  • 1. The hair cells on adaxial surface turned out to be penetration way of Gleosporium Thea sinensis in tea plant. 2. The most parts of Gleosporium Thea sinensis generated in tea plant were young leaves with first-fifth leaves from tip of shoot. 3. The proper temperature for spore germination on leaf is $25-27^{\circ}C$, soaked by water for 12 hours. 4. For prevention of Gleosporium Thea sinensis the drug-spay in tea plant would be the most effective when sprayed at this temperature range, and the control of Gleosporium Thea sinensis would be possible through selection of tea plant with few hairs. 5. The treatment of bordeaux mixture to prevent growth of conidiospore was 48.3% more effective than in control plot which were not sprayed. 6. The effect of sprayed bordeaux mixture decreased to about 28.5% after one week of spray.

Identification and Antibiotic Susceptibility Test of Bacteria from In Vitro Cultures of Pinellia ternata and Tea Plant (반하 및 차나무의 기내배양시 발생하는 세균의 동정 및 항생제 감수성 검정)

  • Contamination of bacterial infection is one of serious problems in in vitro culture system of root crops. From the contaminated tubes over 140 of petiole cultures of Pinellia ternata, a medicinal plant, 4 genera 8species 48 strains of bacteria, including Aeromonas and Pseudomonas, were isolated and identified and another 8 strains were not fully identified. Most of them were motile Gram positive bacteria as in common in early stage of in vitro cultures. Six strains of bacteria, 5 of Gram negative, including Enterobacter, and 1 of Gram positive, were identified from the embryonic axes cultures of tea plant. From the susceptibility test to pre-screened 5 antibiotics, all of the bacteria except for 2 species of Pseudomonas were susceptible to cefotaxime 60∼100mg/L. While 60mg/L erythromycin only was effective to Pseudomonas. Combination of erythromycin 20mg/L and cefotaxime 60mg/L totally suppressed the growth of all bacterial strains tested. Susceptibility test of bacteria from tea embryonic axes cultures showed similar results. Combination of erythromycin 35mg/L and cefotaxime 60mg/L was effective to 15 bacterial strains and partially effective to 1 unidentified.

Comparison of Major Infestations between Conventional Tea Growing and Organic Tea Growing at Sulloc Tea Plantation in Jeju Island

  • The occurrence of major pest infestation was compared between conventional tea plantation and organic tea plantation at Sulloc tea garden in Dosun-dong, Seogwipo-si, Jeju-do from 2002 to 2009. Tetranychus kanzawai was observed a lot in the second year, but it waned from late June. There was not much difference between conventional farming and organic farming in terms of pest density. Empoasca onukii was infested in the second year of organic farming compared with conventional farming, which highlighted the fact that second year of organic farming requires a special care. Scirotothrips dorsalis was highly dense in the second and third year of conventional farming, but its occurrence was lowered when the farming technique was shifted to organic farming. The number of Homona magnanima peaked 4 times each year. In 2008, the first year of organic farming, saw high occurrence of 771.2 per trap per year. In 2009, the second year, the population per trap dropped to 80, showing a great variance depending on year. The occurrence of Caloptilia theivora peaked 5 times annually. In 2008, the first year of organic farming, an average of 2,779 pests per trap was found, and in the following year, 4,143 pests were observed. It showed that density rose in organic growing period.

Influence of Mineral Salts on Shoot Growth and Metabolite Biosynthesis in Tea Tree (Camellia sinensis L.)

  • Effects of mineral salts (N, P, K, $Ca^{2+}$, $Mg^{2+}$, and $Fe^{3+}$) on the shoot growth and metabolite production of tea tree were studied using in vitro culture techniques. Among mineral s alts, ${H_2PO_4}^-$ was the most important for enhanced growth rate of tea tree, while $Mg^{2+}$ and $Ca^{2+}$ did not affect plant growth. Removal of ${NH_4}^+$ and $NO_3$ from the culture medium enhanced shoot multiplication compared to other treatments. Metabolite production was variable depending on mineral types and concentration. Removal of $Ca^{2+}$ decreased the production of caffeine; however, other treatments did not influence its production. $Ca^{2+}$, ${NH_4}^+$ and $Fe^{3+}$ were important factors for catechin production in tea tree. These results can be used as the basis for development of technical soil controls suitable for tea tree cultivation in the future.

