Nonpolar taxoides extracted from a large-scale cell culture of Taxus chinensis were isolated through the normal and reverse phase column chromatographies, and their compounds were identified via NMR spectroscopy. The complete separation method was systematically established and described. In dichloromethane, dissolved paclitaxel and other taxoids with hexane were precipitated during the purification of paclitaxel from the plant cell culture of T. chinensis through a large-scale process while the relatively nonpolar taxane derivatives remained dissolved in the hexane phase. 13-Deoxy baccatin III (I), baccatin VI (II), taxchinin I (III), $2{\alpha}$, $5{\alpha}$, $10{\beta}$, $14{\beta}$-tetraacetoxy-4(20), 11-taxadiene(IV), 1-deoxy baccatinVI(V), and taxayuntin C (VI) were isolated through column chromatography and identified via NMR spectroscopy. Compounds I and IV were found to the major components, aside from paclitaxel, in the plant cell culture of T. chinensis. The concentrations of I and IV were compared with the that concentration of the paclitaxel in each of plant cell culture. The possible applications of compounds I, II, IV, and V were discussed.
A study of forest vegetation in Mt. $T\v{o}kyu$ National Park was investigated by ordination technique. By TWINSPAN(Two-Way Indicator Species Analysis) method, 10 groups were recognized as follows: pinus densiflora, Quercus variabilis, Quercus serrata, Quercus mongolica-Rhododendron schlippenbachii, Quercus mongolica-Abies koreana, Quercus mongolica-Acer pseudo-sieboldi-amum, Quercus mongolica-Symplocos chinensis for. pilosa, Carpinus laxiflora, Fraxinus mandshurica and Taxus cuspidata groups. The floristic composition of these groups showed high correlation to soil moisture(r=0.831), altitude(r=0.784), topography(r=-0.722), organic matter(r=0.642), and pH(r=-0.509) among various environmental factors. According to the results of CCA(Canonical Correspondence Analysis) Pinus densiflora group and Quercus variabilis group were situated in a xeric area at a lower altitude where soil nutrients were poor compared with the other groups. Fraxinus mandshurica group was distributed throughout the valley with high soil moisture and good nutrients, Quercus serrata group and Carpinus laxiflora group were found in the low altitude region with good nutrients, Quercus mongolica group, at the high altitude region with good nutrients, and Quercus mongolica-Acer koreana and Taxus cuspidata at higher altitudes(1, 400-1600 m).
In this study, a tandem water and hexane washing process was developed to improve the recovery efficiency of paclitaxel derived from Taxus chinensis. The polar impurities contained in the sample were effectively removed by washing with water at a sample/water ratio of 1:40 (w/v) for 10 min. In addition, the non-polar impurities were effectively removed by washing with hexane at a sample/hexane ratio of 1:160 (w/v) for 20 min. A high purity of paclitaxel (>30.0%) was obtained in a short operating time (~30 min) by sequential washing with water and hexane.
Endophytic fungi occupy various ecological niches, which reinforces their diversity. As few studies have investigated the endophytic fungi of alpine conifers, we focused on four species of alpine conifers in this study-Abies nephrolepis, Pinus pumila, Taxus cuspidata var. nana, and Thuja koraiensis-and examined them for endophytic fungi. A total of 108 endophytic fungi were isolated. There were four taxa in A. nephrolepis, 12 in P. pumila, 18 in T. cuspidata var. nana, and 17 in T. koraiensis; these were divided into five classes: Agaricomycetes (3.2%), Dothideomycetes (29.0%), Leotiomycetes (15.0%), Sordariomycetes (41.9%), and Orbiliomycetes (1.6%). The most prevalent fungi were Sydowia polyspora (22.7%) and Xylariaceae sp. (22.7%) in P. pumila, Phomopsis juglandina (16.1%) in T. cuspidate var. nana, and Thuja-endophytes sp. 1 (70.0%) in T. koraiensis. However, there was no dominant species growing in A. nephrolepis. Some host plants were analyzed using next-generation sequencing. We obtained 4618 reads for A. nephrolepis and 2268 reads for T. koraiensis. At the genus level, the top three endophytic fungi were Ophiostomataceae_uc (64.6%), Nectriaceae_uc (15.5%), and unclassified organism (18.0%) in A. nephrolepis and Nectriaceae_uc (41.9%), Ophiostomataceae_uc (41.8%), and Magnaporthaceae_uc (9.2%) in T. koraiensis. Our results show that there are different communities of endophytic fungi among different host plants, even if the host plants are in the same region. Such ecological niches are important in terms of the ecological restoration of alpine conifers.
