A New Technique Development for Measuring Plastic Strain of Precision Machined Surface (정밀가공면의 소성스트레인 측정을 위한 새로운 기법의 개발)

  • A plastically deformed layer in the precision machined surface affects in various forms the physical properties of machined components such as the fatigue strength, the dimensional instability, microcracks and the stress corrosion cracking. These physical properties, so called surface integrity, are very important for designing highly stressed and critically loaded components. Typical plastic strains in the precision machined surface are very difficult to measure, since they are located within a very short distance from the surface and they change very rapidly. A new way is suggested to determine the residual strain in plastically deformed materials by analyzing the plastically deformed layer after a subsequent recrystallization process. This investigation is to explore a new technique for measuring plastic strain in machining applications, and in particular, to and the effect of cutting parameters(rake angle, depth of cut, specific cutting energy), on the plastic strains and strain energy.

Immunological Characterization of Bacillus thuringiensis Antigens (Bacillus thuringiensis 항원들의 면역학적 분석)

  • This study was carried out to immunologically characterize Bacillus thuringiensis (B.t) antigens. Protein patterns of ultrasonicated- antigens of B. thuringiensis subspecies using SDS- PAGE revealed marked similarities among all the strains analyzed except for the difference between quantative variations of bands and some protein antigens. The comparison of the protein patterns showed that the protein antigen of 45 kilodalton (kd) was common in 11 strains and that the difference between B. thuringiensis subsp. canadensis and galleriae was noticed in quantative variations of bands despite of ambiguous serogrouping, suggesting a useful method for identification. All strains examined showed similar antigenic patterns in SDS-PAGE, while immunodominant bands differed in antigenic reactivity in western blot using polyclonal antibodies. Polyclonal antibody to B. thuringiensis subsp. thuringiensis and israelensis in indirect immunofluorescence assay reacted with flagella and cell surface antigens. The present study indicates that SDS-PAGE and western blot analysis may be used as tools for differentiation and identification of B. thuringiensis subspecies.

Diversity of Leuconostocs on Garlic Surface, an Extreme Environment

  • Thirty-nine strains of Leuconostocs found to be tolerant to $10\%$ or more garlic were selected for further identification, by comparing their whole-cell protein pattern, 16S rRNA gene (first 530 bases) sequence, cellular fatty acid composition, and carbon source metabolism. Two isolates were Identified as Leuconostoc mesenteroides and 32 others as Leuconostoc citreum. Five other strains belonging to a cluster could not be allocated to the existing species. 16S rRNA gene sequence and cellular fatty acid composition of the unidentified bacteria exhibited close similarity with Leuconostoc argentinum. The unidentified isolates were not allocated to L. argentinum, because they formed polysaccharide from sucrose, while L. argentinum strains do not. Leuconostocs tolerant to high concentration of garlic were found predominantly on garlic surface, an extreme environment which is unfit for most of other microorganisms.

Identification of Some Phellinus spp.

  • Four strains of Phellinus spp. was identified based on internal transcribed spacer(ITS) region of rDNA sequence analysis and morphological characteristics. Basidiocarps of all strains were effused-reflexed and hymenial surface was poroid. Hyphal system was dimitic and basidiospore was globose to ellipsoid. The amplification of ITS regions produced a DNA fragment of 500 to 780 by in all strains examined. The determined sequences were analyzed for the reconstruction of phylogenetic tree. From these results, Phellinus sp. KM-1, KM-2, and KM-4 was identified as P. hartigii, P. baumii, and P. linteus, respectively.

  • The applicability of a novel incremental-iterative technique with 2D differential/integral quadrature method (DIQM) in analyzing the nonlinear behavior of Bi-directional functionally graded (BDFG) porous plate based on neutral surface is verified in the present works. A formulation of four variables high shear deformation theory is used to describe the kinematic relations with respect to neutral surface rather than mid-plane. Bi-directional material distributions are presented by power functions through both thickness and axial directions. Porosities and voids are distributed by different cosine functions. The large deformations are included within the sense of nonlinear von Kármán strains. The integro-differential equilibrium equations with associated modified boundary conditions are solved numerically and iteratively by using 2D DIQM. Model validations and parametric analysis are depicted to present the influence of neutral axis, nonlinear strains, gradation indices, elastic foundations, and modified boundary conditions on the static deflection in addition to normal and shear stresses. The proposed model is effective in analyzing the static behavior of many real applications in nuclear reactors, marine and aerospace structures with large deformations.

Technological Characteristics and Safety of Enterococcus faecium Isolates from Meju, a Traditional Korean Fermented Soybean Food (메주 유래 Enterococcus faecium 균주의 기능적 특성 및 안전성)

  • In this study, we assessed the technological characteristics and safety of 88 Enterococcus faecium strains isolated from meju; the strains possess the glutamate decarboxylase gene gadA/B involved in γ-aminobutyric acid production. The study was conducted to evaluate the possibility of introducing E. faecium meju isolates as food fermentation starters. We observed that a NaCl concentration of 6% (w/v) facilitated the growth and acid production of all strains. At a NaCl concentration of 7%, 21 strains (24%) exhibited a low growth rate, 72 strains (82%) a weak acid production, and 16 strains (18%) showed no acid production. All strains exhibited protease activity at a NaCl concentration of 4%. At a NaCl concentration of 5%, 86 strains exhibited weak activity, and one strain showed no protease activity. We could not detect any lipase activity in the investigated strains. None of the strains exhibited an acquired antibiotic resistance to the seven antibiotics tested in the present study, namely ampicillin, chloramphenicol, ciprofloxacin, gentamicin, penicillin G, tetracycline, and vancomycin. We could identify the Enterococcus endocarditis antigen gene efaA and the tyrosine decarboxylase gene tdc contributing to tyramine production, in 88 meju isolates. We could not detect the Enterococcus surface protein gene esp, which is specifically possessed by human-originated E. faecium strains, in any of the 88 strains tested in the study.

