A Study on Synthesis of Ca and Mg Compounds from Dolomite with Salt Additional React (MgCl2·6H2O) (염 첨가 반응(MgCl2·6H2O)을 이용하여 백운석으로부터 Ca 화합물과 Mg 화합물 합성에 관한 연구)

  • In order to utilize dolomite as a calcium/magnesium compound material, it was prepared highly reactive calcined dolomite(CaO·MgO) using a microwave kiln (950 ℃, 60 min). The experiment was performed according to the standard of the hydration test (ASTM C 110) and hydration reactivity was analyzed as medium reactivity (max 74.1 ℃, 5 min). Experiments were performed with calcined dolomite and salt (MgCl2·6H2O) (a) 1:1, (b) 1:1.5, and (c) 1:2 wt% based on the hydration reaction of calcined dolomite. The result of X-ray diffraction analysis confirmed that MgO of calcined dolomite increased to Mg(OH)2 as the salt addition ratio increased. After the separating reaction, calcium was stirred at 80 ℃, 24 hr that produced CaCl2 of white crystal. XRD results, it was confirmed calcium chloride hydrate (CaCl2·(H2O)x) and CaO of calcined dolomite and salt additional reaction was separated into CaCl2. And it was synthesized with Ca(OH)2 99 wt% by NaOH adding reaction to the CaCl2 solution, and the synthesized Ca(OH)2 was manufactured CaO through the heat treatment process. In order to prepare calcium carbonate, CaCO3 was synthesized by adding Na2CO3 to CaCl2 solution, and the shape was analyzed in cubic form with a purity of 99 wt%.

  • [ $Al_2TiO_5$ ]has a high refractive index and good solubility of the chromophore in the $Al_2TiO_5$ lattice, which allows this structure to be a good candidate for the development of new ceramic pigments. However, pure $Al_2TiO_5$ is well known to decompose on firing at $900{\sim}1100^{\circ}C$. However, this process can be inhibited by the incorporation of certain metal cations into its crystalline lattice. In this study, the synthesis of gray ceramic pigment was performed by doping cobalt on the $Al_2TiO_5$ crystal structure. The $Al_2TiO_5$ was synthesized using $Al_2O_3$ and $TiO_2$, and doped with $Co_3O_4$ as a chromophore material. In order to prevent the thermal decomposition during the cooling procedure, MgO was added to samples by 0.05 mole, 0.1 mole, and 0.15 mole as a stabilizer. The samples were fired at $1500^{\circ}C$ for 2 hours and cooled naturally. The crystal structure, solubility limit, and color of the synthesized pigment were analyzed using XRD, Raman spectroscopy, UV, and UV-vis. $Al_2O_3$ was available for the formation of $CoAl_2O_4$, which should also be considered in order to explain the small amount of this phase detected in the sample with the higher $Co^{2+}$ content (${\geq}$ 0.03 mole). It was found that the solubility limit of $Co^{2+}$ in the $Al_2TiO_5$ crystal was 0.02 mole% through an analysis of Raman spectroscopy. Through the addition of a pigment with 0.02 mole% of $Co^{2+}$ to lime-barium glaze, stabilized gray color pigments with 66.54, -2.35, and 4.68 as CIE-$L^*a^*b^*$ were synthesized.

Effect of Solvent Fractions from Methanol Extract of Doenjang on Inhibition of Growth and DNA Synthesis of Human Cancer Cells. (인체 암세포 성장 및 DNA 합성 억제에 미치는 된장 분획물의 영향)

  • Growth and DNA synthesis inhibitory effects of doenjang methanol extract and its solvent fractions on AGS human gastric adenocarcinoma cells, Hep 3B human hepatocellular carcinoma cells, HT-29 human colon cancer cells and MG-63 human osteosarcoma cells were studied. The treatment of doenjang methanol extract ($ 200{\mu}g/ml $) with the AGS, Hep 3B, HT-29 and MG-63 cancer cells after 6 days of incubation inhibited the growth of cancer cells by $32\%$, $51\%$, $84\%$ and $33\%$, respectively. To separate active compounds of doenjang, doenjang methanol extract was fractionated with dichloromethane, ethylacetate, and buthanol. Among the solvent fractions, the dichloromethane and ethylacetate fractions showed the highest growth inhibitory effects on various cancer cells. For example, the dichloromethane and ethylacetate fractions ($200a{\mu}g/ml$) sig-nificantly inhibited the growth of various cancer cells by $89\∼96\%$ and$62\∼86\%$, respectively. DNA synthesis of AGS and Hep 3B cancer cells was significantly inhibited by adding dichloromethane fraction ($200{\mu}g/ml$) up to $94\%$ and $80\%$, respectively. Similarly, the ethylacetate fraction ($ 200\mug/ml $) showed a $ 95\% $ inhibition rate of DNA synthesis in AGS cells. These results suggest that the dichloromethane and ethylacetate fractions have specific active compounds, which will explain this anticancer effect of doenjang.

