Pathological Pictures of Pasteurella Pneumonia in Swine (돈역성폐염(豚疫性肺炎)에 관한 병리학적연구(病理學的硏究))

  • The pneumonic lungs of 51 pigs, from which the presence of pasteurella organisms was confirmed by bipolar staining, were examined pathologically. The numbers of pigs in each age group were 22 (43.1%) in 3-4 month group, 20 (39.2%) in 1-2 month group, 7 (13.7%) in 5-6 month group, and 2 (4.0%) in group of more than one year. The lungs of 16 pigs which were regarded as pasteurella pneumonia without any other manifestations were studied pathogically. Grossly, the affected lungs showed pulmonary edema, lobular consolidation and interlobular edema. Pigs over 3 months of age frequently showed chronic condition in which the entire lobe was involved as confluent pneumonia. In such pneumonic lungs, infarction and focal necrosis of the lung parenchyma and deposition of fibrinous exudate on the pleura were encountered. Histologically, the alveolar spaces were filled with fibrinous and leukocytic exudates. The interlobular septae showed marked edema and fibrinous exudate. The process of organization was frequently observed in chronic cases.

Inactivation of Pathogenic Escherichia coli Using Crude Extract of Immunized Silkworm (면역유도누에 추출물을 이용한 병원성 대장균의 불활성화)

  • Swine diarrhea is a livestock disease that causes huge economic losses to pig farms. In general, diarrhea occurs because of the proliferation of pathogenic Escherichia coli (E. coli). The toxins produced by the proliferated E. coli cause edema in pigs. Although the proliferation of these coliforms can be prevented by using a vaccine, the vaccines containing chemically produced dead bacteria are not very effective, making it difficult to control the proliferation of E. coli. Therefore, there is a need to develop new, more effective vaccines. In this study, we prepared killed F4+ and F18ab+ E. coli, which induce diarrhea and edema in pigs, using the extracts of immune-induced silkworms containing antimicrobial peptides and examined their availability as a killed-bacteria vaccine. First, the antimicrobial activity analysis of the prepared immune-induced silkworm extract was conducted using the radial diffusion assay. The results showed high activity against both F4+ and F18ab+ E. coli. The production efficiency of E. coli dead cells was determined using the colony-counting method. The concentration of the E. coli dead cells was the highest (50 mg/ml) when treated at 4℃. In addition, the analysis of the prepared dead cells using a transmission electron microscope confirmed that E. coli leaked out of the cytoplasm and the cell membrane remained intact. Therefore, F4+ and F18ab+ E. coli produced using immune-induced silkworms extract are considered to be highly available as bacterial ghost vaccines that can help prevent swine diarrhea and the resulting edema.

Pathological findings on encephalomyocarditis virus infections of swine in Korea (국내에서 발생한 돼지 뇌심근염 바이러스 감염증의 병리학적 관찰)

  • Pathological studies by light and electron microscope were carried out on the twenty piglets naturally affected by encephalomyocarditis virus infection. Gross findings included pale or yellow, small necrotic foci on myocardium, together with pulmonary edema and liver congestion in some cases. On light microscopy, nonsuppurative interstitial endocarcitis, epicarditis and myocarditis, myocardial infarction, and dystrophic calcification or fibtosis were observed in heart. Perivascular cuffings, gliosis and nonsuppurative meningitis were appeared in brain. Focal or diffuse necrosis with mononuclear cell infiltration in lacrimal gland and multifocal necrosis in liver were observed in some cases. Congestion and edema of lung, hyperemia, hemorrhage and deletion of lymphocytes of lymph nodes and spleen were recognized. On electron microscopy, severe swelling and vacuolization of mitochondria and sarcoplasmic reticulum, large intracellular vacuolation and edema, separation and fragmentation of myofibrils were observed. Virus particles were seen in the sarcoplasm of degenerated cardiac muscle cell.

  • During the studies on the esophagogastric ulcers in swine, Anisakis type larvae were found associated with stomach lesions. 1. Of the 1,531 pigs examined, 11(0.7%) were infested with the parasite in the stomach. 2. The parasite was identified as Anisakis type I larvae, according to the morphological characteristics. 3. On gross examination, the parasites were seen penetrating gastric mucosa, resulting in hemorrhagic ulcers. 4. Main histopathological features were edema, massive eosinophilic infiltrations around the parasite, necrosis and perivascular eosinophilic accumulations in the submucosa.

