산업용 고구마 개발을 위한 유전자원 현황 및 전망 (Genetic resources of sweetpotato for industrial use)

  • In many countries including China and U.S., researchers are developing methods to use sweetpotato as raw material for biofuel. We consider the sweetpotato is not only a source of green fuel, it eventually will provide various material including paper, adhesives, biodegradable plastics, and secondary metabolites. Sweetpotato is one of the high efficiency crop because it yields more calories per unit area than either maize or potato, and it requires the shortest growing cycle of the root crops grown in the tropics. Sweetpotato is the most useful crop for the coming starchbased industry era. Sweetpotato genetic resources are collected, characterized, evaluated, and maintained by U.S., China, Japan, and the International Potato Center. New varieties of sweetpotato using the proper genetic resources and molecular breeding will be developed to cope with the global food and energy in 21st century.

Expression Analysis of Sweetpotato Sporamin Genes in Response to Infection with the Root-Knot Nematode Meloidogyne incognita

  • Sweetpotato (Ipomoea batatas [L.]) is a globally important root crop cultivated for food and industrial processes. The crop is susceptible to the root-knot nematode (RKN) Meloidogyne incognita, a major plant-parasitic RKN that reduces the yield and quality of sweetpotato. Previous transcriptomic and proteomic analyses identified several genes that displayed differential expression patterns in susceptible and resistant cultivars in response to M. incognita infection. Among these, several sporamin genes were identified for RKN resilience. Sporamin is a storage protein primarily found in sweetpotato and morning glory (Ipomoea nil). In this study, transcriptional analysis was employed to investigate the role of sporamin genes in the defense response of sweetpotato against RKN infection in three susceptible and three resistant cultivars. Twenty-three sporamin genes were identified in sweetpotato and classified as group A or group B sporamin genes based on comparisons with characterized sweetpotato and Japanese morning glory sporamins. Two group A sporamin genes showed significantly elevated levels of expression in resistant but not in susceptible cultivars. These results suggest that the elevated expression of specific sporamin genes may play a crucial role in protecting sweetpotato roots from RKN infection.

고구마 품질평가 현황과 전망 (Current Status and Prospect of Quality Evaluation in Sweetpotato)

  • The sweetpotato, Ipomoea batatas L. (Lam.), is one of the important summer upland crops in Korea and has been used as human food, industrial yaw material and vegetable. Sweetpotato has been consumed for human foods such as boiled, roasted, fried or salad etc. It should be developed for higher quality as a snack or health food, primarily through improving the eating and marketing qualities as well as nutritional value. Its quality after cooking or processing is a complex one combining the aroma, taste, texture and fiber content. The other important qualities for consumers are root shape, size, skin color, flesh color, insect and disease resistance, nutritional components and safety from phytoalexins(toxic stress metabolites) etc. Korean people generally prefer to red skin color, round or elliptic shape and dry texture, yellow flesh color of sweetpotato which is high in starch content including vitamins and nutrients. The almost factors of quality components of sweetpotato are genetically controlled by breeder, but postharvest handlings and marketing management for making high quality goods should be done thoroughly according to the quality evaluation criteria of sweetpotato from the moment of harvest until shipping them to the market by farmers and the other users. This paper describes current status and prospects of the quality evaluations and researches in sweetpotato roots in Korea.

고구마의 생산과정에서 발생하는 탄소배출량 산정 및 전과정평가 (Estimation of Carbon Emission and LCA (Life Cycle Assessment) From Sweetpotato (Ipomoea batatas L.) Production System)

  • 고구마 생산체계의 탄소성적을 평가하기 위하여 LCI database 구축하고 전과정 영향평가를 통한 잠재적 환경영향을 평가하였다. 인벤토리 목록구축을 위한 자료 수집 결과 고구마의 투입물 중 유기질비료의 투입비가 71% 매우 높았고, 화학비료는 22%, 투입되는 에너지의 6%의 순이었다. 유기질 비료 투입량은 3.26E-01 kg $kg^{-1}$ sweetpotato, 무기질비료는 1.02E-01 kg $kg^{-1}$ sweetpotato, 고구마를 재배할 때 발생되는 직접배출 ($CO_2$, $CH_3$, $N_2O$)은 2.47E-02 kg $kg^{-1}$ sweetpotato였다. 탄소성적은 4.05E-01 kg $CO_2$-eq. $kg^{-1}$ sweetpotato이며, $CO_2$의 배출량이 2.88E-01 kg $CO_2$-eq. $kg^{-1}$ sweetpotato로 전체 온실가스배출 중 71%를 점유하였고, $CH_4$ 18%, $N_2O$가 11%였다. 이들의 공정별 기여도 분석결과 $CO_2$는 비료생산공정과 고구마생산에서 주로 발생하였고, 기여도는 약 32%, 28%였다. $N_2O$는 고구마를 재배할 때 가장 많이 발생되었고, 기여도는 약 90%였다. 전과정 영향평가 결과 비료생산은 모든 영향범주에서 가장 큰 기여도를 나타내었다. GWP (지구온난화범주)에서 고구마재배에 의한 기여가 약 12% 정도였으며, 비료생산은 약 90%였다. GWP와 POCP (광화학산화물생성) 범주의 특성화 값은 각각 4.05E-01 $CO_2$-eq. $kg^{-1}$, 5.08E-05 kg $C_2H_4$-eq. $kg^{-1}$이었다.

