Anti-oxidant Effects of the Water Extracts from the Inonotus Obliquus against Cisplatin- Induced Damage in HEI-OC1 Cells (차가버섯 물 추출물의 cisplatin에 의해 유도된 HEI-OC1세포 손상에 대한 항산화효과)

  • The medicinal mushroom Inonotus obliquus is a traditional and widely used multi-functional fungus. In this study, we have investigated whether Inonotus obliquus (Chaga mushroom) extracts exerts anti-oxidant effects on cisplatin-induced cytotoxicity in auditory cell line, HEI-OC1 cells. First of all, Chaga extracts has no harmful effects on viability of HEI-OC1 cells in the dose range of $65{\sim}125{\mu}g/m{\ell}$. Moreover, it shows cyto-protective effects on the cells treated with cisplatin-induced cytotoxicity in HEI-OC1 cells and the damage of hair cells arrays of the rat primary organ of Corti explants in the presence of cisplatin. Pretreatment with Chaga extracts inhibited the cell death, reactive oxygen species generation (ROS), lipid peroxidation induced by cisplatin. These effects were associated with the induction of antioxidant enzyme by Chaga extracts. We further investigated the effects of Chaga extracts on expression of antioxidant enzymes such as Cu, Zn superoxide dismutase (SOD 1) and Mn SOD (SOD 2) by RT-PCR. In addition, Chaga extracts shows SOD activity and SOD protein expression in cisplatin treated group induced similar to control group. Taken together, these results indicate that Chaga extracts can prevent cisplatin-induced cytotoxicity by radical-scavenging activity (SOD activity) in HEI-OC1 cells. It might be an effective as antioxidant and further studies on the chemo-preventive mechanisms of Inonotus obliquus are needed.

Study of Fermentation Extract Made from Rice Bran and Dendropanax on the Whitening Effects in B16F10 Cell Line (미강·황칠 발효추출물이 B16F10 세포의 미백에 미치는 효과)

  • The purpose of this study is to research the whitening effects of fermentation extract made from the Rice bran and Dendropanax(FRD) Fermentation conditions were as follows; 1) Dendropanaxand and Rice bran were blended in a ratio of 1 to 1, 2) a weight of sugar was 10% of the total weight, 3) an amount of enzyme was 0.1%, and 4) a temperature was 20℃. It has been fermented for 90 days. In order to observe the whitening effects of FRD, the author measured the cell viability and the inhibition rate of the melanin biosynthesis, the activity of tyrosinase and SOD (superoxide dismutase) in malignant melanoma, B16F10 cells. As a result, FRD significantly inhibited the cell viability of B16F10 in more than 500 ㎍/㎖. FRD significantly suppressed the generation of melanin, and that induced by α-melanocyte stimulating hormone (α-MSH) in more than 1,000 ㎍/㎖. FRD significantly decreased the activity of tyrosinase and that induced by α-melanocyte stimulating hormone (α-MSH) in more than 500 ㎍/㎖. FRD did not changed the activity of SOD in dose dependent manner. Therefore, the author considered that the fermentation extract made from a Rice bran and Dendropanax will be able to produce high value-added products, if used as a commercial. Anti-oxidant and Hepatoprotective Effect of White Ginsengs in H2O2-Treated HepG2 Cells

  • The antioxidant activity of white ginseng was not recorded in Korea Functional Food Code, while its activity of red ginsengs was recorded. The aim of this study was to evaluate the antioxidant and hepato protective effect of different ginsengs in H2O2-treated HepG2 cells. White and red ginseng were prepared from longitudinal section of the same fresh ginseng (4-year old). The whole parts of white and red ginsengs were separately extracted with 70% ethanol and distilled water respectively, at 70 ℃ to obtain therapeutic ginseng extracts namely, WDH (distilled water extract of white ginseng), WEH (70% ethanol extract of white ginseng), RDH (distilled water extract of red ginseng) and REH (70% ethanol extract of red ginseng). In this work, we have investigated the DPPH, hydroxyl radical, Fe2+-chelating activity, intracellular ROS scavenging capacity and lipid peroxidation of different ginsengs. All these extracts showed a dose dependent free-radical scavenging capacity and a ROS generation as well as lipid peroxidation was significantly reduced by treatment with bioactive extracts of white ginsengs (WDH) than red ginsengs. Additionally, white ginseng extracts (WDH) has dramatically increased intracellular antioxidant enzyme activities like superoxide dismutase and catalase in H2O2-treated HepG2 cells. All these results explain that administration of white ginseng is useful as herbal medicine than red ginseng for chemoprevention of liver damage.

