Can Moringa oleifera Be Used as a Protein Supplement for Ruminants?

  • The possibility of using Moringa oleifera as a ruminant protein supplement was investigated by comparison between nutritive and anti-nutritive value of its different morphological parts with that of conventionally used Leucaena leucocephala leaf meal (LL). Parameters determined were chemical composition, rumen degradable protein (RDP), acid detergent insoluble protein (ADIP), pepsin soluble protein (PESP), non-protein nitrogen (NPN) total soluble protein (TSP) and protein potentially digested in the intestine (PDI). Total phenols (TP) and total extractable tannins (TET) were also evaluated as anti-nutritive factors. In vitro gas production characteristics were measured and organic matter digestibility (OMD) was estimated basing on 24 h-gas production. Crude protein content ranged from 265-308 g/kg DM in M. oleifera leaves (MOL) and seed cake (MOC) respectively. Leucaena leucocephala and Moringa oleifera soft twigs and leaves (MOLSTL) had CP content of 236 and 195 g/kg DM while Moringa oleifera soft twigs alone (MOST) and Moringa oleifera bucks (MOB) had 160, 114 and 69.3 g/kg DM respectively. RDP was highest in (MOC) (181 g/kg DM) followed by (MOL) (177 g/kg DM) and was lowest in MOB (40 g/kg DM). The proportion of the protein that was not available to the animal (ADIP) was (p<0.05) higher in MOL and MOC (72 and 73 g/kg DM) respectively and lowest in LL (29 g/kg DM). The PDI was high in LL (74 g/kg DM) followed by MOC (55 g/kg DM) then MOL (16 g/kg DM). PESP was highest (p<0.05) in MOC followed by MOL then LL (273, 200 and 163 g/kg DM respectively). MOC exhibited highest NPN content (116 g/kg DM) and was lowest in MOB (18 g/kg DM) (p<0.05). Highly (p<0.05) TSP was observed in MOC and MOL (308 and 265 g/kg DM respectively) followed by LL (236 g/kg DM). MOL had negligible TET (20 g/kg DM) when compared with about 70 g/kg DM in LL. Highly (p<0.05) b and a+b values were observed for MOLSTL (602 and 691 g/kg DM respectively) followed by MOL (490 and 538 g/kg DM). Highest c value was observed in MOSTL followed by MOC and MOL (0.064, 0.056 and 0.053 rate/hour) respectively. OMD was highest (p<0.05) for MOSTL followed by MOC and then MOL (579, 579 and 562 g/kg DM respectively). LL exhibited lower (p<0.05) OMD (467 g/kg DM). It was concluded from this study that the high crude protein content in MOL and MOLST could be well utilized by ruminant animals and increase animal performance however, high proportion of unavailable protein to the lower gut of animals and high rumen degradable protein due to negligible tannin content render it a relatively poor protein supplement for ruminants. MOC can be a best alternative protein supplement to leaves and leaves and soft twigs for ruminants.

Effects of Tropical High Tannin Non Legume and Low Tannin Legume Browse Mixtures on Fermentation Parameters and Methanogenesis Using Gas Production Technique

