Biological Constraints in Algal Biotechnology

  • In the past decade, considerable progress has been made in developing the appropriate biotechnology for microalgal mass cultivation aimed at establishing a new agro-industry. This review points out the main biological constraints affecting algal biotechnology outdoors and the requirements for making this biotechnology economically viable. One of them is the availability of a wide variety of algal species and improved strains that favorably respond to varying environmental conditions existing outdoors. It is thus just a matter of time and effort before a new methodology like genetic engineering can and will be applied in this field as well. The study of stress physiology and adaptation of microalgae has also an important application in further development of the biotechnology for mass culturing of microalgae. In outdoor cultures, cells are exposed to severe changes in light and temperature much faster than the time scale re-quired for the cells to acclimate. A better understanding of those parameters and the ability to rapidly monitor those conditions will provide the growers with a better knowledge on how to optimize growth and productivity. Induction of accumulation of high value products is associated with stress conditions. Understanding the physiological response may help in providing a better production system for the desired product and, at a later stage, give an insight of the potential for genetic modification of desired strains. The potential use of microalgae as part of a biological system for bioremediation/detoxification and wastewater treatment is also associated with growing the cells under stress conditions. Important developments in monitoring and feedback control of the culture behavior through application of on-line chlorophyll fluorescence technique are in progress. Understanding the process associated with those unique environmental conditions may help in choosing the right culture conditions as well as selecting strains in order to improve the efficiency of the biological process.

  • Recently, owing to global warming, average summer temperatures are increasing and the number of hot days is increasing is increasing, which leads to an increase in heat stroke. In particular, outdoor workers directly exposed to the heat are at higher risk of heat stroke; therefore, preventing heat-related illnesses and managing safety have become important. Although various wearable devices have been developed to prevent heat stroke for outdoor workers, applying various sensors to the safety helmets that workers must wear is an excellent alternative. In this study, we developed a smart helmet that measures various vital signs of the wearer such as body temperature, heart rate, and sweat rate; external environmental signals such as temperature and humidity; and movement signals of the wearer such as roll and pitch angles. The smart helmet can acquire the various data by connecting with a smartphone application. Environmental data can check the status of heat wave advisory, and the individual vital signs can monitor the health of workers. In addition, we developed an algorithm that classifies the risk of heat-related illness as normal and abnormal by inputting a set of vital signs of the wearer using a support vector machine technique, which is a machine learning technique that allows for rapid binary classification with high reliability. Furthermore, the classified results suggest that the safety manager can supervise the prevention of heat stroke by receiving feedback from the control system.

사분파장 위상 간섭계 폴리머 광집적회로 기반 광전류센서의 온도 안정성 향상 연구 (Improvement of Thermal Stability of Optical Current Sensors Based on Polymeric Optical Integrated Circuits for Quadrature Phase Interferometry)

  • 광섬유가 지니는 패러데이 효과를 이용하여 전류의 세기를 측정하는 광전류 센서 소자를 설계 및 제작하였다. 센서 소자의 동작 안정성을 향상시키기 위하여 편광회전 반사 간섭계와 사분 파장 위상 간섭계 구조를 도입하였다. 이 복잡한 구조를 구성하는 다양한 광소자들은 하나의 폴리머 광집적회로로 구성하여 작은 크기로 제작되었다. 본 구조를 이용하면 외부에서 별도의 바이어스 피드백 제어가 필요 없는 상태에서 전류를 측정하는 센싱 동작을 수행할 수 있다. 또한 온도변화나 외부진동으로 인한 광센서 특성 변화를 제거하여 안정적인 특성을 유지하는 광전류센서를 구현할 수 있다. 그러나 패러데이 효과를 결정짓는 베르데상수는 온도에 따라 미소한 값의 변화를 가지고 있다. 본 연구에서는 이 변화로 인한 광전류센서의 온도의존성을 극복하기 위한 연구를 수행하였다. 편광회전 반사 간섭계의 부품인 광섬유 사분 파장판의 길이를 최적값으로부터 벗어나는 상태로 맞추어 줌으로써 베르데 상수의 온도의존성에 의해 나타나는 광전류센서의 스케일 팩터 변화를 보상해줄 수 있었다. 온도변화를 보상한 광전류센서는 주변 온도를 상온에서 85℃로 올리는 동안, 센서 측정 신호의 온도 의존성이 0.2% 이내로 유지되는 것을 확인했다.