고구마 유전체 연구현황 및 전망 (Current status of sweetpotato genomics research)

  • 고구마는 척박한 환경에서도 생육이 가능한 세계 7대 농작물로 식량뿐만 아니라 사료용, 전분 등의 산업용으로도 중요하다. 최근 고구마는 항산화물질, 식이섬유질 등을 고함유하는 건강식품으로 각광을 받고 있다. 그러나 고구마 유전체 해독에 관한 연구는 고구마의 중요도에 비해 많이 이루어지지 않고 있다. 본 총설의 목적은 고구마 유전체 연구 동향분석을 통하여 유전체 해독 연구의 효율성 증대 및 유용형질 유전자의 실용화 연구를 위한 기반구축을 모색하는데 있다. 최근 NGS 분석을 통한 동식물 유전체해독이 급진적으로 많이 이루어지고 있다. 고구마 유전체 해독의 경우는 다배수성 문제와 이질유전체 문제로 유전체 완전해독 연구가 이루어지지 않고 있으며 반면 전사체 분석 연구는 활발히 이루어지고 있는 실정이다. 최근 2015년 일본 연구자들에 의해 2배체 고구마의 유전체 해독 초안이 보고되었다. 한중일 고구마 연구협의회(Trilateral Research Association of Sweetpotato, TRAS)에 의해 6배체 고구마 Xushu 18의 유전자지도 작성 및 유전체 해독 연구가 2014년부터 이루어지고 있다. 빌게이츠재단(Bill & Melinda Gates Foundation)은 사하라사막 남쪽 아프리카지역의 기근과 영양문제를 해결하기 위해 고구마 유전체 기반 분자육종을 위한 분자도구 개발에 관한 프로젝트를 미국을 중심으로 한 컨소시엄을 구성하여 출범하였다. 고구마 유전체 해독과정 중에 분석된 고구마 엽록체 유전체 분석을 통하여 진화학적 해석연구가 이루어지고 있다. 본 총설을 통하여 고구마 유전체 해독 연구동향을 살펴보았다. 이러한 연구 동향 분석은 고구마의 생산성 및 기능성 향상 등의 실용화 연구를 수행하는 연구자들에게 최근의 연구현황을 제공할 수 있을 것이며 세계적인 식량, 에너지, 환경문제의 해결에 크게 기여 할 것으로 생각된다.

Morphological Traits of S598A Sweetpotato as an Industrial Starch Crop

  • Sweetpotato is one of the important starch crops, current more considered as an industrial crop rather than food because it has higher starch content (over 80% of biomass), it is used for bio resources for industrial area. In this study, we generated S598A (a mutant gene of oat phytochrome A) sweetpotato plant using Agrobacterium-transformation method. Morphological characteristics of S598A plant were compared with the wild type sweetpotato, S598A had darker green leaves, increased chlorophyll content higher than to two-fold, delayed leaf senescence, shorter plant height (60% shorter than that of the wild type), more number of leaves and petioles about 1.8-fold, shorter petiole length (30% shorter), 1.2-fold more branches and 1.6-fold thicker stem diameters. From this study, S598A plants with such phenotypic characteristics might be able to use the solar energy efficiently, to have increased tolerance to biotic and abiotic stresses and finally to increase productivity (not only starch yield but also root biomass yield). S598A sweetpotato lines are under field trials.

Use of Single-leaf Cutting in the Study of the Expression of Starch Synthesis and Modification Genes in Sweetpotato

  • The evaluation of source potential and sink strength is the generally large and laborious sample size required to adequately assess anyone of the parameters in field-grown sweetpotato. For this purpose we used the rooted single-leaf cuttings with petioles, because the source and sink organs are restricted in this system. The rooted single-leaf cutting of sweetpotato provides a unique source-sink model system, and is established within about 50 days after planting. In this study, the sink potential of sweetpotato tubers was examined based on the expression of genes for starch synthesis (AGPase) and modification (SBEII and GBSSI) in single rooted leaf plant. The gene expression patterns of GBSSI, SBEII and AGPase at various developmental stages and in different types of root tissues presented. These results suggest that the rooted single-rooted method can be used an ideal model system to study physiological and biochemical mechanisms in sweetpotato.

