Characterization of Antibiotic Resistance of Aeromonas spp. and Pseudomonas spp. Isolated from Domestic Aquatic Animals (국내 수산생물로부터 분리된 Aeromonas spp. 및 Pseudomonas spp.의 항생제 내성에 관한 특성 분석)

  • Aeromonas spp. and Pseudomonas spp. are opportunistic pathogens widely distributed in the aquatic environment. To test the antibiotic susceptibility, the MIC of the 18 antibiotics mainly used in aquaculture were measured. Aeromonas spp. and Pseudomonas spp. straoms had different resistance patterns against most antibiotics. The MIC of tetracycline for four Aeromonas spp. strains (10.5%) was < 0.25 ㎍/mL. However, 0.5-4 ㎍/mL tetracycline inhibited most Pseudomonas spp. strains. The tet resistance performance of 14 genes including tet(B), tet(E), and tet(M) were investigated. Investigating, the tetracycline resistance gene of 38 Aeromonas spp. strains detected tet(A) in 21 strains (55.3%). Two Pseudomonas spp. strains showed high MIC values and no inhibition zone. tet gene analysis detected tet(D) in only one strain (5%).

Efficient Control of Human G-CSF Gene Expression in the Primary Culture Cell using a FIV-Tet-On Vector System (FIV-Tet-On Vector System을 이용한 hG-CSF 유전자의 효율적인 발현 조절)

  • In this study, using FIV-based lentivirus vector system, we tried to express hG-CSF in tetracycline-controllable manner. hG-CSF influences the proliferation, differentiation, and survival of cells in the neutrophil lineage. To enhance stability and translation of hG-CSF transcript, WPRE sequence was also introduced into FIV-Tet-On vector at downstream region of either the hG-CSF gene or the sequence encoding rtTA. Primary culture cells (CEF, chicken embryonic fibroblast; PFF, procine fetal fibroblast) infected with the recombinant FIV were cultured in the medium supplemented with or without doxycycline for 48 hours, and induction efficiency was measured by comparing the hG-CSF gene expression level using quantitative real-time PCR, Western blot and ELISA. Higher hG-CSF expression and tighter expression control were observed from the vector in which the WPRE sequence was placed at downstream of the hG-CSF (in CEF) or rtTA (in PEE) gene. This FIV-Tet-On vector system may be helpful in solving serious physiological disturbance problems which has continuously hampered successful production of transgenic animals and gene therapy.

Distribution of Tetracycline-Resistance Genes detected from isolates of cultured fishes in Gyeonggi-do (경기도 내 양식어류에서 분리한 병원성 세균의 Tetracycline 내성 유전자 분포)

  • Tetracycline (TC) is one of the antibiotics used for treatment of bacterial infection in Korea. Inadequate usage and abuse cause the resistance to antibiotics, like Tetracycline, Erythromycin, and Fluoroquinolone. It can also affect severe economic loss in aquaculture field in Korea. We isolated 101 bacterial samples from diseased fish at aquaculture sites in Gyeonggi-do during 2015~2018. Minimum inhibitory concentration (MIC) method has been used to determine distribution and to identify bacterial isolates resistant to antibiotics including Oxytetracycline (OTC), Ampicillin (AMP), Clindamycin (CLI), Enrofloxacin (ENRO), Gentamycin (GEN). TC resistant isolates were confirmed antibiotic resistant genes by conventional PCR. Bacterial isolates were identified as Aeromonas spp. (43.5%), Pseudomonas spp. (4.0%) and Vibrio spp. (5.0%). It was confirmed that multi-resistant isolates (77.2%) were predominant over single-resistant one (22.8%). TC resistant genes like tet(A), tet(D), tet(E), tet(G), tet(M), and tet(S) were detected and tet(A) was the most prevalent. Aeromonas spp. is a dominant strain in bacterial infections in fishes of aquaculture sites, and further investigation on various antibiotic resistance genes will be needed for clear understanding of aquaculture sites in Gyeonggi-do.

