}$ assuming the C:Chl a ratio of 25. It suggests tht approximately 1.3% of the fixed carbon by phytoplankton appears to be deposited in the bottom sediments.

Contrary to an intact rock, the jointed rock mass shows strain-dependent deformation characteristics (elastic modulus and damping ratio). The maximum elastic modulus of a rock mass can be obtained from an elastic wave-based exploration in a small strain level and applied to seismic analyses. However, the assessment and application of the non-linear characteristics of rock masses in a small to medium strain level ($10^{-4}{\sim}0.5%$) have not been carried out yet. A non-linear dynamic analysis module is newly developed for FLAC3D to simulate strain-dependent shear modulus degradation and damping ratio amplification characteristics. The developed module is verified by analyzing the change of the Ricker wave propagation. Strain-dependent non-linear characteristics are obtained from disks of cored samples using a rock mass dynamic testing apparatus which can evaluate wave propagation characteristics in a jointed rock column. Using the experimental results and the developed non-linear dynamic module, seismic analyses are performed for the intersection of a shaft and an inclined tunnel. The numerical results show that vertical and horizontal displacements of non-linear analyses are larger than those of linear analyses. Also, non-linear analyses induce bigger bending compressive stresses acting on the lining. The bending compressive stress concentrates at the intersection part. The fundamental understanding of a strain-dependent jointed rock mass behavior is achieved in this study and the analytical procedure suggested can be effectively applied to field designs and analyses.

Improving the detection performance of facilities corresponding to the sensors of autonomous vehicles helps driving safety. In the road and transportation field, research is being conducted to improve the detection performance of sensors by road infrastructure or facilities. As part of this on the development of autonomous driving support infrastructure, the shape of traffic cones and drums to ensure sufficient LiDAR detection performance even rainy conditions and maintain the line-of-sight guidance function in construction zones improvement effect. The principle was to increase reflection performance and ensure no significant difference in shape from existing facilities. Traffic cones were manufactured in square pyramid shapes instead of cones, and drums were manufactured in hexagonal and octagonal pillar shapes instead of cylinders. LiDAR detection data for the facility was confirmed on a clear day and with 20 mm/h and 40 mm/h rainfall. The detection performance of the square pyramid-shaped traffic cone and octagonal column-shaped drum was to the existing facility. On the other hand, deviations occurred due to repeated measurements, and significance could not be confirmed through statistical analysis. By reflecting these results, future studies will seek a form in which data can be obtained uniformly despite the diversity of measurement environments.
The structural gene of rabbit hemoglobin was cloned into Pst I site of pBR322 in E. coli. The complementary DNA (cDNA) was synthesized from rabbit globin mRNA with avian myeloblastosis viral reverse transcriptase, and then RNA was destroyed at pH 11. The double stranded cDNA was synthesized with both Klenow fragment of E. coli DNA polymerase I and reverse transcriptase and then the hairpin loop was opened with Sl nuclease. Double stranded cDNA was subsequently tailed with dCTP and annealed to dGMP-tailed vector DNA. After transformation and initial screening of appropriate clones by plasmid size, the cloned colonies were identified by in situ colony hybridization using by plasmid size, the cloned colonies were identified by in situ colony hybridization using $[^32P]$-labeled cDNA probes and characterized the inserts with restriction endonucleases. The expression of cloned globin gene was investigated by standard radioimmunoassay using rat anti-rabbit Hb serum as primary antibody and goat antirat IgG serum as secondary antibody. The result suggested that the chimeric proteins (the part of $\\beta$-lactamase from the vector pBR322 and globin from rabbit) were supposedly produced in E. coli and the product had the antigenic determinant of rabbit hemoglobin.
The multiple 7S globulins composed of two fractions (A and B) in the electrophoresis with Davis' method were isolated at different stages of the soybean seed development. Electrophoresis of their subunits liberated in PAWU solvent [phenol-acetic acid-water (2 : 1 : 1) solution plus 5M urea] yielded 4 major bands. Observation of both the electrophoretic bands of the multiple 7S fractions(7S-A and 7S-B) and those of their subunits was suggestive of a similarity of the subunit pattern between two 7S fractions. The two fractions in multiple 7S globulins were isolated with DEAE-Sephadex A-50 column$(2.0{\sim}100cm)$ chromatography. They were separated into 2 fractions in a linear gradient concentration of 0.28 to 0.40M NaCl with phosphate buffer (pH 7.8) containing 10mM ${\beta}-mercaptoethanol$(ME). The isolated protein was dissociated into subunits with two different solvent systems; in PAWU solvent and in Tris-HCl buffer(pH 8.0) containing 1% sodium dodecyl sulfate (SDS) and 40mM ME. The dissociated subunits were subjected to electrophoresis in PAWU-treated 7.5% acrylamide gel and in 1% SDS-treated 5.6% acrylamide gel. In PAWU gel electrophoresis, total 7S globulin was separated into 5 major bands, two of which were occupied in common by two 7S fractions(7S-A and 7S-B). In SDS gel electrophoresis, total 7S globulin was separated into 7 major bands, three of which were overlapped with the subunit of the two 7S fractions. The above results alluded us to the presence of a common and/or similar subunit between the multiple 7S globulins.