In Vitro and In Vivo Inhibitory Effects of Gaseous Chlorine Dioxide Against Diaporthe batatas Isolated from Stored Sweetpotato

  • Chlorine dioxide ($ClO_2$) can be used as an alternative disinfectant for controlling fungal contamination during postharvest storage. In this study, we tested the in vitro and in vivo inhibitory effects of gaseous $ClO_2$ against Diaporthe batatas SP-d1, the causal agent of sweetpotato dry rot. In in vitro tests, spore suspensions of SP-d1 spread on acidified potato dextrose agar were treated with various $ClO_2$ concentrations (1-20 ppm) for 0-60 min. Fungal growth was significantly inhibited at 1 ppm of $ClO_2$ treatment for 30 min, and completely inhibited at 20 ppm. In in vivo tests, spore suspensions were drop-inoculated onto sweetpotato slices, followed by $ClO_2$ treatment with different concentrations and durations. Lesion diameters were not significantly different between the tested $ClO_2$ concentrations; however, lesion diameters significantly decreased upon increasing the exposure time. Similarly, fungal populations decreased at the tested $ClO_2$ concentrations over time. However, the sliced tissue itself hardened after 60-min $ClO_2$ treatments, especially at 20 ppm of $ClO_2$. When sweetpotato roots were dip-inoculated in spore suspensions for 10 min prior to treatment with 20 and 40 ppm of $ClO_2$ for 0-60 min, fungal populations decreased with increasing $ClO_2$ concentrations. Taken together, these results showed that gaseous $ClO_2$ could significantly inhibit D. batatas growth and dry rot development in sweetpotato. Overall, gaseous $ClO_2$ could be used to control this fungal disease during the postharvest storage of sweetpotato.

  • Chlorine dioxide ($ClO_2$) has been widely used as an effective disinfectant to control fungal contamination during postharvest crop storage. In this study, Fusarium oxysporum f. sp. batatas SP-f6 from the black rot symptom of sweetpotato was isolated and identified using phylogenetic analysis of elongation factor 1-${\alpha}$ gene; we further examined the in vitro and in vivo inhibitory activities of $ClO_2$ gas against the fungus. In the in vitro medium tests, fungal population was significantly inhibited upon increasing the concentration and exposure time. In in vivo tests, spore suspensions were drop-inoculated onto sweetpotato slices, followed by treatment using various $ClO_2$ concentrations and treatment times to assess fungus-induced disease development in the slices. Lesion diameters decreased at the tested $ClO_2$ concentrations over time. When sweetpotato roots were dip-inoculated in spore suspensions prior to treatment with 20 and 40 ppm of $ClO_2$ for 0-60 min, fungal populations significantly decreased at the tested concentrations for 30-60 min. Taken together, these results showed that $ClO_2$ gas can effectively inhibit fungal growth and disease development caused by F. oxysporum f. sp. batatas on sweetpotato. Therefore, $ClO_2$ gas may be used as a sanitizer to control this fungus during postharvest storage of sweetpotato.

Characteristics of Growth and Yield by Varieties of Sweetpotato(Ipomoea Batatas L.) Cultivated in Paddy Field

  • In recent, a demand for sweetpotato cultivation technology to expand the cultivated area of field crops in paddy fields is increasing. This research was carried out to establish suitable varieties and cultivation techniques for mass production of sweetpotato for processing raw materials. For the selection of varieties suitable for cultivation in rice fields for each processing purpose, 12 varieties in 2018 (8 varieties for starch, 4 as dried, chips and beverages), and 6 varieties in 2019 (4 varieties for starch including 'Daeyumi'; chips, semi-dried 'Pungwonmi'; beverage and coloring 'Shinjami') were used. Sweetpotato stems were planted in mid-May and harvested after 120 days to investigate the yield. Results revealed that the yield of sweetpotato (2019) for starch production, varied with variety as 'Gogeonmi' 3,926 > 'Jinhongmi' 3,428 > 'Daeyumi' 2,873 > 'Singeonmi' 2,752 kg/10a. The starch content was 20.2% in 'Daeyumi', 18.2 in 'Gogeonmi', 21.2 in 'Singeonmi', and 20.6% in 'Jinghongmi'. The total amount of starch was higher in 'Daeyumi' (730 kg/10a) and 'Gogeonmi' (731 kg/lOa). The yield of chips and edible varieties 'Pungwonmi' was 4,688 kg/10a. The yield of 'Shinjami' of purple variety such as beverages and powder was 3,139 kg/10a. Aaa result of evaluation sweetpotato yield by waterlogging treatments on different growing stages in paddy fields, the yields of 'Daeyumi' and 'Jinhongmi' varieties treated with waterlogging at the storage root formation stage decreased by 11.8% and 11.7%, respectively, compared to the control. In the case of waterlogging treatment at storage root swelling stage, Both varieties showed the lowest yield reduction at 7.0% and 4.8%, respectively. Based on these results, stable production and substitution effect of processing raw materials can be expected by cultivating sweetpotato varieties suitable for paddy cultivation.
