Stability of Ascorbic Acid in the Solutions of Sugars and Amino Acids (당질과 아미노산이 Ascorbic Acid 의 안정도에 미치는 영향)

  • As sugar and amino acid were added to the ascorbic acid solution the content of ascorbic acid was quantitatively determined by 2, 4-dinitrophenyl hydrazine method. The residual ascorbic acid was shown to increase slightly when sorbose, rhamnose or mannose was added to the ascorbic acid solution whereas residual ascorbic acid was shown to decrease in time to the addition of other sugars. The effects of amino acid to the ascorbic acid solution were found that monoamino-mono, or dicarboxylic acids and aromatic amino acids increased the residual ascorbic acidity whereas diamino-monocarboxylic acids and sulfur containing amino acids decreased the residual ascorbic acidity.

Effects of Nitrogen and Sulfur Application on Yield and contents of Amino Acids Containing Sulfur of Rice (질소(窒素) 및 유황시용(硫黃施用)이 수도(水稻)의 수량(收量)과 함유황(含硫黃)아미노산(酸) 조성(組成)에 미치는 영향(影響))

  • A pot experiment was carried out using Seomjin-byeo and Samgang-byeo to investigate the effects of nitrogen and sulfur on the growth, yields, and sulfur containing amino acid content of rice. The yields and number of tillers at 50 days after transplanting were increased with the increase of the amounts of nitrogen and sulfur, but N/S ratio was decreased in both varieties. There were a positive correlations between yield and contents of nitrogen and sulfur in straw and brown rice, but a negative correlation between N/S ratio and yields at harvesting stage. Crude protein contents increased with the increase of amounts of nitrogen and sulfur. contents of amino acid containing sulfur in brown rice were positively correlated with the sulfur and nitrogen contents in rice straw.

Single Hydrolysis Method for the Amino Acid Determination in Foods and Composite Dishes (식품의 아미노산 정량을 위한 단일가수분해 방법의 개발)

  • For the complete and accurate amino acid determination of protein and food samples, 3 different hydrolysis procedures have been conducted in parallel for each sample, which include the alkaline hydrolysis for tryptophan determination, performic acid oxidation prior to the acid hydrolysis for the determination of cysteine and cystine, and the 6N HCl hydrolysis for the determination of the rest of amino acids. In the present study, amino acid concentrations obtained from the modified single hydrolysis procedure were compared with the values from the conventional hydrolysis procedures in casein and nine food and composite dish samples. In most of the samples tested, the modified single hydrolysis procedure gave significantly higher values of cysteins and cystein compared to the performic acid oxidation method, but resulted in a considerable destruction of tryptophan in food and composited dish samples. There was no consistent difference in the rest of amino acid concentrations between the two hydrolysis systems. Therefore, for complete amino acid determination of various foods and composite dishes, the single hydrolysis method may replace the 6N HCl hydrolysis and performic acid oxidation methods, and thereby reduces 3 hydrolyses to 2 steps with much higher recoveries of the sulfur containing amino acids.

  • The amino acid composition of human and cow's milk represents a standard of reference in infantnutrition. The amino acid compositions determined by automatic amino acid analyzer, Yanagimote Model LC-5. Protein in the human and cow's milk were found to be hydrolyzed to yield free amino acids. Qualitative data for free amino acids in the milk are as follows: 1) Amounts of acidic amino acids such as glutamic and aspartic acid in cow's milk were obserbed to be about 2 times compared with human milk and it is considered that the abundance in these amino acids may contribute significantly to the specific flavor of cow's milk. 2) It is much interesting that in the human milk the contents of sulfur-containing amino acids were high comparatively better than cow's milk; cystine was found to be 3 times ana methionine, 2 times. 3) In the human milk a high content of some essential amino acids such as threonine, isoleucine and leucine was demonstrated and a specific flavor sweet amino acids. 4) Large amounts of basic amino acid such as histidine was found to occur in human milk and arginine in cow's milk.