The purpose of this experiment lies in finding the most appropriate cutting condition of Taxus cuspidata Sieb. et Zucc. which has been considered valuable tree species in gardening. Statistical analysis was focused on the comparison of the average ratios of rooting between greenhouses and fields, based on the observation rooted autogenis. To conduct this survey, hardwood cutting of Taxus cuspidata Sieb. et Zucc. were performed at both places of experiment above on April 20, 1982 with rooting results calculated on October 31, 1982. After formation of cuttings, it was soaked in IBA 200 ppm for 12 hours and than put in a bed. A summary of the result is presented as follows; 1) Cutting at the greenhouse showed higher average rate of rooting than at fields. Same tendency was found regardless of the differences in treatment. 2) It was clear that IBA-treatment contributed remarkably to causing higher average rooting rate. As an evidence, when twenty centimeter cuttings were inserted sandy media, IBA-treated area resulted in eighty six percent of average rooting rate, as compared with only twenty three percent in nontreatment case. In case of field cutting, IBA-treatment brought about fifty three percent of rooting in comparison with eleven percent nontreatment. 3) When sandy soil, loam and brown soil were separately used as cutting media, the highest rooting rate was found in case of sandy soil, without any difference between the two experimental places above. 4) As a result of the analysis to seek the impact of the length of cuttings on rooting, the range of length form fifteen to twenty centimeter was apparently most appropriate. It was also found that the rate of rooting declined beyond twenty five centimeter. 5) Two kinds of rooting pattern were observed. One was the case that callus cell lump was created on the lower cut side of cuttings. Importantly, root radical were formed inside the lump to influence the germination of root system. The other relates to the case that adventitious root which look like lateral roots appeared at the stem region. In abstract, first, sandy soil was effectively recommended in case of hardwood cutting in April. Second, the most appropriate length of cuttings ranged between fifteen and twenty centimeters. Third, high density IBA treatment was clearly effective. Forth, for proper environmental management, both pre-disinfection of sail by sterilizer and maintenance of high relative humidity were essentially required.
Distribution types of native conifers (Juniperus chinensis, Pinus parviflora, Tusga sieboldii and Taxus cuspidata var. latifolia) were studied by phytosociological investigation and ZM method in Ulleung Island, South Korea. Two main types were divided maritime vegetation (Juniperus chinensis forest) and mountain vegetation (Taxus cuspidata var. latifolia forest and Pinus parviflora-Tusga sieboldii forest). The former was divided into sea cliff distribution (J-SC) and sea ridge distribution (J-SR) type. The latter was classified 7 distribution types; Taxus cuspidata var. latifolia forest was rock distribution (Ta-R) and mountain slope distribution (Ta-MS) type, and Pinus parviflora-Tusga sieboldii forest was rock distribution (P T-R), upper and ridge distribution (P T-UR, 3 units sub-types:1sub, 2sub, 3sub), and Mountain slope distribution (P T-MS) type. It was considered that J-SC, Ta- R, and P T-R were maintained by topographic climax, but J-SR, Ta-MS, P T-UR and P T-MS were the process of vegetation succession. Distribution types of topographic climax are entrusted to process of vegetation succession. Types in the process of vegetation succession will be needed tending of forest to promote saplings growth and seedlings germination. Especially in order to restore Tsuga sieboldii forest should be afforest and make forest gap because It is mid shade tolerant tree and purity percentage of its seed is 1~2%. The enzyme squalene synthase (EC 2.5.1.21) catalyzes a reductive dimerization of two farnesyl diphosphate (FPP) molecules into squalene, a key precursor for the sterol and triterpene biosynthesis. A full-length cDNA encoding squalene synthase (designated as TcSqS) was isolated from Taxus cuspidata, a kind of important medicinal plants producing potent anti-cancer drug, taxol. The full-length cDNA of TcSqS was 1765 bp and contained a 1230 bp open reading frame (ORF) encoding a polypeptide of 409 amino acids. Bioinformatic analysis revealed that the deduced TcSqS protein had high similarity with other plant squalene synthases and a predicted crystal structure similar to other class I isoprenoid biosynthetic enzymes. Southern blot analysis revealed that there was one copy of TcSqS gene in the genome of T. cuspidata. Semi-quantitative RT-PCR analysis and northern blotting analysis showed that TcSqS expressed constitutively in all tested tissues, with the highest expression in roots. The promoter region of TcSqS was also isolated by genomic walking and analysis showed that several cis-acting elements were present in the promoter region. The results of treatment experiments by different signaling components including methyl-jasmonate, salicylic acid and gibberellin revealed that the TcSqS expression level of treated cells had a prominent diversity to that of control, which was consistent with the prediction results of TcSqS promoter region in the PlantCARE database.