  • Single-layer polymer green electroluminescent devices were fabricated with novel material synthesis by using moleculely-dispersed TTA and NIDI into the polymer PC(B79) emitter layer doped with C6 fluorescent dye which has low operating voltage and high quantum efficiency. A EL cell structure of glass substrate/indium-tin-oxide/PC:TTA:NIDI:C6/Ca/Al was employed and compared with various low work function cathode electrodes Ca and Mg metals. By adjusting the concentration of the fluorescent dye C6, low turn-on voltage of 2.4V was obtained, maximum quantum efficiency of 0.52% at 0.08mole% has been improved by about a factor of ~50 times in comparison with the undoped cell. The PL and EL colors can't be turned by changing the concentration of the C6 dopant. PL emission peaking was obtained at 495nm and EL emission peaking at 520nm with FWHM ~70nm

Antitumor Toxic Protein Abrin and Abrus Agglutinin

  • Abrus agglutinin was purified from the kernels of Abrus precatorius by Sepharose 4B affinity column chromatography followed by Sephadex G-100 gel filtration column chromatography. About 1.25 g of abrus agglutinin was obtained from 1 kg of the kernels. The LD$_{50}$ of abrus agglutinin is 5 mg/kg of body weight, which is less toxic than that of abrin, 20$\mu\textrm{g}$/kg body weight. The amino acid sequence of abrus agglutinin was determined by protein sequencing techniques and deduced from the nucleotide sequence of a cDNA clone encoding full length of abrus agglutinin. There are 258 residues, 2 residues and 267 residues in the A-chain, the linker peptide and the B-chain of abrus agglutinin, respectively. Abrus agglutinin had high homology to abrin-a (77.8%). The 13 amino acid residues involved in catalytic function, which are highly conserved among abrin and ricin, were also conserved within abrus agglutinin. The protein synthesis inhibitory activity of abrus agglutinin ($IC_{50}$/ = 3.5 nM) was weaker than that of abrin-a (0.05 nM). By molecular modeling followed by site-directed mutagenesis showed that Pro199 of abrus agglutinin A-chain located in amphipathic helix H and corresponding to Asn200 of abrin A-chain, can induce bending of helix H. This bending would presumably affect the binding of abrus agglutinin A-chain to its target sequence GpApGpAp, in the tetraloop structure of 285 r-RNA subunit and this could be one of major factors contributing to the relatively weak protein synthesis inhibitory activity and toxicity of abrus agglutinin.n.

  • Leuconostoc mesenteroides NRRL B-1149 produces various glucoseyltransferases for the synthesis of dextran, levan and glucose-1-phosphate using sucrose as a substrate. A sucrose phosphorylase (1149SPase) was purified from L. mesenteroides NRRL B-1149 culture by using hollow fiber filtration (30 kDa cut off), Toyopearl DEAE 650 M column chromatography and following two times of DEAE-Sepharose column chromatographies. The specific activity of the purified 1149SPase was 25.7 (U/mg) with $16\%$ yield. The 1149SPase showed a molecular size of 56 kDa on denatured $10\%$ SDS-PAGE. The N-terminal amino acid sequence of the enzyme was MEIQNKAM. The optimum pH and temperature of this enzyme were 6.2~6.5 and 37^{circ}C, respectively. It had an apparent K_{m} of 6.0 mM and K_{cat} of 1.62/s for sucrose. 1149SPase crystal was formed by hanging drop diffusion technique using 20 mM calcium chloride dihydrate, 100 mM sodium acetate trihydrate pH 4.6 and $30\%$ 2-methyl-2,4-pentanediol as vaporizing and reservation solution. The 1149SPase catalyzes transferring of glucose from isomaltose or sucrose to salicin and salicyl alcohol by disproportionation reaction or acceptor reaction and synthesized two acceptor products, respectively.

Alterations of Antioxidant Status and Mitochondrial Succinate Dehydrogenase Activity in the Liver of Wistar Strain Albino Rats Treated with by Ethanol Extracts of Annona senegalensis Pers (Annonaceae) Stem Bark

