Cornus officinalis Methanol Extract Upregulates Melanogenesis in Melan-a Cells

  • Cornus officinalis is widely distributed in Korea, and its fruit has been used to make as herbal drug for traditional medicine in Korea, Japan, and China because of its tonic, analgesic, and diuretic properties. However, the effects of C. officinalis methanol extract (COME) on melanogenesis remain poorly understood. We evaluated the melanogenic capability of COME in melan-a cells, which are immortalized mouse melanocytes. COME increased melanin synthesis in a dose-dependent manner. Treatment with $12.5{\mu}g/mL$ of COME significantly increased melanin content by 36.1% (p < 0.001) to a level even higher than that (31.6%) of 3-isobutyl-1-methyl-xanthine, a well-known pigmentation agent. COME also upregulated tyrosinase activity and its messenger RNA and protein expression. In addition, COME upregulated the expression of tyrosinase-related proteins 1 and 2 and microphthalmia-associated transcription factor-M messenger RNA expression. These results imply that COME may be appropriate for development as a natural product to treat hair graying.

Recent NMR developments for pharmaceutical research

  • NMR spectrometer has been regarded as essential tool for structure elucidation in variable scientific field as like organic synthesis, natural product and macro protein research. Also NMR can be applied for defining dynamic behavior like ligand and receptor binding. One of advantage of research with NMR is that to be great confident to confirm structure and the measured sample could be recovered. Nevertheless NMR also has a weak points than other spectroscopic methods that require a lot of time for interpreting acquired spectrum and running time due to low sensitivity. For last two decade Bruker has developed hardware and software solution for overcome those weak points. In order to overcome low sensitivity Bruker introduced Cryo and Micro diameter probe head technology. And researcher can reduce the time for routine spectrum processing and interpretation works due to lots of introductions in software solutions for quantification, identification and statistics analysis. Improvement of Skin Photoaging by Polysaccharide Extract Derived from Tremella fuciformis (White Jelly Mushroom)

  • Chronic ultraviolet (UV) radiation causes photoaging, which represents skin damage, disrupts skin barrier function, and promotes wrinkle formation. We investigated that the polysaccharide extract of an edible basidiomycetous white jelly mushroom, Tremella fuciformis, (TF-Glucan®) exhibited statistically photoprotective activity by inhibiting matrix metalloproteases (MMPs) and increasing collagen synthesis, and an anti-inflammatory activity by inhibiting nitric oxide and pro-inflammatory cytokines at the concentrations of less than 1000 ㎍/ml, which is not cytotoxic (p < 0.05). Additionally, TF-Glucan® increased the expression of involucrin and filaggrin to prevent the disruption of UVB-induced barrier function (p < 0.05). TF-Glucan® was assessed as a safe material by the human primary skin irritation (1, 3, 5%), human repeated insult patch test (no sensitization at 5%), 3T3 NRU phototoxicity assay (no phototoxicity, PIF < 2, MPE < 0.1), eye irritation test test by BCOP (no category, IVIS ≤ 3) and local lymph node assay (negative at 10, 25, 50%) for identifying potential skin sensitizing. These results suggest that TF-Glucan® may be useful as an anti-photoaging ingredient for developing cosmeceuticals.

Anti-obesitic Effect of Orostachys japonicus in Rats Model fed a Hyperlipidemic Diet

  • We investigated the effect of Orostachys japonicus extracts on serum lipids, leptin and insulin level in hyperlipidemic rats. Also, diacylglycerol acyltransferase (DGAT) activity and thiobarbituric acid reactive substance (TBARS) were assessed. Inhibitory effect of DGAT related to triglyceride synthesis emerged approximately 96% in EtOAc fraction and showed 90% and 67%, respectively, in CHCl3 and BuOH fractions. Furthermore, the EtOAc and BuOH fractions inhibited 81% and 77%, respectively, in glycerol-3-phosphate acyl transferase (GPAT). Hyperlipidemia and obesity marker, contents of leptin and insulin on serum of hyperlipidemic rats, decreased 50% and 25%, respectively, compared with control group in treated EtOAc fraction. The oxidative stress marker, a concentration of TBARS, showed decrease of approximately 30% in treated EtOAc fraction. Moreover, high density lipoprotein (HDL)-cholesterol contents on serum of rats fed a hyperlipidemic diet were increased 10% and low density lipoprotein (LDL)-cholesterol decreased 50% as well as triglyceride amount of feces multiplied approximately two times more than control group in treated EtOAc fraction. The data suggest that the fractions of O. japonicus may be a potent biomaterial for treatment of hyperlipidemia or obesity.