Rhynchosia volubilis Lour. and Beta vulgaris Modulate Extracts Regulate UV-Induced Retinal Pigment Epithelial Cell and Eye Damage in Mice (약콩, 비트 추출물의 자외선에 의한 망막 상피세포와 마우스의 눈 손상 조절 효능)

  • Ultraviolet (UV)-induced damage plays a major role in ocular diseases, such as cataracts and retinal degeneration. UV irradiation can generate free radicals including reactive oxygen species (ROS), which are known to cause lipid peroxidation of cellular membranes. It has also been shown that UV can damage DNA directly and induce apoptosis. Rhynchosia volubilis Loureiro (the small black bean or yak-kong, RV) and Beta bulgaris (beet, BB) are used as health supplements. In this study, we explored the protective effects of RV and BB against UVA-induced damage in human pigment epithelial (ARPE-19) cells and in mice. RV and BB mixture and their effective constituents (cyanidin, delphidin, petunidin glycosides) improved cell viability and suppressed intracelluar ROS generation. Phosphorylation of p38 mitogen-activated protein kinase (MAPK), c-Jun N-terminal kinase (JNK), Erk1/2 was analyzed by immunoblotting. RV and BB mixture inhibited UVA-induced phosphorylation of p38 MAPK, JNK, Erk1/2 in APRE-19 cells. RV and BB treatment also showed protective effects on ocular damage in UVA-irradiated mice by increasing the levels of endogenous antioxidants such as superoxide dismutase and glutathione. RV and BB have the potential to be used in a range of ocular diseases and conditions, based on in vitro and in vivo study.

Antioxidative Effects of Scutellariae Radix Aaquaacupuncture Solution on Lipid Peroxidation Induced by Free Radicals (자유기에 의한 지질과산화 반응에 대한 황금 약침액의 항산화 효능)

  • Scutellariae radix, has been used as a natural drug for fever, inflammation, cataract, and liver disease in traditional medicine. This study was performed in order to investigate the antioxidative effects of Scutellariae radix aqua-acupuncture solution (SRAS) on lipid peroxidation by free radicals. Lipid peroxidation levels were determined by TBA method during the autoxidation of linoleic acid. In this linoleic acid autoxidation system, SRAS markedly exhibited antioxidant activity, which inhibited 89% of linoleic acid peroxidation. SRAS showed scavenging effects on ${\alpha},{\alpha}-diphenyl-{\beta}-picrylhydrazyl$(DPPH) radical, inhibited superoxide generation in xanthine-xathine oxidase system, and also inhibited lipid peroxidation of rat liver tissue by hydroxyl radical derived from $H_2O_2-FE^{+2}$ system. These effects were similar to those of $dl-{\alpha}-tocopherol$, BHA and BHT. In addition, SRAS protected the cell death induced by ter-butyl hydroperoxide (t-BHP) and significantly increased cell viability in the normal rat liver cell (Ac2F). On the basis of these results, it is suggested that SRAS might play a protective role in lipid peroxidation by free radicals.

