Evaluation of Antioxidant and Antidiabetic Activities of Superior Potato Prepared by Various Cooking Methods (다양한 조리 방법에 따른 수미감자의 항산화 및 항당뇨 활성 평가)

  • This study examined the antioxidant compounds and functional properties of superior potato (SP) after various cooking methods (sun drying: SD, roasting after sun drying: SR, hot-air drying: HD, roasting after hot-air drying: HR, boiling: B, roasting: R, and air-frying: AF) and compared with uncooked SP. As a result, the total phenolic contents (TPC) in the hot water extract (HWE) were the highest in SR. B showed the highest TPC in the ethanol extract (EE). In the α-glucosidase inhibitory activity, S was the highest in the HWE, whereas R was the highest in the EE. At 50 ㎍/mL concentration, AF showed the highest oxygen radical absorbance capacity in both HWE and EE compared to others. SP protected HepG2 cells against H2O2-induced DNA damage. Based on the results, the antioxidant compounds and antioxidant activity differed by the cooking method. Thus, the findings from the current study may provide suitable SP cooking methods for attaining their maximum biological activity.

Detection of Potato Spindle Tuber Viroid Using RT-PCR Technique (RT-PCR 기법을 이용한 감자 걀쭉 바이로이드 (Potato Spindle Tuber Viroid)의 검정)

  • Potato spindle tuber viroid(PSTVd) RNAs were isolated from PSTVd-inoculated potato cv. Superioc and carried out RT-RCR with reverse transcriptase and PSTVd specifie primer pair desigened to amplify the 356 nucleotides of PSTVd genome. As a result, 356 nucleotides PCR products were amplified from PSTVd-inoculated potato cv. Superior. The 356 nucleotides DNA fragment was indeed the PSTVd geneby sequencing analysis. PSTVd could be successfully detected from infected leaf and tuber tissue of potato by using RT-PCR technique. Especially PSTVd was more effectively detected when both downstream and upstream primer were used than only downstream primer was used in RT reaction.

Inhibition of Experimental Gastric Ulcer by Potato Tubers and the Starch

  • In an attempt to establish anti-ulcerogenic activity of potato tubers, inhibitory activity against ethanol- and indomethacin-induced gastric ulcer models in rats was evaluated for the first time. From several varieties of potato tubers including Solanum tuberosum L. cv. Superior (white skin and fresh potato) and two new varieties of (Bora valley and Gogu valley), raw potato juice was prepared and the starch was obtained from each juice by filtration and drying. Upon oral administration to rats, raw potato juice showed more or less inhibitory activity. The starch showed higher and dose-dependent inhibitory activity, suggesting that the active ingredient in raw potato juice may be the starch. Particularly, the starch obtained from the tubers of new potato variety, "Bora valley," with purple color, showed the highest inhibitory activity (62.4% and 37.1% inhibition of ulcer index at 500 mg/kg), while omeprazole (proton pump inhibitor) used as a reference drug showed 74.4% and 75.7% inhibition at 20 mg/kg against ethanol- and indomethacin-induced ulcer formation, respectively. The present study provides a first evidence of anti-ulcerogenicity of raw potato juice and the starch. Especially, the starch from "Bora valley" strongly inhibited ulcer formation in rats. Considering that these are food components, they may be safely used for anti-ulcerogenic nutraceuticals.

Effect of Root Zone Cooling on Growth Responses and Tuberization of Hydroponically Grown 'Superior' Potato (Solanum tuberosum) in Summer

  • A potato (Solanum tuberosum L. cv. Superior) cultivar was grown in aeroponic cultivation system to investigate the effect of root zone cooling in summer. Based on their nutrient uptake, growth responses, and tuberization, the possibilities for potato seed production were determined. Although shoot growth and early tuberization increased in the conventional non-cooling root zone system (root zone temperature of $25\pm2^{\circ}C$), stolen growth, photosynthesis, transpiration rate and number of tubers produced were higher in the cooling root zone system ($20\pm2^{\circ}C$) than in the non-cooling system. Increasing root zone temperature above $25^{\circ}C$ stimulated absorption of K more than T-N, P, Ca, Fe and Mn. On the other hand, root zone temperatures in the range of $20^{\circ}C$ to $25^{\circ}C$ did not affect Mg contents. The lower uptake and supply to leaves of T-N, Fe and Mn at the high root zone temperature promoted early tuberization and advanced haulm senescence. The results stress the importance of keeping root zone temperature to as low as below 20, particularly in summer under temperate Bone.

Seasonal Incidence of Potato virus f Infection on Potato Cultivars for the Double Crops in Korea (2기작 감자 품종의 재배 시기별 PVY 감염 정도 조사)

  • One of major potato viruses is Potato virus Y (PVY) in Korea. In the southern part of Korea, potatoes have been grown as double crops in a year by using cv. 'Dejima' and 'Chubak' due to very short dormancy. However, they have caused a serious problem such as a rapid degeneration. It has been thought that the degeneration is affected by the high incidence of PVY in neighboring potato fields. Therefore, the investigation of factors causing the degeneration is very important in the production of healthy seed potato. In this study, the PVY reinfection rates of several potato varieties and the different seed sources of cv. 'Chubak' have been investigated. Results show that the lowest infection rate of PVY among four potato cultivars derived from minitubers is cv. 'Superior'. The others are in order of 'Dejima', 'Atlantic' and 'Chubak'. Also, the incidences of PVY differ significantly when several seed sources are examined. When the seed potatoes (G2, the progeny of microtuber) as spring potato crops are planted in area without potato field nearby, the infection rate of PVY is as low as that of microtubers. However, PVY incidence in the progenies of minitubers as fall potato crops largely increases. Therefore, the best way of potato production under double cropping system is to use the healthy seed potato produced in area without potato field and plant relatively resistant cultivar such as Dejima.

