The 543 microbial strains were isolated from the rhizosphere in ginseng root rot-suppressive soil. From these, isolate KGM-100 was finally selected for the screening of powerful antagonistic bacterium for the soil-born Fusarium solani causing root rot of many important crops. The isolate KGM-100 was identified as Pseudomonas aeruginosa. Antibiotics produced by the P. aeruginosa KGM-100 were partially characterized as heat-stable and low-molecular weight antibiotics. The strain also produced siderophore, which was assumed to be pseudobactin. Pot test showed approximately 30-50% biological control effects when Fusarium solani-infected soil was treated with the P. aeruginosa KGM-100.
Antagonistic Promicromonospora sp. KH-28 isolated from a suppressive soil could produced a chitinase and a antifungal antibiotic for the biocontrol ability. The chitinase and the antibiotic appeared to inhibit plant pathogens of Fusarium oxysporum. Phytophthora capsici, Alternaria kiki, fusarium solani, Stemphylium sp., and Psudomonas fluorescens. the antibiotic produced from the strain was identified as a antifungal substance of 503 dalton having a pyrimidine skeleton with an aliphatic side chain. The Promicromonospora sp. KH-28 was able to suppress effectively F. oxysporum derived-fusarium wilt of red-pepper plant in the pot in vivo test.
We have studied some effects of the SOEUMIN-HYANGSAYANGYUI-TANG (S.H.Y.) on the C.N.S. and on the intestines. Several empirical remarks, depending on the different treatments, are investigated throughout this study as follows; 1) The treatment, using atropine bromide, represses the effect of the S.H.Y. on the contractive force of intestines. 2) The treatment, using pheniramine maleate, seems not be able to repress the effect of the S.H.Y. on the contractive force of intestines. 3) The treatment, using cyproheptadine, seems not be able to repress the effect of the S.H.Y. on the contractive force of intestines. 4) The analgesic effect as well as the antipyretic effect are remarked while the acetic acid is applied. 5) The suppressive action on the convulsion, induced by strychine, is not observed. However the significant effect on the convulsion induced by picrotoxin is remarked. 6) The sedactive effect of the S.H.Y., using the Rotor rod test, is not obviously observed.
Ten species of herbae, which have been used to treat cancers in Chinese medicine, were tested to investigate their mutagenicity or antimutagenicity in S. typhimurium TA97, TA98, TA100, TA1535, and TA1538. Scolopendra centipede was weakly active in reversion of the frameshift mutation in S. typhimurium TA97 strain and the base-pair substitution in TA100 and TA1535 strains. Other herbae such as Coix lachryma, Dianthus superbus, Tricanthoshse kirilowii, Eupatorium formosanum, Lithospermum erythrorhizon, Ansaema japonicum, Curcuma zedoaria, Helicteres angustifolia, and Euonymus sieboldianus did not show any of the mutagenic potential, regardless of the metabolic activation with rat hepatic microsomal fraction. Dianthus superbus, Eupatorium formosanum, and Euonymus sieboldianus exhibited suppressive activities on microbial mutagenesis of N-methyl-N'-nitrosoguanidine, a base-pair substitution mutagen, in TA1535 and TA100 tester strains. The antimutagenic activities of Dianthus superbus and Euonymus sieboldianus appeared to be dose-dependant.
Ethanol, total saponin and petroleum ether extract of Panax ginseng C.A. Meyer were tested for their anticlastogenic effects against induction of micronuclei by cyclophosphamide and benzo(a)pyrene in mice. Ginseng petroleum ether extract (GPEE) showed the highest suppressive effect among three extracts. GPEE was also tested to compare their anticlastogenic effect against several well-known carcinogens: such as mitomycin C, 7, 12-dimethyl benzo(a)anthracene, ethyl methane sulfonate, dimethylnitrosamine and busulfan. GPEE showed the anticlastogenic effect against most of carcinogens, although there were no significant effects against 7, 12-dimethyl benzo(a) anthracene, dimethyl nitrosoamine and busulfan-induced micronuclei.
In order to compare the suppressive effect of galangin on the genotoxicity by N-methyl-N-nitrosourea (MNU) or benzo[a]pyrene B(a)P, in vivo micronycleus test using mouse peripheral blood and in vitro sister chromatid exchange(SCE) test using mouse spleen lymphocytes were performed. MNU or B(a)P-induced micronucleated reticulocytes in vivo was decreased by the simultaneous treatment of galangin. MNU or B(a)P-induced SCEs in vitro was also decreased by the simultaneous treatment of galangin. On the other hand, the determinations of [$^3$H]MNU-induced total DNA binding and methylated DNA were performed to find out the mechanism of action. [$^3$H]MNU-induced total DNA binding was inhibited by the treatment of galangin in calf thymus DNA. HPLC analysis of DNA hydrolysates showed that galangin caused a decrease of 7-methyl guanine and $O^{6}$-methyl guanine in calf thymus DNA. To elucidate the action mechanism of galangin against B(a)P, alteration of B(a)P metabolism was studied. Galangin inhibited B(a)P metabolism in the presence of S-9 mix and decreased B(a)P-DNA binding in calf thymus DNA with S-9 mix.