Antioxidative and Protective Effects of Haeganjeon Extract on Oxidative Damage of Hepatocytes (해간전(解肝煎)의 항산화(抗酸化) 활성(活性) 및 간세포(肝細胞)의 산화적(酸化的) 손상(損傷)에 대한 보호효과(保護效果))

  • Objectives: Haeganjeon(HGJ) has been used for the treatment of liver disease in traditional medicine. The present study was carried out to evaluate the antioxidant and protective effects of HGJ extract on oxidative damage of hepatocytes by tert-butyl hydroperoxide(t-BHP). Methods: In the linoleic acid water-alcohol system, the levels of lipid peroxide(LPO) were determined by TBA method. The scavenging effect of HGJ on ${\alpha},{\alpha}-diphenyl-{\beta}-picrylhydrazyl$(DPPH) radical was determined according to the method of Hatano. In the Fenton system(ferrous ion reaction with hydrogen peroxide), the levels of hydroxyl radical induced LPO in rat liver homogenate were determined according to the method of TBA. Inhibitory effect of HGJ on superoxide generation was measured by xanthine-xanthine oxidase system. In order to evaluate antioxidative activity of HGJ in the liver cell, cultured normal rat liver cells(Ac2F) were prepared and incubated with or without HGJ. After 18hr, cells placed in DMEM medium without serum, and then incubated with 1mM tert-butyl hydroperoxide(t-BHP) for 2hrs. Viable cells were detected by MTT assay. Conclusions: In the linoleic acid autoxidation system, HGJ extract significantly inhibited the time course of the lipid peroxidation. These effects were similar to those of BHA HGJ extracts showed about 70% scavenging effect on DPPH radical. And HGJ extract inhibited the lipid peroxide formation in rat liver homogenate induced by hydroxyl radical derived from Fenton system. In addition, HGJ extract protected the cell death induced by t-BHP and significantly increased cell viability in the normal rat liver cell. These result indicated that HGJ extract might playa protective role against oxidative hepatic cell injury by means of free radical scavenger.

Antioxidant and Anti-inflammatory Effects of Fractions from Pruni persicae Flos (도화(桃花, Pruni persicae Flos) 분획물의 항산화 및 항염증 효과)

  • Objectives : The solvent extracts of Pruni persicae Flos were investigated for the activities of anti-oxidant and anti-inflammatory effects to apply as a functional ingredient for cosmetic products. Methods : In this study, the fractions of P. persicae Flos were extracted with 70.0% acetone and purified using Sephadex LH-20 column chromatography. As a result, eight fractions were isolated. We performed MTT assay, total polyphenol contents, DPPH free radical scavenging assay, SOD-like activity, xanthine oxidase inhibition assay, astringent activity assay, hyaluronidase inhibition assay and the production of nitric oxide. Results : For anti-oxidant effects, the electron donating ability of fraction (Fr.) 2-5, Fr.-8 isolated from P. persicae Flos was above 90.0% at 100 ppm respectively. The superoxide dismutase (SOD) - like activity of Fr.-5 isolated from P. persicae Flos was 92.1% at 1,000 ppm. The xanthine oxidase inhibitory effect of Fr.-6 isolated from P. persicae Flos was about 83.3% at 1,000 ppm. Hyaluronidase inhibition activity related to the anti-inflammation effect was 94.0% for Fr.-4 isolated from P. persicae Flos at 500 ppm. In the anti-inflammation effect, the Fr.-4 isolated from P. persicae Flos inhibited the generation of nitric oxide. Conclusions : All these findings suggested that the fractions of P. persicae Flos has a great potential as a cosmeceutical ingredient with a anti-oxidant and anti-inflammatory effects.

The Effect of Baek-Nae-Jang-Bang(BNJB) to control of making cataract (白內障方이 白內障 形成 抑制에 미치는 영향에 關한 實驗的 硏究)

