하이아미의 정조 및 현미 추출물의 in vitro 항산화 및 항암활성 (Antioxidant and Antitumor Activities of Ethanol Extracts from Unhulled and Hulled Rice Hiami (Oryza sativa L. cv. Hiami))

  • 하이아미의 정조와 현미상태에 대해 일품벼를 대조구로 하여 항산화성분의 함량과 에탄올 추출물에 대한 in vitro 항산화활성 및 항암활성을 측정하였다. 하이아미의 정조 및 현미 추출물의 총 폴리페놀 함량은 각각 6.31 및 3.75 mg/g으로 나타났으며, 일품은 각각 5.66 및 3.47 mg/g으로 나타나 하이아미가 약간 높은 것을 알 수 있었다. 하이아미의 정조와 현미 추출물의 $\gamma$-oryzanol 함량은 각각 33.53 및 39.47 mg/100 g으로 나타났으며, 일품의 경우 각각 24.33 및 28.68 mg/100 g으로 나타나 일품에 비해 하이아미에서 높은 함량을 보이는 것으로 나타났다. 하이아미와 일품벼 정조 및 현미의 70% 에탄올 추출물에 대한 DPPH radical, hydrogen peroxide, superoxide radical 및 hydroxyl radical 소거활성을 5 mg/mL의 농도에서 측정한 결과 하이아미가 일품에 비해 높은 것으로 나타났으며, 정조 추출물이 현미 추출물보다 높은 radical 소거활성을 보이는 것으로 나타났다. 70% 에탄올 추출물에 대한 in vitro 항암활성을 측정한 결과 정조 및 현미 추출물의 억제 활성은 용량 의존적으로 증가하는 것으로 나타났으며, 특히 하이아미는 위암 및 유방암세포에 높은 활성을 보였으며, 일품은 대장암 및 간암세포에 대해 높은 활성을 보였다.

한약재에탄올 추출물의 항산화 효과 (The Antioxidative Effect of Ethanol Extracts from Lithospermum erythrorhizon Siebold & Zucc., Xanthium strumarium Linn, and Lonicera japonica)

  • 본 연구에서는 한약 재료로 널리 이용되고 있는 3종의 약용식물(지치, 창이자, 인동초)을 기능성 소재로 이용하기 위하여 총 페놀함량, DPPH radical 소거능, xanthine oxidase 저해능 등의 실험을 통해 한약재의 항산화 활성을 살펴보았다. 지치, 창이자, 인동초의 수율은 각 7.01%, 2.92%, 7.95%, 총 페놀함량은 각 $4.3{\pm}0.1{\mu}g/ml$, $5.4{\pm}0.1{\mu}g/ml$, $4.6{\pm}0.1{\mu}g/ml$를 함유하고 있었다. DPPH radical 소거능은 인동초가 0.24 mg/ml에서 $EC_{50}$이 나타났으며 나머지 2종 모두 높은 DPPH radical 소거능을 보였다. Xanthine oxidase 저해능은 0.01 mg/ml의 농도에서 한약재 3종 모두 대조구로 사용된 ascorbic acid보다 더 높은 저해 활성을 나타내었다. 그 외에도 Superoxide radical 소거활성은 지치와 인동초가 50 mg/ml의 농도에서 80% 이상의 소거활성을 보였으며, OH radical 소거활성은 50 mg/ml의 농도에서 40% 이상의 소거활성이 나타났다. 이상의 결과로부터 지치, 창이자, 인동초 모두 높은 항산화 활성을 가지고 있으며, 기능성 소재로 활용 가능함을 확인하였다.

홍화씨 추출물의 in vitro 항산화 및 ethanol로 손상을 유도한 C6 신경교세포 보호 효과 (Protective effects of Carthamus tinctorius L. seed on C6 glial cells treated with ethanol)

  • 만성 알코올 섭취는 산화적 스트레스 및 치매와 같은 신경퇴행성질환을 발병시킨다. 본 연구는 홍화씨 추출물의 in vitro 항산화 활성과 ethanol로 손상을 유도한 신경교세포에서의 보호 효과를 확인하기 위해 수행되었다. 홍화씨 추출물은 50, 100, 250, 500 ㎍/mL의 농도에서 농도 의존적으로 in vitro에서 1,1-diphenyl-2-prcrylhydrazy (DPPH), hydroxyl radical (·OH), superoxide radical 및 nitric oxide radical 소거능이 증가하였다. 특히 DPPH 및 ·OH radical 소거능 측정 결과, 각각 500 ㎍/mL, 100 ㎍/mL의 농도에서 80% 이상의 radical 소거능을 나타내었다. 홍화씨 추출물의 신경교세포 보호 효과를 확인하기 위해, C6 신경교세포에 500 mM ethanol을 처리하여 산화적 손상을 유도하였다. Ethanol을 처리한 C6 신경교세포는 세포 생존율의 감소 및 reactive oxygen species (ROS) 생성량이 증가하여 세포 손상을 나타내었다. 반면 홍화씨 추출물을 처리하였을 때 세포 생존율 증가 및 ROS 생성 억제를 통해 ethanol로 유도된 신경교세포 손상에 대한 보호 효과를 나타내었다. 본 연구를 통해, 홍화씨는 알코올로 유도된 신경교세포의 산화적 스트레스에 대한 보호 효과가 있는 것으로 사료된다.

