Characterization of Materials for Retort Processing in Oyster Porridge (레토르트 굴죽 제조를 위한 원료의 가공적성)

  • The effect of mixing ratio of materials and their retort processing conditions on the physical properties and preference of oyster porridge were investigated. Rice gave adequate viscosity (about 800 cp) to the porridge as it was used 10~12% level and half of it was ground. Potato starch, waxy corn starch, and Perfectamyl AC showed small variation in viscosity unrelated to the temperature, that was a good condition for high quality porridge. Especially, waxy corn starch had a good property for retort porridge because its viscosity was maintained high during the processing and decreased to preferable one after processing. Purity CSC, modified starch gave gum-like texture and didn't cause water-separation much after freezing and thawing treatment of the porridge. Therefore, it was good for enhancing the physical property of the porridge as a co-additive of waxy corn starch when it was used at quarter level of waxy corn starch. Xanthan gum increased dispersibility of materials and it was useful for stabilizing physical quality of the porridge without affecting preference when it was used below 0.2% concentration. As a result, the optimal material-mixing ratio was determined as rice 10% (50% of it was ground), waxy corn starch 1.5%, Purity CSC 0.5%, xmthan gum 0.2%, salt 0.3%, and water 87.5%. Oyster's porridge, retort processed with this recipe, showed stable physical property after 6 month storage at room temperature.

  • Objective: This study was designed to investigate the survival rate of vitrified mouse blastocysts depending on the presence or absence of sucrose in vitrification solution. Methods: Mouse two-cell embryos were collected and cultured to blastocysts. Two vitrification solutions were prepared. The control solution was composed of 25% glycerol, 25% ethylene glycol, and 0.5 M sucrose (G25E250.5S) containing 2.5 mL glycerol, 2.5 mL ethylene glycol, 2 mL SSS, and 0.855 g sucrose in 5 mL PB1. The experimental solution was composed of 25% glycerol and 25% ethylene glycol (G25E25) and contained 2.5 mL glycerol and 2.5 mL ethylene glycol in 5 mL PB1. Artificial shrinkage was conducted by aspirating the blastocoelic fluid using an ICSI pipette. To examine the effect of sucrose in the vitrification solution on the survival rate of mouse blastocysts, the shrunken-equilibrated blastocysts were rehydrated or vitrified after being exposed to one of the two vitrification solutions. After exposure and the vitrification-thawing process, the re-expansion rate and hatching rate were evaluated after 6 hours of in vitro culture. Results: The re-expansion rate of mouse blastocysts exposed to vitrification solution with and without sucrose were not different in the experimental solution (without sucrose) (98%) and the control solution (with sucrose) (92%) (p>0.05). The hatching rate was higher in the experimental solution (95%) than in the control solution (88%), but did not differ across two treatments (p>0.05). The re-expansion rate of mouse blastocysts vitrified in the control solution was 92% and 94%, respectively (p>0.05), and the hatching rate was higher in the experimental solution (90%) than in the control solution (74%) (p<0.05). Conclusion: Sucrose need not be added in vitrification solution for freezing of artificially shrunken mouse blastocysts.

Processing of Sausage Using Duck Mechanically Deboned Meat (오리 기계발골육을 이용한 Sausage 제조)

  • In this paper we dealt with processing of sausage using duck mechanically deboned meat(duck-MDM). The results may be summarized as follows : 1) after semi-thawing of freezing duck-MDM at $25^{\circ}C$ for 3 hours and cutting as thin, 2) alkali washing at low temperature for 4 hours by 0.2% NaHCO$_3$and 0.15% NaCl, 3) curing at low temperature for 4 hours, 4) after washing and dehydrating(moisture 80%), 5) grinding at low temperature for 55 minutes by silent cutter such as 1st grinding for 10 minutes added only dehydrated meat, 2rd grinding for 30 minutes added salt in 1st grinded meat and 3rd grinding for 15 minutes added other additives, 6) after quick casing in PVDC film and heating at 9$0^{\circ}C$ for 80 minutes, 7) cooling to below room temperature. The additives added at 3rd grinding process were Polymix-CA(0.3%), Polymix-CS(0.3%), polyphosphate(0.3%), sugar(4.2%), potato starch(8.0%), pyre-phosphates(0.3%), isolated soy protein(7.0%), MSG(0.2%), onion powder(0.5%), garlic powder(0.1%), nutmeg (1.5%), potassium sorbate(<0.1%), food red no.40(0.0075%), egg albumin(7.0%) and gluten(3.0%).