  • Numerous ethnomedicinal uses have been attributed to different parts of Annona senegalensis (ASE), including its uses as food and food additives. The present study investigated toxicological and antioxidant effects of 28 days administration of ethanol extracts of ASE stem bark to Wistar strain albino rats. Acute toxicity test was done to determine lethal dose in Wistar rats while sub-acute toxicity test was conducted on rats divided into four groups (A - control, B - 50 mg/kg, C - 100 mg/kg, D - 150 mg/kg, respectively and treated for 28 days. Oxidative stress markers in liver and kidney as well as hepatic succinate dehydrogenase activity in the mitochondrial and post mitochondrial fractions (PMF) were evaluated. The $LD_{50}$ value of ASE was > 2,000 mg/kg. White blood cell counts gradually increased, but red blood cell counts and haematocrits level decreased significantly (p < 0.05) by about 50%. Liver enzymes in the serum and mitochondrial succinate dehydrogenase activity increased significantly (p < 0.05). Superoxide dismutase and catalase activities also increased in liver mitochondria and PMF while malondialdehyde (MDA) and reduced glutathione levels increased only in the PMF. Furthermore, only MDA levels increased significantly in the kidney after 28 days extract administration. Histopathological examination showed hepatic necrosis and no obvious signs of nephrotoxicity. Anona senegalensis is relatively safe, but prolonged ingestion could induce oxidative stress and impair ATP synthesis through the modulation of the activity of mitochondrial succinate dehydrogenase.

THE H$\beta$ INDEX AND THE AGES OF OLD STELLAR SYSTEMS

  • The $H{\beta}$ and some metal line indices, such as Mg2, Fe52 of single-age and single-metallicity populations are computed based on the method of evolutionary population synthesis, with careful consideration of the variation of the horizontal-branch morphology with metallicity and age. We find (a) that while metal lines are little af-fected, the $H{\beta}$ index is severely enhanced (up to 30%)by the presence of the blue horizontal-branch stars, frustrating the current age-estimations from this index with out careful consideration of these stars, and (b) that there is a systematic trend in the sense that the globular clusters in giant elliptical galaxies appear to be older than those in our Galaxy by several billion years. We also calculate these indices for the stellar populations with a metallicity spread, by adopting metallicity distribution functions predicted by chemical evolution models. The comparison of the models with the observed indices of the central regions of the early-type galaxies yields the results (a) that the ages of the giant elliptical galaxies would be older than the previous estimations by several billion years, and (b) that there is a considerable age spread among elliptical galaxies, in the sense that the giant elliptical galaxies are older than small ones.

Melanogenesis-Inhibitory Effects of Ethyl Acetate Fraction from Diospyros lotus L. Leaf Extract (고욤(Diospyros lotus L.)잎 Ethyl Acetate 분획물의 멜라닌형성 억제 효과)

  • Diospyros lotus has been cultivated for its edible fruits and leaves which are considered for its medicinal importance. The aim of this study was to evaluate the anti-melanogenesis of ethyl acetate (EA) fractions from D. lotus leaves in B16 cells. The order of the total polyphenol content with regard to the different solvent fractions from D. lotus leaves was EA>butanol>metahanol>chloroform>n-hexane. The major compounds of EA fraction from D. lotus leaves by HPLC analysis were myricitrin and myricetin. Cellular TYR activity and melanin content in response to treatment with 100 mg/mL of EA fraction was inhibited more strongly than group treated with arbutin. Further, EA fraction exhibited significant anti-melanogenesis effects by reducing the levels of microphthalima-associated transcription factor (MITE), inhibiting the synthesis of TYR, tyrosinase-related protein-1 (TRP-1) and TRP-2. Therefore, EA fractions from D. lotus leaves may be a good source of skin-whitening agents in the future development of medicine-based trouble skin therapy.

Effects of Using Monensin and Different Levels of Crude Protein on Milk Production, Blood Metabolites and Digestion of Dairy Cows

  • Twenty-four Holstein dairy cows were used to evaluate the single and combined effects of different levels of crude protein (CP) and monensin treatment during early lactation on blood metabolites, milk yield and digestion of dairy cows. The experiment was designed as a completely randomized block with a $3{\times}2$ factorial arrangement of treatments. The factors were three concentrations of CP supplement (19.5, 21.4, and 23.4% of dry matter) and two levels of monensin (0 and 350 mg per cow per day). The experiment consisted of three phases and each phase was 3 wk in length. Monensin did not affect milk yield, lactose, solids-non-fat (SNF), blood glucose, triglyceride and DMI, but increased blood cholesterol, blood urea nitrogen (BUN), insulin and reduced blood ${\beta}$-hydroxybutyrate (BHBA), milk fat and protein percentage. Monensin premix significantly decreased rumen ammonia, but rumen pH and microbial protein synthesis were not affected by monensin treatment. Increasing dietary CP improved milk and protein production, but did not alter the other components of milk. Digestibility of NDF, ADF, CP were improved by increasing dietary CP. Increasing dietary CP from 19.5 to 21.4% had no significant effect on ruminal ammonia, but increasing CP to 23.4% significantly increased ruminal ammonia. There was a linear relationship between level of crude protein in the diet and volume of urine excretion. Microbial protein synthesis was affected by increasing CP level; in this way maximum protein synthesis was achieved at 23.4% CP.
