Supercritical Fluid Extraction of Physiologically Active materials from Agaricus blazei Fruiting Bodies (Agaricus blazei 자실체로부터 초임계 유체를 이용한 생리활성물질 추출공정)

  • The supercritical fluid extraction(SFE) technique was applied for the isolation and purification of nonpolar physiologically acitve mateials from Agaricus blazei fruiting bodies. the qualitative analysis of extract was accomplished by gas chromatography-mass spectrometer(GC-MS) and extract was determined as linoleic acid(cis-9 cis-12-octadecadienoic acid) In order to obtain the optimum operating conditions of supercritical fluid extraction process the various temperatures and pressures were applied for process operation. From the comparison of exraction efficiencies $50^{\circ}C and 200 kg_f/cm^2$ were determined as optimum conditions.

Physiological Activity of Supercritical Poria cocos back Extract and Its Skin Delivery Application using Epidermal Penetrating Peptide (초임계 복령피 추출물의 생리활성 및 경피투과 펩티드를 이용한 경피 약물전달의 응용)

  • In this study, Poria cocos bark were extracted by supercritical process, and anti-inflammatory, whitening, and antioxidant effects were measured in comparison with ethanol extract. Also, An effective percutaneous permeation method using a selected formulation of the extract and a drug delivery peptide was proposed. Pachymic acid, known as the anti-cancer and anti-inflammatory compound of the ventricle, is an indicator component and the HPLC analysis shows that the supercritical extract of the pericardium is more than twice that of the Poria cocos bark extract. In order to confirm antioxidative effect of Bombyx mori, DPPH scavenging ability and ABTS scavenging ability test showed that the ethanol extract of Poria cocos Back had lower concentration than the supercritical extract of Poria cocos back. However, RAW 264.7 Measurements of Nitric oxide (NO) production in cells showed lower NO production at the same concentration than the Poria cocos back ethanol extract. In addition, after 72 hours of processing of $20{\mu}g/mL$ of the Poria cocos back extract in B16 melanoma cells, both the intracellular and extracellular melanin extract were effective and the supercritical extract was lower melanin content. No toxicity was observed at the concentration of $800{\mu}g/mL$ in RAW 264.7 cells used in NO production experiments. However, in B16 melanoma cells, even at $50{\mu}g/mL$, both Poria cocos back ethanol extract and supercritical extract showed a survival rate of less than 60%. The liposome formulation and drug delivery peptides were shown to be useful for percutaneous permeation of Supercritical Extract of Poria cocos back using a liposome formulation and a drug delivery peptide. it is expected that there will be great potential for development as a variety of cosmetic materials for Poria cocos back.

Anti-Biofilm Activity of Origanum Vulgare Supercritical Fluid Extracts and Cosmetic Active Ingredients Development (오레가노 초임계추출물의 황색포도상구균 바이오필름 형성 억제능을 이용한 기능성 화장품 소재의 개발)

  • In this study, oregano was extracted by supercritical extraction and hydrothermal extraction method. In vitro experiments such as antimicrobial and antioxidant activity test were performed. As a result of the disc diffusion method, only the supercritical extracts formed a clear zone. The MIC for S. aureus was found only in the supercritical fluid extracts and it was 1000 ㎍/mL. The hydrothermal extract's MIC is 125 ㎍/mL for C. acnes. Through biofilm inhibition assay, we found that the supercritical fluid oregano extracts inhibit the biofilm of S. aureus by more than 70% even at low concentrations of 125 ㎍/mL. On the other hand, the antioxidant ability of the hydrothermal extract was better than that of the supercritical fluid extracts. Furthermore, we tried to make a skincare ingredient for atopic dermatitis by utilizing the S. aureus biofilm inhibitory ability of oregano supercritical fluid extracts. Liposome was used to overcome the low solubility of the oregano supercritical fluid extracts and increase stability.

Tow-stage Extraction of Milk Fat by Supercritical Carbon Dioxide

  • To develop mil fat fractions with desirable physico-chemical properties, anhydrous milk fat (AMF) was fractionated by one- and two-stage extractions using supercritical $CO_2$(SC-$CO_2$). Tow-stage extraction of AMF was performed by first producing tow fractions, an extract and a residue at 4$0^{\circ}C$/241bar, which were subsequently used as the feed for an extraction at 6$0^{\circ}C$/241bar and 4$0^{\circ}C$/345bar, and separated into five and four fractions, respectively, based one extraction time. These fractions were quantified and analyzed for fatty acids and physico-chemical properties. SHort-chain (C4~C8) fatty acids in extract fractions from an extract were 200~150% compared with those of the original AF. Long-chain (C14~C18) fatty acids in extract fractions from a residue were 118~141%. The ratio of unsaturated fatty acids in the residue fraction was 131%. Melting point ranged from 22 to 43$^{\circ}C$, iodine value 21.8 to 36.9, and saponification value 255 to 221 in the extract and residue fractions. SC-$CO_2$ fractionation of AMF by two-stage extraction offers the possibility of developing ractions with discrete fatty acid compositions and physico-chemical properties such as melting point, iodine value and saponification value.