  • Comparing and analysing the productivity and the main ingredients of persimmon leaves by cultivars were conducted to improve the quality and productivity of the per-simmon leaf tea. As a result of the survey, Dungsi was decidedly good in growth co-ndition of survival rate, stem diameter and the number of shoots etc. Also , DUngsi had the highest leaf productivity of 567 leaves and the leaf weight of 1.98 kg per one plant after three years of planting . On accounts of productivity, ingredient analysis and panel test etc. we collectively through that Dungsi was the best cutltivar for the persimmon leaf tea.

Characterization of dihydroflavonol 4-reductase cDNA in tea [Camellia sinensis (L.) O. Kuntze]

  • Tea leaves are major source of catechins—antioxidant flavonoids. Dihydroflavonol 4-reductase (DFR, EC 1.1.1.219) is one of the important enzymes that catalyzes the reduction of dihydroflavonols to leucoanthocyanins, a key ''late'' step in the biosynthesis of catechins. This manuscript reports characterization of DFR from tea (CsDFR) that comprised 1,413 bp full-length cDNA with ORF of 1,044 bp (115-1,158) and encoding a protein of 347 amino acids. Sequence comparison of CsDFR with earlier reported DFR sequences in a database indicated conservation of 69-87% among amino acid residues. In silico analysis revealed CsDFR to be a membrane-localized protein with a domain (between 16 and 218 amino acids) resembling the NAD-dependent epimerase/dehydratase family. The theoretical molecular weight and isoelectric point of the deduced amino sequence of CsDFR were 38.67 kDa and 6.22, respectively. Upon expression of CsDFR in E. coli, recombinant protein was found to be functional and showed specific activity of 42.85 nmol $min^{-1}$ mg $protein^{-1}$. Expression of CsDFR was maximum in younger rather than older leaves. Expression was down-regulated in response to drought stress and abscisic acid, unaffected by gibberellic acid treatment, but up-regulated in response to wounding, with concomitant modulation of catechins content. This is the first report of functionality of recombinant CsDFR and its expression in tea.

Blister blight of Tea (Thea sinensis) Caused by Exobasidium vexans in Korea (Exobasidium vexans에 의한 차나무 떡병)

  • Blister blight was occurred on tea (Thea sinensis) in a farmer's tea garden located in Sicheon-myon, Sancheong-gun, Gyeongnam province, Korea. The typical symptoms of the disease were appeared on the leaves and bud. The early symptom were started with small yellowish spots then infected area became upheave and covered with white blister which the hymenium de.veloped from under neath of epidermis. Generally, the infected leaves and buds were appeared like rice cake. Colonies formed on PDA were developed white, later turned milk white. Basidia were 2 sterigma, and 30${\sim}$53 ${\times}$ 3.1${\sim}$5.8 ${\mu}m$ in size. Basidiospores were ellipsoid curved, coloriess, 1-celled and 11${\sim}$14.7 ${\times}$ 4${\sim}$5.4 ${\mu}m$ in size. The optimum temperature for growth of the fungus was about $25^{\circ}C$ on PDA and PSA. This is the first report on the blister blight of tea caused by Exobasidium vexans in Korea.