POLLUTION PREVENTION : ENGINEERING DESIGN AT MACRO-, MESO-, AND MICROSCALES

  • Billions of tons of industrial waste are generated annually in industrialized countries. Managing and legally disposing of these wastes costs tens to hundreds of billions of dollars each year, and these costs have been increasing rapidly. The escalation is likely to continue as emission standards become even more stringent around the world. In the face of these rapidly rising costs and rapidly increasing performance standards, traditional end-of-pipe approaches to waste management have become less attractive. The most economical waste management alternatives in many cases have become recycling of the waste or the redesign of chemical processes and products so that wastes are prevented or put to productive use. These strategies of recycling or reducing waste at the source have collectively come to be known as pollution prevention. The engineering challenges associated with pollution prevention are substantial. This presentation will categorize the challenges in three levels. At the most macroscopic level, the flow of materials in our industrial economy, from natural resource extraction to consumer product disposal, can be redesigned. Currently, most of our raw materials are virgin natural resources that are used once, then discarded. Studies in what has come to be called industrial ecology examine the material efficiency of large-scale industrial systems and attempt to improve that efficiency. A second level of engineering challenges is found at the scale of individual industrial facilities, where chemical processes and products can be redesigned so that waste is reduced. Finally, on a molecular level, chemical synthesis pathways, combustion reaction pathways, and other material fabrication procedures can be redesigned to reduce emissions of pollution and unwanted by-products. All of these design activities, shown in Figure 1, have the potential to prevent pollution. All involve the tools of engineering, and in particular, chemical engineering.

Inhibitory Effect of Benzofuran Compound on Cyclooxygenase

  • Alpha-viniferin was previously isolated as a cyclooxygenase (COX)-2 inhibitor from Carex humilis (Cyperaceae) and is an oligomeric stilbene compound with benzofuran (BF) moieties in its chemical structure. In the present study, a chemically synthetic BF compound, named as 3,3-dimethyl-2,3,4,6,7,8,9,10,11,12,13,14,15,16,17,18-hexadecahydro-1H-benzo[b] cyclopentadeca[d]furan-1-one, was discovered to inhibit bacterial lipo polysaccharide (LPS)-induced prostaglandin $E_2$ $(PGE_2)$ production in macrophages RAW 264.7. The BF compound exhibited a selectively preferred inhibitory effect on COX-2 activity over COX-1 activity. Furthermore, BF compound inhibited LPS-induced COX-2 expression at transcription level. As a down-regulatory mechanism of COX-2 expression shown by BF compound, suppression of nuclear factor $(NF)-{\kappa}B$ activation has been demonstrated. BF compound inhibited LPS-induced $NF-{\kappa}B$ transcriptional activity and nuclear translocation of $NF-{\kappa}B$ p65, in parallel, but did not affect LPS-induced degradation of inhibitory ${\kappa}B{\alpha}$ protein $(I{\kappa}B{\alpha})$. Taken together, anti-inflammatory effect of BF compound on $PGE_2$ production was ascribed by its down-regulatory action on LPS-induced COX-2 synthesis in addition to inhibitory action on enzyme activity of COX-2.

저온 수열법에 의한 헥토라이트 합성 (Synthesis of Hectorite by Hydrothemal Method)

  • 물유리, 수산화 마그네슘, 리튬염을 이용하여 $90{\pm}5^{\circ}C$에서 2단계로 수열반응시켜 팽윤성이 우수한 $12\;{\AA}$ 헥토라이트를 합성하였다. 합성과정은 우선 $SiO_{2}$ 성분을 약 30% 함유한 물유리와 수산화 마그네슘을 화학양론적 조성으로 물에 혼합하고 교반시키면서 pH를 $6{\sim}8$로 유지시켰다. 그 후 수용액을 $90{\pm}5^{\circ}C$의 온도에서 1차로 반응시켜 슬러리 형태의 전구체(precursor)를 제조하였으며 이것을 세척하여 과잉염을 제거하였다. 이때, 리튬 (ie, LiCl)을 팔면체 치환용 이온으로 혼합하였다. 위와 같이 제조된 수용액을 약 10시간 동안 위와 동일한 온도에서 2차로 반응시켜 겔 형태의 헥토라이트를 생성시켰다. 합성된 헥토라이트의 분말 X-선 회절패턴은 자연산 헥토라이트와 일치하였고 FE-SEM으로 관찰한 결과, 직경 50 nm의 균질한 입자로 이루어져 있었다. 이온교환능력과 팽윤성을 측정한 결과, 각각 90 cmol/kg, $60{\sim}70\;ml/2\;g$으로 확인되었으며 에칠렌글리콜 처리 후, 저면간격은 $12\;{\AA}$에서 $17.4\;{\AA}$으로 이동하였다.

