Weathering Properties in Deposits of Fluvial Terrace at Bukhan River, Central Korea (북한강 하안단구 퇴적층의 풍화 특성)

  • Fluvial terraces is poorly developed along Bukhan River in Central Korea. Altitude from riverbed of T1 terraces are 18-29m, T2 terraces 2539m, respectively. Rubification index of T2 is 0.66, T1 is 0.54, and thickness of gravel weathering rind on gneiss of T2 are 14.0mm, granites of T2 are $\infty$, gneiss of T1 are 5.0mm and granites of T2 are 8.0mm, because weathering in deposits of T2 terraces, older than T1, is severer than T1 terraces. Since deposits in T2 have more active and longer weathering than T1, SiO$_2$/Al$_2$O$_3$ is 3.32 in T2 and 4.06 in T1, and SiO$_2$/R$_2$O$_3$ is 2.64 in T2 and 3.19 in T1. CIA(Chemical Index of Alteration) is 87.85% in T2 and 85.88% in T1. Kaolinite and halloysite are founded in deposits of T2 indicating high weathering, and are founded gibbsite made tv eluviation of kaolinite. However, deposits of T1 have no kaolinite, and are found plagioclase, weak mineral in weathering process. Comparing to previous researches by estimated age as altitude from riverbed, rubification index, thickness of gravel weathering rind, element contents and mineral composition, forming age of T2 terraces in Bukhan River are estimated in marine oxygen isotope stage 6 (130-190ka), and T1 terraces are marine oxygen isotope stage 4(59-74ka).

Serial Survey on Group A beta-Hemolytic Streptococcal Carrier Rate and Serotyping in Elementary School Children in 1996~1998 (3년간(1996~1998) 초등학생의 A군 연쇄구균 보균율과 혈청학적 분류에 관한 연구)

  • Purpose : The accuracy of bacteriologic diagnosis of beta-hemolytic streptococcal pharyngotonsillitis depends on the degree of carrier rate in that area where the throat swabs are obtained and the evaluation of serological T typing as an epidemiologic marker is important to understand epidemiology of streptococcal infection. The purpose of this study is to know the carrier rates of group A streptococcus in normal children form four different areas and to find out the epidemiologic characteristic in distribution of the serotypes for 3 years. Method : Throat swabs were obtained from the tonsillar fossa of normal school children in four different areas(Uljin, Seoul, Osan, Kunsan) from March to May 1996, in Uljin in April 1997, and in Uljin in April 1998. The samples were plated on a 5% sheep blood agar plate and incubated overnight at $37^{\circ}C$ before examination for the presence of beta-hemolytic colonies. All isolated beta-hemolytic streptococcus were grouped and serotyped by T agglutination. Results : The carrier rate of beta-hemolytic streptococci and group A streptococci in 1996 were 27.6%, 18.6% at Uljin; 16.4%, 2.7% at Seoul; 33.0%, 26.0% at Osan; 20.0%, 12.3% at Kunsan, respectively. Among 1,192 normal school children from 4 different areas, we obtained 179 strains of group A streptococci. Fifty two percent of the strains were typable by T agglutination in 1996. Common T-type in 1996 were NT, T1, T3, T2 at Uljin; T12, T25 at Seoul; NT, T6, T28 at Osan; T25, T4, NT, T5 at Kunsan, in decreasing order, respectively. At Uljin, T1, T3, T25 accounted for 69% of strains in 1996, T1, T12, T25 accounted for 70% in 1997, and T12, T4 accounted for 88% in 1998. Conclusion : Higher carrier rates were found in Uljin and Osan, where there are a lower population density with scanty of medical facilities compared with another areas. We supposed that low carrier rates is likely to be related to antibiotic abuse or some epidemiologic factor. The periodic and seasonal serotyping analysis is important in monitoring and understanding the epidemiologic patterns of group A streptococci.

