Dog spermatozoa were recovered from the caudae epididymides of 23 domestic dogs which were 11 pure breed and 12 mix-breed dogs ranging in age from 0.6 to 3 years. The experimental designs were as follows: 1) the effect of chilling to 0. 10 or 37$^{\circ}C$. 2) the kinetics of chilling injury at 0 or 4$^{\circ}C$, and 3) the effect of sugars at $0^{\circ}C$. Viable spermatozoa were recovered by percoll gradient separation and adjusted to 5${\times}$10$^{7}$ spermatozoa/ml. In experiment 1, spermatozoa were diluted with 0.33 M glucose supplemented with 3% BSA (G-BSA) at 1:2 dilution. Spermatozoa were loaded into straws and exposed at 0, 10 or 37$^{\circ}C$ for 30 min. In experiment 2, spermatozoa were prepared as the experiment 1 and exposed for 0.5, 5, 15, or 30 min at 0 or 4$^{\circ}C$. In experiment 3, spermatozoa were diluted with different sugars (0.33 M galactose, glucose, fructose, mannitol, lactose, sucrose, raffinose) and cooled to $0^{\circ}C$ for 30 min. Sperm membrane integrity, motility and acrosome integrity were assayed after rewarming at 37$^{\circ}C$ for 5 min. Sperm motility and membrane integrity abruptly decreased with decreasing temperature but acrosome integrity gradually decreased (P<0.05). Sperm motility was more sensitive to cold shock than membrane integrity and acrosome integrity. Spermatozoa cooled to $0^{\circ}C$ were more damaged than those at 4$^{\circ}C$. Sperm motility was not different among exposed times at both. 0 and 4$^{\circ}C$. However, membrane integrity of spermatozoa exposed for 30 min at both 0 and 4$^{\circ}C$ was significantly lower (P<0.05). Spermatozoa diluted in 0.33 M fructose or galactose showed lower motility and higher morphological abnormality with coiled tail at $0^{\circ}C$. These sperm characteristics were strongly related. These results indicate that dog epididymal spermatozoa are relatively sensitive to rapid cooling and higher morphological abnormality at $0^{\circ}C$ was shown in spermatozoa diluted in fructose and galactose.

As an aquatic ecotoxicity test method, a bioassay using the inhibition of sporualtion of the green macroalga, Ulva pertusa, has been developed. Optimal test conditions determined for photon irradiance, pH, salinity and temperature were $100\;{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, $7{\sim}9$, $25{\sim}35\;psu$ and $15{\sim}20^{\circ}C$, respectively. The validity of the test endpoint was evaluated by assessing the toxicity of four metals (Cd, Cu, Pb, Zn) and elutriates of sewage or waste sludge collected from 9 different locations. When the metals were assayed, the $EC_{50}$ values indicated the following toxicity rankings: Cu ($0.062\;mg{\cdot}L^{-1}$) > Cd ($0.208\;mg{\cdot}L^{-1}$) > Pb ($0.718\;mg{\cdot}L^{-1}$) > Zn ($0.776\;mg{\cdot}L^{-1}$). When compared with other commonly used bioassays of metal pollution listed on US ECOTOX database, the sporualtion test proved to be the most sensitive. Ulva sporulation was significantly inhibited in all elutriates with the greatest and least effects observed in elutriates of sludge from industrial waste ($EC_{50}=6.78%$) and filtration bed ($EC_{50}=15.0%$), respectively. The results of the Spearman rank correlation analysis for $EC_{50}$ data versus the concentrations of toxicants in the sludge presented a significant correlation between toxicity and four heavy metals(Cd, Cu, Pb, Zn). The method described here is sensitive to toxicants, simple to use, easy to interpret and economical. It is also easy to procure samples and maintain cultures. The present method would therefore probably make a useful assessment of aquatic toxicity of a wide range of toxicants. In addition, the genus Ulva has a wide geographical distribution and species have similar reproductive processes, so the test method would have a potential application worldwide.