  • In vitro experiments were conducted to evaluate the suitability of several mixtures of high tanniniferous non legumes with low tanniniferous legumes on in vitro gas production (IVGP), dry matter degradation, Ammonia-N, methane production and microbial population. Eight treatments were examined in a randomized complete block design using four non-legumes and two legumes (Carallia integerrima${\times}$Leucaena leucocephala (LL) (Trt 1), C. integerrima${\times}$Gliricidia sepium (GS) (Trt 2), Aporosa lindeliyana${\times}$LL (Trt 3), A. lindeliyana${\times}$GS (Trt 4), Ceiba perntandra${\times}$LL (Trt 5), C. perntandra${\times}$GS (Trt 6), Artocarpus heterophyllus${\times}$LL (Trt 7), A. heterophyllus${\times}$GS (Trt 8). The condensed tannin (CT) content of non legumes ranged from 6.2% (Carallia integerrima) to 4.9% (Ceiba perntandra) while the CT of legumes were 1.58% (Leucaena leucocephala) and 0.78% (Gliricidia sepium). Forage mixtures contained more than 14% of crude protein (CP) while the CT content ranged from 2.8% to 4.0% respectively. Differences (p<0.05) were observed in in vitro gas production (IGVP) within treatments over a 48 h period dominated by C. perntandra${\times}$G. sepium (Trt 6). The net gas production (p<0.05) was also high with Trt6 followed by A. heterophyllus${\times}$L. leucocephala (Trt 7) and A. heterophyllus${\times}$G. sepium (Trt 8). Highest (p>0.05) NH3-N (ml/200 mg DM) production was observed with the A. heterophyllus${\times}$G. sepium (Trt 8) mixture which may be attributed with it's highest CP content. The correlation between IVGP and CT was 0.675 while IVGP and CP was 0.610. In vitro dry matter degradation (IVDMD) was highest in Trt 8 as well. Methane production ranged from 2.57 to 4.79 (ml/200 mg DM) to be synonimous with IVGP. A higher bacteria population (p<0.05) was found in C. perntandra${\times}$G. sepium (Trt 6) followed by Artocarpus heterophyllus+G. sepium (Trt 8) and the same trend was observed with the protozoa population as well. The results show that supplementing high tannin non leguminous forages by incremental substitution of legume forage increased gas production parameters, NH3-N, IVDMD and microbial population in the fermentation liquid. Methane production was not significantly affected by the presence of CT or different levels of CP in forage mixtures. Among non legumes, Ceiba perntandra and Artocarpus heterophyllus performed better in mixture with L. leucocephala and G. sepium.

Wine quality grading by near infrared spectroscopy.

  • The ability to accurately assess wine quality is important during the wine making process, particularly when allocating batches of wines to styles determined by consumer requirements. Grape payments are often determined by the quality category of the wine that is produced from them. Wine quality, in terms of sensory characteristics, is normally a subjective measure, performed by experienced winemakers, wine competition judges or winetasting panellists. By nature, such assessments can be biased by individual preferences and may be subject to day-to-day variation. Taste and aroma compounds are often present in concentrations below the detection limit of near infrared (NIR) spectroscopy but the more abundant organic compounds offer potential for objective quality grading by this technique. Samples were drawn from one of Australia's major wine shows and from BRL Hardy's post-vintage wine quality allocation tastings. The samples were scanned in transmission mode with a FOSS NIR Systems 6500, over the wavelength range 400-2500 ㎚. Data analysis was performed with the Vision chemometrics package. With samples from the allocation tastings, the best correlations between NIR spectra and tasting data were obtained with dry red wines. These calibrations used loadings in the wavelengths related to anthocyanins, ethanol and possibly tannins. Anthocyanins are a group of compounds responsible for colour in red wines - restricting the wavelengths to those relating to anthocyanins produced calibrations of similar accuracy to those using the full wavelength range. This was particularly marked with Merlot, a variety that tends to have relatively lower anthocyanin levels than Cabernet Sauvignon and Shiraz. For dry white wines, calibrations appeared to be more dependent on ethanol characteristics of the spectrum, implying that quality correlated with fruit maturity. The correlations between NIR spectra and sensory data obtained using the wine show samples were less significant in general. This may be related to the fact that within most classes in the show, the samples may span vintages, glowing areas and winemaking styles, even though they may be made from only one grape variety. For dry red wines, the best calibrations were obtained with a class of Pinot Noir - a variety that tends to be produced in limited areas in Australia and would represent the least matrix variation. Good correlations were obtained with a tawny port class - these wines are sweet, fortified wines, that are aged for long periods in wooden barrels. During the ageing process Maillard browning compounds are formed and the water is lost through the barrels in preference to ethanol, producing “concentrated” darkly coloured wines with high alcohol content. These calibrations indicated heaviest loadings in the water regions of the spectrum, suggesting that “concentration” of the wines was important, whilst the visible and alcohol regions of the spectrum also featured as important factors. NIR calibrations based on sensory scores will always be difficult to obtain due to variation between individual winetasters. Nevertheless, these results warrant further investigation and may provide valuable Insight into the main parameters affecting wine quality.

