The effects of newly formed synthetic peptide on bone regeneration in rat calvarial defects

  • Purpose: Significant interest has emerged in the design of cell scaffolds that incorporate peptide sequences that correspond to known signaling domains in extracellular matrix and bone morphogenetic protein. The purpose of this study was to evaluate the bone regenerative effects of the synthetic peptide in a critical-size rat calvarial defect model. Methods: Eight millimeter diameter standardized, circular, transosseus defects created on the cranium of forty rats were implanted with synthetic peptide, collagen, or both synthetic peptide and collagen. No material was was implanted the control group. The healing of each group was evaluated histologically and histomorphometrically after 2- and 8-week healing intervals. Results: Surgical implantation of the synthetic peptide and collagen resulted in enhanced local bone formation at both 2 and 8 weeks compared to the control group. When the experimental groups were compared to each other, they showed a similar pattern of bone formation. The defect closure and new bone area were significantly different in synthetic peptide and collagen group at 8 weeks. Conclusions: Concerning the advantages of biomaterials, synthetic peptide can be an effective biomaterial for damaged periodontal regeneration.

Sinus augmentation using rhBMP-2-loaded synthetic bone substitute with simultaneous implant placement in rabbits

  • Purpose: The aim of this study was to determine the effect of recombinant human bone morphogenetic protein-2 (rhBMP-2)-loaded synthetic bone substitute on implants that were simultaneously placed with sinus augmentation in rabbits. Methods: In this study, a circular access window was prepared in the maxillary sinus of rabbits (n=5) for a bone graft around an implant (${\varnothing}3{\times}6mm$) that was simultaneously placed anterior to the window. Synthetic bone substitute loaded with rhBMP-2 was placed on one side of the sinus to form the experimental group, and saline-soaked synthetic bone substitute was placed on the other side of the sinus to form the control group. After 4 weeks, sections were obtained for analysis by micro-computed tomography and histology. Results: Volumetric analysis showed that the median amount of newly formed bone was significantly greater in the BMP group than in the control group ($51.6mm^3$ and $46.6mm^3$, respectively; P=0.019). In the histometric analysis, the osseointegration height was also significantly greater in the BMP group at the medial surface of the implant (5.2 mm and 4.3 mm, respectively; P=0.037). Conclusions: In conclusion, an implant simultaneously placed with sinus augmentation using rhBMP-2-loaded synthetic bone substitute can be successfully osseointegrated, even when only a limited bone height is available during the early stage of healing.

  • 몇몇의 정형외과 질환에서 골이식은 필수적이며, 합성 골물질은 골이식물로서 널리 쓰여지고 있다. 한편, 골형성 단백질과 섬유아세포 성장 인자와 같은 골 유도 단백질은 골아세포의 분화 및 증식을 촉진시킬 수 있다. 이러한 물질들의 조합은 조기에 골형성을 촉진시킬 수 있어 수의임상에서 널리 사용되고 있다. 이번 증례보고에서는 합성 골물질과 골유도 단백질을 조합하여 대형 골결손부를 성공적으로 치료한 개의 2 증례를 보고하고자 한다.

Consequence of Synthetic Bone Substitute Used for Alveolar Cleft Graft Reconstruction (Preliminary Clinical Study)

  • Background The outcome of alveolar grafting with synthetic bone substitute (Osteon III) in various bone defect volumes is highlighted. Methods A prospective study was accomplished on 55 patients (6-13 years of age) with unilateral alveolar bone cleft. Osteon III, consisting of hydroxyapatite and tricalcium phosphate, is used to reconstruct the defect. Alveolus defect diameter was calculated before surgery (V1), after 3 months (V2), and finally after 6 months (V3) postsurgery. In the t-test, a significant difference and correlation between V1, V2, and V3 are stated. A p-value of 0.01 is considered a significant difference between parameters. Results The degree of cleft is divided into three categories: small (9 cases), medium (20 patients), and large (26 cases).The bone volume of the clefted site is divided into three steps: volume 1: (mean 18.1091 mm3); step 2: after 3 months, volume 2 resembles the amount of unhealed defect (mean 0.5109 mm3); and the final bone volume assessment is made after 6 months (22.5455 mm3). Both show statistically significant differences in bone volume formation. Conclusion An alloplastic bone substitute can also be used as a graft material because of its unlimited bone retrieval. Osteon III can be used to reconstruct the alveolar cleft smoothly and effectively.