Eight comprehensive oceanographic cruises on a squared $30{\times}30\;km$ area have been made to investigate the short and long-term impacts on the water qualities due to the sand mining operations at Exclusive Economic Zone (EEZ) in the central Yellow Sea from 2004 to 2007. The area was categorized to 'Sand Mining Zone', 'Potentially Affected Zone', and 'Reference Zone'. The investigation covered suspended solids, nutrients (nitrate, nitrite, ammonium, phosphate), and chlorophyll-a in seawater and several parameters such as water temperature, salinity, pH, and ORP. Additionally, several intensive water collections were made to trace the suspended solids and other parameters along the turbid water by sand mining activities. The comprehensive investigation showed that suspended solids, nitrate, chlorophyll-a and ORP be sensitively responding parameters of seawater by sand mining operations. The intensive collection of seawater near the sand mining operation revealed that each parameter show different distribution pattern: suspended solids showed an oval-shaped distribution of the north-south direction of 8 km wide and the east-west direction of 5 km wide at the surface and bottom layers. On the other hand, phosphate showed so narrow distribution not to traceable. Also ammonium showed a limited distribution, but its boundary was connected to the high nitrate and chlorophyll-a concentrations with high N/P ratios. From the last 4 years of the comprehensive and intensive investigations, we found that suspended solids, ammonium, nitrate, chlorophyll-a, and ORP revealed the sensitive parameters of water quality for tracing the sand mining operations in seawater. Especially suspended solids and ORP would be useful tracers for monitoring the water qualities of remote area like EEZ in Yellow Sea.
The loss of coolant accident based on a double-ended cold leg break is analyzed with the discharge coefficient (Ca) of 0.4. This analysis covers the whole transient period from the start of depressurization to the complete refilling of the core by using RELAP4/MOD6-EM and RELAP4/ MOD6-HOT CHANNEL for the system thermal-hydraulics and the fuel performance during the blowdown phase respectively, and RELAP4/MOD6-FLOOD and TOODEE2 during the reflood phase. A simple analytical method has been developed to account for the lower plenum filling by approximating steam-water countercurrent flows and superheated wall effects at the downcomer during the refill period. Based on the informations. at the time of EOB (end-of-bypass), the refill duration time and the initial reflooding temperature were estimated and compared with the results from the RELAP4/MOD6, resulting in a good agreement. In addition, some parametric studies on the EOB were performed. The form loss coefficient between upper head and upper downcomer was found to be sensitive to the occurrence of the spurious EOB. Appropriate form loss coefficients should be taken into account to avoid the flow oscillations at the downcomer. The analyses with the six and three volume core nodalizations, respectively, show much similar trends in the system thermal-hydraulic performance, but the former case is recommended to obtain good results.
Atomistic origins of carbon solubility into silicates are essential to understand the effect of carbon on the properties of silicates and evolution of the Earth system through igneous and volcanic processes. Here, we investigate the atomic structure and NMR properties of dissolved carbon in enstatite using Raman spectroscopy and quantum chemical calculations. Raman spectrum for enstatite synthesized with 2.4. wt% of amorphous carbon at 1.5 GPa and $1,400^{\circ}C$ shows vibrational modes of enstatite, but does not show any vibrational modes of $CO_2$ or ${CO_3}^{2-}$. The result indicates low solubility of carbon into enstatite at a given pressure and temperature conditions. Because $^{13}C$ NMR chemical shift is sensitive to local atomic structure around carbon and we calculated $^{13}C$ NMR chemical shielding tensors for C substituted enstatite cluster as well as molecular $CO_2$ using quantum chemical calculations to give insights into $^{13}C$ NMR chemical shifts of carbon in enstatite. The result shows that $^{13}C$ NMR chemical shift of $CO_2$ is 125 ppm, consistent with previous studies. Calculated $^{13}C$ NMR chemical shift of C is ~254 ppm. The current calculation will alllow us to assign potential $^{13}C$ NMR spectra for the enstatite dissolved with carbon and thus may be useful in exploring the atomic environment of carbon.
