Three swine farms which were suffering from slight economic loss due to suckling piglets' diarrhea, were selected to apply commercialized multivalent vaccine for sow use; $SUISENG^{(R)}$ (Hipra, Spain). Farms were pre-diagnosed with clinical symptoms and molecular detection of C. perfringens Type A and C and E. coli pili by PCR. Sows were vaccinated twice 2 ml of the vaccine at 6 and 3 weeks ante partum intramuscularly according to the manufacturer's instruction. All vaccinated sows did not show any adverse reaction or clinical signs; hypersensitivity, fever, granuloma or abscess on the injection site, appetite loss, and so on. Also, no reproductive disorder was appeared in vaccinated sows compared with non-vaccinated control sows. The results suggested that piglets born from vaccinated sows show significantly better performance in regard of the diarrhea index and mean daily weight gain compared with piglets from non-vaccinated sows. Therefore, the commercial vaccine for the prevention of neonatal diarrhea is found to be effective in reducing diarrhea in the first suckling period of piglets after birth.
Porcine epidemic diarrhea (PED), transmissible gastroenteritis (TGE) are an acute viral enteritis. colibacillosis by E coli is a microbial enteritic disease in suckling piglets[1]. These infectious intestinal diarrheal diseases cause severe diarrhea to suckling piglets, so that lead to enormous economical loss in swine-product industries. Ig-Top (AD Biotech, Korea) is a immunomodulator with IgY the specific yolk-antibody for PED, TGE and E. coli and oligosaccharide. Reproductive performance of gilts and sows in ordinary piggeries was investigated with special reference to litter size, piglets weaned per litter, the cause of preweaning mortality and the incidence of diarrhea. Biochemical properties of Escherichia coli isolated from piglets with diarrhea were also determined. The results obtained are summarised as follows: 1. Of the 2,226 gilts and sows farrowed, average number of piglets born alived and weaned piglets per litter were 8.9 and 7.1, respectively, indicating that both gilts and sows lost more than 20% of all their piglets prior to weaning. 2. The causes of preweaning mortality in order of prevalence were diarrhea (39.3%), pneumonia (20.0%), crushing (13.8%), starvation (11.0%) and born weak (10.3%). 3. Incidence of 3 diarrheal syndromes of piglets, i.e. 1 week diarrhea, 3 week diarrhea and post-weaning diarrhea were 18.4%, 66.1% and 15.5%, respectively, showing that most farms were suffering from so called 'white scours' in piglets 14~28 days old. 4. Biochemical properties of 268 cultures of E. coli isolated from diarrheal piglets were tested and compared with those of standard strains of porcine origin. All those properties of isolates were matched to standard E. coli teated while variable results were obtained with haemolytic capabilities of cultures tested. 5. Of 147 isolates 16 cultures (10.9%) were identified as colicin producers.
Molecular diagnostic techniques have been used to identify porcine epidemic diarrhea virus (PEDV), a causative agent of acute enteritis in swine, but they were difficult to be petformed and time-consuming. To detect PEDV in a rapid and easy way, we developed biotinylated cDNA probe for N gene encoding the nucleoproteins of PEDV. Formalin-fixed and paraffin-embedded tissues from 24 naturally infected pigs were used for the experiment. The ISH produced a positive reaction in all cases. When intestinal tissues were hybridized with PEDV probe, strong signals were seen in the villus enterocytes of the jejunum and ileum. Hybridization signals were also found in the duodenum from one pig and in colon from dnother. In conclusion, ISH with a biotinylated cDNA probe was provided to be a useful diagnostic method for detecting PEDV effectively in routinely processed tissue sections.
Immunohistochemical study on the intestinal tissues obtained from the 21 pigs of the 14 terms in Korea in which the clinical and epidemiological features had indicated the possible outbreaks of porcine epidemic diarrhea(PED) was performed using the indirect immunofluorescence test and/or the immunoperoxidase method in order to detect PED viral antigens in the infected cells of the intestines, and histopathological features were described as well. By immunohistochemical analysis, PED viral antigens were detected in the epithelial cells covering the small intestinal villi and recognized slightly in the cells lining the colonic surface epithelium as well. Occasional fluorescence was also seen in a few intestinal crypt epithelium. On light microscopy, the piglets with PED showed marked villous atrophy and fusion, and severe enterocyte degeneration and desquamation. On the other hand, the older pigs more than 4 week old age was mild villous atrophy and fusion, severe villous epithelial cell proliferation, and moderate mononuclear cell infiltration.
Porcine epidemic diarrhea virus (PEDV) is the causative agent of porcine epidemic diarrhea (PED) and causes a considerable economic loss in swine industry. In this study, the virucidal efficacy of the disinfectant composed to n-alkyl-dimethyl-benzyl-ammonium chloride (n-ADBAC) was investigated against PEDV. A virucidal efficacy was determined with the viability of PEDV contacted with the disinfectant in Vero cells. The disinfectant and PEDV were reacted on the hard water (HW) or organic matter suspension (OM) condition. On HW condition, PEDV was inactivated with 50 fold dilutions of the disinfectant. When the antiviral effect on OM condition was evaluated, the antiviral activity of the disinfectant showed on 10 fold dilutions against PEDV. As the disinfectant possesses the virucidal efficacy against PEDV, the disinfectant solution can be used to limit the spread of animal viral diseases.