온도 증가와 염도 감소에 따른 갯벌토양에서 메탄발생량의 변화 (Changes in Methane Production in Coastal Mud Flat under Different Temperature and Salinity)

  • 전 지구적 기후변화로 인하여 습지에서 일어나는 다양한 종류의 생지화학적인 순환에 큰 변화가 관측되고 있다. 특히, 갯벌의 경우 온도변화에 의한 직접적인 환경변화 뿐 아니라 해수면의 상승과 강수량의 변화라는 요인에 의해 간접적인 영향도 받을 것으로 예측된다. 본 논문에서는 온도와 해수의 염도 변화에 따라 갯벌에서 발생하는 메탄양의 변화를 실험실 규모의 실험을 통하여 규명하고자 했다. 시료는 겨울철 강화도 동검도 갯벌에서 채취하여 2 종류의 실험을 수행하였다. 첫 번째 실험에서는 0-5 cm, 5-10 cm 깊이의 토양을 자연 상태의 염도와 50 % 희석한 염도 조건 하에서 20 일간 배양하며 2일 간격으로 메탄 발생량을 측정하였다. 두 번째 실험에서는 5-10 cm 깊이의 토양을 $5^{\circ}C$, $15^{\circ}C$ 에서 자연 상태의 염도와 50 % 희석한 염도 조건 하에서 31일간 배양하며 발생하는 메탄의 양을 하루 간격으로 측정하였다. 첫 번째 실험의 결과 5-10 cm 깊이의 토양에서 메탄이 더 많이 발생하며 염도의 영향은 배양 후반부로 가면서 크게 나타남을 알 수 있었다. 두 번째 실험 결과에서는 $15^{\circ}C$에서 $5^{\circ}C$에서 보다 더 많은 메탄이 발생하였고, 50 % 희석한 염도 조건에서 자연 상태의 염도 조건에서 보다 많은 메탄이 발생하는 것을 관찰할 수 있었다. 이러한 결과는 갯벌에서의 메탄 생성율은 온도 상승과 해수의 화학적 특성의 변화에 따라 많은 영향을 받음을 보여준다. 또한 전 지구적 기후변화로 야기될 온도나 해수 염도의 변화가 메탄의 발생에 영향을 미쳐, 생물학적 되먹임 기작에도 변화가 일어날 것으로 사료된다.

전자식 구치료 시스템 개발 연구 (Development of Programmable and Rechargeable Moxibustion Device)

  • Objectives : This study was to develop programmable and rechargeable electronic moxibustion device as an interdisciplinary research focusing on traditional Korean medicine. Methods : This device deployed several advances on safety and conveniences with the advantage of programmable heat stimulation, heating material and recharging method. The gradient of heat stimulation was programmed with the reference of previous studies and own measurements. The heat curve reached its efficacious stimulus temperature after three minutes, and maintained until 13 minutes with $42^{\circ}C$. We also incorporated feedback from field hospital experiences with the help of hospital staffs. Results : This device can prevent damages from skin burn and fire accidents, and control the smoke, smell and residue of moxa itself. Conclusions : This study will contribute for the development and improvement of efficacious and safe treatment methods for the traditional Korean medicine.

소아환자의 Vancomycin에 대한 약물사용 평가 (II) -제한항균제로 사용규제 강화 후의 효과 (Drug Use Evaluation of Vancomycin in Pediatric Patients (II) -The effect of Approval for Vancomycin Use)

  • The 'Pharmacy and Therapeutic Committee' decided to restrict the use of vancomycin which was categorized into restricted antimicrobials, among general, reserved and restricted antimicrobials. The committee also established prescribing guidelines of vancomycin in Seoul National University Hospital, May, 1991. Especially, the restricted antimicrobials should be used after approval by infectious disease specialist physician. A retrospective drug use evaluation (DUE) on vancomycin has been conducted to compare with the previous vancomycin DUE study in 1990. 'Criteria for DUE on vancomycin' was modified from Am J Hosp Pharm. Total 65 charts of patients were retrospectively reviewed from July 1991 to June 1992 in Seoul National University Children's Hospital. The justification of use was improved from 56% to 75% comparing with the previous study. In analyzing process indicators, several criteria including body temperature monitoring, WBC monitoring and use of concomitant antibiotics were well documented, but serum creatinine monitoring, culture and sensitivity test and level monitoring were infrequently performed, while the accepted level has been improved. Accepted level for appropriate initial dosage and duration of therapy were decreased. In outcome analysis, blood culture after discontinuing the drug was relatively well documented compared with the previous study. As the results, the approval vancomycin use was shown to be effective and rational in antibiotic therapy. And it is suggested that the above findings should be communicated to the medical staff, and a active intervention, such as feedback control, also be necessary for rational drug use.