Amino Acid Profiles of Tropical Legumes, Cooper (Glycine wightii), Tinaroo (Neonotonia wightii) and Siratro (Macroptilium atropurpureum), at Pre-blooming and Blooming Stages

  • An experiment was conducted to investigate amino acid composition of three tropical legumes (Cooper (Glycine wightii), Tinaroo (Neonotonia wightii) and Siratro (Macroptilium atropurpureum)) at two different stages (pre-blooming and blooming stages). Chemical composition and totally 16 amino acids of these plants were analysed for comparison of their composition among species at different growing stages and characterizing the amino acid pattern of these legumes. Crude protein content of the plants ranged from 16% to 27% on a dry matter basis. The total amount of 16 amino acids analyzed in this experiment was highest at 89.7 mg/16 g N in Cooper at pre-blooming and lowest at 80.9 mg/16 g N in Glycine at blooming stage. Total amount of amino acids in each legume species tended to slightly decrease with their maturity but no statistical difference was found. The percentage of aspartic acid, glutamic acid and proline in the total amount of amino acids was dominant at 9% to 13%, and that of methionine was less than 1.6%. In this experiment it was concluded that three tropical legumes were rich in crude protein content and characterized by 16 different amino acids with lower sulfur-containing amino acid as methionine.

Trifunctional Amino Acid Cobalt(Ⅲ) Complexes of N,N'-Diethylethylenediamine-N,N'-di-α-butyrato Ligand

  • Cobalt(Ⅲ) complexes of trifunctional amino acid and N,N'-diethylethylenediamine-N,N'-di-α-butyrate(deedba), s-cis-[Co(deedba)(L-aa)] (L-aa=S-methyl-L-cysteine, L-aspartic acid, L-glutamic acid) have been prepared from the reaction between the s-cis-[Co(deedba)(Cl2)]- complex and the corresponding amino acid. The amino acids have been found to coordinate through the amine and carboxylate groups. The S-methyl-L-cystene is coordinated not by the sulfur donor atom, but by the nitrogen and oxgen donor atoms, and the L-aspartic and L-glutamic acids are coordinated to the cobalt(Ⅲ) ion via formation of the five-membered glycinate chelate ring. Relatively small optical activity shown by the complexes is due to the chiral center present in the amino acids.

Regulation of Branched-Chain, and Sulfur-Containing Amino Acid Metabolism by Glutathione during Ultradian Metabolic Oscillation of Saccharomyces cerevisiae

  • Autonomous ultradian metabolic oscillation (T$\simeq$50 min) was detected in an aerobic chemostat culture of Saccharomyces cerevisiae. A pulse injection of GSH (a reduced form of glutathione) into the culture induced a perturbation in metabolic oscillation, with respiratory inhibition caused by $H_2S$ burst pro-duction. As the production of $H_2S$ in the culture was controlled by different amino acids, we attempted to characterize the effects of GSH on amino acid metabolism, particularly with regard to branched chain and sulfur-containing amino acids. During stable metabolic oscillation, concentrations of intra-cellular glutamate, aspartate, threonine, valine, leucine, isoleucine, and cysteine were observed to oscil-late with the same periods of dissolved $O_2$ oscillation, although the oscillation amplitudes and maximal phases were shown to differ. The methionine concentration was stably maintained at 0.05 mM. When GSH (100 $\mu$M) was injected into the culture, cellular levels of branched chain amino acids increased dramatically with continuous $H_2S$production, whereas the cysteine and methionine concentrations were noticeably reduced. These results indicate that GSH-dependent perturbation occurs as the result of the promotion of branched chain amino acid synthesis and an attenuation of cysteine and methionine synthesis, both of which activate the generation of $H_2S$. In a low sulfate medium containing 2.5 mM sulfate, the GSH injections did not result in perturbations of dissolved $O_2$ NAD(P)H redox oscillations without burst $H_2S$ production. This suggests that GSH-dependent perturbation is intimately linked with the metabolism of branched-chain amino acids and $H_2S$ generation, rather than with direct GSH-GSSG redox control.

