As a series of studies to investigate the effect of immunosuppression on Ascaris suum infection in undefinitive hosts, a delicate relationship between host and parasite, in the present studies golden hamsters were alloted to experiment 1(normal undefinitive host group) and experiment 2(immunosuppressive group treated with prednisolone acetate) and inoculated with a single dose of 1,500 embryonated Ascaris suum eggs. The recovery rates, sizes and features of the larvae and immunological responses in the hamsters were chronologically monitored according to somatic migration. In both experiments, the larvae failed to develop into the adults, but the more and larger larvae were observed for a longer period from experiment 2 in comparison with experiment 1. The numbers of the mast cells in the small intestinal mucosa and mesenteric lymph nodes, of the goblet cells in the small intestinal mucosa and of T-cells in the mesenteric lymph nodes, spleens and cardiac blood from experiment 2 were fewer than those from the experiment 1. In general, increasing of these cells followed by expulsion of the worms in the both groups. Profound leukopenia due to lymphopenia was found through trial period in experiment 2. Considering the experimental results, development or expulsion mechanism of somatic migrant larvae may be related to lymphopenia and temporary increasing tendency of the mast cells, the goblet cells and T-cells. In addition, patent infection of A suum in the hamsters was not obviously observed in spite of immunosuppression by prednisolone acetate.
In the present study, the effect of primary infection to reinfection with Ascaris suum larvae was experimented in mouse model. Mice were challenged with 1,000 infective stage eggs of Ascaris suum. The embryonated eggs were directly introduced into stomach of mice. Reinfection was performed at 50 days after the primary infection with same method as primary infection. Mice were sacrificed 3, 5, 7, 10, 15 and 20 days after infection in both groups respectively. Larvae collected from livers and lungs with Baermann's apparatus were enumerated and measured after sacrifice. Sera of mice were also collected at same time. The results of the experiment were as follows: With antigen prepared from coelomic fluid of adult Ascaris suum and sera collected from mice before reinfection, the production of antibody in experimental mice was confirmed by the gel-diffusion technique. In the livers of reinfected mice, the larvae were recovered up to 10 days after challenge, otherwhile in the primary infected mice, the larvae were observed up to 7 days. The maximum number of larvae were observed in the lungs of primary infected mice on 10 days after inoculation. In the lungs of reinfected mice, maximum number of larvae were recovered on 7 days after, only few larvae were recovered on 10 days after reinfection. As regards the growth of the larvae, the third stage larvae, over $500{\mu\textrm{m}}$ in length, appeared in livers at 5 days after reinfection, but it couldn't be found on 7 days and 10 days after challenge. The third stage larvae continuously developed were observed in lungs of mice from 5 days after reinfection. In conclusion, it was found that development of larvae in livers of immune mice were probably repressed by the immune mechanisms being rises in livers and defence mechanism is also acting by interfering with the process of larval penetration into the lung from the liver.

The recovery rates, sizes and morphology of the larvae and immunological responses in the rabbits were chronologically monitored according to somatic migration. In both experiments, the larvae failed to develop into the adults, but young adults in the experiment 2 grew somewhat faster and survied later than those in the experiment 1. The mast cells of small intestinal mucosa and mesenteric lymph nodes and the goblet cells of small intestinal mucosa in the worm detected cases of experiment 2 decreased remarkably in number comparing with those of experiment 1. Considering the experimental results. the expulsion mechanism of somatic migrant larvae may he related to the temporary increasing tendency of the mast cells, the goblet cells, T-cells of mesenteric lymph nodes and spleens, eosinophils in peripheral blood, degranulation rates of peritoneal mast cells and the migration inhibition rates of leucocytes. In addition, patent infection of A suum in the rabbits was not obviously observed despite of immunosuppression by prednisolone acetate.
