Protective Effect of Resveratrol on the Oxidative Stress-Induced Inhibition of Gap Junctional Intercellular Communication in HaCaT Keratinocytes

  • The aim of this study was to investigate the effect of resveratrol on the oxidative stress-induced inhibition of gap junctional intercellular communication in HaCaT keratinocytes. Anti-oxidative activity of resveratrol was measured by $\alpha,\alpha$-diphenyl-$\beta$-picrylhydrazyl assay and dichlorodihydrofluorescein diacetate oxidation assay. Gap junctional intercellular communication in HaCaT keratinocytes was assessed using the scrape loading/dye transfer technique. Western blots and reverse transcription-polymerase chain reaction were also analyzed for connexin 43 protein and mRNA expression, respectively. Resveratrol scavenged directly the stable $\alpha,\alpha$-diphenyl-$\beta$-picrylhydrazyl radical over a concentration range of 4 mg/ml ($78.2{\pm}2.7$% of control) to 500 mg/ml ($29.9{\pm}4.2$% of control) and decreased the intracellular reactive oxygen species induced by ultraviolet A (UVA) irradiation ($89.3{\pm}1.1$% of UVA group), ultraviolet B (UVB) irradiation ($70.9{\pm}1.7$% of UVB group) and 12-0-tet-radecanoylphorbol-13-acetate (TPA, $48.3{\pm}1.1$% of TPA group), respectively. UVA irradiation and TPA markedly reduced gap junctional intercellular communication, which was restored by resveratrol. There were no significant differences in the level of connexin 43 protein and mRNA expression among any of the experimental groups. Our data suggests that resveratrol has the protective effect on the oxidative stress-induced inhibition of gap junctional intercellular communication in HaCaT keratinocytes, and this protection is likely due to the scavenging of reactive oxygen species.

Effects of Resveratrol and Essential Oils on Growth Performance, Immunity, Digestibility and Fecal Microbial Shedding in Challenged Piglets

  • A study was conducted to evaluate the effects of resveratrol and essential oils from medicinal plants on the growth performance, immunity, digestibility, and fecal microbial shedding of weaned piglets. A total of 48 weaned piglets (8 kg initial weight, 28-d-old) were randomly allotted to four dietary treatments with 3 replications of 4 piglets each. The dietary treatments were NC (negative control; basal diet), PC (positive control; basal diet+0.002% apramycin), T1 (basal diet+0.2% resveratrol), and T2 (basal diet+0.0125% essential oil blend). All piglets were orally challenged with 5 ml culture fluid containing $2.3{\times}10^8$ cfu/ml of Escherichia coli KCTC 2571 and $5.9{\times}10^8$ cfu/ml Salmonella enterica serover Typhimurium. The PC group (p<0.05) showed the highest average daily gain (ADG) and average daily feed intake (ADFI) throughout the experimental period, although feed conversion ratio (FCR) was improved in the T1 group (p>0.05). Serum IgG level was increased in the T1 group, whereas TNF-${\alpha}$ levels was reduced in the supplemented groups compared to control (p<0.05). The PC diet improved the dry matter (DM) digestibility, whereas PC and T2 diets improved nitrogen (N) digestibility compared to NC and T1 diets (p<0.05). Fecal Salmonella and E. coli counts were reduced in all treatment groups compared to control (p<0.05). Fecal Lactobacillus spp. count was increased in the T2 group compared to others (p<0.05). Dietary treatments had no significant effect on fecal Bacillus spp. count throughout the entire experimental period. Based on these results, resveratrol showed strong potential as antibiotic alternatives for reversing the adverse effects of weaning stress on growth performance, immunity and microbial environment in E. coli and Salmonella-challenged piglets.

