Studies on Anti-Cancerous Substances from Higher Plants in East Asian Region

  • To date many kinds of compounds have been obtained from plants kingdom as antineoplastic and anti-cancerous agents. However, there is no special type of compounds for cancer therapy. Various types of substances are effective for various types of cancers and tumors: for instance, alkaloids. lignans, terpenes and steroids etc. Curcumol obtained from Curcuma aromatica was tested and noticed to be effective against cancer of the uterine cervix clinically. Oridonin isolated from Rabdosia ssp. is now investigate for clinical trials in China. Moreover camptothecine isolated from Camptotheca acuminata is also antineoplastic alkaloid, but is very toxic. Chemical modification has been tried to decrease its toxicity This compound is now using as clinical agent. Harringtonin was investigated as an anticancerous drug in China. Taxol, a compound with a taxane ring isolated from the bark of Taxus brevifotia. has been demonstrated to have substantial anticancer activity in patients with solid tumors refractory standard chemotherapy. Supply of this drug has severely limited full exploration of its antineoplastic potential Some efforts are continued in National Cancer Institute(NCI) Washington for surveying various Taxus species for optimal taxol content, improvement in semi-synthesis from baccatin 111, improvement in method of extraction, and development of alternative renewable resources. Further, there are many compounds which have been reported as antineoplastic agents. On the other hand, we have screened on higher plants collected In Japan, China, Korea. Southeast Asia and South America for antineoplastic activity, which has been done using Sarcoma 180 ascites in mice, P388 Iymphocytic leukemia In mice, Chinese hamster lung V-79 cells, P388 cells and nasopharynx carcinoma(KB) cells in our laboratory, as primary screening. In this meeting, 1 will present on antitumor and cytotoxic substances of the higher plants(Rubis cordifolia, Ailanthus vilmoriniana, Aster tataricus, Taxus cuspidata var. nana, Cephalotaxus harringtonia var. drupacea, etc.) selected from above screening tests.

Studies on the Production of (10-Deacetyl) Baccatin III in Cell Cultures of Taxus baccata Pendula (주목세포 배양에 의한 (10-Deacetyl) Baccatin III 생산 연구)

  • Enhanced production of (10-deacetyl) baccatin III and related taxanes was observed in suspension cultures of Taxus baccata Pendula. six % of initial glucose and sucrose concentration increased 10-deacetyl baccatin III production 3.5 and 2.5 times, respectively. Methyl jasmonate, as an elicitor, increased taxane production. Time course changes of taxane production after methyl jasmonate addition showed that baccatin II and 10-deacetyl baccatin III were detected first and paclitaxel, 10-deacetyl taxol and cephalomanine were produced in sequence. Feeding experiments with $500{\mu}M$ of benzoic acid increased 10-deacetyl baccatin III production 10 times. Baccatin III production was also increased 8 times by feeding of $500{\mu}M$ of lysine as a precursor.

Effect of Sulfur Dioxide Gas and Abscisic Acid Pretreatment on Physiological Susceptibility and Growth Damages of Coniferous Seedlings (아황산(亞黃酸)가스 및 ABA 전처리(前處理)가 침엽수(針葉樹) 유묘(幼苗)의 생장피해(生長被害)와 생리적(生理的) 감수성(感受性)에 미치는 영향(影響))

  • This study was conducted to find out the effect of $SO_2$ gas on growth of tree species and the effect of pretreatment of ABA to Pinus koraiensis. Abies holophylla and Taeus cuspidata on the susceptibility for $SO_2$ gas damage. The survey was carried out on the water contents of leaf and shoot, and the acidities of bark and leaf, and the chlorophyll and water soluble sulfur contents, and the changes of transpiration rates on selected seedlings exposed to $SO_2$ gas at seven levels : 0ppm, 0.5ppm, 1ppm, 2ppm, 4ppm, 26.4ppm ABA, 56.8ppm ABA. The results obtained from this research are as follows : 1. The water contents of leaf and shoot of the three tree species decreased with increasing concentrations of $SO_2$ gas and the relative susceptibility of Abies holophylla was larger than those of Taxus cuspidata and Pinus koraiensis because moisture diminution rates were higher in leaves and shoots of Abies holophylla than those of other species. 2. The acidities of bark and leaf of the three tree species increased with increasing concentrations of $SO_2$ gas. The relative susceptibility of Pinus koraiensis in bark acidity was larger than those of Abies holopfaylla and Toxus cuspidata. Abies holophylla was more susceptive than Pinus koraiensis and Taxus cuspidata. 3. Water soluble sulfur contents of the three tree species increased with increasing concentration of $SO_2$ gas. The relative susceptibility of Taxus cuspidata in water soluble sulfur contents was lager than those of Pinus koraiensis and Abies holophylla. 4. Leaf chlorophyll contents of the three tree species decreased with increasing concentrations of $SO_2$ gas. The order of the relative susceptibility in leaf chlorophyll contents was Taxus cuspidata, Abies holophylla and Pinus koraiensis. 5. Correlation coefficients among the measurement were highly significant for $SO_2$ treatment in all three tree species. 6. The transpiration rates of Abies holophylla and Taxus cuspidata did not change until up to 220 minutes after initiation of $SO_2$ gas fumigation, and changed a little during the time between 220 minutes and 270 minutes. On the other hand, Pinus koraiensis continued to change in transpiration rates little by little from the start of $SO_2$ gas treatment up to 270 minutes, meaning that Pinus koraiensis is less sensitive to ABA. 7. When 2ppm of $SO_2$ was treated. ABA pretreatment effect was detected on the six measurement parameters. ABA affected the resistance of plants by inducing stomata closure. 8. The order of relative susceptibility of the three tree species to $SO_2$ gas fumigation was Abies holophylla, Taxus cuspidata and Pinus koraiensis. Pinus koraiensis was more resistant to $SO_2$ gas than Abies holophylla and Taxus cuspidata.