Swine proliferative enteritis(SPE) caused by inかsoma intracellularis is a common enteric disaese of grower and finisher pig. Swine affected with SPE show variable clinical signs including diarrhea, weight loss, aberrant growth and death. The characteristic lesion of ileitis at necropsy is marked thickening of the last section of the small intestine. The inner lining of the thickened intestine proliferates almost like a cancer and curved rod bacteria(L intracellularis) are always seen inside the intestinal wall. Infected swine shed the organism in the feces. Isolation and growth of pure L intracellularis in vitro requires a suitable cell culture. This procedure is difficult and not a practical means of diagnosis, thus the polymerase chain reaction(PCR) test of feces can be used to determine whether a pig is shedding the infective organism. A sensitive assay based on amplification of a 319bp DffA fragment of the L intracellularis of Swine proliferative enteritis was attempted for the detection of the organism in the 62 feces of swine. L intracellularis was identified on three herds and detected in 6 fecal samples, representing a infection rate of 9.7%. The PCR was very sensitive and specific on the individual level. The PCR technique could be very useful for the diagnosis of this disease.
돼지 설사병은 부종병과 함께 양돈 농가에 막대한 경제적 손실을 입히는 가축질병으로 알려져 있으며, 병원성 대장균의 증식에 의하여 설사 및 대장균 생성 독소에 의한 부종병이 동반된다. 본 연구에서는 항균펩타이드를 포함하는 면역유도누에의 조추출물을 이용하여 돼지 설사 및 부종을 유발하는 F4+ 및 F18ab+ 대장균의 사균체를 제작하고 사균백신으로서 이용가능성을 검토하였다. 면역유도누에 추출물의 항균활성을 환확산법을 통해 분석한 결과 F4+ 및 F18ab+ 대장균에 대하여 높은 활성을 나타내었으며, 평판계수법을 이용하여 대장균의 사균체 생산효율을 분석한 결과 50 mg/ml의 농도를 4℃에서 처리했을 때 가장 높게 나타났다. 또한 제작된 사균체에 대해서 투과전자현미경을 이용하여 분석한 결과 세포질은 유출되고 세포막만이 남아있는 대장균이 확인되었다. 따라서 면역유도누에 추출물을 이용하여 제작된 F4+ 및 F18ab+ 대장균은 부종병 예방을 위한 사균백신으로서 이용가능성이 매우 높을 것으로 판단된다.
Pathogenic Escherichia coli is the cause of a wide range of diseases in pigs, including diarrhea, edema disease, and septicemia. Diarrhea caused E. coli may result in significant economic losses, making pathogenic E. coli an important pathogen for the swine industry. This study investigated the prevalence of virulence factor genes, antimicrobial resistance phenotypes, and resistance genes in E. coli isolated from feces of piglets in Korea between 2017 and 2020. As a result, 119 pathogenic E. coli isolates were obtained from 601 fecal samples. The F4 adhesin gene and the STb enterotoxin gene were commonly present in E. coli isolated from diarrhea samples. The dominant virulotypes of isolates from diarrhea samples were STb, Stx2e, and F4:LT:STb. More than 80% of the screened isolates were resistant to ampicillin, sulfisoxazole, chloramphenicol, or tetracycline. To confirm the resistance mechanisms for β-lactam or quinolone, we investigated the genotypic factors of resistance. Each of the ceftiofur-resistant E. coli produced an extended-spectrum β-lactamase encoded by blaCTX-M-14, blaCTX-M-27, and blaCTX-M-55. And all ciprofloxacin-resistant E. coli harbored mutations in quinoloneresistance-determining-regions. In addition, some of the ciprofloxacin-resistant E. coli contained the plasmid-mediated-quinolone-resistance genes such as qepA, qnrB1, or qnrD. This study has confirmed that the F4 fimbria and the STb enterotoxin are the most predominant in pathogenic E. coli isolated from piglets with diarrhea in Korea and there is a great need for responsible and prudent use of antimicrobials to treat colibacillosis.
Swine influenza virus (SIV) or swine-origin influenza virus (S-OIV) is endemic in swine, and classified into influenza A and influenza C but not influenza B. Swine influenza A includes H1N1, H1N2, H3N1, H3N2 and H2N3 subtypes. Infection of SIV occurs in only swine and that of S-OIV is rare in human. What human can be infected with S-OIV is called as zoonotic swine flu. Pandemic 2009 swine influenza H1N1 virus (2009 H1N1) was emerged in Mexico, America and Canada and spread worldwide. The triple-reassortant H1N1 resulting from antigenic drift was contained with HA, NA and PB1 of human or swine influenza virus, PB2 and PA polymerase of avian influenza virus, and M, NP and NS of swine influenza virus, The 2009 H1N1 enables to transmit to human and swine. The symptoms and signs in human infected with 2009 H1N1 virus are fever, cough and sore throat, pneumonia as well as diarrhea and vomiting. Co-infection with other viruses and bacteria such as Streptococcus pneumoniae can occur high mortality in high-risk population. 2009 H1N1 virus was easily differentiated from seasonal flu by real time RT-PCR which contributed rapid and confirmed diagnosis. The 2009 H1N1 virus was treated with NA inhibitors such as oseltamivir (Tamiflu) and zanamivir (Relenza) but not with adamantanes such as amantadine and rimantadine. Evolution of influenza virus has continued in various hosts. Development of a more effective vaccine against influenza prototypes is needed to protect new influenza infection such as H5 and H7 subtypes to infect to multi-organ and cause high pathogenicity.
