Visualization of Water-uptake Process in Excised Roots of Arabidopsis using Synchrotron X-ray Imaging Technique (가속기 X선 영상기법을 이용한 애기장대 뿌리털의 물 흡수과정 가시화)

  • Water-uptake through roots, is an essential process of the water flow in plants. Its visualization is very useful for understanding sap flow dynamics at whole plant level. In this study, the tips of Arabidopsis' root hairs were excised and exposed to repeated dehydration and rehydration processes. The water-refilling through individual xylem vessels was visualized using the synchrotron X-ray micro-imaging technique. The high temporal resolution ($2\;{\mu}m$) and beam intensity of the X-ray source allowed to acquisition of consecutive X-ray images of the water-refilling process up to 10 frames/sec. Various flow patterns were observed and the ascending speed of the water-air interfaces was analyzed. The relation between the water-rising height and ascending speed was also analyzed. The present results would provide better alternative for investigating sap flows in roots.

  • 최근 포항 방사광 가속기 연구소에 미세구조 X-선 영상 실험을 위한 5C1 방사광(Synchrtoron Radiation) 빔라인이 건설되었다. 광대역의 에너지 스펙트럼을 가진 방사광 X-선이 물체를 투과한 후 CdWO$_{4}$ scintillator에 의해 가시광선으로 바뀌고, 그 빛을 CCD 카메라로 받아들여 영상을 획득하게 된다. 방사광 X-선은 일반 의료진단용 X-선에 비하여 위상이 일치하고, 평행하며, 그 양이 풍부한 특성들을 갖고 있다. 방사광 영상시스템과 X-선 유방촬영 시스템에서 영상을 획득하여 영상특성들을 비교, 분석하였다. 고-분해능 X-선 시험 패턴(20 line pairs mm$^{-1}$), 유방촬영 팬텀, 파라핀에 고정한 인체 유방암조직과 포르말린에 고정한 인체 유방암조직, 그리고 capillary tube내 micro-bubbles등의 방사광 영상은 기존의 X-선 유방촬영시스템에서 얻은 영상보다 분해능이 뛰어나고 영상질도 우수하였다. 방사광 X-선 영상시스템은 micrometer 공간 분해능 영상을 획득할 수 있어 많은 기초분야의 영상연구와 의료영상분야에서도 활발하게 활용될 것으로 기대된다.

Competition between Phase Separation and Crystallization in a PCL/PEG Polymer Blend Captured by Synchronized SAXS, WAXS, and DSC

  • We conducted simultaneous, small-angle, X-ray scattering/differential scanning calorimetry (SAXS/DSC) and simultaneous, wide-angle, X-ray scattering (WAXS)/DSC measurements for a polymer blend of poly($\varepsilon$-caprolactone)/poly(ethylene glycol)(PCL/PEG). The time-dependent SAXS/DSC and WAXS/DSC results, measured while the system was quenched below the melting temperature of PCL from a melting state, revealed the competitive behavior between liquid-liquid phase separation and crystallization in the polymer blend. The time-dependent structural evolution extracted from the SAXS/WAXS/DSC results can be characterized by the following four stages in the PCL crystallization process: the induction (I), nucleation (II), growth (III), and late (IV) stages. The influence of the liquid-liquid phase separation on the crystallization of PCL was also observed by phase-contrast microscope and polarized microscope with 1/4$\lambda$ compensator.

Phase Transition Study on Graphite at Room Temperature (고압하에서 방사광을 이용한 흑연에 대한 연구)

  • High pressure X-ray diffraction study was carried out on a polycrystalline graphite to investigate the phase transition(s) at room temperature. Energy dispersive X-ray diffraction method was employed using a Mao-Bell type diamond anvil cell with an Wiggler synchrotron Radiation at the National Synchrotron Light Source. Sodium chloride power was used as the internal pressure sensor for the high pressure determinations as well as the pressure medium for quasihydrostatic pressure environment. Graphite transforms into a hexagonal didose not agree with the previously reported observations and this phase persists when pressure is released down to 0.1 MPa. This result dose not agree with the previously reported observations and this discrepancy would be due to the kinetics in phase transition as well as the uniaxially oriented pressure field in the diamond anvil cell.

Interfacial Diffusion in Fe/Cr Magnetic Multilayers Studied by Synchrotron X-ray Techniques (방사광 x-선 기법에 의한 다층형 Fe/Cr 자성박막의 계면확산 연구)

  • We have studied the interfacial diffusion of Fe/Cr multilayers using synchrotron x-ray techniques, such as x-ray reflectivity, extended x-ray absorption fine structures (EXAFS), and high-resolution x-ray scattering. The results of x-ray reflectivity indicated that the interfacial roughness of Fe/Cr multilayers increased with the Cr-layer thickness. The Fourier transform (FT) of EXAFS data clearly showed that the Fe atoms dominantly diffused into the stable Cr layers at the Fe/Cr interface. The results of high-resolution x-ray scattering supported the interfacial diffusion of Fe atoms. Out study revealed that the dominantly interfacial diffusion of Fe atoms into the Cr layers effects the interfacial roughness of the Fe/Cr multilayers.