Verticillium Wilt of Potato Caused by Verticillium albo-atrum in Daegwallyong Area in Korea

  • Verticillium wilt was first observed in 2001 on potatoes (Solanum tuberosum) cv. Superior at Daegwallyong area, one of the major seed potato producing areas in Korea. The wilted potato plants showed typical symptoms including gradual yellowing and interveinal necrosis. There was discoloration in the vascular tissues of the infected stems which turned light brown. Fungal isolates from discolored vascular tissues were whitish to creamy with folding on potato dextrose agar medium, where they used to produce resting dark mycelia but no micro-sclerotia. Conidiophores were septate with side branches, swelled at the base, and arranged in a whorl. Conidia were 2.5-11.2$\times$2.0-4.5 $\mu\textrm{m}$ um in size and were borne in small clusters at the tips of phialides. Optimal temperature range for mycelial growth was $25-30^{\circ}C$. Based on these cultural and morphological characteristics, the fungus was identified as Verticillium albo-atrum Reink & Berth. Pathogenicity tests by root dipping method revealed that the fungus caused the same symptoms as observed in naturally infected potato plants. This is the first report of Verticillium wilt on potato caused by Verticillium albo-atrum in Korea.

  • To understand the properties of the cauliflower mosaic virus (CaMV) 35S promoter and the effect of the deleted maize alcohol dehydrogenase I-S (Adhl-S) intron 1 on the expression of the CaMV $35S{\beta}-glucuronidase$ (GUS) gene in potato (Solanum tuberosum L. cv. Superior), we constructed a chimeric gene and transferred it into potato with Agrobacterium tumefaciens mediated method. The pLS201, a gene transfer vector of 17.7 kilobase pairs, was composed of the CaMV 35S promoter, the 249 base pairs of deleted maize Adhl-S intron 1, the GUS reporter gene, and the kanamycin resistance gene as a selectable marker for transformation. The GUS activity was examined by histochemical and spectrophotometric assay in transformed potato plants. The GUS activity was found primarily around the vascular tissue cells in stem and root. In the spectorophotometric assay, the level of GUS activity of transgenic potato transformed with CaMV 35S/249 bp of intron 1 fragment-GUS (pLS201) was compared with that of potato transformed with CaMV 35S-GUS (pBI121). The quantitative spectrophotometric assay showed that the level of GUS activity in potato transformed with pLS201 was higher in leaf, stem and root by 30-, 34- and 42-fold, respectively than those in potato transformed with pBI121. This results indicate that the inclusion of the deleted maize Adhl-S intron 1 resulted in increament of the GUS gene expression in transgenic potato.potato.

Comparison of Physicochemical Properties of Several Korean Potato Starches (품종별 한국산 감자의 전분 특성 비교(I))

  • Granular shapes and size, physicochemical properties and gelatinization properties of potato starches from Irish cobber (the dry type), Dejima (the moist type), Shepody and Superior were investigated. The starch granule was round or oval and the size of Irish cobbler and Shepody starch were $11~30\mu\textrm{m}$ and those of Superior and Dejima were$ 21~40\mu\textrm{m}$. Blue value, amylose content, relative crystallinity and density of Irish cobbler were the highest, but that of Dejima were the lowest. Water binding capacity, swelling power, transmittance. peak viscosity and breakdown of amylogram of Superior starch were the highest, but that of Irish cobbler starch were the lowest. Gelatinization temperature of DSC of Irish cobbler starch was the highest, but that of Superior starch was the lowest.

Effect of Low Dose ${\gamma}$ Radiation on the Dormancy and Growth of in vitro Microtuvers of Potato(Solanum Tuberosum L.) (저선량 ${\gamma}$선이 기냐 생산된 감자 소괴경의 휴면과 생에 미치는 효과)

  • To observe the stimulating effect of low dose ${\gamma}$ radiation on the dormancy breaking and growth, microtubers induced in vitro of two potato (Solanum tuberosum L. cv. Dejima and cv. Superior) cultivars with different storage duration were irradiated with ${\gamma}$ radiation at the dose of $0.5{\sim}30$ Gy. Sprouting rate, growth and tuber yield of ‘Dejima’ microtuber were increased by ${\gamma}$ radiation in the range of $2{\sim}16$ Gy. In the microtuber of ‘Superior’, the sprouting rate was promoted by 2 Gy and 4 Gy irradiation, and the growth and tuber yield by 0.5 Gy and 4 Gy irradiation. There were not that much difference in chlorophyll content of potato plantlet by the low dose ${\gamma}$ irradiation. These results suggested that low dose ${\gamma}$ radiation stimulated the dormancy breaking and potato growth.
