Purification and Characterization of Nonmitochondrial Citrate Synthase from Saccharomyces cerevisiae (Saccharomyces cerevisiae의 Nonmitochondrial Citrate synthase 분리 및 특성)

  • Citrate synthase 1 (mitochondrial) and citrate synthase 2 (nonmitochondrial) were purified from Saccharomyces cerevisiae. The physical and enzymatic characteristics of citrate synthase 2 were ananlyzed in comparison with citrate synthase 1. Both isoenzymes were shown to be dimeric proteins of identical subunits, and the molecular weights of the subunits were estimated to be 48.3kDa for citrate synthase 1 and 47.0kDa for citrate synthase 2, respectively. The optimal pH value for enzyme activity was pH 7.5 for both isoenzymes. However, the optimal temperature for the activity was strikingly different; while the activity of citrate synthase 1 reached its peak at 65.deg.C, that of citrate synthase 2 was maximal at 40.deg.C. Citrate synthase 2 showed much lower thermal and pH stability than citrate synthase 1. In addition, citrate synthase 2 was affected much more by the metal ions such as $Zn^{2+}$ , $Mn^{2+ , and $Co^{2+} than citrate synthase 1. Among the several possible regulatory metabolites tested, ATP showed the strongest inhibitory effect on both enzymes. ADP and NADH were found to have greater effect on citrate synthase 2 than on citrate synthase 1. Kinetic analysis revealed that citrate synthase 2 has approximately 7- and 3.5-fold lower affinity to acetyl CoA and to oxaloacetate, respectively, than citrate synthase 1.

  • Glyphosate had no effect on 5-enolpyruvylshikimate-3-phosphate synthase(EPSP-synthase) biosynthesis per se. But it inhibited clealy the activity of EPSP-synthase. EPSP-synthase seemed to be synthesized as a higher molecular weight(54 kDa) presusor protein and to be transported into plastid. The apparent molecular weight of mature EPSP-synthase in plastid is 45 kDa. Thus, the molecular size of transit peptide appeared to be about 9 kDa. The etiolation for 48 h after glyphosate application did not exhibit the inhibition of translocating level of EPSP-synthase across chloroplast envelope in actively growing meristematic leaves. But even when the plants were etiolated 2 hr after glyphosate treatment, a complete inhibition did not occur at least within 12 hr, i.e. 2 hr after beginning light period, suggesting that EPSP-synthase biosynthesis appeared to be not completely light dependent and the level of EPSP-synthase translocation to chloroplast could be controlled by an unknown regulatory mechanism of light dependent herbicidal effect of glyphosate.

  • 5-Enolpyruvylshikimate 3-phosphate(EPSP) synthase is the sixth enzyme of the shikimate pathway that synthesizes aromatic amino acids. The enzyme is a primary target for the glyphos'lte which is a broad-spectrum and environmetally safe herbicide. As a first step toward development of glyphpsate-resistant EPSP synthase, the EPSP synthase gene(aroA) was amplified by polymerase chain reaction and cluned into pET-25b vector. In this construct. designated pET-aro, the aroA gene is expressed under control of strong T7 promoter. and the EPSP synthase is produced as a fusion protein with pelB leader at N-terminus and HSV-tag and His-tag at C-terminus. When the pET-aro clone was induced to produce the enzyme, it was found that the EPSP synthase was successfully exported to peri plasmic space. The periplasmic transport was greatly dependent on the induction temperatures. Among the induction temperatures examined($25^{\circ}C$, $30^{\circ}C$, $34^{\circ}C$ and $37^{\circ}C$). induction at $34^{\circ}C$ gave rise to maximal periplasmic transport. The recomhinant EPSP synthase could have been purified hy $Ni^{2+}$ -affinity chromatography using the His-tag. and detected hy anti-HSV -tag antibody. The recombinant EPSP synthase also hound to phosphocellulose resin and was eluted hy shikimate 3-phosphate and phosphoenolpyruvate. as expected. The recombinant EPSP synthase purified from phosphocellulose resin showed typical EPSP synthase activity.

  • The 3-Deoxy-D-arabinoheptulosonate 7-phosphate (DAHP) synthase is the first enzyme of aromatic amino acid-, folic acid-, and phenazine-1-carboxylic acid biosynthetic pathways. DAHP synthase of Bacillus sp. B-6 that produces phenazine-1-carboxylic acid was feedback inhibited by two intermediary metabolites of aromatic amino acid biosynthetic pathways, prephenate and chorismate, but not by other metabolites, such as anthranilic acid, shikimic acid, p-aminobenzoic acid, and 3-hydroxyanthranilic acid. DAHP synthase of Bacillus sp. B-6 was not inhibited by end products, such as aromatic amino acids, folic acid, and phenazine-1-carboxylic acid. The inhibition of DAHP synthase by prephenate and chorismate was non-competitive with respect to erythrose 4-phosphate and phosphoenolpyruvate. Prephenate and chorismate inhibited 50% of the DAHP synthase activity at concentrations of $2{\times}10^{-5}\;M$ and $1.2{\times}10^{-4}\;M$, respectively The synthesis of DAHP synthase of Bacillus sp. B-6 was not repressed by exogenous aromatic amino acids, folic acid, and phenazine 1-carboxylic acid, single or in combinations.