GJIC of HaCaT keratinocyte was assessed using the scrape loading/dye transfer technique. Resveratrol scavenged directly the stable DPPH radical over a concentration range of 4 mg/$\mell$ (78.2 $\pm$ 2.7% of control) to 500 mg/$\mell$ (29.9 $\pm$ 4.2% of control) and prevent to increase the intracellular fluorescence induced by oxidative stress significantly. Ultraviolet A irradiation (UVA) and 12-O-tetradecanoylphorbol-13-acetate markedly reduced GJIC, which was restored by resveratrol. There were no significant differences in the level of Connexin 43 protein and mRNA expression among any of the experimental groups. Our data suggests that resveratrol has the protective effect on the oxidative stress-induced inhibition of gap junctional intercellular communication in HaCaT keratinocyte and this protection is likely due to the scavenging of reactive oxygen species.

Resveratrol inhibits the protein expression of transcription factors related adipocyte differentiation and the activity of matrix metalloproteinase in mouse fibroblast 3T3-L1 preadipocytes

  • This study attempted to investigate the effects of resveratrol on the differentiation of adipocytes. After cells were treated with various concentrations of resveratrol (0, 10, 20, and 40 ${\mu}mol/L$), adipocyte proliferation, the protein expression of transcription factors, and MMPs' activities were determined. Cell proliferation was inhibited more within 4 days of incubation (P<0.05), and lipid accumulation in adipocyte was significantly inhibited by 93.8%, 92.4% and 91.5%, respectively, after two days of 10, 20, and 40 ${\mu}mol/L$ resveratrol treatment (P<0.05). Six days of incubation with the three resveratrol concentrations caused a significantly decreases of 63%, 59.9%, and 25.1% GPDH activity as a dose-dependent response. The triglyceride concentration also decreased significantly with the increase of resveratrol concentration (P<0.05). The protein expression of CCAAT/enhancer-binding protein (C/$EBP{\beta}$) was decreased significantly by 56% and 30% while $PPAR{\gamma}$ was significantly reduced by 57% and 15% with resveratrol treatments of 20 and 40 ${\mu}mol/L$, respectively (P<0.05). The protein expression of C/$EBP{\alpha}$ was decreased by 83%, 74%, and 38% to increased dosage levels, with significance determined for this decrease from 20 ${\mu}mol/L$ of resveratrol. The protein expression of fatty acid binding protein (FABP4) was decreased significantly by 88%, 72%, and 46% with the increase of resveratrol concentration. The activity of MMP-2 was decreased significantly by 84%, 70%, and 63% while MMP-9 activity was decreased significantly by 74%, 62%, and 39% with the increased resveratrol concentrations of 10, 20, and 40 ${\mu}mol/L$, respectively (P<0.05).

Resveratrol and clofarabine induces a preferential apoptosis-activating effect on malignant mesothelioma cells by Mcl-1 down-regulation and caspase-3 activation

  • We previously demonstrated that resveratrol and clofarabine elicited a marked cytotoxicity on malignant mesothelioma (MM) MSTO-211H cells but not on the corresponding normal mesothelial MeT-5A cells. Little is known of the possible molecules that could be used to predict preferential chemosensitivity on MSTO-211H cells. Resveratrol and clofarabine induced downregulation of Mcl-1 protein level in MSTO-211H cells. Treatment of cells with cycloheximide in the presence of proteasome inhibitor MG132 suggested that Mcl-1 protein levels were regulated at the post-translational step. The siRNA-based knockdown of Mcl-1 in MSTO-211H cells triggered more growth-inhibiting and apoptosis-inducing effects with the resultant cleavages of procaspase-3 and its substrate PARP, increased caspase-3/7 activity, and increased percentage of apoptotic propensities. However, the majority of the observed changes were not shown in MeT-5A cells. Collectively, these studies indicate that the preferential activation of caspase cascade in malignant cells might have important applications as a therapeutic target for MM.