'Bring to Lab' of 19 Novel Species Among 60 Isolates Retrieved from a Freshwater Pond

  • We report here on the cultivation of numerous novel bacterial species from a eutrophic freshwater pond. A total of 60 strains, 15 strains per each culture medium, were obtained from the surface of a eutrophic freshwater pond by employing a conventional dilution-plating method with four different kinds of culture media, including R2A, 1/10R2A, PCA, and 1/10PCA. Among the 60 strains isolated, 27 strains showed less than 97% 16S rRNA gene sequence similarities to validly published species, and thus they are considered to comprise 19 novel species. Of the 27 strains assigned to the novel species, the majority of the strains (20 strains) were affiliated with the Alphaproteobacteria and Betaproteobacteria. The remaining 7 strains were affiliated with the Gammaproteobacteria, Firmicutes, Actinobacteria, and Deinococci. Because we have isolated 19 novel species from a usual freshwater pond using a conventional culturing technique, our results suggest that an unexplored ecosystem, even if it looks like a common ecosystem found elsewhere, harbors diverse unidentified microbes, which will be definitely further characterized.

Relationship between Hydrophobicity and Pellicle Formation in a Film Strain of Hansenula beijerinckii FY-5 Isolated from Apple Wine (사과주(酒) 산막효묘(産膜酵母) Hansenula beijerinckii FY-5 의 소수성(疏水性)과 산막성(産膜性)과의 관련성(關聯性))

  • Relationship between cell surface hydrophobicity and pellicle formation was studied in a film strain isolated from stored apple wine and identified as Hansenula beijerinckii FY-5. In the media containing non-ionic surface-active agents the pellicle formation of strain FY-5 was efficiently repressed, whereas growth of the yeast was possible, and also cell surface hydrophobicity was greatly decreased by the addition of these agents. These results indicate that a pellicle formation factor, which keeps yeast cells floating on the medium surface, is necessary for the pellicle formation, and surely this factor is the hydrophobicity of the cell surface. The pellicle formation in the film strains was abundant with the increase of the cell surface hydrophobicity, whereas the non-film strains had less hydrophobicity as compared with the film strains. Ethanol, as a sole carbon source, efficiently increased hydrophobicity more than glucose, and the hydrophobicity was lowered with the rise of pH. In the experiments of time course, the hydrophobicity was increased in proportion to cell growth, and was maximum during the stationary phase.

A Rapid Serotyping of Hydrophobic Strains of Mycobacterium acrofulaceum by Fluorescent Anti-Complement Technique (형광항보체법(螢光抗補體법)에 의한 Mycobacterium scrofulaceum 소수성균주(疏水性菌株)의 형별동정(型別同定))

  • In the ecology and epidemiologic studies on various serotypes of atypical mycobacteria(AM), Schaefer's bacterial agglutination test(BA) provided the basis of the serologic procedures. Recently, attempts have been made to modify and to simplify the Schaefer's BA such as a slide agglutination test(Engel & Beerwald, 1970), a "simplified" BA(Reznikov & Leggo, 1972), an agglutination inhibition test(Richards & Eacret, 1972) and "micromethod"(Thoen et al., 1975). The BA, however, was not widely applied as a routine laboratory test mainly because it requires much times and labors to perform and partley because it is not applicable to hydrophobic strains either often encountered in the isolation of AM in the clinical bacteriology or stock strains maintained in the laboratory. On the contrary, fluorescent antibody technique with mycobacteria may have advantages over the BA because it is far more simpler in serologic procedures and is applicable to all strains of mycobacteria regardless of smooth or rough types of cultures. At the present, it is well known that the type-specific antigens are lacking on the surface of rough type of AM compared to that on smooth type of strain, but the antigenicity on the surface of the hydrophobic strains of AM which resulted from a series of subculture and the strain in the laboratory for 3 to 6 months has not been clarified. In this study, an attempt to serotype the hydrophobic strains of M. scrofulaceum serotype 41, 42 and 43 by fluorescent anti-complement(FAC) technique was made. The FAC technique with mycobacteria was also described in detail. In the summary, the complement fixing antibody titres of reference sera to smooth types of homologous serotype was highest, but the antibody titres of reference sera to hydrophobic strains of serotypes, 41, 42 and 43 gave two-to 8-folds lower than those to smooth type of strains. Although the sensitivity of type-specific antigens on the hydrophobic strains to reference sera was much lower, using the two units of reference sera determined by titration with hydrophobic strains, three serotypes, i. e., 41, 42 and 43 were specifically differentiated one another by FAC technique. This result indicated that the hydrophobic strains which were maintained in the laboratory at least for 6 months still retain type-specific antigen detectable by FAC technique.

Recent Development of Automated Strain Measurement System for Sheet Metal Parts (판재 변형률 자동측정시스템의 발전)

  • It is reasonable to use the stereo vision and image processing technique to digitize 3D coordinates of grid points and to evaluate surface strains on a sheet metal parts. However this method has its intrinsic problems such as the difficulty in enhancement of bad images inevitable error due to digital image resolution of camera and frame grabber unreliability of strains and thickness evaluated from coarse grid on the corner area with large curvature and the limitation of the area that can be measured at a time. Therefore it is still hard to measure strain distribution over the entire surface of a medium,- or large-sized stamped part at a time even by using an automated strain measurement system. In this study the curvature correction algorithm based on the grid refinement and the geometry assembling algorithm based on the global error minimization (GEM) scheme are suggested. Several applications are presented to show the reliability and efficiency of these algorithms.