A Study on the Possibility of Produced by Supercritical Fluid Extraction from Lycii Fructus Seed for Cosmetic Ingredients (구기자 종자 초임계유체 추출물의 화장품소재로서의 가능성 평가)

  • The purpose of this study was to investigate the possibility of applying cosmetic material about extracted from Lycii Fructus seed supercritical fluid. This study was conducted to evaluate the anti-melanogenesis effect and antimicrobial activity about the extracts from Lycii Fructus seed. These researches studied for emulsion physical stability of pH, viscosity, particle size from emulsion containing Lycii Fructus seed extract. As a result, the supercritical fluid extract from Lycii Fructus seed significantly inhibited melanin synthesis by 81.86 % at the concentration $750{\mu}g/mL$. Antimicrobial effects of extract was determined against Staphylococcus aureus, Candida albicans, Escherichia coli. except by Staphylococcus epidermidis. The physical stability of viscosity and pH on the emulsion containing Lycii Fructus seed extract were stable for 28 days. Emulsion containing Lycii Fructus seed extract did not change particles at observation into optical microscope. These results suggest that extracts from Lycii Fructus seed may have value as the potential cosmetic formulation.

  • Various factors affecting the supercritical carbon dioxide extraction of daidzein from soybean were studied. Daidzein was not extracted with pure supercritical carbon dioxide. The ethanol was an efficient modifier for supercritical carbon dioxide to extract daidzein. The extraction efficiency increased as the pressure increased up to 300 bar. At $35^{\circ}C$ and 300 bar, 93% of daidzein was extracted with supercritical carbon dioxide modified with 15% of ethanol.

  • This study is about a technique for obtaining collagen by extracting fat by treating collagen-containing liposuction effluent in the presence of supercritical fluid. Using a supercritical solvent, a collagen extract could be obtained from animal-derived fat in a short time (about 6 hours), and about 2-3% of collagen by mass compared to the raw material could be obtained. The presence of collagen in the extract obtained by supercritical extraction was confirmed by SDS-PAGE, and it was confirmed that it was type 1 collagen having a relatively large molecular weight. In addition, the growth factors of IGF-1, bFGF, VEGF and NGF were analyzed to find out which growth factors were present in the collagen obtained by supercritical extraction, and it was found that these growth factors were contained in the extract. There was no significant difference in DNA content per mg of sample before and after supercritical treatment. Further in-depth studies are likely to be needed on decellularization technology using the supercritical process. In conclusion, the extracellular matrix obtained through the solvent extraction process using a supercritical fluid contains growth factors above a certain amount even after decellularization and removal of fat, so that it was found that not only biocompatibility is greatly increased, but also tissue regeneration can be rapidly induced.

Enrichment of Coffee Flavors with Supercritical Carbon Dioxide (초임계 이산화탄소를 이용한 커피의 향기 성분 증진)

  • In this study, the supercritical $CO_2$ extraction was applied for the enrichment of coffee flavors. The extraction efficiency of coffee flavors was dependent on the pressure and temperature and optimized as 350 bar and 80$^\circ$C. Five flavors of high aroma values were analyzed. The flavors extracted by the supercritical $CO_2$ extraction were 76.6 times higher than those by the traditional method, espresso extraction. The modified headspace system proved the enhanced efficiency of supercritical $CO_2$ extraction. The coffee beverage containing the extract of supercritical $CO_2$ resulted 2.3 times of flavor enrichment when it was compared to that without the supercritical extract.

The Effect of Anti-microbial and the Inhibitory Effect of Biofilm Formation and Inflammatory Factors Production of Perillae semen Supercritical Fluid Extracts (초임계 자소자추출물의 항균효과와 바이오필름, 염증매개인자 생성 억제 효능)

  • In this study, we assessed anti-oxidant activity, anti-microbial, inhibition of biofilm formation and inflammatory factors(nitric oxide, interleukin-6, interleukin-8) inhibitory effect of Perillae semen hydrothermal extract(PSW) and three kinds of Perillae semen supercritical fluid extract(PSSs) extracted by controlling temperature with no variation of pressure. Compared with PSW, PSSs had significantly lower minimal inhibitory concentrations(MICs) against Staphylococcus aureus(S. aureus) and the ability of PSSs to inhibit formation of biofilm was also superior. PSSs reduce the production of inflammatory mediator and inflammatory cytokines significantly compared to PSW. We suggest, therefore, Perillae semen supercritical fluid 45℃ extract which showed the best anti-microbial, inhibition of biofilm formation, and inhibition of inflammatory factors production among the supercritical fluid extracts could be used for protecting patients with atopic dermatitis from pruritus and transepidermal water loss as a functional ingredient from nature.

Antimicrobial Activity of Ginger(Zingiber officinale Roscoe) Oleoresin by Supercritical Fluid Extraction (초임계유체 추출에 의한 생강(Zingiber officinale Roscoe) Oleoresin의 항균활성)

  • The study indicated that antimicrobial activity about gram positive and gram negative bacteria of ginger-oleoresin(GO) extract with the condition of ethanol and supercritical fluid extractions. As the concentration of extraction increases, the clear zone of GO ethanol extract also increased dependently. This led the antimicrobial activity of gram positive bacteria to take bigger place than gram negative bacteria especially in Listeria monocytogenes. There was a high antimicrobial activity in E-III treatment where the ratio of the ginger powder extract to ethanol extraction was 1:6. It was quite effective to treat the antimicrobial activity of GO ethanol extract under $80^{\circ}C$ and there was not big difference in the intervals which were the extraction time - 1 to 7 hours. The antimicrobial activity of supercritical fluid extract seemed to take the biggest place in Listeria monocytogenes. From the supercritical fluid extract, it was shown the strong ability of antimicrobial activity in the condition with 100 bar $35^{\circ}C$, 250 bar $35^{\circ}C$ and 250 bar $65^{\circ}C$. Furthermore, according to the case of solvent extract, there was not any significant difference in the antimicrobial activity with condition of extraction. However, there was significant antimicrobial activity in E-III treatment of 100 bar and 500 bar of extraction pressure, and $35^{\circ}C$ and $65^{\circ}C$ of extraction temperature.