Livestock excretes are frequently used in organic farming to produce vegetables and crops. However, zoonotic parasite eggs can be contaminated into such products and people can be infected with parasites of animal origin. Sporadic zoonotic infections with Ascaris suum have been reported even in developed countries, such as North America, Denmark, and the United Kingdom. Ascaris eggs are resistant to most adverse environmental conditions, and pigs and humans become infected by ingestion of fecally excreted eggs through contaminated food, water, or soil. This article discusses the resistant nature of ascarid eggs against harsh environment, and conditions of kimchi to be safe from consuming viable A. suum eggs.
This study was carried out to investigate the prevalence of Ascaris suum infection of slaughtered pigs in Chungju and Eumseong area from January to June in 2007. The percentage of pigs and farms detected milk spot, adult worm, and adult worm in detected milk spot were 4.2% and 23.8%, 0.2% and 9.3%, and 4.9% and 39%, respectively. The prevalence of A suum infection from April to June 2007 was lower than that of January to March 2007. There is not enough of difference in farms' infection rate based on their breeding type.
마우스에의 돼지회충(Ascaris strum) 또는 선모충(Trichinella spiralis) 감염이 sRBC에 대한 면역능에 미치는 영향을 조사한 다음 전처치로서 선모충을 감염시키거나 또는 cyclophosphamide를 투여하고 나서 돼지회충란을 경구투여하여 그 감염상태와 소장점막내 비만세포와의 관련성 그 리고 비특이 세포성 및 체액성 면역능을 함께 관찰하였다. 마우스에 1,000개의 돼지회충란을 경구투여한 바 지연형 과민반응과 로제트 형성능은 시일이 경과됨에 따라 점점 감소하기 시작하여 각각 5주, 6주에 최저치에 이른 다음 점점 상승하여 10주에 원상으로 복귀하였으며, 적현구 응집소가 및 용현소가는 점점 상승하여 3주에 최고치에 이른 다음 그 후 정상으로 복귀하였으며, 말초힐액내 호산구 및 소장접막내 비만세포 출현율은 각각 4주 및 2주에 최고치를 보였다. 한편, 유충은 1주후에 폐와 간으로부터 총 21.97%가 누수되었다. 마우스에 300마리의 선모충의 감염유충을 경구투여한 바 지연형 과민반응과 로제트 형성능은 시일이 경과 됨에 따라 점점 낮아져 각각 30일 및 21일에 전저치에 이른 다음 그 후부터 다소 높아지다가 70일 및 80일에 다시 일시적으로 낮아졌다. 그리고, 적현구 응집소가 및 용혈소가도 각각 21일 및 90일 에 다른 기보다 낮았으나, 말초혈액내 호산구 및 소장점막내 비만세포 출현율은 각각 40일째 및 14 일째에 다른 기보다 높았다. Cyclophosphamide를 400 mg/kg의 용량으로 마우스의 복륙 내에 투여한 바 체중이나 비장의 중량, 지연형 과민반응, 로제트 형성능, 적혈구 응집소가 및 용혈소가, 백현구 총수, 소장점막내 비만세포 출현율은 투여 후 1일에 비하여 5일에 현저하게 저하되었다가 10일에 다 시 증가하여 1일과 거의 비슷한 수준으로 복귀하였다. 말포혈액내 호산구 출현율은 시일이 경과함 에 따라 점점 낮아지는 경향이었다. 한편, 마우스의 복강 내에 cyclophosphamide를 투여한 다음 1일, 5일, 10일에 각각 1,000개의 감염성 돼지회충란을 투여하고 난 후 7일의 유충각수률은 1일 7.07%, 5일 14.94%, 10일 10.1%이었으며 대조군은 8.02%이었다 유모충을 마우스에 감작감염시키고 나서 각각 30일 및 70일 간격을 두고 감염성 돼지회충란을 도전감염시킨 바 참작감염후 37일과 도전감염후 7일의 시점에 있어서 지연형 과민반응과 로제트 형성능은 고도로 저하하였는데 반하여 소장점막내 비만세포 출현율은 고도로, 말초혈액내 호산구출현율, 적혈구 응집소가 및 용혈소가는 상당히 증가하였다. 이 시점에 있어서의 유충은 대조가 22.18% 회수되었는데 비하여 9.3%밖에 회수되지 않았다. 한편, 감작감염후 77일과 도전감염후 7일의 시점에 있어서 sRBC에 대한 면역능에 미치는 영향은 전자의 양상과 비슷하였는데 대조에 비하여 지연형 과민반응과 로제트 형성능이 현저하게 저하되었다. 이 시점에 있어서의 유충회수율은 대조가 10.5% 이었는데 비하여 8.3%이었다.