  • 풍력 및 태양광 발전시스템은 화석에너지의 고갈에 대한 대체에너지로 각광을 받고 있으며, 환경오염을 발생하지 않고 무한정으로 사용할 수 있으나 풍속과 태양광의 변화에 따른 안정성의 문제가 발생한다. 풍력발전시스템의 경우 태풍과 급속한 풍속의 변화에 의해 시스템의 안정성 문제가 발생한다. 본 논문에서는 풍속을 이용한 피드백 제어를 기초로 하는 자동강풍제어기를 포함하는 풍력 발전시스템을 구성하였으며, 이를 다양한 조건의 실험을 통하여 입증하였다. 태양전지 어레이의 최대 출력을 위한 MPPT제어와 고르지 못한 DC 전압을 정류하기 위하여 buck-boost컨버터를 사용하였고, 실험을 동하여 시스템 출력전류 리플 저감의 결과를 확인하였다.

온라인 위생교육프로그램의 지속적 제공이 어린이급식소 위생관리 수행도에 미치는 영향 (The Effect of Continuous Provision of On-line Hygiene Education Program on Hygiene Management Performance of Children's Cafeterias)

  • The purpose of this study was to investigate the effect of developing and continuously providing on-line hygiene education programs on the improvement of hygiene conditions in children's cafeterias. As a result of the 2020 sanitation and safety checklist analysis, 6 items (personal hygiene, separate use, clean ventilation, temperature control of refrigerators, country of origin, food distribution) were derived and on-line hygiene education programs for each of 6 items were produced. ① Customized educational materials and self-inspection checklists were provided to 208 children's cafeterias. After that, educational videos were provided through Kakao Talk twice a week for 6 months, and they were made available for viewing at any time through YouTube upload, ③ Kakao Talk Through this, a quiz related to the educational video was conducted to give feedback for interaction with the cook. As a result of analyzing the total hygiene and safety checklist score of all registered facility catering centers by visit order, in 2020 it was 82.8 points/100 points, but in 2021, it was 84.2 points (1st round), 89.3 points (2nd round), 91.4 points (3 points) The score improved significantly (p<0.001) as the on-line hygiene education program continued. As a result, significant (p<0.001) changes were observed in the items of 'Knife, chopping board' and 'Sanitation clothes, sanitary hat, sanitary shoes, apron, and sanitary gloves', confirming a clear improvement effect. Therefore, it is considered that the on-line hygiene education program will play a positive role in showing a lasting effect on improving hygiene management in children's cafeterias.

Molecular Analysis of Growth Factor and Clock Gene Expression in the Livers of Rats with Streptozotocin-Induced Diabetes

  • Many biological systems are regulated by an intricate set of feedback loops that oscillate with a circadian rhythm of roughly 24 h. This circadian clock mediates an increase in body temperature, heart rate, blood pressure, and cortisol secretion early in the day. Recent studies have shown changes in the amplitude of the circadian clock in the hearts and livers of streptozotocin (STZ)-treated rats. It is therefore important to examine the relationships between circadian clock genes and growth factors and their effects on diabetic phenomena in animal models as well as in human patients. In this study, we sought to determine whether diurnal variation in organ development and the regulation of metabolism, including growth and development during the juvenile period in rats, exists as a mechanism for anticipating and responding to the environment. Also, we examined the relationship between changes in growth factor expression in the liver and clock-controlled protein synthesis and turnover, which are important in cellular growth. Specifically, we assessed the expression patterns of several clock genes, including Per1, Per2, Clock, Bmal1, Cry1 and Cry2 and growth factors such as insulin-like growth factor (IGF)-1 and -2 and transforming growth factor (TGF)-${\beta}1$ in rats with STZ-induced diabetes. Growth factor and clock gene expression in the liver at 1 week post-induction was clearly increased compared to the level in control rats. In contrast, the expression patterns of the genes were similar to those observed after 5 weeks in the STZ-treated rats. The increase in gene expression is likely a compensatory change in response to the obstruction of insulin function during the initial phase of induction. However, as the period of induction was extended, the expression of the compensatory genes decreased to the control level. This is likely the result of decreased insulin secretion due to the destruction of beta cells in the pancreas by STZ.
