Studies on the constituents of syringa oblata var. dilatata(Nakai) Rehder (Syringa oblata var. dilata (Nakai) Rehder의 성분 연구)

  • Syringa oblata var dilatata (Nakai) Rehder is a oleaceous, deciduous shrub indigenous in the calcareous zones of Korea. The author researched the constituents of the leaves of the plant, especially the bitter principles which may be used as amara. According to the literature a bitter principle Syringin and Mannitol were isolated from the leaves of S. vulgaris L. And later on B. Power indicated that syringin and mannitol were widely distributed in oleaceous plants. The fresh leaves of the plant wer extracted with hot water, the filtered clear liquid was mixed with solution of lead acetate. The lead precipitate was filtered off and the filtered liquid was freed from lead by H$_{4}$S. The filtrate thus obtained was evaporated, and from the residue colorless needles, M. P. 166.deg., were obtained. It was soluble in water, in hot alcohol, and insoluble in ether and had a sweet taste. The results of the elementar analysis, M. P. and other characteristics agreed with that of d-mannitol. Finally it was proved to be identical with d-mannitol through conversion of it into Hexaacetyl mannitol, M. P. 124.deg., Triformal mannitol, M. P. 227.deg., Tribenzal mannitol, M. P. 224.deg., which exhibited no depression, when mixed with authentic specimens. The experiments to isolate bitter principles of the plant are in progress.

Chemical Constituents of the Twigs of Paulownia coreana (오동나무 가지의 성분)

  • Three triterpenoids, one sterol glycoside and a phenylpropanoid glycoside were isolated from the n-BuOH soulble fraction of Paulownia coreana twigs. On the basis of spectral data, the structure of isolated compounds were identified as pomolic acid (1), euscaphic acid (2), arjunic acid (3), daucosterol (4), and syringin (5), respectively. All compounds are isolated from this plant for the first time.

Establishment of HPLC-UV Analysis Method Validation for Simultaneous Analysis of Standard Compounds of Oplopanax elatus Nakai Stem (HPLC-UV를 이용한 땃두릅나무 줄기의 지표 성분 동시 분석법 확립)

  • In our previous study, we found uracil, adenosine, protocatechuic acid, syringin (eleutheroside B) and scoparone (6, 7-dimethoxycoumarin) in the Oplopanax elatus Nakai Stem. High-performance liquid chromatography (HPLC) -UV was used to quality and quantify the internal marker compounds in the O. elatus extract after validation of method with linearity, limit of detection (LOD), limit of quantitation (LOQ), accuracy and precision. The specificity assessment visually confirmed that the substance was detected without the introduction of other substances. The established method showed high linearity of the calibration curve and coefficient of correlation ($R^2$) of over the 0.999. HPLC was reported as five standard compounds equivalent using the following linear equation based on the calibration curve. The accuracy of measurement was 84.34 ~ 119.74% and the relative standard deviation (RSD) value was 0.28 ~ 1.60%. In addition, our established method showed high repeatability. The RSD value was 1.10 ~ 6.81%. So, we found the amount of the internal marker compounds in the O. elatus extract. These results demonstrated that can be used to quality evaluation of the O. elatus.

Two Cyanidin compound from the Fruits of Acanthopanax divaricatus var. albeofructus

  • Acanthopanax divaricatus var. albeofructus is one of the indigenous medicinal plant and the fruits of Acanthopanax spp. used as a remedial for "wipe out evil wind". Two anthocyanin were isolated from the fruits of Acanthopanax divaricatus var. albeofructus. Their structures were elucidated as cyanidin 3-lathyroside (1) and cyanidin 3-galactoside (2) by chemicophysical and spectroscopic analysis. And also, four chemical, syringin, chlorogenic acid, caffeic acid and acanthoside D were identified. Both anthocyanide were isolated for the first time from Acanthopanax species. cyanidin 3-lathyroside is one of the rare anthocyanin in natural resources.