Antimicrobial resistance of Escherichia coli isolated from wild birds in Daegu (대구지역 야생조류에서 분리된 대장균의 항생제 내성 조사)

  • This study was aimed to investigate occurrence and the antimicrobial resistance of Escherichia coli isolates obtained from the feces of wild birds in Daegu. In total, 98 E. coli isolates (17.9%) were obtained from 547 fecal samples of wild birds. The E. coli carried by the birds showed a relatively high rate of antimicrobial resistance to tetracycline (27.6%) and ampicillin (21.4%). Drug resistance of the isolates to the others (penicillins, cephems, carbapenems, aminoglycosides, quinolones, sulfonamides and phenicols) resulted in the rates less than 20%, and all isolates were susceptible to imipenem, ciprofloxacin, cefotetan, and amikacin. Approximately, 45% E. coli among the isolates were resistant to one or more drugs tested. The higher rate of tetracycline resistance led us to determine the prevalence of the tet genes (tetA, tetB, tetC, tetD and tetE) in the tetracycline-resistant E. coli isolates by using PCR. All isolates of the tetracycline-resistant E. coli contained at least one or more of these tet genes examined. The most prevalent one was tetA (59.3%), and followed by tetB (7.4%) when tested with the selected 5 tet genes. Except tetA and tetB, however, the remaining tet genes (tetC, tetD, and tetE) tested were not found in this study. Nine isolates among the tetracycline-resistant E. coli contained the two (tetA and tetB) determinants of tetracycline resistance, simultaneously.

  • Aquaculture practices to ensure greater production, such as high density breeding and excessive feeding, are become stressors that raise the prevalence of diseases. Accordingly, increasingly large volumes of antibiotics are used more frequently each year. Long term use antibiotics can generate resistant bacteria, which interrupt treatments and cause a potential transfer to human bodies. Thus, antibiotic resistance is of importance in public health. Tetracycline (Tc) is one of the typical medicines used in the aquaculture drugs, which has a wide range of application including gram-positive and gram-negative bacteria. In the examination of 153 strains isolated from olive flounder (Paralichthys olivaceus) farms located in Jeju in 2016, it turned out that a total of 84 strains were resistant to Tc or oxytetracycline (OTC). The extent to which the strains are resistant to Tc and OTC was confirmed through MIC test, mostly within the range of 25 to $100{\mu}g/m{\ell}$. Twelve different types of tet genes were detected using single and multiplex PCR in the 84 Tc-resistant strains. The PCR was used to find tet(K), tet(M), tet(O), and tet(S), which are known to exist primarily in gram positive strains. According to the results, - tet(S) is the most dominant gene in 49 strains of Streptococcus parauberis, accounting for 63.2%. And there were two strains that have two different types of resistant genes. The multiplex PCR was used to detect tet(A), tet(B), tet(C), tet(D), tet(E), and tet(G), which are commonly found in gram-negative strains. Each of tet(B), tet(D), and tet(B)&(M) was found in a strain presumed to be Vibrio sp., and only tet(D) was found in 10 Edwardsiella tarda strains.

Induction of cancer cell-specific death via MMP2 promoterdependent Bax expression

  • Controlled gene expression in specific cells is a valuable tool for gene therapy. We attempted to determine whether the lentivirus-mediated Tet-On inducible system could be applied to cancer gene therapy. In order to select the genes that induce cancer cell death, we compared the ability of the known pro-apoptotreic genes, Bax and tBid, and a cell cycle inhibitor, p21cip1/waf1, and determined that Bax was the most effective. For the cancer cell-specific expression of $rtTA2^S$-M2, we tested the matrix metalloproteinase-2 (MMP-2) promoter and determined that it is highly expressed in cancer cell lines, including SNU475 cells. The co-transduction of two lentiviruses that contain sequences for TRE-Bax and $rtTA2^S$-M2, the expression of which is controlled by the MMP-2 promoter, resulted in the specific cell death of SNU475, whereas other cells with low MMP-2 expression did not evidence significant cell death. Our data indicate that the lentivirus-mediated Tet-On system using the cancer-specific promoter is applicable for cancer gene therapy.