피혁산업 부산물에 의한 용존 중금속 제거 특성 (Characteristics of Heavy Metal Removal from Aqueous Solutions using Leather Industry by-products)

  • 본 연구에서는 가죽 생산 공정 중 원피 무게의 50% 이상 고형 폐기물로 발생하는 콜라겐 단백질의 섬유 기재에 미모사(Catechol Tannin) 및 채스트넛(Pyrogallol Tannin)과 같은 식물성 탄닌물질을 다양한 조건에서 고정화시킨 10 가지 다른 종류의 바이오 흡착제를 제조하였다. 제조한 각 바이오 흡착제들의 중금속 제거용 흡착제로서의 성능 평가를 위하여 Cu(II), Cd(II), Zn(II), Pb(II), Cr(III) 이온을 함유한 인공 오염수를 사용하여 다양한 반응 조건에서의 회분식 실험을 실시하였으며, 중금속들의 제거 특성을 규명하였다. 미모사를 탄닌물질로 사용하여 콜라겐에 고정화 반응을 시켰을 때 섬유 번들 내부로의 미모사의 침투력은 나프탈렌계 침투제의 주입량에 비례 하였다; 3% ${\geq}$ 1.5% > 0%. 모든 바이오 흡착제들에서 pH 3.0 이하에서는 중금속 이온들의 제거는 거의 일어나지 않았으나 pH 3.0 이상에서 중금속의 제거율이 급격히 발생하였으며 Zn(II)을 제외한 나머지 중금속이온들은 pH 6.0 이상에서는 거의 완전히 제거되었다. Cr(III)의 경우에는 바이오 흡착제 종류별 제거량이 매우 유사한 경향을 나타내었으나 Cu(II), Zn(II), Pb(II)의 제거에서는 축합형 탄닌(미모사)에 비해 이온결합이 가능한 다량의 카르복실기를 함유한 가수분해형 탄닌(채스트넛)을 사용하여 고정화시킨 바이오 흡착제에서 높은 제거능을 보였다. S10 바이오 흡착제에 대한 Pb(II) 및 Cu(II) 중금속 이온들의 흡착은 이온농도가 1,000배 변화거나 경쟁이온 화학종을 일하전 및 이하전 화학종을 사용하였을 때 영향을 받지 않았다.

한국산 감잎로부터 Polyphenol계 생리활성물질 분리 (Isolation of Polyphenol Compounds from the Leaves of Korean Persimmon (Diospyrus kaki L. Folium))

  • 옛부터 민간 약제로 사용했던 감나무 잎을 식품, 화장품 기능 생물 신소재로 활용하기 위해 감나무잎의 주성분으로 알려진 폴리페놀 화합물을 분리하고 생리활성 기능을 조사하였다. 감나무 잎을 60%의 아세톤으로 추출한 용액을 동결 건조시켜 농도별로 조제하고 glucosyltransferase, tyrosinase 효소저해를 관찰한 결과 glucoslytransferase는 $1.8{\times}10^{-1}$ mg/ml에서 82.84% 저해를, tyrosinase는 0.8 mg/ml에서 21,65% 저해 활성이 확인되었다. Sephadex G-50으로 gel filtration한 결과 fraction 1, 2, 3, 4, 5로 분획되었고 이중 fraction 1, 2에서 효소 저해 활성을 나타내었다. 활성 분획물의 물질군을 확인하기 위해 proteinase K 처리와 멸균처리한 결과 효소저해에 영향은 없어 활성물질군은 단백질군이 아니며, UV spectrum에서 자외선 영역에 최대 흡광치를 보여주므로 페놀링을 갖는 화합물로 추정하게 되었다. 추정화합물을 Sephadex LH-20, MCI-gel 및 Bondapak $C_{18}$ column에서 분리한 결과 F1 fraction에서 compound $1{\sim}4$가지 4종류의 화합물, F2 fraction에서는 compounds 5, 6, F3 fraction에서 compound 7, 8, 9를 순수 정제하여 재결정하였다. 분리된 화합물은 TLC 발색반응에서 flavan-3-ol 골격을 기본으로 하는 축합탄닌 반응을 보였고 전개정도에서 monomer가 2종류, dimer가 3종류, trimer가 1종류로 확인되었다.