Resveratrol의 첨가가 육계의 생산성, 혈액 생화학 특성, 혈액 내 면역글로불린과 혈액 내 항산화 인자에 미치는 영향 (Effects of Dietary Resveratrol on Growth Performance, Blood Biochemical Parameter, Immunoglobulin, and Blood Antioxidant Activity in Broiler Chicks)

  • 본 연구는 육계사료 내 resveratrol의 첨가가 육계의 생산성, 도체율 및 조직 중량, 혈액 생화학적 성상, 면역 및 혈액 항산화 활성에 미치는 영향을 조사하기 위하여 수행하였다. 공시계는 1일령 육계 수평아리 (Ross ${\times}$ Ross 308) 320수를 선별하여 육계초기(0~3주령)와 육계후기(3~5주령)의 35일 동안 시험을 실시하였다. 시험설계는 8처리구(C(-), 대조군; C(+), 항생제 처리군; ${\alpha}$-tocopherol 20 IU 처리군; ${\alpha}$-tocopherol 200 IU 처리군; resveratrol 20 ppm 처리군; resveratrol 200 ppm 처리군; methylated resveratrol 20 ppm 처리군; methylated resveratrol 200 ppm 처리군)를 처리구당 4반복, 반복당 10수씩으로 나누어 완전임의 배치하였다. 육계의 사료섭취량과 사료요구율은 처리구 사이에서 유의차가 없었으나, 종료체중과 증체량은 항생제와 resveratrol 및 methylated reveratrol 처리구에서 무항생제와 ${\alpha}$-tocopherol 처리구에 비해 유의적으로 높았다(P<0.05). 도체율과 조직 중량은 처리구간 유의적인 차이가 없었으나, 간과 F낭 무게는 항생제와 resveratrol 20 ppm 및 methylated resveratrol 처리구에서 낮게 나타났으며, 췌장의 무게는 resvertrol과 methylated resveratrol 처리구에서 높게 나타났다. 맹장 미생물 균총(total microbes, Coliform bacteria, Salmonella, lactic acid bacteria)은 resveratrol과 methylated resveratrol 처리구가 무항생제와 항생제 처리구 및 ${\alpha}$-tocopherol 처리구와 비교하여 차이를 보이지 않았다. 육계 혈액 내 creatinine, BUN, total protein, albumin, AST 그리고 ALT는 처리구 사이에서 유의적인 차이를 보이지 않았으며, globulin 함량은 resveratrol 200 ppm과 methylated resveratrol 20 ppm 처리구에서 무항생제 처리구에 비해 낮은 수치를 보였으나, 처리구 사이에 유의차는 없었다(P<0.05). IgA, IgG와 IgM는 항생제 처리구와 reveratrol 처리구에서 무항생제와 ${\alpha}$-tocopherol 처리구에 비해 유의적으로 감소하였다(P<0.05). SOD 유사 활성은 resveratrol 처리구에서 다른 처리구들에 비해 높게 나타났으며(P<0.05), MDA 함량은 유의적인 차이가 없었다. 따라서 뽕나무 유래 resveratrol은 육계 생산성 개선 및 건강성 유지를 통해 천연 항생제 대체물질로 이용 가능성이 시사된다.

3T3-L1 세포에서 Resveratrol과 Epigallocatechin Gallate(EGCG)의 지방세포 분화 억제에 미치는 시너지 효과 (Synergistic Anti-adipogenic Effects of Resveratrol and Epigallocatechin Gallate in 3T3-L1 Adipocytes)

  • Resveratrol (RVT) and epigallocatechin gallate (EGCG) individually inhibit adipogenesis in 3T3-L1 adipocytes. The objective was to examine the possibility of interaction between RVT and EGCG, resulting in enhanced inhibition of adipogenesis in 3T3-L1 adipocytes. Preadipocytes were treated with RVT and EGCG individually at 6.25 or $25{\mu}M$ (RVT6.25 or RVT25) and 12.5 or $50{\mu}M$ (EGCG12.5 or EGCG50) and in combination (RVT6.25 + EGCG12.5 and RVT25 + EGCG50). RVT25 as an individual compound decreased lipid accumulation in 3T3-L1 adipocytes by 24%, and RVT25 + EGCG50 further decreased lipid accumulation by 77%. In addition, exposure of 3T3-L1 adipocytes to RVT6.25 + EGCG12.5 and RVT25 + EGCG50 combinations resulted in an enhanced increase of adiponectin release and inhibition of leptin release. Quantitative analysis revealed that the combination of tested materials (RVT6.25 + EGCG12.5 and RVT25 + EGCG50) decreased the expression levels of C/EBP${\alpha}$, PPAR${\gamma}2$, and aP2. These results indicate that the combined treatments with RVT and EGCG produce synergistic effects on inhibiting adipogenesis in 3T3-L1 adipocytes. The overall results suggested that the combining RVT and EGCG might be more capable of exerting antiobesity effects than each individual compound by itself.

