Identification of Novel Salt Stress-responsive Genes Using the Activation Tagging System in Arabidopsis (애기장대에서 activation tagging system을 이용한 새로운 고염 스트레스 반응 유전자의 동정)

  • Abiotic stresses limit the growth and productivity of plants. Cellular adaptation to abiotic stresses requires coordinated regulation in gene expression directed by complex mechanisms. This study used the activation tagging system to identify novel salt stress-responsive genes. The study selected 9 activation tagging lines that showed salt stress-tolerant phenotypes during their germination stages. Thermal asymmetric interlaced-PCR (TAIL-PCR) was used to identify the T-DNA tagging sites on the Arabidopsis genome in selected activation tagging lines, including AT7508, AT7512, AT7527, AT7544, AT7548, and AT7556. RT-PCR analysis showed that ClpC2/HSP93-III (At3g48870), plant thionin family (At2g20605), anti-muellerian hormone type-2 receptor (At3g50685), vacuolar iron transporter family protein (At4g27870), and microtubule-associated protein (At5g16730) were activated in AT7508, AT7512, AT7527, AT7544, and AT7556, respectively. Interestingly, in AT7548, both the genes adjacent to the T-DNA insertion site were activated: Arabinogalactan protein 13 (AGP13) (At4g26320) and F-box/RNI-like/FBD-like domains-containing protein (At4g26340). All of the seven genes were newly identified as salt stress-responsive genes from this study. Among them, the expression of ClpC2/HSP93-III, AGP13, F-box/RNI-like/FBD-like domains-containing protein gene, and microtubule-associated protein gene were increased under salt-stress condition. In addition, AT7508, AT7527, and AT7544 were more tolerant to salt stress than wild type at seedling development stage, functionally validating the screening results of the activation tagging lines. Taken together, our results demonstrate that the activation tagging system is useful for identifying novel stress-responsive genes.

The Variation of Tagging Contrast-to-Noise Ratio (CNR) of SPAMM Image by Modulation of Tagline Spacing

  • Myocardial tagging technique such as spatial modulation of magnetization (SPAMM) allows the study of myocardial motion with high accuracy. Tagging contrast of such a tagging images can affect to the accuracy of the estimation of tag intersection in order to analyze the myocardial motion. Tagging contrast can be affected by tagline spacing. The aim of this study was to investigate the relationship between tagline spacing of SPAMM image and tagging contrast-to-noise ratio (CNR) experimentally. One healthy volunteer was undergone electrocardiographically triggered MR imaging with SPAMM-based tagging pulse sequence at a 1.5T MR scanner (Gyroscan Intera, Philips Medical System, Netherland). Horizontally modulated stripe patterns were imposed with a range from 3.6mm to 9.6mm of tagline spacing. Images of the left ventricle (LV) wall were acquired at the mid-ventricle level during cardiac cycle with FEEPI (TR/TE/FA=5.8/2.2/10). Tagging CNR for each image was calculated with a software which developed in our group. During contraction, tagging CNR was more rapidly decreased in case of short tagline spacing than in case of long tagline spacing. In the same heart phase, CNR was increased corresponding with tag line spacing. Especially, at the fully contracted heart phase, CNR was more rapidly increased than the other heart phases as a function of tagline spacing.

TagPlus: A Retrieval System using Synonym Tag in Folksonomy (TagPlus: 폭소노미에서 동의어 태그를 이용한 검색 시스템)

  • Collaborative tagging describes the process by which many users add metadata in the form of keywords to shared content. Recently, collaborative tagging has grown in popularity on the web, on sites that allow users to tag bookmarks, photographs, videos and other content. In this paper, we analyze the structure and basic knowledge of collaborative tagging systems as well as their dynamical aspects. We also present a retrieval system, TagPlus, using synonym tag that is derived from WordNet database. Specifically, TagPlus, a synonym tag based system has users retrieve images from Flickr system. The proposed system show the images tagged by not only the tag that users input but also the synonyms that are synonyms with the tag.

WalkieTagging : Efficient Speech-Based Video Annotation Method for Smart Devices (워키태깅 : 스마트폰 환경에서 음성기반의 효과적인 영상 콘텐츠 어노테이션 방법에 관한 연구)

  • The rapid growth and dissemination of touch-based mobile devices such as smart phones and tablet PCs, gives numerous benefits to people using a variety of multimedia contents. Due to its portability, it enables users to watch a soccer game, search video from YouTube, and sometimes tag on contents on the road. However, the limited screen size of mobile devices and touch-based character input methods based on this, are still major problems of searching and tagging multimedia contents. In this paper, we propose WalkieTagging, which provides a much more intuitive way than that of previous one. Just like any other previous video tagging services, WalkieTagging, as a voice-based annotation service, supports inserting detailed annotation data including start time, duration, tags, with little effort of users. To evaluate our methods, we developed the Android-based WalkieTagging application and performed user study via a two-week. Through our experiments by a total of 46 people, we observed that experiment participator think our system is more convenient and useful than that of touch-based one. Consequently, we found out that voice-based annotation methods can provide users with much convenience and satisfaction than that of touch-based methods in the mobile environments.