Background: Oral squamous cell carcinoma (OSCC) is the sixth most common malignancy worldwide. Cancer development and progression require inactivation of tumor suppressor genes and activation of proto-oncogenes. The well recognized mechanism of action demonstrated for chemotherapeutic agents is induction of apoptosis via reactivation of p53. In this context, we evaluate the efficacy of IV and oral routes of our novel PH and temperature sensitive doxorubicin-methotrexate-loaded nanoparticles (DOX-MTX NP) in affecting p53 profile in an OSCC rat model. Methods: In this study, 120 male rats were divided into 8 groups of 15 animals each. The new formulated DOX-MTX NP and free doxorubicin were IV and orally given to rats with 4-nitroquinoline-1-oxide induced OSCC. Results: Results showed that both DOX and DOX-MTX-NP caused significant increase in mRNA levels of P53 compared to the untreated group (p<0.000). With both DOX and DOX-MTX NP, the IV mode was more effective than the oral (gavage) route (p<0.000). Surprisingly, in oral mode, p53 mRNA was not affected in DOX treated groups (p>0.05), Nonetheless, both IV and oral administration of MTX-DOX NP showed superior activity (~3 fold) over free DOX in reactivation of p53 in OSCC (p<0.000). The effectiveness of oral route in group treated with nanodrug accounts for the enhanced bioavailability of nanoparticulated DOX-MTX compared to free DOX. Moreover, in treated groups, tumor stage was markedly related to the amount of p53 mRNA (p<0.05). Conclusion: Both oral and IV application of our novel nanodrug possesses superior activity over free DOX-in up-regulation of p53 in a OSCC model and this increase in p53 level associated with less aggressive tumors in our study. Although, impressive results obtained with IV form of nanodrug (-21 fold increase in p53 mRNA level) but both forms of nanodrug are effective in OSCC, with less toxicity normal cells.

To minimize the damage caused by summer season such as high temperature and red tide, this study was conducted to investigate the effect of feeding and starvation on the survival rate, growth, growth restoration and physiological response of red seabream Pagrus major which were reared in marine net cage ($6m{\times}6m{\times}6m$). The feeding group was fed throughout the experiment for 10 weeks, whereas starvation group was not fed for 6 weeks and subsequent refeeding for 4 weeks. Survival rates of feeding and starvation groups were 94.4% and 98.1% respectively. The growth rate of starvation group was significantly lower than feeding group during starvation period, but recovered rapidly after feeding. The nutritional status such as albumin, total protein, total cholesterol, triglyceride also showed similar tendency to the growth data. Hematocrit and hemoglobin levels were higher in the feeding group than the starvation group during starvation period. Aspartate aminotransferase, glucose and total cholesterol were rapidly increased at the same time (in 5 week) in feeding group, whereas starvation group did not significant difference during starvation period. It was shown that the feeding group is more sensitive to stress than the starvation group. Thus, starvation can be applied as a rearing management plan of net cage fish farm in summer season.