Alzheimer's disease (AD) is the most common neurodegenerative disease without known ways to cure. A key neuropathologic manifestation of the disease is extracellular deposition of beta-amyloid peptide (Ab). Specific mechanisms underlying the development of the disease have not yet been fully understood. In this study, we investigated effects of 4-O-methylhonokiol on memory dysfunction in APP/PS1 double transgenic mice. 4-O-methylhonokiol (1 mg/kg for 3 month) significantly reduced deficit in learning and memory of the transgenic mice, as determined by the Morris water maze test and step-through passive avoidance test. Our biochemical analysis suggested that 4-O-methylhonokiol ameliorated $A{\beta}$ accumulation in the cortex and hippocampus via reduction in beta-site APP-cleaving enzyme 1 expression. In addition, 4-O-methylhonokiol attenuated lipid peroxidation and elevated glutathione peroxidase activity in the double transgenic mice brains. Thus, suppressive effects of 4-O-methylhonokiol on $A{\beta}$ generation and oxidative stress in the brains of transgenic mice may be responsible for the enhancement in cognitive function. These results suggest that the natural compound has potential to intervene memory deficit and progressive neurodegeneration in AD patients.
Petroleum ether extracts of panax ginseng C.A. Meyer (GPEE) were tested for the evaluation of anticlastogenic effects against benzo(a)pyrene-induced micronucleated polychromatic erythrocytes using mouse bone marrow micronucleus test. When the GPEE was singly administered before benzo(a)pyrene injection, GPEE showed significant anticlastogenic effect at $50{\sim}200\;mg/kg$. When the GPEE was multiply administered for 5 consecutive days before benzo(a)pyrene injection, GPEE showed potent anticlastogenic effect, even at the low doses, $5{\sim}50\;mg/kg/day$. As a control experiment, GPEE was administered without benzo(a)pyrene injection to demonstrate a clastogenic effect of this extract. When the range of $1{\sim}200\;mg/kg/day$ for 5 consecutive days was administered to mice, it was found that there was no increase of MNPCEs frequency.
Objectives: We are aimed to identify anti-tumor effects of Curcuma aromatics on some kinds of cancer cells through molecular biologic methods. Materials & Methods: We used 4 kinds of cancer cell lines such as lung cancer cells(AS49), cervical cancer cells(HeLa), glioma cancer cells(A172) and prostate cancer cells(PC3). We treated the boiled extract of Curcuma aromatica $5{\mu}g,\;10{\mu}g$ to cultural media(ml) for 24 hours. We measured the cytotoxicitv on 4 kinds of cancer cells through tryphan blue exclusion test and the suppressive effect on viability of 4 kinds of cancer cells via MTT assay. We measured change of mitochondria membrane potential via flow cytometry. The quantitative RT-PCR was used to examine the effect on the revelation of Bcl-2 and Bax which are genes related to apoptosis. We examined the effect on the revelation of Bcl-2 Protein and Bar protein by western blot analysis. Results : In the experiment of tryphan blue exclusion test, the extract of Curcuma aromatica showed more significant killing effect on AS49, HeLa than the control group with density dependent manner, which was statistically significant. In the experiment of MTT assay the extract of Curcuma aromatica showed more suppressive effect on viability of A549, HeLa than the control group with density dependent manner, which was statistically significant. Curcuma aromatica induced apoptosis by decreasing the membrane potential of mitochondria in A549, HeLa. In the experiment of the revelation of genes related to apoptosis, the revelation of Bcl-2 decreased and the revelation of Bax increased in A549, HeLa treated with Curcuma aromatica with dose dependent manner. In the experiment of the revelation of protein related to apoptosis, the protein levels of Bcl-2 decreased and the protein levels of Bax increased in AS49, HeLa treated with Curcuma aromatica with dose dependent manner. Conclusions: From this study, we can infer that Curcuma aromatica has anti-tumor effect on lung cancer cells and uterine carcinoma cells but not on glioma cells and prostate cancer cells.
토양 우점능이 강한 생물학적 방제제 제조를 위해 경북지역 토양에서 길항균주를 분리하고 이들 중 Fusarium oxysporum, Phytophthora capsici 에 강력한 길항능을 보이는 Pseudomonas sp. 4059 를 선발, 동정하였다. Pseudomonas sp. 4059 의 시들음 병균 Fusarium oxysporum, 고추 역병균 Phytophthora capsici 에 대한 길항기작은 내열성 저분자의 항생물질과 철이온을 특이적으로 흡착하는 siderophore의 생산에 의한 것이다. Pseudomonas sp. 4059 는 항진균성 항생물질 Phenazine 생산 유전자를 소유하며 Salkowski test에 양성인 옥신류 생산도 한다는 것을 확인하였다. Pseudomonas sp. 4059 는 bioochemical tests, API test, MicroLogTM system을 통해 Pseudomonas fluorescens (biotype A)으로 98% 상동성을 보였으므로 이를 Pseudomonas fluorescens (biotype A) 4059 로 명명하였다. 선발된 길항균 Pseudomonas fluorescens (biotype A) 4059는 고추를 기주식물로 하였을 때 고추역병균인 Phytophthora capsici가 원인이 되는 고추역병을 in vivo 상에서도 충분히 억제할 수 있는 생물방제능을 나타내었다.
