Culture Characteristics and Optimal Conditions for Mycelial Growth of Calocybe indica (Calocybe indica의 배양적 특성과 균사 배양 적합 조건 설정)

  • Calocybe indica is an edible mushroom commercially cultivated in India and other tropical countries. In this study, the culture characteristics and optimal conditions of milky mushroom strains were determined. The growth temperature and pH range of milky mushrooms was extensively investigated between 15-35 ℃ and pH 3-11. For efficient cultivation, 20 types of nutrient sources were selected that consisted of one of 21 types of carbon sources, 6 organic nitrogen sources, 6 inorganic nitrogen sources, 13 amino acids, 6 organic acids and 12 inorganic salts. The impact of each of the selected nutrition sources and their concentration on growth was investigated. The optimum pH and temperature were determined to be pH 6.0 and 15 ℃, respectively. The optimum concentration of medium elements for the mycelial growth of C. indica was determined to be as follows: carbon source, 2% maltose; organic nitrogen source, 1% yeast extract; inorganic nitrogen source, 0.1% NaNO3; amino acid, 0.7% asparagine; organic acid, 0.07% acetic acid; inorganic salt, 0.07 mM MnSO4.

Fruit Yield and Quality Evaluation of Sweet Pepper (Capsicum annuum L.) F1 Hybrids Derived from Inbred Lines

  • Sweet pepper inbred lines (KNU1003, KNU1006, KNU1007, KNU1009, KNU1015, KNU1017 and KNU2006) developed at Kangwon National University (KNU) through conventional means, inbred lines (5AVS1, 5AVS2, 5AVS3, 5AVS5, 5AVS7 and 5AVS8) collected at Rural Development Administration (RDA) and inbred lines (SP12, SP27 and SP14) derived from anther culture were used as female parents and anther culture derived homozygous lines (SP9, SP10, SP14, SP24, SP25, SP27, SP30, SP32, SP34, SP38, SP43, SP45 and SP51) were used as male parents to produce $F_1$ hybrids. A total of 37 $F_1$ hybrids were evaluated for fruit yield and quality characters in summer season, 2007. Variation in fruit number, fruit weight, fruit yield per plant and fruit volume was observed among the $F_1$ hybrids. Superiority on yield over standard/commercial varieties were differed among $F_1$ hybrids. Hybrid $5AVS8{\times}SP45$ exhibited highest heterosis over Special (16.5%) and Fiesta (24.7%). Fruit quality characters (fruit length, fruit width, pericarp thickness, total soluble solid, fruit shape and fruit color) were varied among the $F_1$ hybrids. Fruit number, fruit weight and fruit volume per plant were correlated with fruit yield. Based on the standard heterosis expressed by the hybrids and quality characters evaluation, $KNU1017{\times}SP27$, $5AVS1{\times}SP43$, $5AVS5{\times}SP27$, $5AVS8{\times}SP45$, $SP12{\times}SP38$ and $SP27{\times}SP25$ hybrids were found to be superior over commercial cultivars and are selected. Inbred lines of these hybrid combinations can be used to produce $F_1$ hybrid seed for commercial production.

  • Artomyces microspora is a genus of coral fungi from the family Auriscalpiaceae that have sporophores which are clavarioid, profusely and pyxidately branched, and devoid of a conspicuous stipe. These fungi can be found in summer and fall. This study aimed to decipher fundamental information regarding optimal growth conditions of Artomyces microsporus mycelia, including pH, temperature, carbon sources, and nitrogen sources. Based on the assessment of colony diameter and mycelial density, the optimal culture medium, temperature, and pH for mycelial growth were found to be PDA, 25 ℃, and pH 5.0, respectively. Furthermore, the study revealed that the optimal carbon and nitrogen sources for mycelial growth were 1% soluble starch and 2% malt extract, respectively. The other suitable inorganic nitrogen sources were deemed to be 0.1% NH4H4PO4 and 0.1% aspartic acid.