Investigation on the Bacterial Diseases of Pig Occurred in Korea (한국에서 발생한 돼지의 세균성 질병 조사)

  • The purposes of this investigations were survey of the occurred bacterial diseases, development of new animal health drug, guidance to formers on the treatment and control methods of diseases. Some series of investigations have been carried out by microbiological, pathological and serological examinations. The results could be summarized as follows. 1. A total of 953 cases of outbreaked swine diseases have been diagnosed in Clinical pathology laboratories, Bayer Vet Res Institute during 8 years (from 1986 to 1993). The high incidence diseases were colibacillosis, pleuropneumonia, streptococcal infection and pasteurellosis in decreasing order. 2. Pleuropneumonia caused by Actinobacillus pleuropneumoniae was the most important respiratory diseases and pasteurellosis by Pasteurella multocide could be confirmed in several cases. 3. Actinobacillus pleuropneumoniae 50 strains were isolated and identified by biochemical and serological tests. In serotyping test, 22 isolated strains were serotype 5, 21 strains as serotype 2, each 2 strains as serotype 3 and 7 by the coagglutination test. 4. Colibacillosis and edema discase caused by Escherichia coli has been the most predominant outbreaked disease in this investigations. The 100 isolates of E coli strains were sensitive to amikacin, colistin, enrofloxacin, gentamycin and trimethoprim -sulfamethoxazole. 5. Swine erysipelas caused by Erysipelothrix rhusiopathiae was confirmed 25 cases as acute septicemic forms. Isolates of E rhusiopathiae were highly sensitive to ampicillin, cephalothin, enrofloxcin, penicillin and tetracycline. 6. The 49 cases of hemorrhagic and necrotic enteritis in piglets were observed and 13 strains of Clostridium perfringens could be isolated and confirmed by biological and serological test. Isolates of Clostridium perfringens type C were highly sensitive to ampicillin, cephalothin, enrofloxacin, penicillin and trimethoprim- sulfamethoxazole. 7. The 14 strains of Streptococcus suis type II could be isolated from meningitis of piplets. 8. Polyserositis caused by Haemophilus parasuis and salmonellosis were observed and confirmed. Also Corynebacterial infections and several parasitosis have been also observed in this investigations.

  • Nephrotic syndrome is a clinical syndrome characterized by heavy proteinuria, hypoalbuminemia, edema and hyperlipidemia. Causes of idiopathic nephrotic syndrome include minimal change nephrotic syndrome (MCNS), focal segmental glomerulosclerosis (FSGS) and mesangial proliferation. Other causes of nephrotic syndrome are rare genetic disorders and secondary diseases associated with drugs, infections, or neoplasia. Since February 2009, a swine-origin H1N1 influenza virus (S-OIV) from Mexico has been spread among humans in unexpected rapidity. S-OIV is markedly different from seasonal influenza, in that many of those affected are previously healthy young people. While pulmonary complications of S-OIV infection have been frequently documented, renal complications have not been as widely recognized. We report a case of 4 year-old boy who had developed nephrotic syndrome after S-OIV infection with good response after steroid treatment.

Natural Occurrence of Fusarium Mycotoxins in Cereals (곡류에서의Fusarium속 곰팡이 독소의 오염)

  • Species of the genus Fusarium are worldwide in their distribution. They are saprophytes as soil inhabitants and parasites of cultivated plants. Some isolates of certain species car capable of producing mycotoxins. Mycotoxicoses, including moldy corn toxicosis, alimentary toxic aleukia, fusariotoxicosis, and rel mold disease are known to be associated with trichothecene-producing fungi. Historically severe epidemic of cereal scab occred in the southern part of Korea in 1963. The epidemic caused a social problem because of not only heavy economic losses but also mycotoxicoses to humans and farm animals. However the toxic principles were remained unknown until some publications in 1980's indicated that trichothecenes and zearalenone were major mycotoxins in Korea cereals contaminated with Fusarium moniliforme have been found to be associated with human and animal disease such as leukoencephalomalacia in horses, pulmonary edema in swine and esophageal cancer in humans. High concentrations of fumonisins have been detected in corn samples from high espohoageal cancer risk areas of South Africa and China. Thus fumonisins have been implicated in the etiolgy of human esophageal cancer in those high incidence countries. Recently increase of imported cereals from foreign countries demands to assess the safety of mycotoxins in Korea. The informations on the natural occurrence of mycotoxins are needed to solve the toxicological problems in humans and farm animals associated with the consumption of moldy cereals.

Pathologic studies for porcine pleuropneumonia and serotypes of the agents in Jeju (제주지역 돼지 흉막폐렴에 대한 병리학적 연구 및 원인체의 혈청형)

  • Actinobacillus (A.) pleuropneumoniae is the etiological agent of a porcine pleuropneumonia and have great economic importance to the global swine industry. For recent 5 years, a total of 50 pleuropneumonia cases of 24 pig farms were selected from pig lungs submitted to the College of Veterinary Medicine, Jeju National University using polymerase chain reaction (PCR) analysis. Collected lungs were fixed in 10% neutral phosphate-buffered formalin and processed for histological examination. Serotypes of A. pleuropneumoniae in pneumonic lesions were analyzed by PCR methods. And the antimicrobial susceptibility of A. pleuropneumoniae isolates was determined by a disc diffusion test. Grossly, unilateral distribution of hemorrhagic or necrotic pneumonic lesions was more common than bilateral distribution in lungs. In peracute or acute cases, histopathologic changes were characterized by necrosis, hemorrhage, neutrophils infiltration, vascular thrombosis, widespread edema and fibrinous exudates. Following the acute response, macrophage infiltration, marked fibrosis around zonal necrotic areas, and marked fibrous pleuritis were characteristic in chronic cases. A total of 50 pleuropneumonia were associated with A. pleuropneumoniae serotype 5 in 46 cases (92%), serotype 2 in 3 cases (6%), and both 2 and 5 in 1 case (2%). More than 90% of collected isolates showed high sensitivity to ceftiofur, amoxicillin, and colistin. However, ampicillin, penicillin, and tylosin showed low susceptibility. The results of this study demonstrated that A. pleuropneumoniae serotype 5 was predominant at porcine pleuropneumonia cases in Jeju.