Isolation of Tetracycline-resistant Lactic Acid Bacteria from Kimchi (김치에서 tetracycline 내성 유산균의 분리)

  • Tetracycline resistant bacterial strains were isolated from 10 batches of Kimchi among 50 batches collected in Taegu restrict. The MIC of tetracycline ranged between 25 and> 100 ㎖/l. Total genomic DNA preparation from all 10 tetracycline resistant lactic acid bacterial isolates were subjected to PCR amplification with class-specific primers for tet(M) and tet(O). In only one isolate, HJ9, tet(M) was detected. By Southern blotting and hybridization with a tet(M)-specific probe, the tet(M) gene of HJ9 isolate could be localized on a plasmid. The partial nucleotide sequence and deduced amino acid sequence of tet(M) of HJ9 showed 90-99% and 94-100% homology to those of Gram positive bacteria, respectively. With sequencing of 16S rRNA, HJ9 isolate from Kimchi was identified as Lactobacillus sakei. From these results, Kimchi can be considered potential vehicle for the spread of antibiotic-resistant lactic acid bacteria along the food chain to the consumer.

Tetracycline and Erythromycin Resistant Mutants of the Mycoplasma pneumoniae Isolated from Patients with Respiratory Diseases (호흡기질환 환자로부터 분리된 Mycoplasma pneumoniae의 tetracycline과 erythromycin에 대한 저항성 변이)

  • One hundred and twenty three strains of Mycoplasma pneumoniae were isolated from patients with respiratory diseases from February 2002 to April 2005 in Busan, Korea. The MICs of tetracycline and erythromycin up to $90\%$ of the 123 M. pneumoniae isolates tested were $0.5\~1.0$, and $0.5\~512{\mu}/ml$, respectively. Plasmid DNA was not isolated from all of the M. pneumoniae isolates. Out of 323 strains of M. pneumoniae, 57 ($46.3\%$) stains contain tetM gene on their chromosomal DNA, and 60 ($48.8\%$) strains were mutated in domain V of 23S rRNA for erythromycin resistance. Out of 63 strains of M. pneumoniae which were not mutated in domain V of 235 rRNA for erythromycin resistance, 36 ($57.1\%$) strains contained tetM gene, and out of 60 strains of M. pneumoniae which were mutated in domain V of 23S rRNA for erythromycin resistance, 21 ($35.0\%$) strains contained tetM gene. These results suggest that the isolation rate of erythromycin and tetracycline resistant M. pneumoniae is higher than those of other countries, and erythromycin and tetracycline are not first choice drug for M. pneumoniae infection in Korea, and it need confirm by a nationwide surveilance of antimicrobial resistance.

Hypermethylation of TET1 Promoter Is a New Diagnosic Marker for Breast Cancer Metastasis

  • Breast cancer metastasis is a major cause of cancer-related death in women. However, markers for diagnosis of breast cancer metastasis are rare. Here, we reported that TET1, a tumor suppressor gene, was downregulated and hypermethylated in highly metastatic breast cancer cell lines. Moreover, silencing of TET1 in breast cancer cells increased the migration and spreading of breast cancer cells. In breast cancer clinical samples, TET1 expression was reduced in LN metastases compared with primary tissues. Besides, the methylation level of the TET1 promoter was increased significantly in LN metastases. Taken together, these findings indicate that promoter hypermethylation may contribute to the downregulation of TET1 and could be used as a promising marker for diagnosis in patients with breast cancer metastasis.

Organization of Antibiotic Resistance Gene Cluster of Multi-Drug Plasmid in Clinically Isolated Salmonella Enteritidis Strain (식중독환자에서 분리한 Salmonella Enteritidis 다제내성 플라스미드의 내성유전자 집락의 구조해석)

  • Clinically isolated Salmonella Enteritidis strain has a multi drug resistance plasmid, which confers ampicillin, chloramphe-nicol, sulfonamide, streptomycin and tetracycline, named pCAST2. We cloned a 7 kb Sacl fragment of pCAST2 which has sulfonamide, streptomycin and tetracycline resistance genes. The 7 kb SacI fragment showed the organization of sulII-strA-strB-tetR-tetA gene cluster which is different from the other clusters reported previously. In this study, we presented the method to detect this cluster by PCR analysis and showed that this cluster was found in Salmonella strains occurred sporadically at Kyungpook province in 2002.