철 기반 촉매의 Fischer-Tropsch 합성에서 γ-Al2O3/SiO2 혼합 지지체 조성의 영향 (Effect of Composition of γ-Al2O3/SiO2 Mixed Support on Fischer-Tropsch Synthesis with Iron Catalyst)

  • Fischer-Tropsch 합성(F-T 합성)은 석탄, 바이오매스, 천연가스 등을 개질하여 얻은 합성 가스(CO, $H_2$)를 촉매를 이용하여 탄화수소로 전환 하는 기술이다. Fischer-Tropsch 합성에 이용되는 촉매는 활성 금속, 조촉매, 지지체로 구성되는데 이들의 종류와 조성은 반응의 활성 및 생성물 선택도에 영향을 미친다. 본 연구에서는 ${\gamma}-Al_2O_3$$SiO_2$ 혼합 지지체의 조성이 Fiscsher-Tropsch 반응의 활성과 생성물 선택도에 미치는 영향을 알아 보기위해, ${\gamma}-Al_2O_3/SiO_2$ 혼합 지지체를(100/0 wt%, 75/25 wt%, 50/50 wt%, 25/75 wt%, 0/100 wt%) 이용하여 함침(impregnation)법으로 철 촉매를 제조하였다. 촉매의 물리적 특성은 질소 물리 흡착 법과 X-선 회절 분석법을 통해 분석 하였고, 고정층 반응기에서 Fischer-Trosch 반응을 $300^{\circ}C$, 20bar에서, 60시간 동안 수행 하였다. 촉매의 물리적 특성 분석 결과 촉매의 BET 표면적은 ${\gamma}-Al_2O_3$의 조성이 감소함에 따라 감소하였으며, 촉매 기공의 부피 및 평균 크기는 지지체 조성이 ${\gamma}-Al_2O_3/SiO_2$ (50/50 wt%)인 경우를 제외 하고 증가하는 경향을 보였다. 또한, X-선 회절 분석법을 통해 ${\alpha}-Fe_2O_3$의 입자 크기를 계산한 결과 ${\gamma}-Al_2O_3$의 조성이 감소함에 따라 입자 크기가 감소 하였다. Fischer-Tropsch 합성 결과 ${\gamma}-Al_2O_3$의 조성이 감소함에 따라 CO 전환율은 감소 하였으며, C1-C4의 선택도는 ${\gamma}-Al_2O_3$의 조성이 25 wt%일 때 까지 감소하였으며 이와 반대로, C5+의 선택도는 ${\gamma}-Al_2O_3$의 조성이 25 wt%일 때 까지 증가 하였다.

Mechanism of the natural product moracin-O derived MO-460 and its targeting protein hnRNPA2B1 on HIF-1α inhibition

  • Hypoxia-inducible factor-$1{\alpha}$ ($HIF-1{\alpha}$) mediates tumor cell adaptation to hypoxic conditions and is a potentially important anticancer therapeutic target. We previously developed a method for synthesizing a benzofuran-based natural product, (R)-(-)-moracin-O, and obtained a novel potent analog, MO-460 that suppresses the accumulation of $HIF-1{\alpha}$ in Hep3B cells. However, the molecular target and underlying mechanism of action of MO-460 remained unclear. In the current study, we identified heterogeneous nuclear ribonucleoprotein A2B1 (hnRNPA2B1) as a molecular target of MO-460. MO-460 inhibits the initiation of $HIF-1{\alpha}$ translation by binding to the C-terminal glycinerich domain of hnRNPA2B1 and inhibiting its subsequent binding to the 3'-untranslated region of $HIF-1{\alpha}$ mRNA. Moreover, MO-460 suppresses $HIF-1{\alpha}$ protein synthesis under hypoxic conditions and induces the accumulation of stress granules. The data provided here suggest that hnRNPA2B1 serves as a crucial molecular target in hypoxiainduced tumor survival and thus offer an avenue for the development of novel anticancer therapies.

Quick Real-time PCR을 이용한 Avian Influenza Virus Subtype H5N1의 신속검출법 (Rapid Detection Method of Avian Influenza Subtype H5N1 using Quick Real-Time PCR)

  • 조류 인플루엔자바이러스(AIV) H5N1 아형을 Real-time PCR법을 이용하여 가장 빠르게 진단할 수 있는 방법을 개발하였다. 검색 대상의 염기서열은 AIV H5N1 아형의 hemagglutinin 유전자 중 가장 상동성이 높은 387 bp의 부위를 선택하였고, 실험의 안전을 위하여 인공합섬의 방법으로 제작하였다. Microchip을 기반으로 한Real-time PCR법을 사용하였으며, 총PCR 반응액의 양을 $1{\mu}l$로, PCR 과정 중 각 단계, 즉 해리, 접합, 신장의 시간을 각1초, 1초, 3초로 하여 총 실험시간을 단축하였다. 진단을 위한 실험과정에서 PCR 및 융점분식에 소요된 최단 시간은 12분28초였으며, 민감도측정에서 최소2.4개의 hemaggutinin 유전자를 기질로 하여 목적한 특이 189 bp의 PCR 산물을 증폭할 수 있었기에, 본 연구에서는 이런 초고속 PCR 실험방식을 Quick Real-time PCR이라 명명하였다. 이 결과들은 가금류 및 사람에게 전파된 AIV H5N1아형의 진단에 적용될 수 있을 뿐 아니라, PCR이 사용되는 다른 신속검색법에도 널리 적용 될 수 있을 것으로 기대한다.