더덕 잎의 파이토케미컬(phytochemicals)과 항산화 활성 (Phytochemicals and Antioxidant Activity of Codonopsis lanceolata Leaves)

  • 더덕 잎의 활용가치를 높이고자 더덕 잎 추출물 내 존재하는 기능성 생리활성 물질과 항산화 활성을 평가하였다. 먼저, 더덕 잎의 생리활성 물질의 존재를 검정하기 위해 파이토케미컬 검색을 한 결과, 더덕 잎 추출물에 환원당, 사포닌, 트라이터페노이드, 타닌 및 플라보노이드와 같은 생리활성 물질들이 존재함을 확인하였다. 이들 파이토케미컬의 존재를 근거로 하여 항산화 활성을 나타내는 것으로 알려진 총 폴리페놀 및 총 플라보노이드 함량을 측정하고, 이를 기반으로 더덕 잎의 항산화 활성을 평가하였다. 250 및 $500{\mu}g/mL$ 더덕 잎 추출물은 DPPH 라디칼 소거 실험에서 각각 43% 및 81%의 강력한 라디칼 소거 효과를 나타냈고, L-아스코브산의 71% 및 95%에 해당하는 환원력을 나타냈다. 또한, 더덕 잎 추출물 62.5, 125 및 $250{\mu}g/mL$에서 각각 73, 90 및 99%의 NO생성 저해활성을 나타내어 항산화 활성을 기반으로 한 항염증 효능의 가능성도 제공하였다. 이를 기반으로 더덕 잎 추출물의 아글리콘 분획을 조제하여 분석한 결과, 플라보노이드 화합물인 루테올린이 함유되어 있음을 확인하였다. 반면, 더덕과 같은 초롱꽃과(Campanulaceae)인 도라지의 항산화 연구에서 도라지 잎과 줄기 용매 분획물 중 줄기의 부탄올 분획물 $500{\mu}g/mL$을 첨가하였을 때 가장 높은 환원력과 화학성분을 보고한 바 있으나 항산화 활성 성분에 대해서는 언급하지 않았다(35). 루테올린이 함유되어 있는 더덕 잎 아글리콘 분획의 항산화 활성을 평가한 결과, 250 및 $500{\mu}g/mL$의 아글리콘 분획이 각각 67% 와 87%의 강력한 라디칼 소거 효과를 나타냈으며, L-아스코브산의 128% 및 141%에 해당하는 우수한 환원력을 나타냄을 확인하였다. 따라서, 우수한 항산화 활성 및 생리활성물질을 함유하고 있는 더덕 잎이 기능성 식품 소재로서의 활용가치가 높을 것으로 기대된다.

Nutritional Evaluation of Bamboo Shoot Shell and Its Effect as Supplementary Feed on Performance of Heifers Offered Ammoniated Rice Straw Diets