Changes in milk production and blood metabolism of lactating dairy cows fed Saccharomyces cerevisiae culture fluid under heat stress

  • In this study, Saccharomyces cerevisiae culture fluid (SCCF) has been added to a diet of lactating dairy cows to attempt to improve the ruminal fermentation and potentially increase the dry matter intake (DMI) and milk yield. This study was conducted to investigate the effects of SCCF on the milk yield and blood biochemistry in lactating cows during the summer. Twenty-four Holstein dairy cows were randomly assigned to one of four treatments: (1) total mixed ration (TMR-1) (Control); (2) TMR-1 supplemented with SCCF (T1); (3) TMR-2 (containing alfalfa hay) (T2); and (4) TMR-2 supplemented with SCCF (T3). SCCF (5 ml/head, 2.0×107 CFU/mL) was mixed with TMRs daily before feeding to dairy cows. The mean daily temperature-humidity index (THI) during this trial was 76.92 ± 0.51 on average and ranged from 73.04 to 81.19. For particle size distribution, TMR-2 had a lower >19 mm fraction and a higher 8-9 mm fraction than TMR-1 (p < 0.05). The type of TMR did not influence the DMI, body weight (BW), milk yield and composition, or blood metabolites. The milk yield and composition were not affected by the SCCF supplementation, but somatic cell counts were reduced by feeding SCCF (p < 0.05). Feeding SCCF significantly increased the DMI but did not affect the milk yield of dairy cows. The NEFA concentration was slightly decreased compared to that in the control and T2 groups without SCCF. Feeding a yeast culture of S. cerevisiae may improve the feed intake, milk quality and energy balance of dairy cows under heat stress.

Severe Root Rot on Hydroponically-Grown Lettuce Caused by Phytophthora drechsleri

  • Phytophthora root rot of lettuce, which has not been reported in Korea before, occurred severely in liquid hydroponic culture. The disease occurred in all seasons and was most severe in summer from June to August, showing over 90% infection rate in some farms. A total of 51 isolates collected from various farms were all identified as Phytophthora drechsleri. The fungus showed strong pathogenicity to lettuce and Chinese cabbage, moderate pathogenicity to cucurbits and tomato, and weak pathogenicity to pepper. However, the fungus was not pathogenic to other leafy vegetables namely: chicory, kale, endive, garland chrysanthemum, spinach beet, and perilla. Among 10 species of Phytophtora inoculated to lettuce, only P. drechsleri and P. cryptogea were found pathogenic.

Development of a Numerical Model for Prediction of the Cooling Load of Nutrient Solution in Hydroponic Greenhouse (수경온실의 양액 냉각부하 예측모델 개발)

  • Cooling of nutrient solution is essential to improve the growth environment of crops in hydroponic culture during summer season in Korea. This study was carried out to provide fundamental data for development of the cooling system satisfying the required cooling load of nutrient solution in hydroponic greenhouse. A numerical model for prediction of the cooling load of nutrient solution in hydroponic greenhouse was developed, and the results by the model showed good agreements with those by experiments. Main factors effecting on cooling load were solar radiation and air temperature in weather data, and conductivity of planting board and area ratio of bed to floor in greenhouse parameters. Using the model developed, the design cooling load of nutrient solution in hydroponic greenhouse of 1,000$m^2$(300pyong) was predicted to be 95,000 kJ/hr in Suwon and the vicinity.

  • Special educations for gifted students have not been given enough attention in Japan with a little exception. Indeed, such educations were sometimes despised in Japan by teachers and parents as well as by boards of education, because one of the features of postwar education system in Japan was an excessive egalitarianism. The other is cramming of knowledge in school education, which is necessary for high school graduates to pass entrance examinations for famous universities such as University of Tokyo, or Kyoko University. However, in 1997, some trials of special educations for gifted students started. The Ministry of Education, Sports, Culture, Science and Technology admitted 'skipping a year to enter universities.' In this paper, the following three topics would be discussed. 1. Enrollment of high school students aged 17 into Chiba University. 2. Summer seminars conducted by Japan Mathematics Foundation of Olympiad. 3. Super Science High School Program funded by the Ministry of Education.