  • The interfacial diffusion in Fe/Cr/MgO(001) multilayers has been studied using synchrotron x-ray techniques, such as x-ray reflectivity, extended x-ray absorption fine structures (EXAFS), and anomalous x-ray scattering (AXS). The results of x-ray reflectivity indicated that the interfacial roughness of Fe/Cr multilayers with Cr-$4{\AA}$-thick was larger than that with Cr-$4{\AA}$-thick. The results of EXAFS indicated that the Fe element dominantly diffuse into the stable Cr layers at the Fe/Cr interface. The AXS was certified the existence of the interdiffused Fe element in the Cr layers. Our study revealed that the rough interface of the Fe/Cr multilayers was caused by the interfacia diffusion of Fe element into the Cr layers.

Crystallization of Ba-ferrite/sapphire(001) Thin Films Studied by Real-Time Synchrotron X-ray Scattering

  • The crystallization of amorphous Ba-ferrite/sapphire(001) thin films was studied in real-time synchrotron x-ray scattering experiments. In the sputter-grown amorphous films, we found the existence of epitaxial $Fe_3O_4$ interfacial crystallites (50-${\AA}$-thick), well aligned $[0.03^circ$full-width at half-maximum (FWHM)] to the sapphire [001] direction. The amorphous precursor was crystallized to epitaxial Ba-ferrite and \alpha-Fe_2O_3$grains in two steps; i) the nucleation of crystalline \alpha-Fe_2O_3$ phase started at $300^circ{C}$ together with the transformation of the $Fe_3O_4$ crystallites to the \alpha-Fe_2O_3$ crystallites, ii) the nucleation of Ba-ferrite phase occurred at temperature above $600^circ{C}$. In the crystallized films irrespective of the film thickness, the crystal domain size of the \alpha-Fe_2O_3$grains was about 250 ${\AA}$ in the film plane, similar to that of the Ba-ferrite grains.

  • High pressure X-ray diffraction study was carried out on a graphite to investigate its compressibility as well as any possible phase transition to the hexagonal diamond structure at room temperature. Energy dispersive X-ray diffraction method was introduced using a Mao-Bell type diamond anvil cell with Synchrotron Radiation. Polycrystalline sodium chloride was compressed together with graphite for the high pressure determinations. Because of the poor resolution of the X-ray diffraction pattern of graphite, its compressibility was estimated to be almost same as that of NaCl by graphite (002) X-ray diffraction peak only. An observation of any new peak from a possible hexagonal diamond phase seems very unplausible for its definite identification based on the present data. Alternative approaches such as an Wiggler Radiation source as well as a Large Volume high pressure apparatus will be necessary for the detailed studies on a graphite in future.

Verification of Microstructure Qualities of ACR-Approved Mammography Phantoms by Refraction-Enhanced Synchrotron Radiation Imaging

  • Images of microcalcification specks showed large variation in conventional radiographs of phantoms which are approved for mammography image quality standard by the American College of Radiology (ACR). This kind of variation is not appropriate for image quality standards because the number of specks are visually counted in images and that number is important in image quality evaluation. Our study using synchrotron radiation (SR) imaging revealed the overlapping of micro-sized air bubble(s) to some specks, and also the structural deformation or crackings. Eight phantoms approved by ACR from two different makers and an air-bubble phantom were examined. SR imaging was performed at a synchrotron radiation facility, SPring-8, in Japan. The image-detector was a fluorescent-screen optical-lens coupling system using a CCD camera with a spatial resolution of 6 $\square$m. Objects when imaged with longer sample-to-detector distance show edge enhancement due to a difference in refraction indices, that is refraction enhancement. Refraction-enhanced SR images revealed that some of specks carried foreign objects, which were proven to be air. In phantoms provided by one maker, attaching/overlapping airs were observed for 62 out of 150 specks (41%) , with a higher incidence for the smallest specks. A speck becomes hardly visible in a conventional radiograph when air(s) overlaps the majority part of a speck, though depending on the size of the air-inclusion and on its configuration. Those airs might have been adsorbed on a speck surface before being embedded and then introduced into the matrix together with specks. Our study using SR imaging has clearly shown the nature of defects in some mammography phantoms which seriously degrade the quality as an image standard.

High Pressure X-Ray Diffraction Study on a Goethite using Synchrotron Radiation (방사광을 이용한 괴타이트에 대한 고압 X-선 회절연구)

  • High pressure X-ray diffraction study was carried out on a natural FeO(OH)-goethite to investigate its compressibility at room temperature. Energy dispersive X-ray diffraction method was employed using Mao-Bell type diamond anvil cell with Synchrotron Radiation. MgO powder was compressed together with goethite for the high pressure determinations. Bulk modullus was determined to be 147.9 GPa by the Birch-Murnaghan equation of state under assumption of K0' of 4. This value was subjected to compare with its structural analogs and related materials.