  • The present study was conducted to examine the feasibility of utilising bamboo (Bambusa arundinacea) shoot shell (BSS) in ruminants. Chemical composition, rumen degradability and some antinutritional compounds were determined for fresh and boiled BSSs to evaluate its feed value and safety. Thirty-two Holstein heifers were allocated to four groups and used to investigate the response in growth rate to supplementing ammoniated rice straw with fresh shell (phase 1) or silage of boiled BSS (phase 2). All animals were offered ammoniated straw ad libitum with 1kg of cotton seed meal (phase 1) or 0.5 kg of cotton seed meal and 0.5 kg of concentrate mixture (phase 2) per head per day. The BSS was supplemented at levels of 0, 3, 6 or 9 kg/d (phase 1) and 0, 5, 10 or 15 kg/d (phase 2) (as fed basis). The BSS was very high in moisture content, and its contents of crude protein and neutral detergent fiber were 13~16% DM and 65~76% DM, respectively; boiling resulting in higher moisture and protein. No hydrocyanic acid was detected in both BSSs and content of tannins was negligible. Rumen degradability of BSS was reasonably high, and with boiling the rapidly degradable fraction decreased, and potentially degradable component increased. Silage of the boiled BSS was slightly lower in both rapidly and slowly degraded fractions than the fresh BSS. Animals consumed all supplemented BSSs without any adverse health problems. Intake of ammoniated straw decreased with the increasing levels of BSS, but total intake was higher in almost all supplementary groups than in the non-BSS. Heifers had a higher growth rate in phase 1 with fresh BSS than in phase 2 with ensiled shell, and daily weight gains were 622, 629, 744 or 690 g in phase 1, and 578, 575, 677 or 635 g in phase 2 at four BSS levels, respectively. For both phases growth rate was significantly higher for the animals in groups 3 and 4 than those in groups 1 and 2 (p<0.01), with little difference between groups 1 and 2 (p>0.05) but significant difference between groups 3 and 4 (p<0.05). Supplementation with BSS also resulted in an improved feed conversion rate, with the least concentrate consumption in group 3 for both phases. It is concluded that the BSS has a high potential nutritional value as indicated by its medium protein content, reasonably high rumen degradability, and that inclusion of BSS in ammoniated rice straw diet is not only safe to animals, but also may improve growth rate of ruminants and feed conversion rate. It may be disadvantageous to use high amounts of BSS in ammoniated straw diets.

Anti-Inflammatory Activity of Oligomeric Proanthocyanidins Via Inhibition of NF-κB and MAPK in LPS-Stimulated MAC-T Cells

  • Oligomeric proanthocyanidins (OPCs), classified as condensed tannins, have significant antioxidation, anti-inflammation and anti-cancer effects. This study was performed to investigate the anti-inflammatory effects of OPCs and the mechanism underlying these effects in lipopolysaccharide (LPS)-stimulated bovine mammary epithelial cells (MAC-T). Real-time PCR and ELISA assays indicated that OPC treatment at 1, 3 and 5 ㎍/ml significantly reduced the mRNA and protein, respectively, of oxidant indicators cyclooxygenase-2 (COX-2) (p < 0.05) and inducible nitric oxide synthase (iNOS) (p < 0.01) as well as inflammation cytokines interleukin (IL)-6 (p < 0.01), IL-1β (p < 0.01) and tumor necrosis factor-α (TNF-α) (p < 0.05) in LPS-induced MAC-T cells. Moreover, OPCs downregulated LPS-induced phosphorylation of p65 and inhibitor of nuclear factor kappa B (NF-κB) (IκB) in the NF-κB signaling pathway (p < 0.01), and they inhibited p65 translocation from the cytoplasm to the nucleus as revealed by immunofluorescence test and western blot. Additionally, OPCs decreased phosphorylation of p38, extracellular signal regulated kinase and c-jun NH2-terminal kinase in the MAPK signaling pathway (p < 0.01). In conclusion, the anti-inflammatory and antioxidant activities of OPCs involve NF-κB and MAPK signaling pathways, thus inhibiting expression of pro-inflammatory factors and oxidation indicators. These findings provide novel experimental evidence for the further practical application of OPCs in prevention and treatment of bovine mastitis.

Protective effects skin keratinocyte of Oenothera biennis on hydrogen peroxide-induced oxidative stress and cell death via Nrf2/Ho1 pathway.