Developmental and survivability according to cryopreservation of in vitro produced bovine embryos cultured by addition of Antioxident cysteine (Cysteine 첨가로 배양된 소 수정란의 발달과 동결성 효과)

  • The aim of the present study was to assess the embryo development and survivability of post-thawed bovine embryos produced in vitro by addition of cysteine. The rates of metaphase II formation were not differed significantly among three groups(TCM199 73.8%, TCM199 with 0.3% cysteine 76.9%, TCM199 with 0.5% cysteine 83.8%, respectively). No difference of cleavage rate(70.6~74.6%) was seen among three culture medium(TCM199 70.6%, CR1aa 71.3%, SOF 74.6%) with 0.5M cysteine. however, Significantly(P<0.05) higher development rate into blastocyst stage by 0.5M cysteine addition was obtained in SOF medium(35.6%) than in TCM199(27.6%) or CR1aa(26.6%), however no significant differences in the cleavage rates were among three culture medium. After frozen the blastocysts cultured with 0.5M cysteine, The re-expansion rates were 61.3%~86.4% among groups, and hatching rates were 26.3%~46.9% among groups, the rates of re-expansion and hatching were significantly(P<0.05) higher in SOF medium(86.4% and 46.9%) than those in TCM199(61.3% and 26.3%) and CR1aa medium(87.1 and 44.4%). After thawing, the blastocyst re-expansion rate was significantly(P<0.05) higher in in vivo (87.1%) and in vitro (70.3%) embryos. In conclusion, our results demonstrate that supplementation of IVM and IVC medium with 0.5M cysteine improved the quality of in vitro production embryo and post- thawed embryo. Future studies comparing these media systems in well-designed trials should be performed.

  • Artificial lightweight aggregates and solids were manufactured with coal ash(fly ash, bottom ash). In order to apply alkali-activated coal ash(fly ash, bottom ash) artificial lightweight aggregate to concrete, several experimental studies were performed. Thus, it can be noticed the optimal mix proportion, basic characteristies, mechanical properties and environmental safety of alkali-activated coal ash(fly ash, bottom ash) solid and alkali-activated coal ash(fly ash, bottom ash) artificial lightweight aggregate. Also, the freezing-thawing test property of concrete using the alkali-activated coal ash(fly ash, bottom ash) artificial lightweight aggregate was investigated. As a result, the optimal mixing proportion of coal ash(fly ash, bottom ash) solid to make alkali-activated artificial lightweight aggregates was cement $10\%$, water glass $15\%$, NaOH $10\%$, $MnO_2\;5\%$. Alkali-activated coal ash(fly ash, bottom ash) solid can achieve compressive strength of 36.4 MPa, at 7-days, after the paste was cured at air curing after moist curing during 24 hours in $50^{\circ}C$. Alkali-activated coal ash(fly ash, bottom ash) artificial lightweight aggregate that do impregnation to polymer was improved $10\%$ crushing strength $150\%$, and was available to concrete.

A Study on the Physical Properties of Interlocking Block with the Contents of the Recycled Aggregate (순환골재 혼합비율에 따른 인터로킹 블록의 물리적 특성에 관한 연구)

  • Recycled aggregates are made from construction wastes, and they have many national and social benefits by saving energy, developing substitute resources, and protecting environment. However, low-quality recycled aggregate with low density and high absorption rate cannot be used for structural concrete aggregate but is used mainly for low added value. Therefore, this study aims to identify the characteristics of the materials of recycled aggregates made after crashing and pulverizing waste concrete. For this, their major physical characteristics of cement content, absolute dry density, absorption rate, etc. were reviewed to make a mix design (draft) for the production of the secondary product and performance evaluation was done on the bending strength, absorption rate, bending strength after freezing and thawing, compressive strength, air-dried gravity, etc. of the test products produced by applying the mix design to compare the results with the quality standards of GR mark. The results of the tests showed that the substitution rate of recycled aggregate increased to 50~90 %, which is of superior quality than the performance standards of GR F 4007. Therefore, it is thought that they can be used for various construction works with certain physical characteristics applicable to the production of secondary concrete products using recycled aggregates.

Effects of ice-binding protein from Leucosporidium on the cryopreservation of boar sperm

  • The aim of this study was performed to evaluate the effects of ice-binding protein from the arctic yeast Leucosporidium (LeIBP) supplementation on cryopreservation of boar sperm. The collected semen was diluted ($1.5{\times}10^8/ml$) in lactose egg yolk (LEY) and cooled at $5^{\circ}C$ for 3 h. The cooled semen was then diluted ($1{\times}10^8/ml$) in LeIBP containing LEY with 9% glycerol and maintained at $5^{\circ}C$ for 30 min. The semen was divided into six experimental groups (control, 0.001, 0.005, 0.01, 0.05 and 0.1 mg/ml of LeIBP). The straws were kept on above the liquid nitrogen ($LN_2$) vapors for 20 minutes and then plunged into $LN_2$. After thawing, computer-assisted sperm analysis was used for sperm motility and flow cytometry was performed to assess the viability, acrosome integrity (FITC-PSA/PI), ROS (DCF/PI), lipid peroxidation (BODIPY C11/PI) and apoptosis (Annexin V/PI), respectively. No significant responses were observed for sperm motility. However, sperm viability was significantly increased on 0.05 and 0.1 mg/ml of LeIBP groups compared to control (P < 0.05). In addition, acrosome integrity was significantly increases LeIBP groups (P < 0.05) and both ROS and lipid peroxidation level were lower in all LeIBP groups than those of control (P < 0.05). On the other hand, a significant higher apoptosis rate was observed in 0.05 and 0.1 mg/ml of LeIBP groups compared to control (P < 0.05). It can be assumed that a supplementation of LeIBP in boar sperm freezing extender is an effective method to increase the sperm qualities after cryopreservation.