  • 본 연구는 5가지 경북약용작물(홍화자, 향부자, 형개, 위유, 작약)의 항산화와 항염증 효과에 대해서 검증하였다. 추출은 70% 에탄올에서 추출하였다. 항산화 활성을 측정하기 위해 총플라보노이드, 폴리페놀 함량을 측정하였고, 추출물의 소거활성은 a-a-Diphenyl-${\beta}$-picrylhydrazyl ($DPPH{\cdot}$), 2,2-azino-bis 3-ethylbenzthiazoline-6-sulfonic acid ($ABTS{\cdot}$), ferric reducing antioxidant power (FRAP), superoxide anion 저해활성으로 측정하였다. 홍화자 추출물에서 높은 총 폴리페놀과 플라보노이드 함량과 뛰어난 $DPPH{\cdot}$, $ABTS{\cdot}$, FRAP, superoxide anion 소거활성을 나타내었다. 또한, 홍화자 추출물에서 가장 높은 NO 생성 저해 활성을 나타냈다. 그 결과 홍화자 추출물은 항산화와 항염증 활성에 효과를 가진 기능성 소재로 사용될 수 있을 것이다.

Streptozotocin 유발 당뇨쥐 신장조직의 항산화계에 미치는 Vitamin E의 영향 (effects of Vitamin E on the Antioxidative Defense System of Kidney in Streptozotocin-Induced Diabetic Rats)

  • The purpose of this study was to investigate the effects of vitamin E on the antioxidative defense system of kidney in streptozotocin induced diabetic rats. Sprague Dawley male rats weighing 100$\pm$10g were randomly assigned to one normal and three STZ induced diabetic groups, which were subdivided into vitamin E free diet(DM 0E group), 40mg vitamin E per kg diet(DM 40E group) and 400mg vitamin E per kg diet(DM 400E group). Vitamin E level of normal group was 40 mg per kg diet. Diabetes was experimentally induced by intravenous injection of 55mg/kg of body weight of streptozotocin(STZ) in citrate buffer(pH 4.3) after 4 weeks feeding of experimental diets. Animals were sacrificed at the 6th day of diabetic states. There were no significant on body weights, food intakes, and food efficiency ratio before the diabetic occurrence. But after the injection of STZ, body weights and food efficiency ratios were significantly decreased and the food intakes was increased. Kidney weights were significantly increased in diabetic groups compared to normal group. However, there were no significant differences among the diabetic groups. Plasma insulin levels of diabetic groups were significantly decreased, whereas, blood sugar levels were increased compared to that of normal group. There were no significant differences among diabetic groups in plasma insulin and glucose levels. Activities of superoxide dismutase(SOD) in DM 0E and DM 40E groups were signi ficantly decreased by 33% and 27%, respectively, compared to normal group. But that of DM 400E group was increased by 35% compared to DM 0E group. Activity of glutathione peroxidase(GSHpx) in DM 0E group was decreased by 20% compared with normal group. GSHpx activity in DM 400E group was increased by 29% compared to normal group. The contents of vitamin E in kidney were 58% and 49% lower in DM 0E and DM 40E group, respectively, than normal group. There was no significant difference in renal vitamin E contents between DM-400E group and normal group. The contents of superoxide radical(O2 ) in kidney were 150% and 98%, respectively, higher in DM 0E and DM 40E groups than normalgroup. DM 400E and normal groups were similar levels in their superoxide radical contents of kidneys. These results indicate that vitamin E functioned as chain breaking antioxidant in kidney such as in other tissues.

해조류 추출물의 라디칼 소거활성 검색과 보라우무의 항산화활성 성분

  • The antioxidant activities of methanol and dichloromethane/acetone extracts of twenty three seaweeds were tested by using 1,1-Diphenyl-2-picryl-hydrazyl(DPPH) at a $100{\mu}g/ml$ concentrations. The dichloromethane/acetone extracts of three seaweeds(Symphyocladia latiuscula, Gloiopoltis furcata, Sagassum thunbergii), were found to be most effective in DPPH radical scavenging activity. The DPPH radical scavenging effect of these seaweeds was Symphyocladia latiuscula(85.82%), Gloiopoltis furcata(82.83%), Sagassum thunbergii(74.05%) in order. These seaweeds were evaluated using the pyrogallol UV-VIS spectrophotometeric method to generate superoxide anion. Among them, the methanol extracts of six seaweeds were showed weak superoxide dismutase-like activities. The dichloromethane/acetone extracts obtained from Symphyocladia latiuscula was fractionated with $CH_2Cl_2$, n-hexane, 15% aq.MeOH, n-BuOH, $H_2O$. The 15% aq.MeOH soluble fraction exhibiting the strongest antioxidative activities was further purified by C18 column flash chromatography and reversed HPLC. The two active principles of Symphyocladia latiuscula were isolated and characterized as 2,3,6-tribromo-4,5-dihydroxybenzyl alcohol(1), 2,3,6-tribromo-4,5-dihydroxybenzyl methyl ether(2).