  • This study was carried out to prove the therapeutic effects of BNJB on the catract. The objects of this study were CXSD mice that spontaneously eye rupture occurred from three weeks after birth and eventually generate cataract within 12 weeks. We applied eyewash made from BNJB to eyes of CXSD mice twice in a day till all the eyes of the negative control showed up the symptoms of a cataract and recorded the increasing patterns of cataractous eyes. To explained the therapeutic effects of the BNJB, We carried out the ex vivo experiment which cultivating eyeballs were offered oxidative stress condition by $0.03{\%}$ $H_2O_2$ during three days. Total co-enzyme was extracted from the cultured eyeballs and used to measure activities of catalase, superoxide dismutase, glutathion S-transferase and content of GSH. The results were obtained as follows: 1. When we treated the catalin to CXSD mouse as a positive control, it represented the delaying effect for cataract generation for 2-3 days compare with negative control. But it seems that it doesn't appropriate as a therapeutic, or delaying agent. 2. In the experimental BNJB group, Cataract formation rate was dramatically reduced by BNJB. This rate was much lower than positive group and means our new formulation for the therapy of cataract has a good potential. 3. In the analysis of individual medicines of BNJB, Mok-Jeok-Cho, Hwang-Lyun and Ha-Go-Cho didn't have a major effect of BNJB. 4. The cataract formation rate was repressed by Bing-Pyun and Jin- Joo-Boon about $40{\%}$ and $50{\%}$, respectively. We can presume that the anti-cataract effect of BNJB was caused by these two medicines. 5. When we surveyed the anti-oxidant activities of BNJB, enzyme activities of GSH, SOD, and Catalase increased about $10{\%},\30{\%}$, and 2.5 folds, respectively.

  • Effects of Taeumjowetang on Lipid Peroxidation by Free Radicals and Oxidative Damage of Hepatocytes by tert-Butyl Hydroperoxide. 1. Purpose The present study was carried out to evaluate the antioxidant effects of Taeumjowetang in vitro. 2. Methods In this study, antioxidant effects of TJT on lipid peroxidation were determined according to the method of TBA. (Abbreviation) TJT : Taeumjowetang, TBA : 2-thiobarbituric acid. 3. Results : 1) TJT inhibited markedly peroxidation of linoleic acid during the autoxidation. 2) TJT inhibited lipid peroxidation induced by hydroxyl radical derived from H2O2-Fe2+ in rat liver homogenate. 3) TJT showed 66% scavenging effect on DPPH radical. 4) TJT exhibited a 25% inhibitory effect on superoxide generation from xanthine-xan thine oxidase system. 5) To investigate the antioxidative effects of TJT on the hepatocytes, cultured normal rat liver cells(Ac2F) were prepared and incubated with or without TJT. After 16~18hr, cells placed in DMEM medium without serum, and then incubated with 1mM t-BHP for 2hr. Viable cells were detected by MTT assay. In this test, TJT protected the cell death induced by t-BHP and significantly increased cell viability in the normal rat liver cell. Anti-oxidative and Anti-inflammatory Effect of Fractionated Extracts of Cynomorium Songaricum (쇄양의 항산화 및 항염증 효과)

  • Oxidative stress and inflammation are important events in the development of chronic inflammatory diseases including arthritis, atherosclerosis, diabetes, hypertension. Cynomorium songaricum (CS) has been used as a traditional Korean herbal medicine, and it is currently used in traditional clinics to treat frequent urination, spermatorrhea, weakness of the sinews and constipation in the folk medicine. The aim of this study was to determine whether fractionated extracts of CS inhibit free radical generation such as DPPH radical, superoxide radical, nitric oxide and peroxynitrite, production of nitrite an index of NO, $PGE_2$, iNOS, COX-2 and pro-inflammatory cytokines in lipopolysaccharide (LPS)-treated RAW 264.7 macrophages. Cytotoxic activity of extracts on RAW 264.7 cells was measured using 5-(3-caroboxymeth-oxyphenyl)-2H-tetra-zolium inner salt (MTS) assay. Our results indicated that the most superior extract which scavenged DPPH radical, reactive oxygen species (ROS) and RNS was CS ethyl acetate extract (CSEA). Moreover, CSEA significantly inhibited the LPS-induced NO, $PGE_2$ production and iNOS, COX-2 expression accompanied by an attenuation of TNF-$\alpha$, IL-$1{\beta}$ and IL-6 formation in macrophages. Furthermore, CSEA treatment also blocked LPS-induced intracellular ROS production and the activation of NF-${\kappa}B$. These findings indicate that CSEA inhibits the production of pro-inflammatory mediators and cytokines via the suppression of ROS production and NF-${\kappa}B$ activation. Take together, these results indicate that CSEA has the potential for use as an natural anti-oxidant and an agent of anti-chronic inflammatory diseases.