Acinetobacter marinus sp. novo and Acinetobacter seohaensis sp. nov., Isolated from Sea Water of the Yellow Sea in Korea

  • Two Gram-negative, nonmotile, coccobacilli, SW-$3^T$ and SW-$100^T$, were isolated from sea water of the Yellow Sea in Korea. Strains SW-$3^T$ and SW-$100^T$ contained ubiquinone-9 (Q-9) as the predominant respiratory lipoquinone and $C_{18:1}\;{\omega}9c$ and $C_{16:0}$ as the major fatty acids. The DNA G+C contents of strains SW-$3^T$ and SW- $100^T$ were 44.1 mol% and 41.9 mol%, respectively. A neighbor-joining tree based on l6S rRNA gene sequences showed that the two isolates fell within the evolutionary radiation enclosed by the genus Acinetobacter. Strains SW-$3^T$ and SW-$100^T$ exhibited a l6S rRNA gene similarity value of 95.7% and a mean DNA-DNA relatedness level of 9.2%. Strain SW-$3^T$ exhibited l6S rRNA gene sequence similarity levels of 93.5-96.9% to the validly described Acinetobacter species and fifteen Acinetobacter genomic species. Strain SW-$100^T$ exhibited l6S rRNA gene sequence similarity levels of less than 97.0% to the other Acinetobacter species except Acinetobacter towneri DSM $14962^T$ (98.0% similarity). Strains SW-$3^T$ and SW-$100^T$ exhibited mean levels of DNA-DNA relatedness of 7.3-l6.7% to the type strains of some phylogenetically related Acinetobacter species. On the basis of phenotypic, phylogenetic, and genetic data, strains SW-$3^T$ and SW-$100^T$ were classified in the genus Acinetobacter as two distinct novel species, for which the names Acinetobacter marinus sp. novo (type strain SW-$3^T$=KCTC $12259^T$=DSM $16312^T$) and Acinetobacter seohaensis sp. novo (type strain SW-$100^T$=KCTC $12260^T$=DSM $16313^T$) are proposed, respectively.

  • For isotopic exchange labelling of 3,5,3'-triiodothyronine (T$_3$) and thyroxine (T$_4$) with radioiodide in the presence of molecular iodine, T$_3$:I$_2$ or T$_4$:I$_2$ molar ratios, pH, and reaction time are. considered to be important factors. A modified labelling and separation method is proposed in present paper, by which T$_3$-$^{125}$ I and T$_4$-$^{125}$ I can be obtained with the mean labelling yields of 45%, and 50%, respectively. The whole reaction products can be separated by adoption of thin-layer chromatography technique using silica gel plate and the solvent system composd of chloroform, methanol and ammonia.

Effects of Pregnancy Stage on Eating Behaviors of Hanwoo Heifers (Bos taurus coreanae) (한우 미경산우 임신 단계가 채식행동에 미치는 영향)

  • The objective of this study was to determine the effect of different pregnancy stages on the eating and ruminating behavior of Hanwoo heifers (Bos taurus coreanae). A total of twelve Hanwoo heifers were divided into four groups depending on pregnancy stage: C: before pregnancy, 9 months old, T1: 3 months pregnant, 16 months old, T2: 6 months pregnant, 19 months old, T3: 9 months pregnant, 22 months old (3 replicates per group). Concentrate of 1% of the body weight was fed to them. Roughage was provided at ad libitum to all treatments. Our results revealed that the intake of roughage was the highest in the T3 group (at 3.68 kg) and the lowest in the C group (at 1.02 kg, p<0.05). Total intake in the order from high to low was: T3 > T2 > T1 > C (p<0.05). The rate of dry matter intake compared to body weight (DMI/BW, %) from high to low order was: T3 > T2 > T1 > C (p<0.05). Eating time, ruminating time, and chewing time were the highest in T3 group (p<0.05). Resting time was the highest in C group (p<0.05). The number of bolus and the number of total chews were significantly higher in T3 compared to those in other groups. However, ruminating time per bolus was not significantly difference among groups. The number of chews per bolus from high to low order was: C > T1 > T3 > T2 (p<0.05). The number of bolus per minute was not significantly different among groups. The number of drinking and the number of defecating were significantly higher in C and T1, respectively (p<0.05). However, the number of urinating had no effect according to pregnancy stage. Eating rate and chewing efficiency were significantly higher (p<0.05) in accordance with pregnancy stage (T3 > T2 > T1 > C). Ruminating efficiency was significantly (p<0.05) higher T1 than that in other groups.

