We have studied the mechanism of action of TCDD on CYP1A1 promoter activity in both Hepa I and MCF-7 cells using transient transfection system with plAl-Luc reporter gene. When HDAC inhibitors, such as trichostatin A, HC toxin and a novel HDAC inhibitor, IN2001 were cotreated with TCDD to the cells transfected with plAl-Luc reporter gene, the basal promoter activity of CYP1A1 was increased by HDAC inhibitors. Also, in MCF-7 human breast cancer cells, HDAC inhibitors, such as IN2001 and trichostatin A increased the basal activity of CYP1A1 promoter but TCDD stimulated CYP1A1 promoter activity was not changed by HDAC inhibitors. And, in stably-transfected Hepa I cells with plAl-Luc, HDAC inhibitors increased the basal promoter activity only.

To evaluate effect of Gamimajeonojahwan(GMOH) water extract on sexual functions, we orally administered GMOH exract(1,100㎎/㎏) for four weeks in male rats impaired by 2,3,7,8-tetrachlorodibenzo-p-dioxin(TCDD) and ketoconazole(KZ). And we investigated the sexual behaviors such as mount latency, mount frequency, intromission latency, intromission frequency, ejaculation period and ejaculation latency. Serum testosterone concentrations also are evaluated in GMOH-and/or vehicle-treated male rats. The results were obtained as follows: GMOH inhibited the increase of intromission latency by TCDD in male rats. GMOH inhibited the decrease of intromission frequency by TCDD in male rats. GMOH inhibited the increase of ejaculation latency by TCDD in male rats. GMOH inhibited the decrease of serum testosterone concentration by TCDD in male rats. GMOH inhibited the decrease of serum testosterone concentration by KZ in male rats. These results suggest that GMOH water extract is active in improving sexual functions and protecting a decrease of testosterone concentrations induced by TCDD and KZ in male rats.

Aryl hydrocarbons, environmental contaminants accumulate in tissue and pose potential risk in human health. 2,3,7,8-Tertachlorodibenzo-p-dioxin (TCDD) is known as a most potent toxicant among aryl hydrocarbons. TCDD elicits numerous toxic responses in experimental animals and human, including hepatic carcinoma, pulmonary and skin tumor in adult rodents, craniofacial abnormality during mouse embryogenesis, chloracne, reproductive abnormality, immunotoxicity, endocrine effects in exposed humans.(omitted)

In order to understand the mechanism of action of TCDD. we have examined the effect of COX-inhibitors on cypla1 activity. We observed the effect of COX-inhibitor on EROD activity in C57BL/6 mouse in vovo. And we also evaluated the effect of COX-inhibitors on cypla1 mRNA. mouse cyplal promoter activity and EROD activity in Hepa cell. When Aspirin were pretreated with 3MC in vivo, the EROD activity that was stimulated by 3MC was inhibited. (omitted)