The objective of this study was to evaluate the performances of the ASBR under critical conditions of solid-liquid separation, caused by extremely high solids concentration, for wider application of the ASBR to various wastes. The ASBRs and completely-mixed daily-fed control runs were operated using a municipal mixed sludge at 35$^{\circ}C$ and 55$^{\circ}C$. Conversion of completely-mixed daily-fed reactor to sequencing batch mode and changes in HRT of all ASBRs were easily achieved without adverse effect, regardless of digestion temperature. Solids accumulation was remarkable in the ASBRs, and directly affected by settleable solids concentration of the feed sludge. Noticeable difference in solids-liquid separation was that flotation thickening occurred in the mesophilic ASBRs, while gravity thickening was a predominant solid-liquid separation process in the thermophilic ASBRS. Solids profiles at the end of thickening step dramatically changed at solid-liquid interface, and slight difference in solids concentrations was observed within thickened sludge bed. Organics removals based on subnatant or supernatant after thickening always exceeded 80% in all reactors. Thickened sludge volume and gas production of the ASBRs affected mutually. Gas production increased as thickened sludge accumulated, and continuous gas evolution during thickening could cause thickened sludge to expand or resuspend. Thickened sludge volume exceeding a predetermined withdrawal level resulted in loss of organic solids as well as biomass during withdrawal step, leading to decrease in gas production ind SRT. Such an adverse mutual effect was significant in gravity thickening, while it was not sensitive in flotation thickening. Changes in organic loading had no significant effect on organic removals and gas production after build-up of solids in the ASBRs.

The concrete wall is the most useful of retaining structure which can obtain the engineering stability, but has problems that is not friendly with nature environment in a fine view, such as poor rear drainage, and shrinkage crack by temperature difference, etc. Because of this problems, the research for a segmental crib retaining wall has been performed. A segmental crib retaining wall is quickly and easily erected because is possible to be erected as the individual members, and is not sensitive to differential settlement and earthquakes. Also, it shows effective drainage and has a friendly advantage with nature environment because of being able to be planted with vines and shrubs in retaining walls The design of crib retaining walls has traditionally been based on classical soil mechanics theories. These theories, originally derived by Rankine(1857) and Coulomb(1776), assume that the wall acts as a rigid body. This assumption results in failure being predicted by either monolithic overturning or base sliding mechanisms. However, the wall consists of individual members which have been created a three dimensional grid. This grid confines an fill mass which becomes part of the wall. The filled wall resists the earth pressure with the same mechanism of classical gravity walls. Because of the flexibility of the individual segment, it allows relative movement between the individual members within the wall. The three dimensional flexible grid leads to stress redistribution when the wall is subjected to external or fill loads. Due to the flexibility and the stress redistribution, the failure of segmental crib wall consists of not only overturing and base sliding but the local deformation and the failure between the segmental members. It has been researched in the field that due to this flexibility and load redistribution, serviceability failure of segmental crib walls is unlikely to be due to overturning or base sliding. Therefore, in this study, the relative displacement appearance of retaining wall due to variation of inclination is measured to examine this behavior characteristics. Also, the behavior characteristics of retaining walls by surcharge load, and location of acting point of retaining wall rear, and the displacement characteristics and deflections are estimated about the existence and nonexistence of Rear Stretcher performing an role in transmitting earth pressure of Header and Stretcher organizing retaining walls. For the effective ethanol fermentation, the high concentration of sugar as the substrate of microbial fermentation is required. The most important reason in the inefficient hydrolysis; the easy deactivation of enzyme by temperature or shear stress and the severe inhibition effects of its products. In our work, we comprehended the kinetic characteristics of cellulose and ${\beta}$-glucosidase in the progress of hydrolysis, and observed the potential inhibitory effects of the hydrolyzed products and the deactivation of enzymes. We also tried to present the kinetic model of enzymatic hydrolysis of cellulose, which is applicable to process at the high concentration of sugar. Cellulase and ,${\beta}$-glucosidase exhibit diverse kinetic behaviors. At a level of only 5g/$\ell$ of glucose, the ${\beta}$-glucosidase activity was reduced by more than 70%. This result means that ${\beta}$-glucosldase was the most severely inhibited by glucose. Also at l0g/$\ell$ of cellobiose, the cellulose lost approximately 70% of its activity. ${\beta}$-glucosldase was more sensitive to deactivation than cellulose by about 1.6 times. The comprehensive kinetic model in the range of confidence was obtained and the agreement between the model prediction and the experimental data was reasonably good, testifying to the validity of the model equations used and the associated parameters.
