A Study on Superoxide Dismutase from various Tissue of the Tricarboxylic acid cycle blocked Rat (Tricarboxylic acid회로를 차단한 흰쥐의 조직에서 Superoxide Dismutase에 관한 연구)

  • This study was carried out to observe the formation of superoxide radicals and the changes in the activities of superoxide dismutase (EC.1.15.1.1.) from the various organs of a rat which was blocked tricarboxylic acid cycle. In order to block the tricarboxylic acid cycle, the beta-fluoroethylacetate was injected into peritoneal cavity of rat and removed the various tissues from the rat at internals of an hour. By tissue extracts being prepared by the method of Weigiger and Fridovich the activities of superoxide dismutase, aconitase, and contents of bliid glucose, citrates, and wuperoxide radicals were determined. The experimental results are summarized as follows: Accumulation of citrates if increased within three hours after treatment in the all tested tissues, especially, in the geart and spleen they are higher than one of other tissues as 12 and 20 times of control. The activities of aconitase are ingibited to 30-35% on an hour after beta-fluoroethylacetate treatment comparing with that of control rat. The content of blood glucose is increased to 1.6 fold of normal value after 5 hours of treatment. In all tested tissues, superoxide radicals are formed in the heart as 0.26 micromoles per gram tissue between one and three hours after treatment. The activities of total superoxide dismutase are increased between one and three hours after treatment in the all tested tissues and one of these enzymes in heart is highest. The activities of superoxide dismutase containing Mn are also increased with an increase of total superoxide dismutase activities.

Effects of Glycerol on the Oxygen Free Radical Reactions and Renal Functions in the Renal Cortex of Rats (Glycerol이 흰쥐 신피질에서의 산소유리기반응과 신기능에 미치는 영향)

  • In an attempt to define the early biochemical determinants that participate in the pathogenesis of glycerol-induced nephrotoxicity, especially focusing on oxygen free radicals, we studied malondialdehyde (MDA) level and the activities of catalase and superoxide dismutase in the renal cortex of rats, and the concentrations of blood urea nitrogen(BUH) and serum creatinine of rats at 24hr after the injection of a 50% solution of glycerol. Sprague-Dawley albino rats weighing 240 to 260 mg were injected intramuscularly with a 50% solution of glycerol(2 mι/kg, 4 mι/kg and 8 mι/kg). The group treated with glycerol showed significantlv higher MDA level and catalase activity, lower SOD activity and higher BUN and serum creatinine concentrations at 24 hr after the injection as compared to those of control group. These results suggest that the excessive oxygen free radicals resulting from the depression of SOD activity is an important determinant in the pathogenesis of glycerol-induced nephrotoxicity.

Modification of Cu,Zn-Superoxide Dismutase by Oxidized Catecholamines

  • Oxidation of catecholamines may contribute to the pathogenesis of Parkinson's disease (PD). The effect of the oxidized products of catecholamines on the modification of Cu,Zn-superoxide dismutase (SOD) was investigated. When Cu,Zn-SOD was incubated with the oxidized 3,4-dihydroxyphenylalanine (DOPA) or dopamine, the protein was induced to be aggregated. The deoxyribose assay showed that hydroxyl radicals were generated during the oxidation of catecholamines in the presence of copper ion. Radical scavengers, azide, N-acetylcysteine, and catalase inhibited the oxidized catecholamine-mediated Cu,Zn-SOD aggregation. Therefore, the results indicate that free radicals may play a role in the aggregation of Cu,Zn-SOD. When Cu,Zn-SOD that had been exposed to catecholamines was subsequently analyzed by an amino acid analysis, the glycine and histidine residues were particularly sensitive. These results suggest that the modification of Cu,Zn-SOD by oxidized catecholamines might induce the perturbation of cellular antioxidant systems and led to a deleterious cell condition.

The Effect of Human Recombinant Superoxide Dismutase Conjugated with Polyethylene Glycol on the Hepatic Toxicity of Acetaminophen (HrSOD-폴리에칠렌 접합체의 아세트아미노펜 간독성에 미치는 영향)

  • The covalent conjugation of human recombinant superoxide dismutase (hrSOD) with trichloros-triazine activated polyethylene glycol (PEG) 5000 formed soluble conjugates with molecular weight of 92KD, which retained $90{\sim}98%$ of original activity with a markedly prolonged plasma half-life of enzyme activity. The effect of hrSOD-PEG conjugates on acetaminophen (ACP)-induced hepatotoxicity was tested in male rats which were pretreated with 3-methylcholanthrene. HrSOD-PEG conjugates inhibited the hepatotoxicity produced by ACP, on the other hand, native hrSOD had no protective effect. The above results indicated that oxygen radicals might participate in the mechanism of the ACP-induced hepatotoxicity and that polymer conjugated-protein drugs with prolonged half-lives could be employed as an effective therapeutic agent.