Screening of Chitin Synthase II Inhibitors from Medicinal and Wild Plants (고등식물로부터 Chitin Synthase II 활성 저해물질의 탐색)

  • Chitin is an important structural component of fungal cell wall and is synthesized by chitin synthase I, II, and III. The chitin synthase II is an essential enzyme for the formation of primary septum in Saccharomyces cerevisiae. Therefore, specific inhibitors of this enzyme might block the formation of fungal cell wall and could be used as effective antifungal agents. To search chitin synthase IIinhibitors from natural products, 67 plants were extracted with methanol and examined for the inhibitory activities against chitin synthase II of S. cerevisiae by our cell free assay system. As a result, the extracts from 16 plants showed more than 70% inhibition at the concentration of $280{\;}\mu\textrm{g}/ml$. Of note, Laurus nobilis (81.4%), Lonicera maackii (81.5%), Berchemia berchemiaefolia (82.9%), Koelreuteria paniculata (87.9%), Chamaecyparis pisifera (86%) and Taxus cuspidata (83.9%) inhibited strogly the chitin synthase IIactivity.

Glyphosate Toxicity: II. EPSP-synthase Activity in Cell Suspension Culture of Corydalis Sempervirens and Lycopersicon Esculentum (Glyphosate 독성(毒性): II. corydalis Sempervirens와 토마토의 세포배양체(細胞培養體)에서 EPSP-synthase의 활성(活性)에 미치는 영향(影響))

  • Glyphosate (N-[phosphonomethyl]glycine) applied to the assimilate-exporting leaves or sprayed to the whole plants of tomato(Lycopersicon esculentum Mil var. Moneymaker) induced the rapid inhibition of 5-enolpyruvyl skimic acid 3-phosphate synthase(EPSP-synthase). It shows that EPSP-synthase activity precedes chlorophyll loss. There is no difference in EPSP-synthase activity between in vivo tomato meristem and cell suspension culture if glyphosate is not applied. The EPSP-synthase activity is in a range of 4 to 6 nkat per mg protein. The inhibition of EPSP-synthase action is induced within 36 h after glyphosate application while the Chl contents were reduced 48 h after the application. In cell suspension culture of tomato and Corydalis (Corydalis sempervirens), a sublethal concentration of glyphosate retards the fresh weight increase and prolonged lag phase. The fresh weight is reached maximal about 14 days after the subculture in the presence of glyphosate. The inhibitory effect of glyphosate on EPSP-synthase is remarkably induced in lag phase.

Acid Secretion and Nitric Oxide Synthase Activity in Gastric Glands Following Hypoxia/Reoxygenation and Acidosis (Hypoxia/Reoxygenation과 Acidosis가 위선세포에서 위산분비와 NO Synthase 활성에 미치는 영향)

  • Acid secretion and NO synthase activity were determined in isolated gastric glands following hypoxia/reoxygenation and acidosis to investigate the involvement of NO in acid secretion. Isolated gastric glands were exposed to hypoxia (30 min)/reoxygenation (1 h) and/or to acidosis (pH 6.0 and 4.0). Acid secretion was measured by the ratio of $[^{14}C]-aminopyrine$ accumulation between intra- and extraglands. NO synthase activity was determined by percent conversion to $[^{14}C]-citrulline\;from\;[^{14}C]L-arginine$, a precursor of NO. The results indicate that dibutyryl cAMP stimulated acid secretion dose-dependently but had no effect on NO synthase activity in basal gastric glands. Hypoxia/reoxygenation significantly suppressed acid secretion both in unstimulated and stimulated gastric glands, which was exaggerated by acidosis. Constitutive NO synthase, activity, not responded to dibutyryl cAMP, was also inhibited by hypoxia/reoxygenation and acidosis. In conclusion, pathologic state of gastric mucosa such as hypoxia/reoxygenation and acidosis suppresses both acid secretion and NO release but the role of NO in acid secretion stimulated by dibutyryl cAMP in basal gastric glands is not significant.

Preparation of Enzyme Source for Screening of Enzyme Inhibitor of $\beta$-1,3-glucan Synthase (베타-1,3-글루칸 합성효소 저해제의 스크리닝을 위한 효소원 제조법)

  • Assay conditions for screening of $\beta$-1,3-glucan synthase inhibitor were evaluated. Cells in the beginning of mid-log phase showed the highest activity of the $\beta$-1,3-glucan synthase. Cells permeabilized with 1% digitonin treatment could be used as a good crude enzyme source for convenient screening of the $\beta$-1,3-glucan synthase inhibitors. Calcofluor white (0.125% in final) and papulacandin B (25 $\mu$g/ml) inhibit 90% and more than 50% of the $\beta$-1,3-glucan synthase activity, respectively. Cells grown at 37$\circ$C showed higher enzyme activity than those of 25$\circ$C. Catalytic factor of the $\beta$-1,3-glucan synthase was solubilized from particulated membrane preparations, holoenzyme, by extracting with 0.00938% CHAPS.

  • We identified a 1,134-bp putative type III polyketide synthase from the sequence analysis of Streptomyces peucetius ATCC 27952, named Sp-RppA, which is characterized as 1,3,6,8-tetrahydroxynaphthalene synthase and shares 33% identity with SCO1206 from S. coelicolor A3(2) and 32% identity with RppA from S. griseus. The 1,3,6,8-tetrahydroxynaphthalene synthase is known to catalyze the sequential decarboxylative condensation, intramolecular cyclization, and aromatization of an oligoketide derived from five units of malonyl-CoA to give 1,3,6,8-tetrahydroxynaphthalene, which spontaneously oxidizes to form 2,5,7-trihydroxy-1,4-naphthoquinone (flaviolin). In this study, we report the in vivo expression and in vitro synthesis of flaviolin from purified gene product (Sp-RppA).