Virulence factors and antimicrobial resistance patterns of pathogenic Escherichia coli isolated from fecal samples of piglets (자돈 분변 유래 병원성 대장균의 병원성 인자 및 항생제 내성 양상)

  • Pathogenic Escherichia coli is the cause of a wide range of diseases in pigs, including diarrhea, edema disease, and septicemia. Diarrhea caused E. coli may result in significant economic losses, making pathogenic E. coli an important pathogen for the swine industry. This study investigated the prevalence of virulence factor genes, antimicrobial resistance phenotypes, and resistance genes in E. coli isolated from feces of piglets in Korea between 2017 and 2020. As a result, 119 pathogenic E. coli isolates were obtained from 601 fecal samples. The F4 adhesin gene and the STb enterotoxin gene were commonly present in E. coli isolated from diarrhea samples. The dominant virulotypes of isolates from diarrhea samples were STb, Stx2e, and F4:LT:STb. More than 80% of the screened isolates were resistant to ampicillin, sulfisoxazole, chloramphenicol, or tetracycline. To confirm the resistance mechanisms for β-lactam or quinolone, we investigated the genotypic factors of resistance. Each of the ceftiofur-resistant E. coli produced an extended-spectrum β-lactamase encoded by blaCTX-M-14, blaCTX-M-27, and blaCTX-M-55. And all ciprofloxacin-resistant E. coli harbored mutations in quinoloneresistance-determining-regions. In addition, some of the ciprofloxacin-resistant E. coli contained the plasmid-mediated-quinolone-resistance genes such as qepA, qnrB1, or qnrD. This study has confirmed that the F4 fimbria and the STb enterotoxin are the most predominant in pathogenic E. coli isolated from piglets with diarrhea in Korea and there is a great need for responsible and prudent use of antimicrobials to treat colibacillosis.

Effect of Antibody Titer on Xenograft Survival in Pig-To-Dog Heterotopic Cardiac Xenotransplantation -Opening of Xenotrasplantation Era- (돼지\longrightarrow개 이소이종심장 이식모델에시 생존에 미치는 항체 역가의 영향 -이종이식시대의 개막-)

  • Xenotransplantation in discordant species results in immediate and irreversible hyperacute rejection due to natural antibodies, IgM. With this, antibody depletion is one option to reduce hyperacute rejection, we investigated the effect of PCPP (postcentrifugal plasmapheresis) on the depletion of natural antibodies and the effect of antibody titer on xenograft survival. Material and Method: Outbred swines (n=4) weighing 10∼20 kg were used as donors and mongrel dogs (n=4) weighing 25∼30 kg were used as recipients. Recipient canines underwent plasmapheresis (COBE TPE Laboratories, Lakewood. CO, USA). Pre-transplantation PCPP was peformed on day -2 and day 0. There were three groups (Group 0: no PCPP, Group 1: 1 pla sma-volume (PV) at day -2 and 2 PV at day 0, Group 2: 2 PV at day -2 and 2 PV at day 0). A swine heart was heterotopically transplanted into a recipient's abdominal infrarenal aorta and inferior vena cava. Mean percent depletion of total IgM and IgG in plasma of the recipients was calculated. Serum albumin, electrolyte, complement activity and coagulation factors were measured. Histopathologic examination of heart specimens was performed. Result: Mean percent depletion of IgM and IgG were 95.7$\pm$1.2%, 80.5$\pm$2.4% in the group 2 at the end of PCPP. The percent depletion of serum albumin concentration was decreased from 2.8 to 1.4 g/㎗ in the group 1 and 3.0 to 1.5 g/㎗ in the group 2. Complement hemolytic activity was decreased in group 1 and 2, but returned to normal level within 24 hours. Complement hemolytic activity was reduced to 10% of pre-PCPP level in group 2. Serum fibrinogen decreased to 20% or less and was recovered within 24 hours in group 2. Antithrombin III decreased but less than fibrinogen. PT and aPTT were sometimes but not always prolonged during plasmapheresis. After plasmapheresis, PT and aPTT were prolonged beyond the measurable level. D-dimer was not found during PCPP, but appeared and maintained from 10 minutes after trasplantation. Graft Survival time was 5 min in group 0, and it was 90$\pm$0 min in the group 2. Histopathologic changes were more typically characterized by edema, hemorrhages, thrombosis in all groups at the end of experiment. Conclusion: PCPP effectively removed immuoglobulins and reduced the titer of natural antibodies, as a result, significantly prololonged swine heart xenograft survival.