To investigate the vegetation structure of mountain valley from Cheonwangbong to Seonneotang, 23 plots($400m^2$) set up with random sampling method were surveyed. Two groups of Abies koreana community, Fraxinus mandshurica-Acer pseudo-sieboldianum community were classified by cluster analysis. Abies koreana was a major woody plant species in the valley area from Cheonwangbong to Mapogpo, and Pinus koraiensis, Acer pseudo-sieboldianum and Sorbus amurensis was partly occupied. High positive correlations was proved between Picea jezoensis and Sorb us amurensis, Taxus cuspidata, Fraxinus sieboldiana, Kalopanax pictus; Betula costata and Betula platyphylla var. japonica, Corn us controversa; Abies koreana and Pinus koraiensis; Stephanandra incisa and Betula ermani, Acer tegmentosum, Prunus sargentii; Acer pseudo-sieboldianum and Betula ermani; Fraxinus mandshurica and Deutzia prunifolia, Acer tschonoskii var. rubripes; Sorbus amurensis and Kalopanax pictus; Betula platyphylla var.japonica and Cornus controversa; Lindera erythrocarpa and Lespedeza maximowiczii, Quercus serrata, Styrax obassia; Lindera obtusi/oba and Abies holophylla; Carpinus laxiflora and Lespedeza maximowiczii; Fraxinus sieboldiana and Acer tschonoskii var. rubripes, Pinus koraiensis, Taxus cuspidata; Acer tschonoskii var. rubripes and Taxus cuspidata; Quercus mongolia and Rhododendron schlippenbachii; Quercus serrata and Styrax obassia, and relatively high negative correlations was proved between Abies koreana and Deutzia prunifolia; Lindera obtusi/oba and Pinus koraiensis. Species diversity(H') of investigated groups was ranged $2.7478{\sim}2.9573$.
Ethylene production and inhibition of ethylene production by Ag ion were investigated in ornamental trees injured by $SO_2$ exposure. Concurrently the sulfur content of leaves was measured to examine the absorption of $SO_2$. The results were summarized as follows: 1. Ethylene production by leaves of Pinus thunbergii and Cedrus deodara showed peak after 2 - day exposures to $SO_2$ when visible injury was not appeared ; whereas in Taxus cuspidata showed continuos increase even after 4 - day exposures and was markedly stimulated at leaves appeared visible injury. The amount of ethylene production by leaves of Cedrus deodara and Taxus cuspidata was higher than those of Pinus thunbergii. 2. Ag ion applied on leaves at concentration of 100 ppm $A_gNO_3$ significantly inhibited ethylene production by leaves of Cedrus deodara either $A_gNO_3$ sprayed before $SO_2$ exposure or after leaves were injured by $SO_2$ exposure. 3. The sulfur content of leaves after 5 - day exposures to $SO_2$ showed higher in Pinus thunbergii and Cedrus deodara than in Taxus cuspidata, but there were no great differences in the sulfur content among untreated species.
This study analyzed on the characteristics of community structure, species composition, population conditions and annual mean radial growth of Taxus cuspidata in the subalpine zone of the Seoraksan, Deokyusan, and Hallasan National Parks. Deokyusan and Hallasan sites had three layers of stratification structure without tree layer in it and four layer in Seoraksan site. The major dominant species in the order of importance value were as follows: T. cuspidata, Acer tschonoskii var. rubripes, Quercus mongolica, Abies koreana and Sorbus commixta. The trees (> 5 cm DBH) of T. cuspidata were extremely high with 986.0 individuals/ha at the Hallasan site. Average DBH class were 42.0 cm at the Hallasan site and mainly showed large class. The populations of seedlings and saplings with 357.3 individuals/ha, and juvenile with 128.6 individuals/ha, as a succession tree, were found to be the highest at the Hallasan site. In the size frequency distribution, the populations of T. cuspidata in Mt. Halla site showed a reverse J-shaped curve and it was estimated that T. cuspidata community of this site might be maintained continuously as a stable state like present state. Annual mean radial growth of T. cuspidata populations at Seoraksan, Deokyusan, and Hallasan sites showed up as 1.27 mm/year, 0.93 mm/year and 0.89 mm/year respectively.