Effects of Sulfur-Containing Amino Acids on Lipid Metabolism in Rats (함유황아미노산이 지질대사에 미치는 영향)

  • The effect of sulfur-containing amino acids on lipid metabolism was studied in rats fed casein as a protein source. Plasma cholesterol, HDL-cholesterol and atherosclerotic index decreased in the cysteine group compared to the methionine group. Plasma triglyceride and phospholipid level were not affected by the supplementation of the sulfur-containing amino acids. The levels of cholesterol and triglyceride in liver decreased by both methionine and cysteine. Cysteine increased the fecal excretion of coprostanol, total neutral steroid and bile acid. The results suggest that plasma cholesterol level is affected by dietary ratio of cysteine/methionine and that the hypocholesterolemic effects of cysteine is, at least in part, through reducing cholesterol absorption from small intestine and through enhancing fecal excretion of bile acids.

Dose-dependent Effects of Betaine on Hepatic Metabolism of Sulfur Amino Acids in Mice (마우스 간의 황함유 아미노산 대사에 미치는 베타인의 용량의존성 영향)

  • Acute betaine treatment induces time-dependent changes in the hepatic glutathione (GSH), cysteine and S-adenosylmethionine (SAM) levels. Our previous study demonstrated that betaine administered $1{\sim}4$ hours prior to sacrifice decreased hepatic GSH levels, but these levels were increased when measured 24 hours following the treatment. The present study was aimed to determine dose-dependent effects of betaine on hepatic metabolism of sulfur amino acid in mice. Mice were sacrificed 2.5 or 24 hours after intraperitoneal treatment with betaine at different dose levels ranging from 50 to 1000 mg/kg. The concentrations of methionine and SAM were increased by a betaine dose of 100 mg/kg, and the concentrations of GSH and cysteine were decreased by a betaine dose of 200 mg/kg at 2.5 hours. These changes were augmented with increasing doses of betaine. At 24 hours following betaine treatment, increased GSH and decreased taurine levels were observed from dose levels of 400 mg/kg. Changes in hepatic activities of cystathionine beta-synthase, gammaglutamylcysteine ligase and cysteine dioxygenase were observed from dose levels of $200{\sim}400$ mg/kg of betaine administered 24 hours prior to sacrifice.

Effects of Fasting on Hepatic Metabolism of Sulfur Amino Acids in Rats (절식이 랫트 간의 황함유 아미노산 대사에 미치는 영향)

  • Food deprivation decreases hepatic glutathione (GSH) levels, which is ascribed to alterations in availability of hepatic cysteine, a rate limiting factor for the GSH synthesis. The present study examines the effects of food deprivation on hepatic metabolism of sulfur amino acid in male rats. In rats fasted for 24 or 48 hours, hepatic GSH levels were decreased from $6.70{\pm}0.16{\mu}mol/g$ liver to $4.02{\pm}0.20$ or $4.06{\pm}0.07{\mu}mol/g$ liver, respectively. Hepatic S-adenosylmethionine levels were also decreased in fasted rats, but S-adenosylhomocysteine levels were increased. Hepatic methionine levels were not changed by food deprivation for 48 hours. On the other hand, hepatic cysteine or taurine levels were increased from $106.2{\pm}4.1$ to $130.0{\pm}2.7$ nmol/g liver or from $2.45{\pm}0.43$ to $5.07{\pm}0.78{\mu}mol/g$ liver, respectively, in 48-hour fasted rats. Activity of cystathionine beta-synthase catalyzed homocysteine to cystathionine, was markedly decreased, but activity of betaine homocysteine methyltransferase was increased in fasted rats, indicating that methylation of homocysteine to methionine is activated. Also activity of cysteine dioxygenase, involved in taurine synthesis, was increased. These results suggested that hepatic methionine levels were maintained in rats fasted for 48 hours through increase in homocysteine methylation, and hepatic GSH may serve as a cysteine supplier reservoir in fasting state.