Antioxidant activity of ethanol extracts from the root and bark of Ulmus davidiana

  • Antioxidant activities of Ulmus davidiana root and bark extracts were evaluated by various antioxidant tests, including DPPH radical-scavenging, nitric oxide-scavenging, superoxide anion radical-scavenging, and ABTS radical-scavenging assays, and superoxide dismutase (SOD)-like activity and reducing power analysis, along with the determination of total phenolic and flavonoid contents. Both the extracts showed strong antioxidant activities by these testing methods. Ulmus davidiana root extract possessed strong reducing power and nitric oxide-scavenging activity, and high scavenging activities against free radicals including the superoxide anion, and the ABTS and DPPH radicals, but a weaker scavenging activity of SOD. In contrast, the Ulmus davidiana bark extract exhibited a strong SOD-like activity, but all the other activities were weak. It was observed that the total phenolic and flavonoid contents of the Ulmus davidiana root extract were higher than those of the Ulmus davidiana bark extract.

Effects of Local Anesthetics on Electron Transport and Generation of Superoxide Radicals in Mitochondria (국소마취제가 Mitochondria에서의 전자이동 및 Superoxide Radicals의 생성에 미치는 영향)

  • Local anesthetics were investigated for their effects on mitochondrial electron transport system, production of superoxide radical from submitochondrial particles and malondialdehyde production through lipid per oxidation. Local anesthetics had various effects on activities of enzymes in electron transport chain. The activities of NADH dehydrogenase, NADH oxidase and NADH-ubiquinone oxidoreductase were effectively inhibited by lidocaine, procaine and dibucaine but slightly influenced by cocaine. The activities of succinate dehydrogenase, succinate-cytochrome c oxidoreductase and succinate-ubiquinone oxidoreductase were inhibited by lidocaine and dibucaine, but the succinate oxidase activity was stimulated by local anesthetics. Both dihydroubiquinone-cytochrome c oxidoreductase and cytochrome c oxidase activities were inhibited by local anesthetics. In these reactions, the response of Complex I segment to local anesthetics was greater than other Complex segments. Local anesthetics inhibited both the superoxide production from submitochondrial particles supplemented with succinate or NADH and the enhanced production of superoxide radicals by antimycin. The malondialdehyde production by oxygen free radicals was inhibited by local anesthetics. These results suggest that the inhibition of superoxide and malondialdehyde production caused by local anesthetics may be brought by suppression of the electron transport in mitochondria at sites in or near complex I segment.

  • This study was undertaken to examine the effect of Jwagyuyeum and Woogyuycum, being known to reinforce Kidney-yin and yang, on the activities of endogenous antioxidant enzymes and the production of oxygen free radicals in the liver and kidney tissues, Alterations in enzyme activities were observed after in vivo treatment in rats, Jwagyuyeum and Woogyuyeum caused a significant increase in the activities of superoxide dismutase(SOD) and catajase Jwagyuyeum significantly increased the activity of glutathione peroxidase in both liver and kidney, but the enzyme activity was not significantly altered by Woogyuyeum. Treatment in vitro of Jwagyuyeum and Woogyuyeum decreased the production of oxygen free radicals in a dose-dependent fashion. These results suggest that Jwagyuyeum and Woogyuyeum stimulate the activities of antioxidant enzymes and inhibit directly the production of oxygen free radicals. These effects of both herbs may contribute to prevent the oxygen free radical-induced impairment of cell function.

Antioxidative Activity of the Extracts from Browned Oak Mushroom (Lentinus edodes) with Unmarketable Quality (시장성이 없는 품질의 갈변 표고버섯(Lentinus edodes) 추출물의 항산화 활성)

  • Physiological functionality of browned oak mushroom was evaluated with focusing its electron-donating ability to DPPH radicals, scavenging ability to superoxide radicals and hydroxyl radicals, and inhibitory activity on lipid peroxidation. The results showed that overall antioxidative activities of browned oak mushroom were superior to those of raw oak mushroom with marketable quality, implying possible involvement of resultant browning reaction products in an increment of antioxidativity. The increased radical-scavenging ability was suggested to mainly be exerted by direct quenching of both superoxide and hydroxyl radicals, not by inhibition of xanthine oxidase activity and $Fe^{2+}$ chelation, respectively. Collectively, these results indicate a possible use of unmarketable browned mushroom as a material for manufacturing various processed functional foods.

Influence of the Donor Side of Photosystem II on the Photogeneration of Superoxide Radicals and Chlorophyll a Fluorescence

  • Direct EPR evidence of the photo-generation of superoxide radicals ( $O_2$$^{-.}$) was obtained by using spin trapping techniques in spinach photosystem II (PSII) membranes. $O_2$$^{-.}$ was detected by following the formation of 5-diethoxyphosphoryl-5-methyl-1 -pyrroline-N-oxide (DEPMPO) superoxide adducts, DEPMPO-OOH. The significant increase of the EPR signal amplitude of DEPMPO-OOH in N$H_2O$H-, CaC $l_2$- and NaCl-treated PSII membranes showed that the oxygen-evolving system has a close relation to the $O_2$$^{-.}$ production. PSII membranes with inactivated donor side could not prevent the $O_2$$^{-.}$ production efficiently. Treatments on PSII donor side also influence the maximum level and the kinetics of Chlorophyll (Chi) a fluorescence. Results suggested that manganese cluster and extrinsic proteins might affect Chi a fluorescence in ways different from that happens at the acceptor side of PSII.SII.SII.