Coinjection of alcohol and air or alcohol flooding only were evaluated with 3-D soil tank for removal of nonaqueous phase liquid (NAPL) contaminant from soil. 70%-ethanol and 40%-isopropanol were used for non-NAPL-swelling alcohol and NAPL-swelling alcohol, respectively. 729 ml-benzene was placed in the 37 liter soil tank. Alcohols were respectively injected from the injection well placed near the bottom of the tank and mobilized free phase NAPL and aqueous phases were then recovered from the extraction well placed in the upper part of the soil tank. Approximately 50% of removed NAPLs were free-phase in all experiments. The results were completely different to the previous soil column experiment results and also implied that alcohol properties did not affect the NAPL removal efficiency in the 3-D soil tank experiment. Air was also co-injected with alcohol to evaluate co-injection effects on NAPL removal. Enhanced NAPL removal effect of co-injection of 70%-ethanol and air was also found even in the 3-D soil tank evaluation. However, co-injection effect of 40%-iso-propanol and air was less apparent. This study determined that the most important parameter governing alcohol flooding for NAPL removal would be extraction capacity to recover NAPL and aqueous phase flowing in the soil. More researches are required for improving recovery efficiency of extraction well in real soil contamination conditions.

The study of bond behavior between concrete and rebar has been performed for a long time. On this study, we tried to analysed variation of bond behaviors quantitatively with varying the strength of concrete. Bond stress which observed below the neutral surface of beam and at connecting part of beam and column is affected by various bond parameters. Resistance of deformed bars which embedded in concrete to the pullout force is divided 1) chemical adhesive force 2) frictional force 3) mechanical resistance of ribs to the concrete and these horizontal components of resistance is being bond strength. We selected the most common and typical variable which is concrete strength among various variables. So we used two kinds of concrete strength like as 25MPa(NSC) and 65MPa(HSC). Tension Test was performed to verify how bond behavior varied with two kinds of concrete strength. Concentration of bond stress was observed at load-end commonly in Tension Test of the initial load stage. At this stage stress distribution was almost coincident at each strength. As tension load added, this stress distribution had difference gradually and movement of pick point of bond stress to free-end and central section was observed. This tendency was observed at first and moving speed was more fast in NSC. At the preceeding result the reason of this phenomenon is considered to discretion of chemical adhesion and local failure of concrete around rebar in load-end direction. Especially, when concrete strength was increased 2.6 times in tension test, ultimate bond strength was increased 1.45 times. In most recent used building codes, bond strength is proportioned to sqare root of concrete compressive strength but comparison of normalized ultimate bond strength was considered that the higher concrete strength is, the lower safety factor of bond strength is in each strength if we use existing building codes. In Tension Test, in case of initial tensile force state, steel tensile stress of central cross section is not different greatly at each strength but tensile force increasing, that of central cross section in NSC was increased remarkably. Namely, tensile force which was shared in concrete in HSC was far greater than that of concrete in NSC at central section.