Assessment of stem cell viability in the initial healing period in rabbits with a cranial bone defect according to the type and form of scaffold

  • Purpose: Increased bone regeneration has been achieved through the use of stem cells in combination with graft material. However, the survival of transplanted stem cells remains a major concern. The purpose of this study was to evaluate the viability of transplanted mesenchymal stem cells (MSCs) at an early time point (24 hours) based on the type and form of the scaffold used, including type I collagen membrane and synthetic bone. Methods: The stem cells were obtained from the periosteum of the otherwise healthy dental patients. Four symmetrical circular defects measuring 6 mm in diameter were made in New Zealand white rabbits using a trephine drill. The defects were grafted with 1) synthetic bone (${\beta}$-tricalcium phosphate/hydroxyapatite [${\beta}-TCP/HA$]) and $1{\times}10^5MSCs$, 2) collagen membrane and $1{\times}10^5MSCs$, 3) ${\beta}-TCP/HA+collagen$ membrane and $1{\times}10^5MSCs$, or 4) ${\beta}-TCP/HA$, a chipped collagen membrane and $1{\times}10^5MSCs$. Cellular viability and the cell migration rate were analyzed. Results: Cells were easily separated from the collagen membrane, but not from synthetic bone. The number of stem cells attached to synthetic bone in groups 1, 3, and 4 seemed to be similar. Cellular viability in group 2 was significantly higher than in the other groups (P<0.05). The cell migration rate was highest in group 2, but this difference was not statistically significant (P>0.05). Conclusions: This study showed that stem cells can be applied when a membrane is used as a scaffold under no or minimal pressure. When space maintenance is needed, stem cells can be loaded onto synthetic bone with a chipped membrane to enhance the survival rate.

Improvement of osteogenic potential of biphasic calcium phosphate bone substitute coated with synthetic cell binding peptide sequences

  • Purpose: The aim of this study was to evaluate the improvement of osteogenic potential of biphasic calcium phosphate (BCP) bone substitute coated with synthetic cell-binding peptide sequences in a standardized rabbit sinus model. Methods: Standardized 6-mm diameter defects were created bilaterally on the maxillary sinus of ten male New Zealand white rabbits, receiving BCP bone substitute coated with synthetic cell binding peptide sequences on one side (experimental group) and BCP bone substitute without coating (control group) on the other side. Histologic and histomorphometric analysis of bone formation was carried out after a healing period of 4 or 8 weeks. Results: Histological analysis revealed signs of new bone formation in both experimental groups (4- and 8-week healing groups) with a statistically significant increase in bone formation in the 4-week healing group compared to the control group. However, no statistically significant difference in bone formation was found between the 8-week healing group and the control group. Conclusions: This study found that BCP bone substitute coated with synthetic cell-binding peptide sequences enhanced osteoinductive potential in a standardized rabbit sinus model and its effectiveness was greater in the 4-week healing group than in the 8-week healing group.

The Effect of Spray-dried Porcine Plasma and Tryptophan on Feed Intake and Performance of Weaning Piglets