일산화탄소 폭로후 고압산소 투여가 흰쥐 심장에서의 malondialdehyde 함량과 산소유리기 반응에 미치는 영향 (Effects of Hyperbaric Oxygen Treatment on the Malondialdehyde Level and Oxygen Free Radical Reactions in the Heart of the Rats Exposed to Carbon Monoxide)

  • ln an attempt to define the effects of hyperbaric oxygen treatment on the lipid peroxidation and oxygen See radical reactions in rats exposed to carbon monoxide, we studied malondialdehyde(MDA) level and activities of catalase and superoxide dismutase in the heart of the rats exposed to carbon monoxide. Male Sprague-Dawley albino rats weighing 240 to 260gm were used. Experimental groups consist of Control group (=breathing with air), HBO group(=exposed to hyperbaric oxygen(HBO, 3ATA, 100%) after air breath), CO group(=exposed to CO(3,970 ppm) after air breath), CO-Air group(=exposed to CO after air breath followed by air breath) and CO-HBO group(=exposed to CO after ai. breath followed HBO treatment). The CO group showed significantly higher MDA level, catalase activity and SOD activity as compared to that of control group. The CO-HBO group showed significantly lower MDA level as compared to that of CO group, and did not show significantly lower catalase activity and SOD activity as compared to that of CO group. These results suggest that the excessive oxygen free radicals is an important determinant in pathogenesis of Co-induced cardiotoxicity and HBO inhibits the lipid peroxidation caused by excessive oxygen free radicals in the heart of the rats exposed to carbon monoxide.

The Effects of Chicken Leg Bone Extract on Antioxidative Properties under Different Heating Condition

  • The aim of this study was to extract chicken leg bone, which is a by-product of industrial poultry processing, using different heating temperatures (80, 90 and $100^{\circ}C$) and durations (5, 10 and 15 min). The pH value, soluble protein content, peptide content and antioxidative properties, including superoxide anion scavenging ability, 1, 1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging ability, reducing capacity and inhibitory activity of linoleic acid peroxidation, were measured. The results showed no significant differences (p>0.05) in pH value among all treatments. Decreased soluble protein content and peptide content were observed in chicken leg bone extract obtained under higher heating temperatures (90 or $100^{\circ}C$) and longer heating durations (10 or 15 min). In antioxidative properties, the extracts which were heated at 90 or $100^{\circ}C$ for 15 min exhibited significantly higher superoxide anion scavenging ability, DPPH free radical scavenging ability, reducing capacity and inhibitory activity of linoleic acid peroxidation (p<0.05).

Sterin C, a New Antioxidant from the Mycelial Culture of the Mushroom Stereum hirsutum

  • Structurally new antioxidative metabolite was isolated from mycelial culture of mushroom Stereum hirsutum. Culture broth was subjected to Diaion HP-20 column chromatography, and 70% aqueous MeOH eluent was extracted with EtOAc. EtOAc extract was purified through silica gel and Sephadex LH-20 column chromatographies, and reversed phase $C_{18}$ HPLC. Compound was revealed to be new dihydroxylated derivative of sterin B with molecular formula of $C_{12}H_{16}O_5$(MW 240) by MS and various NMR spectral data analyses, and designated as sterin C. Sterin C showed superoxide radical-scavenging activity with $EC_{50}$ value of 0.31 mM.

Evaluation of antioxidant properties of a new compound, pyrogallol-phloroglucinol-6,6'-bieckol isolated from brown algae, Ecklonia cava

  • In this study, antioxidant and free radical scavenging activities of the natural antioxidative compound, pyrogallol-phloroglucinol-6,6'-bieckol (PPB) isolated from brown algae, Ecklonia cava was assessed in vitro by measuring the radical scavenging activities (DPPH, alkyl, hydroxyl, and superoxide) using electron spin resonance (ESR) spectrometry, intracellular reactive oxygen species (ROS) scavenging activity, and DNA damage assay. According to the results of these experiments, the scavenging activity PPB against difference radicals was in the following order: DPPH, alkyl, hydroxyl, and superoxide radicals ($IC_{50}$; 0.90, 2.54, 62.93 and $109.05{\mu}M$). The antioxidant activities of PPB were higher than that of the commercial antioxidant, ascorbic acid. Furthermore, PPB effectively inhibited DNA damage induced by $H_2O_2$. These results suggest that the natural antioxidative compound, PPB, can be used by the natural food industry.