  • 특허제도는 발명자에게 발명의 공개에 대한 대가로 일정기간 독점권을 인정하여 기술개발에 투자한 자금을 회수할 수 있는 기회를 제공하고 공개된 발명은 새로운 기술개발을 위하여 누구나 이용할 수 있도록 하여 기술발전을 촉진할 뿐만아니라 이미 개발된 발명에 대한 중복투자, 연구를 하는 낭비를 방지하기 위한 목적을 가진다. 본 조사는 2020년 12월 31일까지의 정부 특허정보넷인 키프리스(www.kipris.or.kr)에서 꽃차 관련 실용신안 특허 내용을 검색하여 정리하였으며 그 결과는 다음과 같았다. 1. 1983년 엘다베리꽃차 제조방법에 대한 것을 시작으로 최근 2020년 12월 31일까지 총 29개가 출원되었다. 2. 출원된 실용신안 특허 총 29개 중에서 소멸 2개(엘다베리/1983, 연꽃/2009), 취하 3개(무궁화/1992, 백화차/2005, 꽃차/2014), 거절 6개(매화/2001, 장미녹차/2007, 혼합꽃녹차/2007, 동결꽃차/2008, 소금꽃차/2015, 꽃차누룽지/2017), 공개 6개(꽃차/2014, 소금꽃차/2015, 꽃차추출물/2017, 기능성꽃차/2018, 꽃차추출물/2018, 금잔화꽃차/2019), 포기 2개(개복숭아 꽃차/2015, 동백꽃차/2017), 등록(녹차꽃차/2006, 사과꽃차/2008, 목련꽃차/2012. 목련꽃차/2014, 칡꽃차/2015, 장미꽃차/2017, 모란꽃차/2017, 꽃차제조방법/2018, 꽃차로스팅장치/2018, 꽃음료/2020)은 10개였다. 3. 출원된 실용신안 특허 총 29개 중에서 개인이 출원한 것이 19개, 회사(아모레퍼시픽, 휴먼코스메틱, 화이통협동조합, (주)꽃다비, 주식회사 플러드)나 기관(한국식품연구원, 고창군), 대학(전남대학교, 동의대학교 2개)이 출원한 것이 10개였다.

The DPPH and Nitrite Scavenging Activity of Flower Tea by Year of Manufacture in Magnolia denudata (백목련 꽃차 제조연도에 따른 유리산소 소거능(DPPH)와 아질산염 소거능 특성)

  • 색과 향과 맛 및 기능성까지 뛰어난 대표적인 꽃차가 백목련이나 기능성에 관한 연구는 아직 매우 미미하다. 본 연구에서는 유리산소 소거능(DPPH)와 아질산염 소거능(Nitrite Scavenging Activity)이 백목련 꽃차가 제조된 2017년부터 2021년까지의 연도에 따라서 차이가 있는 지를 알아보고자 수행하였으며 그 결과는 다음과 같았다. 1. DPPH는 측정농도(0.5g/100ml=500ppm)에서 제조연도가 2017년 것은 87.8±0.0, 2018년 것은 87.5±0.1, 2019년 것은 85.6±0.6, 2020년 것은 88.5±0.2, 2021년 것은 86.1±0.1으로 2020년에 제조한 것이 가장 높은 함량을 보였다. RC50이란 소거능력이 50%를 환원시킬 수 있는 환원율을 의미하며 낮을수록 적은 양으로 환원이 가능하여 함량이 높다고 할 수 있으며 RC50값은 2017년이 140,2, 2018년이 165.0, 2019년이 178.6, 2020년이 112.2, 2021년이 151.4으로 2020년이 가장 뛰어난 활성을 보였다. 2. 아질산염 소거능은 측정농도(0.5g/100ml=500ppm)에서 제조연도가 2017년 것은 31.8±1.9, 2018년 것은 21.1±4.6, 2019년 것은 18.2±1.9, 2020년 것은 31.0±1.4, 2021년 것은 19.3±5.7의 수치를 보여 2017년과 2020년이 높은 활성을 보였다.

Quantitative Analysis of the Flavonoid Content in the Leaves of Boehmeria nivea and Related Commercial Products

  • Content analysis of flavonoids (epicatechin, epicatechin gallate, and rutin) present in the leaves of Boehmeria nivea (originating from Geumsan-myeon, Biin-myeon, Hansan-myeon, and Baeksu-eup) and their commercial products (ramie tteok, ramie songpyeon, ramie bory-tteok, and ramie tea) was conducted by HPLC. The content of epicatechin, epicatechin gallate, and rutin was highest in the leaves of B. nivea from Geumsan-myeon (0.138 mg/g), Baeksu-eup (1.654 mg/g) and Geumsan-myeon (12.205 mg/g), respectively. With respect to commercial products, the content of epicatechin and epicatechin gallate was highest in ramie tea, with concentrations of 1.879 and 1.090 mg/g, respectively. Given these flavonoid concentrations, B. nivea leaf extracts have the potential to be used as additives in natural medicinal products, health supplements, and beverages.