Study on the Usability of Vitrified Mouse Hatched Blastocysts in Embryo Transfer (초자화 동결된 생쥐 완전탈출 배반포기배 이식에 관한 유용성 검토)

  • This study was carried out to confirm whether the in vivo developmental potential of mouse hatched blastocysts (HBs) can be obtained by vitrification method using the cryoprotectant EFS35. The HBs ($\theta$ 130$\mu\textrm{m}$) were cultured in vitro until day 5 from zygotes produced in vivo and were equilibrated in 10% ethylene glycol(EG) for 5 min, and then were exposed or vitrified in EFS35 (35% EG, 18% Ficoll and 0.3M sucrose). After 30 min thawing, re-expanding HBs were transferred into one or both uterine horns of pseudopregnant recipients on day 3 (4~6 embryos /horn). Pregnancy rates of recipients and implantation were assessed by autopsy on day 15 of gestation. The results obtained in these experiments were summarized as follows : After thaw-ing, in vitro survival of HBs was not significantly different between exposed (65.5%) and vitrified(54.5%) group. Also, when the in vivo development potential was examined, total implantation was not different between control (58.5% ) and vitrified (41.0%) group, although the live fetus formation of vitrified group (24.0%) was significantly lower than that of control (58.3%) group (p< 0.05). These results suggested that vitrification freezing method of mouse HBs using EFS35 can be used to make wide the utility of embryo transfer of the more embryos produced in vitro.

Durability and Bioassay of a Sulfur Polymer Surface Protecting Agent for Concrete Structures (콘크리트 구조물용 유황폴리머 표면보호재의 내구성능 및 생물독성)

  • In this study, to examine the use of sulfur polymer as a coating agent for concrete, durability and hazard evaluations were performed. The result of the evaluation indicated that the chemical resistance of the coating agent for concrete was outstanding against acidic, base, and alkaline solutions. The evaluation of the bond strength after an accelerated weathering test depending on the mixing condition indicated that the most outstanding strength characteristic was obtained when silica powder and fly ash were mixed at the same time. The bond strength exceeded 1 MPa in every mixing condition even after the repeated hot and cold treatment of the coating agent specimen for concrete, and the SFS mix proportion showed the highest bond strength. The examination of the accelerated carbonation and chloride ion penetration resistance of the concrete coated with the coating agent indicated that the specimen coated with the coating agent using silica powder as a filler showed the most outstanding durability. When a fish toxicity test was performed to examine the hazard of the use of the functional polymer as a coating agent for concrete, the functional polymer was found to have no effect on the organisms. When the chemical resistance, freezing and thawing resistance, carbonation, and chloride ion penetration resistance of the coating agent were considered, substituting silica powder and fly ash as the fillers of the functional polymer by 20%, respectively, was the optimal level in the range of this study.

Probe of Unfilled Sheath of Prestressed-Concrete Girder Bridge Using Impact-Echo Method (충격탄성파법을 이용한 PC형교의 쉬이스 내부 그라우트 미충전부 탐사)

  • Sheaths are arranged in the web and lower part of prestressed-concrete girder bridges, and prestressing wires and concrete are indirectly bonded together by filling interior parts of the sheaths. However, when the filling is not sufficient, water can penetrate the interior parts of the sheaths and, thereby, prestressing wires can be corroded or transverse cracks would occur around sheaths by the freezing and thawing action of the penetrated water. Therefore, it is an important element in maintenance management of structures to find out the sheaths unfilled with grout early after construction. In this paper, in order not to damage bridge members, the impact-echo method with a new approach in application is used to probe sheaths unfilled with grout for real structures. The location of sheaths is first estimated with reinforcing bar probing instrument of radar type and the measurement locations of sheaths are determined. By sending elastic wave to the side of girders and receiving the response, the location of the unfilled part of a sheath was estimated from the difference between high frequency peak and twice the resonance frequency indicating thickness. To verify the location of void estimated by the impact-echo method, pictures were taken by an industrial fiber scope after drilling a hole.