  • Oenothera biennis, commonly known as evening primrose, a potential source of natural bioactive substances: flavonoids, steroids, tannins, fatty acids and terpenoids responsible for a diverse range of pharmacological functions. However, whether extract prepared from aerial part of O. biennis (APOB) protects skin against oxidative stress remains unknown. To investigate the protective effects of APOB against oxidative stress-induced cellular damage and elucidated the underlying mechanisms in the HaCaT human skin keratinocytes. Our results revealed that treatment with APOB prior to hydrogen peroxide ($H_2O_2$) exposure significantly increased viability, and the highest DPPH radical-scavenging activities and reducing power of HaCaT cells. APOB also effectively attenuated H2O2-induced comet tail formation and inhibited the $H_2O_2$-induced phosphorylation levels of the histone ${\gamma}H2AX$, as well as the number of apoptotic bodies and Annexin V-positive cells. In addition, APOB exhibited scavenging activity against intracellular reactive oxygen species (ROS) accumulation and restored the mitochondrial membrane potential loss by $H_2O_2$. Moreover, $H_2O_2$ enhanced the cleavage of caspase-3 and degradation of poly (ADP-ribose)-polymerase (PARP), a typical substrate protein of activated caspase-3, as well as DNA fragmentation; however, these events were almost totally reversed by pretreatment with APOB. Furthermore, APOB increased the levels of heme oxygenase-1 (HO-1), which is a potent antioxidant enzyme, associated with the induction of nuclear factor-erythroid 2-related factor 2 (Nrf2). According to our data, APOB is able to protect HaCaT cells from $H_2O_2$-induced DNA damage and cell death through blocking cellular damage related to oxidative stress through a mechanism that would affect ROS elimination and activating the Nri2/HO-1 signaling pathway.

The Effects of Feeding Acacia saligna on Feed Intake, Nitrogen Balance and Rumen Metabolism in Sheep

  • The aim of this study was to determine the feeding value to sheep of Acacia saligna grown under temperate conditions. Pen trials were undertaken to determine the effects of feeding A. saligna, which had been grown in a Mediterranean environment, on feed intake, nitrogen balance and rumen metabolism in sheep. Sheep were given ad libitum access to A. saligna with or without supplementation with PEG 4,000 or PEG 6,000. PEG 4000 appears to be the major detannification agent used in trials involving high tannin feed despite the fact that PEG 6000 has been shown to be more effective, in vitro. For this reason it was of interest to compare the two, in vivo. Dry matter intake was greater (p<0.05) in sheep supplemented with either PEG 4,000 or PEG 6,000 compared to the control. There was no difference, however, in intake between those supplemented with either PEG 4,000 or 6,000. Although animals were not weighed throughout the trial, a loss in body condition was obvious, in particular in the control group. Intake of N was greater (p<0.05) in sheep supplemented with either PEG 4,000 or PEG 6,000 than in the control. There was no difference in N intake between those supplemented with either PEG 4,000 or PEG 6,000. There were no significant differences in either the faecal or urinary N output between any of the treatment groups and all treatment groups were in negative N balance. Neither the average nor maximum pH of ruminal fluid of the control group was different to those supplemented with PEG. The minimum pH for the control group, however, was significantly higher (p<0.05) than for either of the PEG treatments. The average and the maximum ammonia levels were lower (p<0.05) in the control group compared with those in either of the PEG treatment groups. For all dietary treatments ruminal ammonia levels were well below the threshold for maximal microbial growth. Feeding A. saligna, without PEG, had a definite defaunating effect on the rumen. It was concluded that A. saligna was inadequate as the sole source of nutrients for sheep, even with the addition of PEG 4,000 or PEG 6,000. The anti-nutritional effects on the animals were largely attributed to the excessive biological activity of the phenolics in the A. saligna leaves. There is a need to determine other supplements that may be complimentary with PEG to enhance the nutritive value of A. saligna to maintain a minimum of animal maintenance.