Effect of Dietary Inuloprebiotics on Performance, Serum Immunoglobulin and Caecal Microflora in Broiler Chickens (이눌로프리바이오틱스의 사료 내 첨가가 육계의 성장능력, 혈액 면역물질 및 맹장 미생물에 미치는 영향)

  • The potential of encapsulated inuloprebiotics from domestic Jerusalem artichokes (Helianthustuberosus) as natural antibacterial growth promotor for an antibiotic replacement in broiler chickens was presently assessed through assays of growth performance, serum immunoglobulin production and influence on caecal microflora. Two hundred-forty, 1-day-old, male broilers (Ross 308) were randomly allotted to four treatments (T1-T4), with three replicate pens per treatment and 20 chicks per pen. Broiler chicks were fed a basal diet (T1: control) or basal diet plus antibiotics (T2: Chlorotetracycline, 0.10%), 300 ppm of the inuloprebiotics (T3), or 450 ppm of the inuloprebiotics (T4) for 35 days. Body weight, dressing percentage or weight of breast and thigh muscles relative to carcass weight of T3 and T4 broiler chickens was significantly (P<0.05) higher than T1 and T2 broiler chickens. The weight of abdominal fat from T3 and T4 broiler chickens were significantly (P<0.05) lower than that of T1 and T2 chickens. Serum immunoglobulins in the T3 and T4 groups were significantly (P<0.05) elevated compared to the T1 and T2 groups. The weight of immune organs, thymus and Bursa of Fabricius relative to live body weight in the T3 and T4 groups were significantly (P<0.05) higher than the T1 and T2 groups. Bifidobacteria and Lactobacillus, which are beneficial bacteria, were present in greater numbers in the caecum of T3 and T4 birds than T1 and T2 groups, whereas potentially harmful Escherichiacoli and Salmonella were present in lower numbers, with differences being significant (P<0.05). These results suggest that a diet supplemented with 300 ppm of inuloprebiotics has potential as an antibiotic replacement for organic livestock feed supplement intended to improve production of broiler chicken.

The increased GUS gene inactivation over generation in Arabidopsis transgenic lines (애기장대 형질전환 식물체의 세대경과에 따른 GUS유전자의 비활성화에 관한 연구)

  • The effect of transgene inactivation in T2, T3 and F2 generations was analyzed in progeny seedlings which had been generated by Agrobacterium (LBA4404/pBI121)-mediated transformation in Arabidopsis thaliana. In a system which investigated in the expression of $\beta$-glucuronidase(GUS)gene in kanamycin-resistant (ke $n^{R}$)seedlings, GUS inactivated seedlings were observed in 5 of 12 tested lines of T2 generation and the frequency of GUS inactivation was approximately 2.3%. Lines with multi-copies of T-DNA exhibited severe GUS gene inactivation with the frequency of 5.8% in T2 generation. In T3 generation lines exhibited GUS gene inactivation with the frequency of 1.3%. In contrast, inactivation increased dramatically up to 12.6% in multi-copy T-DNA line. A similar phenomenon was also found in F2 progeny from a transgenic line which had been crossed with wild-type Arabidopsis plant, WS-O (GUS gene inactivation frequency 9.9%). These results indicate that the foreign gene introduced into the plant was inactivated progressively in its transmission during subsequent generations and the transgenic line with multi-copies of T-DNA tended to show more increased inactivation.