Ion channels are important targets of anthelmintic agents. In this study, we identified 3 types of ion channels in Ascaris suum tissue incorporated into planar lipid bilayers using an electrophysiological technique. The most frequent channel was a large-conductance cation channel (209 pS), which accounted for 64.5% of channels incorporated (n=60). Its open-state probability (Po) was ~0.3 in the voltage range of -60~+60 mV. A substate was observed at 55% of the main-state. The permeability ratio of Cl- to K+ (PCl/PK) was ~0.5 and PNa/PK was 0.81 in both states. Another type of cation channel was recorded in 7.5% of channels incorporated (n=7) and discriminated from the large-conductance cation channel by its smaller conductance (55.3 pS). Its Po was low at all voltages tested (~0.1). The third type was an anion channel recorded in 27.9% of channels incorporated (n=26). Its conductance was 39.0 pS and PCl/PK was 8.6±0.8. Po was ~1.0 at all tested potentials. In summary, we identified 2 types of cation and 1 type of anion channels in Ascaris suum. Gating of these channels did not much vary with voltage and their ionic selectivity is rather low. Their molecular nature, functions, and potentials as anthelmintic drug targets remain to be studied further.
Cecropins (Cec) are antibacterial peptides and their expression is induced in a pig intestinal parasite Ascaris suum by bacterial infection. To explore the usefulness of its activity as an antibiotic, CecP4 cDNA was prepared and cloned into the pPICZ B expression vector and followed by the integration into AOX1 locus in Pichia pastoris. The supernatants from cell culture were collected after methanol induction and concentrated for the test of antimicrobial activity. The recombinant P. patoris having CecP4 showed antimicrobial activity when tested against Staphyllococcus aureus in disc diffusion assay. We selected one of the CecP4 clones (CecP4-2) and performed further studies with it. The growth of recombinant P. pastoris was optimized using various concentration of methanol, and it was found that 2% methanol in the culture induced more antibacterial activity, compared to 1% methanol. We extended the test of antimicrobial activity by applying the concentrated supernatant of CecP4 culture to Pseudomonas aeruginosa and E. coli respectively. Recombinant CecP4 also showed antimicrobial activity against both Pseudomona and E. coli, suggesting the broad spectrum of its antimicrobial activity. After improvements for the scale-up, it will be feasible to use recombinant CecP4 for supplementation to the feed to control microbial infections in young animals, such as piglets.
This study aimed to examine the distribution of gastrointestinal parasitic infections in domestic pigs in the Republic of Korea. From May 2020 to October 2021, 364 pig fecal samples were collected from 75 farms in 7 Provinces and microscopically examined. A total of 170 (46.7%) pigs were infected with at least one of the following parasites: Balantioides coli, strongyles, Ascaris suum, Trichuris suis, and coccidia. By parasite species, B. coli, strongyles, A. suum, T. suis, and coccidia oocysts or eggs were detected in 144 (39.6%), 24 (6.6%), 14 (3.8%), 4 (1.1%), and 1 (0.3%) samples, respectively. One hundred fifty-four, 15, and 1 cases showed single, double, and triple infections, respectively. Of the swine fecal samples from 75 farms, 69 specimens (92.0%) were infected with 1 or more parasites. All surveyed farms across the country exhibited a positive rate of over 30%, among which the highest positive rate was 65.0% in Chungcheongnamdo, and Jeollabuk-do was followed by 61.9%. Winter showed a statistically lower prevalence than other seasons. This study showed that gastrointestinal parasites are prevalent in pigs in Korea, although the diversity of parasites is low.