Isolation and Biological Activity of $Resveratrol-3-O-{\beta}-D-Glucoside$ in Transgenic Rehmannia glutinosa L. Transformed by Peanut Resveratrol Synthase Gene (RS3)

  • Resveratrol, which is both a phytoalexin with antifungal activity and a phytochemical associated with reduced cancer risk and reduced cardiovascular disease, is synthesized in a limited number of plant species including peanut. Resveratrol synthesis is catalyzed by the enzyme stilbene synthase including resveratrol synthase (RS). Resveratrol synthase gene (RS3) obtained from peanut, Arachis hypogaea, Fabaceae has been transferred into chinese foxglove, Rehmannia glutinosa by using Agrobacterium mediated transformation. RS t-DNA introduced to chinese foxglove (R. glutinosa L) by transformation and its reaction product, $resveratrol-3-O-{\beta}-D-glucoside$ was isolated and characterized using HPLC. Also its biological effects was tested in inhibition of the lipid peroxidation of mouse LDL by glycosylated stilbenes derivatives obtained from transgenic plants. $Resveratrol-3-O-{\beta}-D-glucoside$ isolated from transgenic R. glutinosa L. showed antimicrobial activity of the growth inhibition zone against Escherichia coli and Salmonella typhimurium. Therefore, this compound can be contributed to be useful as a phytoalexin for plant health as well as a phytochemical for human health.

Agrobacterium-mediated Transformation of Rehmannia glutinosa L. with Resveratrol Gene (RS3) of Peanut

  • The objectives of this study were to establish the genetic transformation system of stilbene synthase in Rehmannia glutinosa. Resveratrol, which is both a phytoalexin with antifungal activity and a phytochemical associated with reduced cancer risk and reduced cardiovascular disease, is synthesized in a limited number of plant species including peanut. Resveratrol synthesis is catalyzed by the enzyme stilbene synthase including resveratrol synthase (RS). Stilbene synthase gene (RS3) obtained from peanut, Arachis hypogaea, Fabaceae has been transferred into chinese foxglove, Rehmannia glutinosa by using Agrobacterium mediated transformation. PCR analysis with RS3 primer confirmed that the targeted gene was introduced into the plant genome, 904 bp in size. Further analyses of identification of transformation using developed other molecular techniques and transgenic plants that RS t-DNA introduced to chinese foxglove (R. glutinosa L) and its reaction product, stilbene such as resveratrol will be isolate and characterize using NMR, MS, and HPLC.

땅콩과 땅콩새싹 추출물의 resveratrol과 aspartic acid 함량분석 및 지방세포분화 억제효능 (Resveratrol and Aspartic acid Contents and Antiadipogenic Effect of Peanut and Peanut Sprout Extracts)

  • 식품산업 활용성을 고려하여 땅콩과 땅콩새싹의 열수와 주정 추출물을 제조하고 추출물내 resveratrol과 aspartic acid 함량을 UPLC-MS/MS를 이용하여 분석한 결과 땅콩새싹의 주정 추출물이 가장 높은 resveratrol과 aspartic acid 함량을 나타내었다. 또한 땅콩새싹의 주정 추출물은 3T3-L1 세포에 처리시 지방세포분화를 유의적으로 억제하였다. 따라서 땅콩새싹의 주정추출물은 기능성식품소재로 활용 가능성이 높을 것으로 판단된다. 향후 이러한 결과를 바탕으로 땅콩의 발아조건 및 추출 시간과 온도에 따른 활성성분 함량을 측정하여 최적의 활성성분 추출조건을 확립하는 연구를 진행할 계획이다.