  • In this paper, we propose a system that automatically assigns user's experience-based emotion tags from wearable sensor data collected in real life. Four types of emotional tags are defined considering the user's own emotions and the information which the user sees and listens to. Based on the collected wearable sensor data from multiple sensors, we have trained a machine learning-based tagging system that combines the known auxiliary tools from the existing affective computing research and assigns emotional tags. In order to show the usefulness of this multi-modality-based emotion tagging system, quantitative and qualitative comparison with the existing single-modality-based emotion recognition approach are performed.

Named Entity Recognition for Patent Documents Based on Conditional Random Fields (조건부 랜덤 필드를 이용한 특허 문서의 개체명 인식)

  • Named entity recognition is required to improve the retrieval accuracy of patent documents or similar patents in the claims and patent descriptions. In this paper, we proposed an automatic named entity recognition for patents by using a conditional random field that is one of the best methods in machine learning research. Named entity recognition system has been constructed from the training set of tagged corpus with 660,000 words and 70,000 words are used as a test set for evaluation. The experiment shows that the accuracy is 93.6% and the Kappa coefficient is 0.67 between manual tagging and automatic tagging system. This figure is better than the Kappa coefficient 0.6 for manually tagged results and it shows that automatic named entity tagging system can be used as a practical tagging for patent documents in replacement of a manual tagging.

Epitope Tagging with a Peptide Derived from the preS2 Region of Hepatitis B Virus Surface Antigen

  • Epitope tagging is the process of fusing a set of amino acid residues that are recognized as an antigenic determinant to a protein of interest. Tagging a protein with an epitope facilitates various immunochemical analyses of the tagged protein with a specific monoclonal antibody. The monoclonal antibody H8 has subtype specificity for an epitope derived from the preS2 region of hepatitis B virus surface antigen. Previous studies on serial deletions of the preS2 region indicated that the preS2 epitope was located in amino acid residues 130~142. To test whether the amino acid sequence in this interval is sufficient to confer on proteins the antigenicity recognizable by the antibody H8, the set of amino acid residues in the interval was tagged to the amino terminal of ${\beta}$-galactosidase and to the carboxyl terminal of the truncated $p56^{lck}$ fragment. The tagged ${\beta}$-galactosidase, expressed in Escherichia coli, maintained the enzymatic activity and was immunoprecipitated efficiently with H8. The tagged $p56^{lck}$ fragment, synthesized in an in vitro translation system, was also immunoprecipitated specifically with H8. These results demonstrate that the amino acid sequence of the preS2 region can be used efficiently for the epitope tagging approach.

  • In this paper, we present a study on data augmentation methods for DNN (Deep Neural Network)-based audio tagging. In this system, an audio signal is converted into a mel-spectrogram and used as an input to the DNN for audio tagging. To cope with the problem associated with a small number of training data, we augment the training samples using time stretching, pitch shifting, dynamic range compression, and block mixing. In this paper, we derive optimal parameters and combinations for the augmentation methods through audio tagging simulations.

The Evaluations of Fish Survival Rate and Fish Movements using the Tagging Monitoring Approach of Passive Integrated Transponders (PIT) (수동형 전자발신장치(Passive Integrated Transponder, PIT) 모니터링 기법 적용에 따른 어종별 생존율 평가 및 어도에서 어류이동성 평가)

  • The objective of this study was to evaluate survival rate and fish movement (migration) using a tagging approach of passive integrated transponder (PIT) in Juksan Weir, which was constructed as a four major river restoration projects. For this study, survival rates of each fish species and the mobility of fish individuals were analyzed during 2 weeks by the insertion of PIT tags to various fish species in the laboratory. According to tagging tests in the laboratory, the survival rate 37.5% (30 survivals of 80 individuals) after the insertion of PIT tags. The survival rate of Carassius auratus and Hemibarbus labeo was 100% and 80% after the insertion of the tags, respectively, whereas it was only 13.3% for Zacco platypus. In the field experiments of Juksan Weir, 6 species and 157 individuals from 8 species (563 individuals) were detected in the fixed automatic data-logging system, indicating a detection rate of 27.9% in the fishway of Juksan Weir. In the meantime, some species with no or low detection rates in the fixed automatic data-logging system were turn out to be stagnant-type species, which prefer stagnant or standing water to live.

  • To aim at improving performance and reflecting user's needs of retrieval, the number of researches has been actively conducted in recent year as the quantity of information and generation of the web pages exceedingly increase. One of alternative approaches can be a tagging system. It makes users be able to provide a representation of metadata including writings, pictures, and movies etc. called tag and be convenient in use of retrieval of internet resources. Tags similar to keywords play a critical role in maintaining target pages. However, they still needs time consuming labors to annotate tags, which sometimes are found to be a hinderance caused by overuse of tagging. In this paper, we present an automatic tagging scheme for a solution of current tagging system conveying drawbacks and inconveniences. To realize the approach, face recognition-based tagging system on SNS is proposed by building a face area detection procedure, linear-based classification and boosting algorithm. The proposed novel approach of tagging service can increase possibilities that utilized SNS more efficiently. Experimental results and performance analysis are shown as well.