  • A new phenylpropanoid was isolated from the roots of Codonopsis ussuriensis (Rupr. et Maxim) Hemsley. It was coloress crystals, mp. 140-142.deg.C and was elucidated as 4-(3-ethoxy-1-propenyl)-2, 6-dimethoxyphenyl-$\beta$-D-glucopyranoside on basis of spectral data analysis.

Antioxidants Isolated from Kalopanax pictus (엄나무 유래 신규 항산화 활성물질)

  • Screening of new antioxidants form oriental medicines resulted in the isolation of a new antioxidative compound and eight known compounds from the stem bark of Kalopanax pictus. On the basis of various spectrosopic studies, the structure of the new compound was determined to be 4-rhamnose-3,5-dimethoxybenzoic acid methly ester. Other known compounds were identified as ferulic acid, 4,5,6,-trihydroxyflavanone, 2', 4',4' -trihydroxychalcone, caffeic acid, coniferyl alcohol, syringin, 1,3-di-O-caffeoylquinic acid. These compounds showed lipid peroxidation inhibitory acitivity in rat liver microsomes and free radical scavenging acitivity.

  • Myristica fragrans Houtt were extracted in 80% aq. MeOH and solvent fractionated sing $CHCl_3$, EtOAc, n-BuOH and water, successively. The n-BuOH fraction gave three phenylpropanoids through application of silica gel column chromatographies. The chemical structures of the phenylpropanoids were determined by the interpretation of several spectral data, including NMR and MS as meso-dihydroguaiaretic acid (1), nectandrin B (2) and syringin methyl ether (3). Compound 1, which was first isolated from this plant by authors, showed inhibitory activities with $60.0{\pm}2.1%\;(100\;{\mu}g/ml),\;42.6{\pm}0.9%\;(140\;{\mu}g/ml)\;and\;12.2{\pm}0.2%\;(200\;{\mu}g/ml)$ on ACAT(acyl-CoA:Cholesterol Acyltransferase), chitin synthase III and HMG-CoA reductase (3-hydroxy-3-methylglutaryl coenzyme A reductase), respectively. Compound 3 showed inhibitory activities with $27.2{\pm}0.9%\;(100\;{\mu}g/ml),\;45.5{\pm}0.8%\;(200\;{\mu}g/ml)$ on ACAT and chitin synthase III.

  • Plants are known as important source in the search for new drugs. The twelve compounds from Viscum album (Loranthaceae), lupeol (1), betulonic acid (2), betulinic acid (3), terminic acid (4), ursolic acid (5), $\beta$-sitosterol (6), $\alpha$-spinasterol (7), oleanolic acid (8), 5-hydroxy-1-(4′-hydoxyphenyl)-7-(4"-hydroxyphenyl)-hepta-1-en-3-on (9), 2′-hydroxy-4′,6′-dimethoxychalcone-4-O-glucoside (10), 2′-hydroxy-4′,6′-dimethoxychalcone-4-O-[apiosyl(l$\longrightarrow$2)]glucoside (11) and syringin (12) were evaluated for their immunomodulatory properties. Compounds 6 and 11 induced the macrophage tumoricidal activity and the lymphocyte blastogenesis. In addition, these compounds stimulated the macrophages to induce the production of TNF-$\alpha$ and NO. These findings suggest that compounds 6 and 11 are modulating various elements of the host immune response.

  • Three limonoids and two phenolics were isolated from stem bark of Pellodendron amurense Rupr. (Rutaceae) and were defined clearly as calodendrolide (1), obacunone (2), limonin (3), methyl 4-hydroxybenzoate (4), and syringin (5) based on NMR spectroscopy results obtained with the aid of X-ray crystallographic analysis. This is the first report on the isolation of calodendrolide (1) from this plant.

On the basis of spectral data, the structure of isolated compounds were identified as scoparone (1), uracil (2), protocatechuic acid (3), syringin (4), and adenosine (5). Among the these compounds, scoparone (1), uracil (2), protocatechuic acid (3), and adenosine (5) were isolated for the first time from this plant.