  • There were three trials involved in this experiment. All piglets in Trial 1 were randomly distributed into the following 4 treatments. Treatment 1. Corn-soybean diet with 5% SDPP. The tryptophan level was 0.237%. Treatment 2. Corn-soybean diet with 10% meat and bone meal. The tryptophan level was 0.177%. Treatment 3. Treatment 1+0.0662% synthetic tryptophan. The total tryptophan level was 0.303. Treatment 4. Treatment 2+0.0662% synthetic tryptophan. The total tryptophan level was 0.236. Piglets in Trial 2 were distributed randomly into the following 4 treatments. Treatment 1: corn-soybean diet+10% meat and bone meal. The total tryptophan level was 0.176%. Treatment 2: corn-soybean diet+10% meat and bone meal+5% SDPP. The total tryptophan level was 0.180%. Treatment 3: Treatment 1 diet+0.004% synthetic tryptophan. The total tryptophan level was 0.180%. Treatment 4: Treatment 1 diet+0.631% synthetic tryptophan. The total tryptophan level was 0.237%. There were 4 treatments in Trial 3. Treatment 1: cornsoybean diet+10% meat and bone meal. The total tryptophan level was 0.176%. Treatment 2: Treatment 1 diet+0.061% synthetic tryptophan. The total tryptophan level was 0.237%. Treatment 3: Treatment 2 diet+0.061% synthetic tryptophan. The total tryptophan level was 0.298%. Treatment 4: corn-soybean diet+10% meat and bone meal+5% SDPP. The total tryptophan level was 0.180%. The results of Trial 1 showed that the piglets ate significantly more (p<0.05) when feed included SDPP in the diet during the first 2 weeks. The feed intake also increased when synthetic tryptophan was added in the 5% meat and bone meal diet; however, the difference did not reach a significant level (p>0.05) during the first 2 weeks. Three weeks onwards the feed intake of 5% meat and bone meal treatment was significantly lower (p<0.05) than for the other three treatments. The results of Trial 2 showed that the feed intake could be significantly improved only when the total tryptophan level reached 0.237%. Piglets in the 5% SDPP treatment had higher feed intake than piglets in 10% meat and bone meal treatment with 0.180% of tryptophan, but did not reach a significant level (p<0.05). Body weight gain also had the same trend as feed intake. The pigs in Treatment 1, the lowest total level of tryptophan treatment (0.176%), had lowest feed intake and weight gain, but the difference did not reach a significant level (p>0.05). The pigs in Treatment 1 of Trial 3 had the lowest feed intake and weight gain (p>0.05). Treatment 2 (0.237%) had the highest average feed intake from Week 1 to Week 5; the second best result was recorded in Treatment 4. As for the weight gain of the piglets in Treatment 4 (5% SDPP), they had a higher average weight during the first 3 weeks. The feed efficiency was better for Treatment 4 (5% SDPP) during the first 2 weeks. The results of these trials showed that both SDPP and tryptophan had a trend to improve the feed intake and weight gain.

Soluble expression and purification of synthetic human bone morphogenetic protein-2 in Escherichia coli

  • A 345-bp gene that encodes human bone morphogenetic protein-2 (hBMP-2) has been synthesized. The codon usage of the resulting gene was modified to include those triplets that are utilized in highly expressed Escherichia coli genes. The hBMP-2 gene was efficiently expressed in E. coli as a soluble and active protein. Since the recombinant hBMP-2 was readily solublized, no further solublization steps were required throughout purification. No additional tagging residues were introduced into the synthetic hBMP-2 gene product. The developed synthetic gene is a promising approach for scaling-up the soluble expression of hBMP-2.

  • As the population ages, the importance of effective bone disease treatments is increasing, highlighting the role of bone grafts. Bone grafts are categorized into natural (autografts, allografts, xenografts) and synthetic (ceramics, polymers). Natural grafts have excellent regenerative abilities but pose biological risks, while synthetic grafts are biocompatible but less effective in regeneration. Various studies aim to enhance the safety and efficacy of bone grafts, significantly altering their surface properties. This review examines these studies and the resulting surface changes, aiming to guide future research and clinical applications.

Tomographic and histometric analysis of autogenous bone block and synthetic hydroxyapatite block grafts without rigid fixation on rabbit calvaria

  • Purpose: The preferred material for bone augmentation beyond the envelope of skeletal bone is the bone block graft, due to its dimensional stability. We evaluated the necessity of rigid fixation for the bone block graft, and compared the bone regeneration and volume maintenance associated with grafting using a synthetic hydroxyapatite block (HAB) and an autogenous bone block (ABB) without rigid fixation on rabbit calvaria over two different periods. Methods: Cylinder-shaped synthetic HAB and ABB were positioned without fixation on the rabbit calvarium (n=16). The animals were sacrificed at 4 or 8 weeks postoperatively, and the grafted materials were analyzed at each healing period using microcomputed tomography and histologic evaluation. Results: Integration of the graft and the recipient bed was observed in all specimens, although minor dislocation of the graft materials from the original position was evident in some specimens (six ABB and ten HAB samples). A tendency toward progressive bone resorption was observed in the grafted ABB but not in the grafted HAB, which maintained an intact appearance. In the HAB group, the area of new bone increased between 4 and 8 weeks postoperatively, but the difference was not statistically significant. Conclusions: The nonfixed HAB was successfully integrated into the recipient bed after both healing periods in the rabbit calvaria. In spite of limited bone formation activity in comparison to ABB, HAB may be a favorable substitute osteoconductive bone material.