Studies on Rooting of Taxus cuspidata Cuttings by Electric Treatment (전기처리에 의한 주목의 삽목발근에 관한 연구)

  • Tow-year-old Taxus cuspidata shoot cuttings were treated with various electrical impacts of cathode on their base and anode on their apex by normal and reverse source. The cuttings were previously treated with 200ppm IBA for 12 hours and the rooting percentage, the length, and the weight of roots were checked. The auxin contents of cuttings were also examined by high-performance liquid chromatography. The results obtained on this study are as follows; 1. Normal treatment, cathode into the base and anode into apex, seemed to accelerate rooting while reverse treatment showed less effective than normal treatment on rooting, but both treatments were more effective than control. 2. The impact of electrical treatment at 30mV for 30min has a remarkable effect on the percentage rooting, the length, and the weight of roots. 3. Root primordia were formed at the basipetal end of cuttings where the end of primary pith ray meets the cambium in control treatment and formed at the basal part of cuttings irregularly in electrical treatment. 4. High-performance liquid chromatography showed electrical treatment was more effective on auxin accumulation than control, and 30mV-30min was the most effective on auxin accumulation.

Effects of Precursors and Exogenous Taxanes on Taxane Production by Cell Suspension Cultures (전구체 및 외부공급 Taxane이 세포배양에 의한 Taxane 생산에 미치는 영향)

  • Effects of three kinds of precursors as well as exogenous taxanes on the production of taxol and its derivatives were investigated in Taxus cuspidata and Taxus brevifolia cell suspension cultures. When geraniol was added as a precursor of geranylgeranyl pyrophosphate to enhance diterpenoid metabolism, production of some taxanes including taxol was enhanced. The time of addition and amount of feeding were found to be important factors. Feeding of camphor and menthol resulted in negative effects on taxol production. Influences of exogenous taxanes on taxane production by cell cultures were found to be very complicated. When taxol, baccatin III, cephalomannine were exogenously added into the culture, production of baccatin III, 7-epi-10-deacetyltaxol, 10-deacetyltaxol was increased respectively.

Notes on Five Endophytic Fungal Species Isolated from Needle Leaves of Conifers in Korea (침엽수의 잎에서 분리한 5종의 국내 미기록 내생균)

  • Diverse endophytic fungi were isolated from surface-sterilized leaves of three species of conifers inhabiting various sites in Korea: Abies nephrolepis, Pinus koraiensis and Taxus cuspidate. The isolates were identified based on morphological characteristics and sequences analysis of both internal transcribed spacer and large subunit regions of rDNA. In this paper, we report on five previously unreported species of endophytic fungi isolated from conifers: Biscogniauxia maritime, Nemania diffusa, Pezicula carpinea, Phomopsis juglandina and Sydowia polyspora.

  • We isolated endophytic fungi from the coniferous leaves of Abies koreana and Taxus cuspidata inhabiting Mt. Hallasan, Korea. The isolated fungal strains were identified based on a phylogenetic analysis using nucleotide sequences of the internal transcribed spacer region, large subunit region of ribosomal DNA, and translation elongation factor region. Results confirmed four prevously unreported endophytic fungi in Korea: Lachnellula hyalina, Ochrocladosporium elatum, Phacidium lacerum, and Phyllosticta cussoniae. In this report, we describe the morphological characteristics of these fungi and the results of the phylogenetic analysis.

Rapid Micropropagation by Stem Node Culture of Japanese Yew (주목의 줄기절간 조직배양에 의한 급속 대량증식)

  • The effect of plant growth regulators on proliferation of shoot from stem node culture of Japanese yew (Taxus cuspidata Sieb. et Zucc.) was studied using Quoirin and Lepoivre (1977) medium. Among the cytokinin tested, BAP, kinetin, and thidiazuron at various concentrations had no effect on shoot multiplication However when zeatin at 5$\times$10$^{-5}$ M was added to the medium, an average of 6 shoots were regenerated per explant after 8 weeks of culture. The ratio of rooting ex vitro was remarkably increased up to 34% by dipping the basal end in 0.5 to 1.0% IBA on talc compared with 3% in vitro rooting. Rooted plantlets were acclimated in greenhouse conditions for one month and successfully transplanted to the field.

Isotherm, Kinetic and Thermodynamic Characteristics for Adsorption of Acenaphthene onto Sylopute (실로퓨트에 의한 아세나프텐 흡착에 관한 등온흡착식, 동역학 및 열역학적 특성)

  • The adsorption characteristics of the major tar compound, acenaphthene, derived from Taxus chinensis by the commercial adsorbent Sylopute were investigated using different parameters such as initial acenaphthene concentration, adsorption temperature, and contact time. Out of Langmuir, Freundlich, Temkin and Dubinin-Radushkevich isotherm models, adsorption data were best described by Langmuir isotherm. The adsorption kinetics was evaluated by pseudo-first-order, pseudo-second-order and intraparticle diffusion models. The pseudo-second-order model was found to explain the adsorption kinetics most effectively. Thermodynamic parameters revealed the feasibility, nonspontaneity and exothermic nature of adsorption. In addition, the isosteric heat of adsorption was independent of surface loading indicating the Sylopute used as an energetically homogeneous surface.