Effect of Olive-Oil Prepared Tomato Powder (OPTP) and Refining Lycopene on the Physicochemical and Sensory Characteristics of Seasoned Raw Pork During Storage (올리브유처리 토마토 분말과 정제 Lycopene이 저장 중 양념육의 물리화학적 및 관능적 특성에 미치는 영향)

  • This study was carried out to determine the effects of olive-oil prepared tomato powder (OPTP) and lycopene on seasoned raw pork. 1.5% OPTP was added in T1 and 3.0% in T2; and 0.03% refining lycopene was added in T3. While in storage, pH values of the treated samples were lower (p<0.05) than those of the control samples. Upon increasing storage, salinity values of OPTP samples increased as well, however, the values of T3 did not change. There were significantly higher TBARS values observed in the OPTP samples compared to control, but no significant difference between T3 and the control samples in TBARS values during storage. VBN values of T2 samples were lower than that of other samples at Day 3 and Day 6 of storage. At Day 9 however, the VBN values of T1 and T2 were significantly higher than those of the control and T3. There was no significant difference between the control and the treated samples in the total plate counts. There were relatively higher redness, yellowness and chromatic values observed in the treated samples than in control. In sensory evaluation, there was no significant difference between the control and the treated samples in color, tenderness, juiciness and overall acceptability.

Physicochemical Quality Characteristics of Pork Patty with Tangerine (Citrus unshiu) Peel (감귤껍질을 첨가한 돈육 patty의 이화학적 품질특성)

  • This study was conducted to investigate the physicochemical quality characteristics of pork patty added with four different amount (T0:0%, T1:0.3%, T2:0.7%, T3:1.0%) of tangerine (Citrus unshiu) peel. There was no significant difference in chemical composition, cooking yield, water holding capacity, moisture retention, fat retention, hardness, springiness, cohesiveness, gumminess, chewiness, VBN content, L-value. In taste, texture, juiciness and palatability, the addition of 0.3%~1.0% tangerine peel in pork patty showed no significant difference on sensory properties compared to the pork patty without tangerine peel. Total polyphenol content was highest in T3, and DPPH radical scavenging activity was highest in T2 and T3 (p<0.001). The TBARS contents decreased as tangerine peel become added (p<0.001). The pH was highest in T0, and was lowest in T3 (p<0.001). The external a-value of T2 and T3 were significantly higher than that of T0 (p<0.01). The external and internal b-value of T2 and T3 were higher than those of T0 (p<0.01). Flavor of T2 and T3 were higher than those of T0 and T1 (p<0.01). In conclusion, the results of this study indicate that an addition of tangerine peel could be utilized as an ingredient in pork patty in promotion of function of tangerine by-products.

  • Objectives : This study wad designed to elucidate the short term effect of Rossa rugosae Radix on proliferation, differentiation and maturation of 3T3-L1 Preadipocyte. Methods : 3T3-L1 preadipocytes obtained from Korean Cell line Bank were cultured in a Dulbecco's modified eagle medium(DMED) culture colution containing 10% fetal bovine serum(FBS) and various concentration of aqueous extract of Rossa rugosae Radix on proliferation, differentiation and maturation of 3T3-L1 preadipocytes were investigate after treatment for 24 hours b measuring MTT, Oil Red O and latate dehydrogenase activity.. Results : The Rossa rugosae Radix extract inhibited significantly the proliferation of 3T3-L1 preadipocytes and tended to increase latate dehydrogenase activity in the media of differentiated 3T3-L1 preadipocytes & matured 3T3-L1 preadipocytes. the extract also inhibit the lipid accumulation of differentiated and maturaion of 3T3-L1 preadipocytes, suggesting that Rossa rugosae Radix has anti-obesity effect: however further in vivo study is needed to demonstrate its pharmacological effects.
